Morphine and its major metabolites, morphine-3-glucuronide and morphine-6-glucuronide, were given intracerebroventricularly (i.c.v.) to rats. The antinociceptive effects were assessed in the tail-flick and hot-plate tests as well as the writhing test. Ventilatory effects were studied in halothane-anaesthetized rats. Based on calculated ED50 values, morphine-6-glucuronide was approximately 200 times more potent that morphine itself in the tail-flick and hot-plate tests. In the writhing test the difference in ED50 was approximately 9-fold. Morphine and morphine-6-glucuronide administered i.c.v.

1. i.c.v. morphine administration (50 micrograms/10 microliters) to sham-lesioned guinea-pigs produces a decrease of body temperature 90 min after injections. 2. This effect was blocked in animals that received i.c.v. 6-OHDA injections 20 days before. 3. On the other hand, morphine injection to sham-lesioned guinea-pigs resulted 30 and 90 min later in suppressed plasma B-END and cortisol concentration. 4. This suppressive effect was increased 20 days after 6-OHDA injections. 5. These results indicate that in the guinea-pig the effects of morphine on both body temperature and plasma levels of B

The effects of pentobarbitone anaesthesia on the cardiovascular changes induced by centrally administered taurine have been investigated in spontaneously hypertensive (SHR) and normotensive rats. Administration of taurine (100-400 micrograms) into the lateral cerebral ventricle (i.c.v.) of anaesthetised SHR and normotensive rats induced a dose-related fall in systemic blood pressure and heart rate, which tended to be of a greater magnitude in the SHR. In anaesthetised rats attached to a ventilator, taurine was not as potent at inducing a fall in systemic blood pressure, approximately double th

Both endogenous opiate peptides and serotonin are known to be involved in blood pressure control. In this work we present results obtained in anaesthetized normotensive rats in which we studied the possible interaction between the central blood pressure effects induced by methionin (met)-enkephalin and serotonergic pathways. Met-enkephalin administered intracerebroventricularly produced a dose-dependent hypotensive action that was prevented by pretreating the animals with naloxone (opioid antagonist), or with methiothepin (serotonergic S1 and S2 receptor antagonist) or 5,7-dihydroxytryptamine

Acute microinfusions of ovine corticotropin releasing factor (CRF) into the third cerebral ventricle (III vent.) of sexually experienced male rats produced a dose-dependent suppression of masculine sexual behaviour. This inhibition of male sexual performance induced by CRF could be reversed by simultaneous infusions of naloxone, the opiate receptor antagonist, which when infused without CRF facilitated copulatory performance. These findings suggest that CRF can exert drastic effects on male reproductive behaviour through mechanisms which involve the activation of opioid pathways within the cen

Cryptococcal meningitis is a life-threatening disease. Headache, vomiting, cranial nerve symptoms and mental changes are the most common symptoms, but as many as 15% may have no symptoms referable to the CNS. For chemotherapy four drugs are available: namely amphotericin B, 5-fluorocytosine, miconazole and ketoconazole. Most cases have been treated by combination of amphotericin B and 5-fluorocytosine. The intrathecal administration of amphotericin B should be considered for patients who fail to respond to the usual intravenous therapy. The case is reported of a patient who died due to hydroce

Rats were taught to self-administer Leu-E (10, 25 and 100 micrograms/microliters) or Met-E (0.5, 10 and 100 micrograms/microliters) through a cannula implanted in the lateral cerebral ventricle (i.c.v.). Their self-injection behaviour was studied before, during and after nociceptive stimulation. In the course of the control period of the experiment, the rats rapidly learned lever pressing for self-injection of enkephalin but they did not increase their self-administration of Leu-E or Met-E during the nociceptive electrical stimulation period. Also studied were the acute effect of i.c.v. enkeph

A new animal model for epilepsy was successfully produced by microinjection of cobaltous chloride into the lateral cerebral ventricle of the rat. The median convulsive dose (CD50) and the median lethal dose (LD50) of CoCl2 was 0.45 microM/10 microliters (0.27-0.77 microM/10 microliters) and 1.07 microM/10 microliters (0.73-1.57 microM/10 microliters), respectively. The behavioral changes, electrocorticogram (ECoG), and the action of 5 classical anticonvulsants were studied using this new model. Seizures induced by cobaltous chloride are clinically similar to those produced by systemic administ

Opiate peptides (beta-endorphin, alpha-endorphin or met-enkephalin) were administered to the lateral cerebral ventricle of adult male rats via single injection into implanted cannulas. Scanning EM of the third ventricle above the median eminence of animals sacrificed approximately one hour after peptide injection revealed numerous free cells above the ependyma. Beta-endorphin induced a significantly greater cellular response than other peptides or control injections. Transmission EM demonstrated that the cells had morphologic characteristics of macrophages. Opiate peptides in cerebrospinal flu

1. The thermoregulatory outputs (including metabolic, respiratory and vasomotor activities) produced by an injection of clonidine or 5-hydroxytryptamine (5-HT) into the third cerebral ventricle of conscious rabbits were assessed at three different ambient temperatures (Ta) of 2, 22 and 32 degrees C. 2. When injected into the third cerebral ventricle, both clonidine and 5-HT produced a dose-dependent hypothermia in rabbits at both 2 and 22 degrees C Ta. The hypothermia was due to a decrease in metabolic heat production (M) at 2 degrees C Ta, while at 22 degrees C Ta the hypothermia was due to c

Release of oxytocin and Substance P-like peptides from the rat posterior pituitary lobe incubated in situ after infusion of hypertonic solution into the third cerebral ventricle. Acta Physiol. Pol., 1978, 29 (1): 9-16. The experiments were carried out on male rats. Under general urethane and chloralose anaesthesia the ventral surface of the hypothalamus and the pituitary gland were exposed by a transpharyngeal approach and the anterior pituitary lobe was entirely removed. The posterior lobe with the remaining neural and partially vascular connections with the hypothalamus was incubated in situ

Vasopressin, or vasopressin antiserum, was injected into a lateral cerebral ventricle of conscious rats. These rats were normally hydrated, cellular dehydrated (NaCl loading) or hypovolemic (polyethylene glycol model). Elevation or reduction of vasopressin in cerebrospinal fluid produced no consistent change in consummatory behavior, urine volume or sodium and potassium excretion. These results show vasopressin in cerebrospinal fluid not to be an absolute requirement for maintenance of hydration or for response to acute volume and osmotic stimuli.

1 The central effects produced by a new anticholinesterase RX72601 injected into the cerebral ventricles or perfused within circumscribed forebrain structures have been investigated in the unanesthetized cat. 2 When injected into the cerebral ventricles in doses of 1-10 mug, RX72601 produced a dose-related hyperthermia together with behavioural and autonomic responses. 3 Experiments designed to localize the site or sites of these actions demonstrated that hyperthermia could be produced from several diencephalic sites but that the other effects were not normally produced from these same sites.

BACKGROUND AND PURPOSE: Gene therapy may show promise for stroke patients, but invasive techniques such as intraventricular or intracerebral injection of therapeutic genes may have limited applicability. The purpose of this study is to develop systemic gene therapy using macrophages infiltrating the infarct to deliver and express the gene.
METHODS: After permanent middle cerebral artery occlusion in rats, an enhanced green fluorescent protein (EGFP) plasmid conjugate in liposomes was injected via the femoral vein. We also constructed a bicistronic plasmid vector for fibroblast growth factor-2

The involvement of the L-arginine-nitric oxide (NO) pathway in brain, in the regulation of drinking behaviour, has been evaluated by injecting L-arginine and N omega-nitro-L-arginine methyl ester (L-NAME) into the lateral cerebral ventricle (i.c.v.). L-Arginine (5 and 10 micrograms/rat), but not D-arginine, was antidipsogenic when administered to 24 hr water-deprived rats but did not change the intake of water in normally hydrated rats. However, L-NAME (5 and 10 micrograms/rat) did antagonize the effect of L-arginine in water-deprived animals but, by itself, did not increase thirst. L-Arginine

To clarify the pathogenesis of normal pressure hydrocephalus (NPH), quantitative measurement of CSF dynamics was attempted using RI techniques. In this study results concerning RI ventriculography and RI transfer test from CSF to plasma were reported. "Barrier ratio" and regional cerebral blood flow study in NPH will be reported elsewhere. Fifty-three patients with chronic communicating hydrocephalus wer devided into two groups. Thus, twenty-seven cases were diagnosed as NPH according to clinical symptoms and signs, and others were considered as mere chronic communicating hydrocephalus without
