Bifidobacterium longum subsp. infantis B8762 modulates the infant gut-lung axis via microbial and metabolic reprogramming.

Respiratory and gastrointestinal infections are leading causes of morbidity in children. Increasing evidence highlights the gut-lung axis as … biosynthesis and enhanced predicted production of immunoregulatory metabolites including butyrate, inosine, and chenodeoxycholic acid. In summary, this study suggests that B8762 modulates the pediatric gut microbiota toward a composition and metabolic profile that supports mucosal barrier integrity and systemic immune regulation. These findings provide m …

RNA editing for the treatment of alpha-1 antitrypsin deficiency.

Alpha-1 antitrypsin deficiency (AATD) is both a gain- and loss-of-function disease that impacts the liver and lung. Most severe cases result from a homozygous missense mutation in the SERPINA1 gene (Z mutation). While current therapies and those in development may ameliorate lung or liver disease, few are designed to address both. We have developed SERPINA1-994, an N-Acetylgalactosamine-conjugated chemically modified oligonucleotide that elicits adenine-to-inosine RNA editing using endogenous adenosine deaminases acting on RNA enzymes, to correct SERPINA1 Z transcripts. We show that SERPINA1-9…

Microbiota-driven uric acid reduction by Lactiplantibacillus plantarum 15-5 through nucleoside catabolism and short-chain fatty acid-mediated host protection.

Hyperuricemia, driven by disrupted purine metabolism, predisposes individuals to hepatic and renal injury. To explore potential microbial interventions, 50 lactic acid bacterial (LAB) strains were isolated from Tibetan fermented foods, and 3 nucleoside-degrading candidates were identified, including Lactiplantibacillus plantarum 15-5, Lactiplantibacillus plantarum YL-2, and Lacticaseibacillus paracasei XS23. Strain 15-5 eliminated 99% of inosine and guanosine within 1 h, surpassing YL-2 and XS23. In hyperuricemic mice, L. plantarum 15-5 reduced serum uric acid by 42.91%, normalized hepatic xan…

Neuron-specific deletion of ADAR1 induces brain malformation and early postnatal lethality.

Adenosine deaminase acting on RNA 1 (ADAR1), which mediates adenosine-to-inosine RNA editing, is expressed as two isoforms, p110 and p150. Deletion of Adar1 p150 in mice results in embryonic lethality caused by aberrant activation of melanoma differentiation-associated protein 5 (MDA5)-mediated sensing of unedited endogenous transcripts, whereas Adar1 p110-specific deficient mice die postnatally through RNA editing-independent mechanisms. ADAR1 mutations cause Aicardi-Goutières syndrome (AGS), a congenital autoinflammatory disease accompanied by encephalopathy with a type I interferon (IFN) si…

TadA-mediated A-to-I mRNA editing rewires redox metabolism to promote dominance of epidemic Klebsiella pneumoniae clones.

Dominant carbapenem-resistant Klebsiella pneumoniae (CRKP) clones have markedly intensified the burden of invasive infections while severely restricting treatment options. Although genomic determinants of these high-risk lineages are well characterized, adaptive regulatory mechanisms beyond fixed DNA variation remain poorly understood. Here we systematically map the adenosine-to-inosine (A-to-I) mRNA editing landscape in CRKP and reveal that clinically dominant lineages possess a constrained and clone-specific landscape of RNA editing. We identify PncR (a previously uncharacterized AraC/XylS f…

ADAR1 and ADAR2 Expression in the Thoracic Aortic Wall Correlates with Aneurysm Severity and Dissection Risk: Insights into A-to-I RNA Editing Dysregulation in Marfan Syndrome-Derived vSMCs.

Adenosine-to-inosine (A-to-I) RNA editing, catalyzed by ADAR1 and ADAR2, is an epitranscriptomic mechanism with emerging roles in cardiovascular disease. We investigated ADAR expression, clinical correlates, A-to-I RNA editing signatures, transcriptome and miRNA targetome in thoracic aortic aneurysm (TAA), focusing on Marfan syndrome (MFS). Immunohistochemical analysis of 58 TAA specimens (18 MFS-associated TAA, 19 tricuspid aortic valve-associated TAA (TAV), 21 bicuspid aortic valve-associated TAA (BAV)) showed the highest ADAR1 and ADAR2 in MFS. ADAR2 in vascular smooth muscle cells (vSMCs) …

Emerging roles of RNA modifications in normal development and congenital craniofacial malformations.

RNA modifications represent a pivotal epitranscriptomic layer modulating gene expression beyond the classic central dogma. Increasing studies have revealed their essential roles in orchestrating mammalian development and contributing to congenital disorders. In this review, we focus on seven well-characterized RNA modifications, including N6-methyladenosine(m6A), N1-methyladenosine(m1A), 5-methylcytosine (m5C), N4-acetylcytosine (ac4C), N7-methylguanosine (m7G), pseudouridine (Ψ), and adenosine-to-inosine (A-to-I) editing, and briefly introduce emerging marks like N1-methylguanosine at positio…

Chemistry and Pharmacological diversity of Benzothiazepine - Excellent pathway to drug discovery.

Recent studies in distinct preclinical tumor models have established the nucleotide synthesis enzyme inosine-5'-monophosphate dehydrogenase (IMPDH) as a viable target for antitumor therapy. IMPDH inhibitors have been used clinically for decades as safe and effective immunosuppressants. However, the potential to repurpose these pharmacological agents for antitumor therapy requires further investigation, including direct comparisons of available compounds. Therefore, we tested structurally distinct IMPDH inhibitors in multiple cell and mouse tumor models of the genetic tumor syndrome tuberous sc…
