Resistance of head louse (Pediculus humanus capitis de Geer) to DDT in Malaysia.

A survey of 308,101 schoolchildren conducted in 10 of 11 states in Peninsular Malaysia revealed that 10.7% have Pediculus humanus capitis infestation. The results of treatment with 1%, 2% and 5% DDT in coconut oil in 374 cases reveal cure rates of 3.5%, 15.8% and 51.3%, respectively, suggesting that the head louse has probably developed resistance to this insecticide in Malaysia.

Protocols for Testing the Toxicity of Novel Insecticidal Chemistries to Mosquitoes.

New classes of insecticides with novel modes of action are needed to control insecticide resistant populations of mosquitoes that transmit diseases such as Zika, dengue and malaria. Assays for rapid, high-throughput analyses of unformulated novel chemistries against mosquito larvae and adults are presented. We describe protocols for single point-dose and dose response assays to evaluate the toxicity of small molecule chemistries to the Aedes aegypti vector of Zika, dengue and yellow fever, the malaria vector, Anopheles gambiae and the northern house mosquito, Culex quinquefasciatus, on contact

An amino acid substitution attributable to insecticide-insensitivity of acetylcholinesterase in a Japanese encephalitis vector mosquito, Culex tritaeniorhynchus.

A cDNA sequence encoding a Drosophila Ace-paralogous acetylcholinesterase (AChE) precursor of 701 amino acid residues was identified as the second AChE gene (Ace2) transcript from Culex tritaeniorhynchus. The Ace2 gene is tightly linked to organophosphorus insecticide (OP)-insensitivity of AChE on chromosome 2. The cDNA sequences were compared between an insecticide-susceptible strain and the resistant strain, TYM, that exhibits a 870-fold decrease in fenitroxon-sensitivity of AChE. Two amino acid substitutions were present in TYM mosquitoes. One is F455W whose homologous position in Torped AC

G3'MTMD3 in the insect GABA receptor subunit, RDL, confers resistance to broflanilide and fluralaner.

Meta-diamides (e.g. broflanilide) and isoxazolines (e.g. fluralaner) are novel insecticides that target the resistant to dieldrin (RDL) subunit of insect γ-aminobutyric acid receptors (GABARs). In this study, we used in silico analysis to identify residues that are critical for the interaction between RDL and these insecticides. Substitution of glycine at the third position (G3') in the third transmembrane domain (TMD3) with methionine (G3'M TMD3), which is present in vertebrate GABARs, had the strongest effect on fluralaner binding. This was confirmed by expression of RDL from the rice stem b

Assessment of a Density-Based Action Threshold for Suppression of Sugarcane Aphids, (Hemiptera: Aphididae), in the Southern High Plains.

In the High Plains, sugarcane aphid, Melanaphis sacchari Zehntner (Hemiptera: Aphididae), colonize grain sorghum, Sorghum bicolor (L) Moench (Cyperales: Poaceae), when it is in reproductive stages and aphid populations increase rapidly. Sugarcane aphid management recommendations based on aphid densities have been implemented in southern United States, but have not been assessed in the High Plains. Therefore, the goal of this work was to validate whether these guidelines can provide effective suppression of the aphids and improve yield. In a 2-y field experiment, sorghum susceptible (DKS44-20)

Characterization of azadirachtin resistance in a laboratory-selected strain of Drosophila. melanogaster.

Azadirachtin, a natural sesquiterpenoid compound, exhibits broad-spectrum insecticidal activity, yet its mechanism of action remains incompletely understood. This study aimed to elucidate its toxicological mechanisms by developing an azadirachtin-resistant Drosophila strain through continuous laboratory selection. After five years of directional screening, we successfully established the Aza-Sel strain with a resistance ratio of 86.7-fold. Genetic analysis revealed that the resistance trait is incompletely recessive and polygenic in nature. Detoxification enzyme and synergism assays demonstrat

RNA interference of Argonaute-1 disrupts ovarian development and enhances insecticide susceptibility in Tribolium castaneum.

BACKGROUND: With the development of rapid resistance, new modes of action for pesticides are needed for insect control, such as RNA interference (RNAi)-based biopesticides targeting essential genes. To explore the function of Argonaute-1 (Ago-1) and potential microRNAs (miRNAs) in ovarian development and their impact on rapid resistance in important storage pest Tribolium castaneum, we knocked down Ago-1 and observed the development of ovarian and insecticide susceptibility. RESULTS: Our results indicated that Ago-1 had a relatively high transcriptional level of adult female T. castaneum duri

The symbiont Wolbachia alleviates pesticide susceptibility in the two-spotted spider mite Tetranychus urticae through enhanced host detoxification pathways.

The two-spotted spider mite (Tetranychus urticae) is one of the most well-known pesticide-resistant agricultural pests, with resistance often attributed to changes such as target-site mutations and detoxification activation. Recent studies show that pesticide resistance can also be influenced by symbionts, but their involvement in this process in spider mites remains uncertain. Here, we found that infection with Wolbachia, a well-known bacterial reproductive manipulator, significantly increased mite survival after exposure to the insecticides abamectin, cyflumetofen, and pyridaben. Wolbachia-i

Uridine diphosphate glucosyltransferases are involved in spinosad resistance in western flower thrips Frankliniella occidentalis (Pergande).

Uridine diphosphate glucosyltransferases (UGTs) play crucial roles in the insect detoxification system and are associated with pesticide resistance. Our previous transcriptomic analysis of spinosad-susceptible (Ivf03) and resistant (NIL-R) Frankliniella occidentalis revealed numerous upregulated UGT genes in the NIL-R strain, suggesting their potential contribution to spinosad resistance. To investigate this hypothesis, here we conducted UGT activity assays and spinosad induction experiments, employing RNA interference (RNAi) techniques for gene function validation. We found significantly elev

Knockdown of the glutamate-gated chloride channel gene decreases emamectin benzoate susceptibility in the fall armyworm, Spodoptera frugiperda.

Emamectin benzoate (EB), a derivative of avermectin, is the primary insecticide used to control the fall armyworm (FAW) in China. However, the specific molecular targets of EB against FAW remain unclear. In this study, we cloned the glutamate-gated chloride channel (GluCl) gene, which is known to be a primary molecular target for avermectin. We first investigated the transcript levels of SfGluCl in FAW and found that the expression level of SfGluCl in the head and nerve cord was significantly higher than that in other tissues. Furthermore, we found that the expression level of SfGluCl was sign

Sequential phase I metabolism of pyrethroids by duplicated CYP6P9 variants results in the loss of the terminal benzene moiety and determines resistance in the malaria mosquito Anopheles funestus.

Pyrethroid resistance in Anopheles funestus is threatening the eradication of malaria. One of the major drivers of pyrethroid resistance in An. funestus are cytochrome P450 monooxygenases CYP6P9a and CYP6P9b, which are found upregulated in resistant An. funestus populations from Sub-Saharan Africa and are known to metabolise pyrethroids. Here, we have functionally expressed CYP6P9a and CYP6P9b variants and investigated their interactions with azole-fungicides and pyrethroids. Some azole fungicides such as prochloraz inhibited CYP6P9a and CYP6P9b at nanomolar concentrations, whereas pyrethroids

First detection of voltage-gated sodium channel mutations in Phlebotomus argentipes collected from Bangladesh.

BACKGROUND & OBJECTIVES: Phlebotomus argentipes is the main vector of visceral leishmaniasis in Bangladesh and is controlled using deltamethrin, a synthetic pyrethroid, through indoor residual spraying (IRS). A mutation at L1014 (leucine at codon 1014) of the voltage-gated sodium channel (VGSC), known as a knockdown resistance (kdr) gene, is thought to be an important pyrethroid resistance mechanism. This study detected mutations at codon 1014, and at codons 1011, 1016, and 1020, which are kdr sites in other insects. The kdr relationship with deltamethrin resistance in P. argentipes from an IR

Detection of Male Oriental Fruit Fly (Diptera: Tephritidae) Susceptibility to Naled- and Fipronil-Intoxicated Methyl Eugenol.

Naled-intoxicated methyl eugenol (ME) is commonly used to control oriental fruit flies, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae), in Taiwan. However, non-responsiveness to ME and pesticide resistance in oriental fruit flies may reduce control efficacy. In this study, mark-recapture experiments were used to analyze the effects of naled-intoxicated ME on field and naled-resistant fly strains. ME non-responsiveness was tested in field, naled-resistant, and susceptible strains and pyrosequencing techniques were used to detect frequencies of point mutations on organophosphate resistant a

DDT-based indoor residual spraying suboptimal for visceral leishmaniasis elimination in India.

Indoor residual spraying (IRS) is used to control visceral leishmaniasis (VL) in India, but it is poorly quality assured. Quality assurance was performed in eight VL endemic districts in Bihar State, India, in 2014. Residual dichlorodiphenyltrichloroethane (DDT) was sampled from walls using Bostik tape discs, and DDT concentrations [grams of active ingredient per square meter (g ai/m(2))] were determined using HPLC. Pre-IRS surveys were performed in three districts, and post-IRS surveys were performed in eight districts. A 20% threshold above and below the target spray of 1.0 g ai/m(2) was def
