Impaired glucose-stimulated insulin secretion in the GK rat is associated with abnormalities in islet nitric oxide production.

We investigated implications of nitric oxide (NO) derived from islet neuronal constitutive NO synthase (ncNOS) and inducible NOS (iNOS) on insulin secretory mechanisms in the mildly diabetic GK rat. Islets from GK rats and Wistar controls were analysed for ncNOS and iNOS by HPLC, immunoblotting and immunocytochemistry in relation to insulin secretion stimulated by glucose or l-arginine in vitro and in vivo. No obvious difference in ncNOS fluorescence in GK vs control islets was seen but freshly isolated GK islets displayed a marked iNOS expression and activity. After incubation at low glucose …

Insulin sensitivity and beta-cell secretion in thalassaemia major with secondary haemochromatosis: assessment by oral glucose tolerance test.

UNLABELLED: Diabetes mellitus in patients with thalassaemia major is caused by secondary haemochromatosis due to transfusional iron overload. The pathogenetic mechanisms leading from siderosis to diabetes are still poorly understood. This study aimed at assessing the influence of insulin resistance and insulin deficiency on that process. Glucose, insulin and C-peptide levels during oral glucose tolerance tests (OGTT) from 36 thalassaemic patients with normal ( n=23), impaired ( n=6), or diabetic glucose tolerance ( n=7) and 32 control subjects were examined. Insulin secretion and insulin sensi…

Insulin secretion and action show differences in impaired fasting glucose and in impaired glucose tolerance in Asian Indians.

BACKGROUND: The aim of the study was to compare the abnormalities in insulin secretion and insulin sensitivity in impaired fasting glucose (IFG) and in impaired glucose tolerance (IGT). METHODS: Glucose tolerance was determined by oral glucose tolerance test in 289 non-diabetic subjects selected for this study (M : F, 160 : 129, mean age 42.2 +/- 11.0 (SD) years, mean body mass index (BMI) 24.9 +/- 4.2 kg/m(2)). Diabetic subjects were excluded from the study. The association of insulin resistance and beta-cell function (calculated by HOMA model) with IFG and IGT was studied using linear and m…

The calcimimetic R-467 potentiates insulin secretion in pancreatic beta cells by activation of a nonspecific cation channel.

The extracellular, G protein-linked Ca(2+)-sensing receptor (CaSR), first identified in the parathyroid gland, is expressed in several tissu … e stimulation on islets, raises the possibility of a totally new class of drugs that will stimulate insulin secretion during hyperglycemia but which will not cause hypoglycemia.

Differential inhibition of insulin and islet amyloid polypeptide secretion by intraislet somatostatin in the isolated perfused human pancreas.

Islet amyloid polypeptide (IAPP) and insulin are co-stored and generally secreted in parallel; however, studies have demonstrated that the IAPP/insulin molar secretory ratio may be altered in response to certain stimuli. Because we previously demonstrated that intraislet somatostatin is an inhibitory regulator of basal insulin secretion in the isolated perfused human pancreas, this study was designed to determine the relative influence on the regulation of IAPP versus insulin secretion. Single-pass perfusion was performed in pancreata obtained from cadaveric organ donors with continuous perfus…

Arrest of pulsatile luteinizing hormone (LH) secretion during insulin-induced hypoglycemia (IIH): improvement by intrahypothalamic perfusion with glucose.

Insulin-induced hypoglycemia (IIH), as with many other acute stressors, restrains the activity of the reproductive axis, reducing luteinizing hormone (LH) release. In adult ovariectomized, steroid-primed rats, we investigated the effect of IIH and of mediobasal intrahypothalamic perfusion with glucose (200 mg/dl) on pulsatile LH secretion. IIH led to a significant decrease in all pulsatility parameters studied using PC-Pulsar analysis, e.g. pulse amplitude and frequency, maximum and baseline LH levels (p < 0.05 versus control), and LH overall mean release (p < 0.01 versus control). Intrahypoth

Effects of taurine on the insulin secretion of rat fetal islets from dams fed a low-protein diet.

An isocaloric low-protein (LP) diet (8% instead of 20% in controls) given to dams during gestation reduces the fractional insulin release of stimulated fetal islets. The LP diet lowers the plasma concentration of taurine in both pregnant rats and their fetuses. This study reports the effect of taurine on the in vitro release of insulin from control and LP fetal islets. Direct stimulation with taurine, methionine or leucine increased the release of insulin from control islets. Nevertheless, no effect on LP islets was observed with either taurine or methionine. The release of insulin from LP isl

Impaired insulin but normal pentagastrin effect on gastric acid secretion in diabetic rats: a role for nitric oxide.

Significant changes in gastrointestinal function, decreased gastric secretion and motility in particular, are often observed in patients with chronic diabetes. The mechanisms leading to those remain unclear. In these studies we evaluated the gastric acid secretory response to insulin and pentagastrin in normal Wistar and streptozotocin diabetic rats. We also sought to determine the role of nitric oxide (NO) in this process. The animals were anesthetized with sodium pentobarbital. Warm saline was perfused through a polyethylene tube placed in the oesophagus and collected from the duodenum at 10

Impact of long-term naltrexone treatment on growth hormone and insulin secretion in hyperandrogenic and normal obese patients.

The growth hormone (GH) response to stimulation tests is impaired in obesity. Moreover, obese patients exhibit a "paradoxical" increase of GH to GH-releasing hormone (GHRH) stimulation after food ingestion; this paradoxical response is reversed by naloxone infusion. On the other hand, beta-endorphin seems to exert profound effects on insulin release. Recent studies also demonstrated an impairment of GH response to several stimuli in polycystic ovary syndrome (PCOS), a condition associated with obesity, hyperinsulinism, and insulin resistance. Chronic inhibition of opioid tone by the opioid ant

[Increase in neuroendocrine secretion granules in submaxillary and parotid glands in patients with non-insulin dependent diabetes mellitus].

The amount of neuroendocrine granules in microgranular cells of salivary glands were investigated in necropsies of 20 non-insulin dependent diabetics (NIDDM) and in 20 non-diabetic subjects with Grimelius and Fontana-Masson stainings. Granules in serous acini, and intercalated ducts were observed in both groups; however, a significant higher number of granules and microgranular cells were observed in NIDDM subjects as compared with non-diabetics. Both parotid and submaxillary glands were significantly heavier in NIDDM than in the non-diabetic group. These granules may be related to immunoreact

[Secretion of atrial natriuretic peptide in patients with non-insulin-dependent diabetes mellitus and effect of autonomic neuropathy].

In order to clarify the mode of atrial natriuretic peptide (ANP) release and the effect of autonomic nerve function on ANP release, we measured plasma ANP concentrations in response to hypertonic saline infusion in patients with non-insulin-dependent diabetes mellitus (NIDDM) having or not having autonomic neuropathy (AN). Studies were made on 72 normal subjects (male 44, female 28; 53.0 +/- 0.9 yr.), and 63 patients with NIDDM (male 36, female 27; 56.9 +/- 2.1 yr.). The patients with NIDDM were divided into two groups: Group A was 48 diabetics without AN, and Group B was 15 diabetics with AN.

Consequences of ventromedial hypothalamic lesions upon insulin and glucagon secretion by subsequently isolated perfused pancreases in the rat.

The existence of a relationship between the ventromedial hypothalamic area (VMH) and the activity of the endocrine pancreas has been shown previously. This relationship has been further tested and extended in the present study, using isolated perfused pancreases from rats previously lesioned (4-7 d) in the VMH. It was found that in isolated pancreases obtained from rats fed ad lib. for 4 d after VMH lesions (i.e., that were hyperphagic), the typical biphasic pattern of insulin secretion was observed following glucose stimulation (20 mM) and that the total insulin output was much greater than t…

ABCA1 as a Gatekeeper of Pancreatic β-Cell Cholesterol Homeostasis: Links Between Lipoprotein Stress, Lipotoxicity, and Insulin Secretion.

Pancreatic β cells require tightly controlled cholesterol distribution to maintain glucose sensing, insulin-granule trafficking, and regulated exocytosis. ATP-binding cassette transporter A1 (ABCA1) exports cellular cholesterol and phospholipids to lipid-poor apolipoprotein A-I and is a central component of β-cell cholesterol homeostasis. β-cell-specific Abca1 deletion causes cholesterol accumulation and impaired glucose-stimulated insulin secretion, whereas human studies of rare ABCA1 loss-of-function mutations have reported heterogeneous secretory phenotypes. Endocrine, metabolic, inflammato…

FAM151A Regulates Insulin Secretion in Pancreatic β-Cells and Is Implicated in Pathogenesis of Type 2 Diabetes Mellitus.

BACKGROUND: The pathophysiology of type 2 diabetes mellitus (T2DM) is characterized by insulin resistance in peripheral tissues and dysfunction of β-cells. However, the molecular mechanisms underlying β-cell dysfunction remain incompletely understood. METHODS: Analysis of single-cell RNA sequencing of T2DM islets was used to elucidate changes in family with sequence similarity 151 member A (Fam151a) expression. Deletion of Fam151a in INS-1E cells and mice was used to investigate the function of this gene in insulin secretion. Metabolic profiling and metabolomics analyses were used to decipher…

Direct electrochemical modulation of the membrane potential enables insulin secretion control in pancreatic β-cells cultured on electrodes.

Interfaces that convert digitally defined signals into biological analog functions (D/A biointerfaces) remain underdeveloped compared with biosensors that digitize biological analog information (A/D biointerfaces). Here, we report direct electrochemical control of insulin secretion through membrane-potential modulation using a stable tetraethyl orthosilicate (TEOS)-incorporated PEDOT:PSS electrode that supports long-term culture of pancreatic β-cells as a model endocrine system. β-cells expressing luminescent insulin (iGL cells) were cultured on the electrode, and secretion dynamics were monit…

TrkB Receptor Antagonism Enhances Insulin Secretion and Increases Pancreatic Islet Size in Rats Fed a Cafeteria-Style Diet.

Background: In recent years, the role of neurotrophins and their receptors in peripheral tissues has been of great interest. At a metabolic level, the brain-derived neurotrophic factor (BDNF) and its receptor trkB have been reported to participate in insulin secretion from the pancreas in response to increases in circulating blood glucose. Objetive: To determines the role of the BDNF-trkB pathway in insulin secretion and pancreatic morphology in rats fed a cafeteria-style diet for 16 weeks. Methods: For the study, male rats of the Wistar strain were divided into three groups as follows: (1) co…

FFAR4-mediated IL-6 release from islet macrophages promotes insulin secretion and is compromised in type-2 diabetes.

The function of islet macrophages is poorly understood. They promote glucose-stimulated insulin secretion (GSIS) in lean mice, however, the underlying mechanism has remained unclear. We show that activation of the free fatty acid receptor FFAR4 on islet macrophages leads to interleukin-6 (IL-6) release and that IL-6 promotes β-cell function. This mechanism is required for GSIS in lean male mice, but does not function anymore in islets from people with obesity and obese type 2 diabetic male mice. In islets from obese mice, FFAR4 downstream signaling in macrophages is strongly reduced, resulting…

Intravenous D‑allose administration improves blood glucose control by maintaining insulin secretion in diabetic mice with transplanted islets.

Although pancreatic islet transplantation outcomes have improved, further refinements are required to extend the insulin withdrawal period. The present study examined whether intravenous D-allose administration improves insulin secretion when pancreatic islets are transplanted into type 1 diabetes model mice. Alterations in casual blood glucose levels, intraperitoneal glucose tolerance test (IPGGT) results, the number of apoptotic cells in the engrafted cells, and caspase 3, heme oxygenase 1 and nitric oxide synthase 2 (NOS2) expression in the engrafted cells were examined using the following …
