The inflammatory UPR-IFNG axis reveals LBP as a survival and immunotherapy-response marker in hepatocellular carcinoma.

BACKGROUND: Liver hepatocellular carcinoma (LIHC) remains difficult to stratify because tumor heterogeneity and an immunosuppressive microenvironment limit immune checkpoint blockade (ICB). The unfolded protein response (UPR) and interferon-gamma (IFNG) signaling connect cellular stress with tumor inflammation, but clinically interpretable biomarkers linking these programs to prognosis and treatment response remain limited. METHODS: We integrated 278,337 cells from three single-cell RNA sequencing (scRNA-seq) datasets with bulk transcriptomic cohorts. Malignant-cell programs were mapped at si

Lifespan-extending interventions induce consistent patterns of fatty acid oxidation in mouse livers.

Aging manifests as progressive deteriorations in homeostasis, requiring systems-level perspectives to investigate the gradual molecular dysregulation of underlying biological processes. Here, we report systemic changes in the molecular regulation of biological processes under multiple lifespan-extending interventions. Differential Rank Conservation (DIRAC) analyses of mouse liver proteomics and transcriptomics data show that mechanistically distinct lifespan-extending interventions (acarbose, 17α-estradiol, rapamycin, and calorie restriction) generally tighten the regulation of biological modu

The Role of SR-BI in sepsis: leveraging mechanistic insights to advance precision steroid therapy.

According to the Surviving Sepsis Campaign, 50.3% of septic shock patients received steroid/glucocorticoid (GC) therapy. However, whether GC therapy is beneficial and who might benefit from it are hotly debated. Initial guidelines recommended GC therapy for septic patients with adrenal insufficiency, but this has since been retracted. Recent studies using animal models of adrenal insufficiency have shed light on the mechanisms, demonstrating that the adrenal stress response is a part of the host response that is essential for control inflammatory response in sepsis and the adrenal insufficienc

Quantitative proteome analysis of an antibiotic resistant Escherichia coli exposed to tetracycline reveals multiple affected metabolic and peptidoglycan processes.

UNLABELLED: Tetracyclines are among the most commonly used antibiotics administrated to farm animals for disease treatment and prevention, contributing to the worldwide increase in antibiotic resistance in animal and human pathogens. Although tetracycline mechanisms of resistance are well known, the role of metabolism in bacterial reaction to antibiotic stress is still an important assignment and could contribute to the understanding of tetracycline related stress response. In this study, spectral counts-based label free quantitative proteomics has been applied to study the response to tetracy

Emotional distress is associated with neuroendocrine-immune remodeling and less favorable neoadjuvant immunotherapy outcomes in oral squamous cell carcinoma.

BACKGROUND: Immunotherapy research, particularly in the neoadjuvant setting, has largely focused on tumor-intrinsic determinants, with limited attention to host-related factors. Emotional distress (ED), a prevalent condition in cancer patients, remains poorly understood in the context of neoadjuvant immunotherapy (NAIT). METHODS: We analyzed a cohort of 68 patients with locally advanced OSCC treated with NAIT. ED was assessed using the EORTC QLQ-C30 to stratify patients, and clinical outcomes were evaluated to determine therapeutic efficacy. Serum neuroendocrine markers were measured to asses

Synergistic melatonin and myo-inositol application reinforces antioxidant defence, osmotic homeostasis, and membrane stability in Western Himalayan apple rootstocks under drought stress.

Drought stress induces profound biochemical and physiological disturbances in apple (Malus × domestica Borkh.), including oxidative imbalance, chlorophyll degradation, osmotic disruption, and membrane injury. This study evaluated the individual and synergistic effects of two key metabolites, melatonin (MT) and myo-inositol (MI) in enhancing drought resilience across five genetically diverse apple rootstocks (MM 111, MM 106, Bud 118, MM 116, and M9). A two-phase experimental framework was implemented, beginning with metabolite dose optimisation under 40% field capacity using the Response Index

Rv3839-Rv3840 links the endogenous heme biosynthesis pathway with Mycobacterium tuberculosis adaptation to nitric oxide and iron limitation stress.

During infection, Mycobacterium tuberculosis (Mtb) encounters multiple environmental stressors, including nitric oxide (NO) and iron limitation, and an ability to mount an integrated response is essential for the bacterium's adaptation and continued survival. Iron-containing prosthetic groups in key enzymes are critical for Mtb sensing and detoxification of NO, and there is significant overlap between NO- and low iron-responsive genes. However, how Mtb adapts to these two stressors concurrently is largely unknown. Here, we find that exposure to NO globally augments expression of low iron-respo

Painful facet joint injury induces neuronal stress activation in the DRG: implications for cellular mechanisms of pain.

The cervical facet joint is implicated as one of the most common sources of chronic neck pain, owing to its rich nociceptive innervation and susceptibility to injurious mechanical loading. Injuries to the facet joint and its ligament can induce inflammation in the joint and spinal cord. Inflammatory molecules which are known to have a role in pain can also stimulate the integrated stress response (ISR). Therefore, we hypothesize that ISR is activated by facet joint injury in a rodent model of pain. To address this hypothesis, we assessed the expression of binding protein (BiP) (also known as g…

Transcriptomic differences in chromatin and cell cycle regulation in A549 cells after irradiation with carbon ions and X-rays.

INTRODUCTION: High-linear energy transfer (LET) radiation such as carbon ions exhibits greater biological effectiveness than conventional low-LET X-rays, but the transcriptional mechanisms underlying this advantage remain incompletely understood. We hypothesized that high-LET radiation induces a qualitatively different transcriptional response rather than simply amplifying low-LET signaling. METHODS: A549 non-small cell lung cancer cells were exposed to equal physical doses (8 Gy) of X-rays or carbon ions (LET 73 keV/µm), and transcriptomic profiling was performed 4 h post-irradiation. Host-microbe signatures of oxidative stress and epithelial apoptosis in bacterial vaginosis during pregnancy.

BACKGROUND: Bacterial vaginosis (BV) is a common vaginal dysbiosis associated with adverse pregnancy outcomes. However, the extent to which microbial indicators of dysbiosis covary with host oxidative stress and epithelial apoptotic responses during pregnancy remains unclear. METHODS: In this analytical cross-sectional study, 220 pregnant women were screened at a referral gynecology clinic in Faisalabad, Pakistan; 168 met the eligibility criteria and had concordant modified Amsel and Nugent classifications (56 BV-positive, 112 BV-negative). Cervicovaginal secretions, vaginal swabs, and serum

Noninvasive monitoring of adrenocortical activity in roe deer (Capreolus capreolus) by measurement of fecal cortisol metabolites.

A method for measuring glucocorticoids noninvasively in feces of roe deer was established and validated. The enzyme immunoassay (EIA) measures 11,17-dioxoandrostanes (11,17-DOA), a group of cortisol metabolites. Such measurement avoids blood sampling and reflects a dampened pattern of diurnal glucocorticoid secretion, providing an integrated measure of adrenocortical activity. After high-performance liquid chromatography, the presence of at least three different immunoreactive 11,17-DOA in the feces of roe deer was demonstrated. The physiological relevance of these fecal cortisol metabolites t

Endoplasmic reticulum stress as a mechanistic link between nutrition and inflammatory bowel disease.

Inflammatory bowel diseases (IBD) are chronic inflammatory disorders that arise from the complex interplay among genetic predisposition, immune dysregulation, environmental factors, and disruption of intestinal epithelial barrier integrity. Increasing evidence suggests that endoplasmic reticulum (ER) stress may represent an important mechanistic pathway in this multifactorial pathogenesis. Experimental evidence indicates that dietary patterns and nutrient-derived factors may modulate ER stress responses. In this review, the role of ER stress in IBD is discussed from a mechanistic perspective,

The Role of Integrated HBV DNA in the Quest for a Cure in HIV/HBV co-infection.

PURPOSE: This review highlights the importance of Hepatitis B virus (HBV) integrated into the host genome (iDNA) to functional cure and how human immunodeficiency virus (HIV) may affect HBV integration. RECENT FINDINGS: Functional cure of chronic hepatitis B infection is characterized by durable loss of hepatitis B surface antigen (HBsAg) from blood after treatment discontinuation. Because HBsAg is transcribed from two intrahepatic sources, covalently closed circular DNA (cccDNA) or iDNA, functional cure requires elimination or silencing of both sources. It is not clear how HIV affects HBV in

Drought and salinity stress remodel Asian rice (Oryza sativa) leaf development through cell-type-specific regulatory programs.

Osmotic stressors impair tissue expansion and slow crop development, which can ultimately reduce yield. The mechanisms through which dividing and differentiating cells perceive, integrate, and respond to stress are not well understood. To explore this knowledge gap, we assessed Asian rice (Oryza sativa) seedling responses to drought and salinity stress by performing single-nucleus transcriptome analyses on developing leaves and by assaying ion distribution using ICP-MS and CoroNa Green staining. With a dataset of > 125 000 high-quality nuclei, we identified transcriptome signatures of developm

Integration of troponin I phosphorylation with cardiac regulatory networks.

We focus here on the modulation of thin filament activity by cardiac troponin I phosphorylation as an integral and adaptive mechanism in cardiac homeostasis and as a mechanism vulnerable to maladaptive response to stress. We discuss a current concept of cardiac troponin I function in the A-band region of the sarcomere and potential signaling to cardiac troponin I in a network involving the ends of the thin filaments at the Z-disk and the M-band regions. The cardiac sarcomere represents a remarkable set of interacting proteins that functions not only as a molecular machine generating the heartb

Oxidative stress and genotoxic effects in gill and kidney of Anguilla anguilla L. exposed to chromium with or without pre-exposure to beta-naphthoflavone.

Fish in the aquatic environment can be subjected to a multipollution state and the occurrence of sequential exposures is an important aspect of eco-toxicological research. In this context, a preceding exposure can affect a toxic response to a subsequent exposure. Therefore, the current study was based on sequential exposures, viz. to a PAH-like compound (beta-naphthoflavone, BNF) followed by a heavy metal (chromium, Cr), focusing on the assessment of oxidative stress responses and their role in induction of genotoxicity. Oxidative stress responses in gill and kidney were investigated in Europe

Heat Stress Induces Metabolic and Physiological Imbalance in Laying Hens, Accompanied by Hepatic Transcriptomic, Cecal Microbial, and Metabolomic Alterations.

Heat stress is a major constraint to productivity and physiological homeostasis in laying hens. This study investigated integrated responses to acute heat stress using a multi-omics approach, including performance traits, serum biochemical parameters, histology, hepatic transcriptomics, cecal metagenomics, and metabolomics. Acute heat stress impaired productive performance, as reflected by changes in egg production and reduced eggshell strength, and induced systemic physiological disturbances, including increased stress- and injury-related blood indicators and disrupted metabolic and electroly

Integrated control of MRSA biological contamination via SarA-targeting γ-mangostin: A green natural antimicrobial strategy.

Methicillin-resistant Staphylococcus aureus (MRSA) remains a major public health threat owing to its pronounced antibiotic resistance, multiple virulence factors, and robust biofilm-forming capacity, yet effective integrated antimicrobial strategies are still lacking. The global regulator SarA serves as a regulatory hub that simultaneously governs these key MRSA hazard determinants. Here, we report a comprehensive platform for the all-round control of MRSA biological contamination, centered on a label-free electrochemical impedance spectroscopy (EIS)-based biosensing system designed to screen

Virtual reality-based stress paradigms and early adversity: a scoping review of mechanisms and stress-response systems.

BACKGROUND/OBJECTIVES: Virtual reality (VR)-based stress paradigms are increasingly used to induce stress in controlled yet ecologically valid settings. However, the diversity of VR-based stress paradigms and their differential engagement of stress-response systems remain insufficiently synthesized. This scoping review aimed to map VR stress paradigms and examine how they engage subjective, autonomic, endocrine, neural, and behavioral responses, while considering the role of early adversity. METHODS: A structured literature search was conducted across Web of Science, ScienceDirect, SpringerLi

TaUSP_6D-5 integrates light signalling and heat stress to regulate thermotolerance and developmental transitions in plants.

Universal Stress Proteins (USPs) constitute a conserved class of stress-responsive proteins that play important roles in helping plants adapt to adverse environmental conditions, including heat, oxidative stress, and endoplasmic reticulum (ER) stress. However, the molecular mechanisms through which USPs integrate environmental and developmental signalling pathways remain poorly understood. In this study, we characterized the wheat Universal Stress Protein TaUSP_6D-5 and identified it as a potential link between light signalling and thermotolerance. Our results demonstrated that TaUSP_6D-5 is l
