Laminin - myCAF interplay promotes α6-integrin-mediated breast cancer cell invasion.

It is well established that the tumor microenvironment (TME) plays a dynamic role in breast cancer progression. Stromal components of the TME, including fibroblasts and the extracellular matrix (ECM), provide biophysical and biochemical cues to drive tumor cell proliferation and invasion. However, how the interplay between activated fibroblast (CAF) subtypes and ECM composition impinges on breast cancer cell behavior is still not well understood. To address this gap, we leveraged microfluidic technologies to engineer a model of the breast TME that incorporates tumor cells, CAFs, and ECM compon

Gemcitabine-induced β4 integrin drives cancer progression and gemcitabine resistance in pancreatic cancer.

BACKGROUND: Pancreatic ductal adenocarcinoma (PDAC) is characterized by high mortality and the frequent emergence of chemoresistance, particularly to gemcitabine (GEM). Recent studies have indicated that tumors exhibit increased malignancy following the acquisition of chemoresistance. However, the underlying molecular mechanisms remain poorly understood. This study aimed to elucidate the mechanisms by which PDAC cells acquire GEM resistance and how this resistance drives cancer progression, focusing on identifying the key molecular drivers involved in these processes. METHODS: We established G

Inflammation drives TGFβ1 activation via the αvβ6 integrin-mechanotransduction pathway in human skin.

Transforming growth factor β (TGFβ) is a critical regulator of skin homeostasis and inflammation, including psoriasis. Multiple mechanisms can regulate TGFβ signaling, such as the activation of latent TGFβ1 through integrin-dependent pathways. We investigated the molecular mechanisms of TGFβ1 activation during cutaneous inflammation using reconstructed human epidermis treated with inflammatory cytokine cocktail. We report αvβ6 integrin upregulation and enhanced binding of TGFβ1 latency-associated peptide (LAP), which, however, did not result in active TGFβ1 release, restricting the TGFβ1 activ

DM9CP-integrin-RhoA pathway promotes the hemocyte phagocytosis in the immune priming of Crassostrea gigas.

DM9 domain-containing proteins (DM9CPs) constitute a novel family of pattern recognition receptors (PRRs) recognizing specific ligands to initiate downstream immune responses. Their involvement in phagocytosis and immune priming is crucial for a comprehensive understanding of their regulatory mechanisms on immune responses. In the present study, the content of CgDM9CP-1 protein in the cell-free hemolymph of Crassostrea gigas was significantly upregulated at 3, 6, and 12 h after Vibrio splendidus stimulation, and CgDM9CP-1 was able to interact with CgIntegrin both in vitro and in vivo. After th

SH3BP5L triggers the RAB11A-regulated integrin recycling network implicated in breast cancer metastasis.

Metastatic progression in aggressive breast cancer (BC) depends on a tightly controlled vesicular recycling network regulated by RAB11, a small guanosine triphosphate enzyme (GTPase). In a cohort of more than 1,000 patients with BC, we identified SH3BP5L as the most highly expressed guanine nucleotide exchange factor (GEF) for RAB11A. High SH3BP5L expression marked an advanced tumor stage, distant metastasis, and poor prognosis, with significant associations in human epidermal growth factor receptor 2-positive (HER2+) and triple-negative breast cancer (TNBC). Using Förster resonance energy tra

Myocardin-related transcription factor A contributes to diabetic nephropathy by activating integrin β2 in macrophages.

AIMS: Diabetic nephropathy (DN) is one of the most severe complications for diabetic patients. Previously it has been reported that myocardin-related transcription factor A contributes to DN-associated renal fibrosis by regulating fibroblast phenotype. In the present study we investigated the contribution of macrophage derived MRTF-A to DN pathogenesis. METHODS AND MATERIALS: Macrophage conditional MRTF-A knockout (CKO) mice were generated by cross-breeding the Mrtfaf/f mice to the LyzM-Cre mice. DN was modeled in mice by high-fat diet (HFD) feeding and streptozotocin (STZ) injection. KEY FI

Conformationally Tuned Cyclic RGD Peptides for Integrin-Subtype-Selective PET/CT Imaging.

Integrin-subtype selectivity remains a challenging for RGD-based imaging probes because of conserved binding interfaces among integrins. Here, we developed a conformational tuning strategy for cyclic RGD peptides by varying the carbon-spacer length and turn-inducing motifs in a 25-member library. Biolayer interferometry screening identified subtype-preferred ligands with nanomolar-level apparent affinities toward αvβ6, αvβ3, and α5β1, including 3ba/3bb/3bd, 3be, and 3bc/3dc, respectively. Circular dichroism suggested that cross-linker geometry and turn motifs modulate the RGD peptides' seconda…

Effects of quantum dots on integrin expression and cellular responses in DU145 prostate cancer cells.

Quantum dots (QDs) are extensively investigated for biomedical applications, including cancer diagnostics and therapy; however, their potential subcytotoxic effects on cancer cell behavior remain incompletely understood. The aim of this study was to assess whether exposure to QDs at non-cytotoxic concentrations (0.1-10 μg/mL) modulates the expression of selected integrins and alters adhesion-related functional properties of DU-145 prostate cancer cells. Cytotoxicity was evaluated using sulforhodamine B, neutral red, and resazurin assays. Changes in integrin expression were analyzed at the mRNA…

Molecular PET imaging of integrin αvβ6 reveals biliary and fibrotic changes in preclinical MASH.

BACKGROUND: The progression of metabolic dysfunction-associated steatohepatitis (MASH) involves complex crosstalk among hepatocellular injury, inflammation, and ductular reaction-a process characterized by biliary epithelial proliferation and fibrotic activation. Integrin αvβ6, an epithelial-specific receptor largely absent in normal liver but induced under chronic injury, plays a pivotal role in activating latent TGF-β and driving fibrogenesis. Here, we investigated αvβ6 as a molecular imaging biomarker in MASH using a PET-based strategy to non-invasively visualize epithelial fibrotic signali

Integrin-linked kinase (ILK) in hematologic malignancies: Bridging molecular mechanisms to therapeutic innovation.

Therapy resistance remains a formidable challenge in hematologic malignancies despite significant advances in targeted therapies. This comprehensive review examines integrin-linked kinase (ILK) as a critical molecular hub at the nexus of cell adhesion, signal transduction, and therapy resistance across leukemias, lymphomas, and multiple myeloma. Unlike in solid tumors, where ILK primarily drives invasion and metastasis, in hematologic malignancies it uniquely mediates microenvironmental protection and therapy resistance through distinct signaling networks. ILK functions as a central mediator c

Reduced platelet formation associated with serine metabolic dysregulation in integrin αIIbβ3-deficient megakaryocytes.

Glanzmann thrombasthenia (GT) is characterized by absent platelet aggregation in response to all agonists except ristocetin and is caused by recessive inactivating variants in ITGA2B or ITGB3. Although patients with GT typically are described as having normal platelet counts, autosomal dominant activating variants in ITGA2B or ITGB3 cause macrothrombocytopenia. Interestingly, in our cohort of 16 patients with GT, 8 consistently exhibited platelet counts at the lower end of the normal range. We studied the role of integrin αIIbβ3 in platelet formation using megakaryocytes (MKs) derived from gen

Integrin β1 signaling at the crossroads of cellular plasticity, the tumor microenvironment, and therapy resistance in medulloblastoma.

Medulloblastoma (MB), the most common malignant pediatric brain tumor, remains associated with relapse and long-term treatment-related toxicities. Tumor-microenvironment interactions increasingly appear to contribute to progression, stemness, and therapy resistance in brain cancers. Integrin β1, a major mediator of cell-extracellular matrix interactions, regulates survival signaling, mechanotransduction, and cancer stem cell maintenance in several tumor types, particularly glioblastoma. This mini-review discusses how developmental programs, extracellular matrix remodeling, hypoxia, and cancer

Integrin-Linked Kinase Suppression Reduces SPARC's Effect on Intraocular Pressure and Extracellular Matrix.

PURPOSE: The molecular pathway for increased outflow resistance in the trabecular meshwork (TM) remains unknown. Secreted protein, acidic and rich in cysteine (SPARC) has been shown to regulate intraocular pressure (IOP) correlating to extracellular matrix (ECM) alterations. In other tissues, SPARC binds integrin-linked kinase (ILK) and regulates ECM organization through signaling cascades. We hypothesized SPARC regulates ECM proteins partly through ILK in the TM. METHODS: Adenovirus carrying cDNA of human SPARC (Ad.SPARC) was used to overexpress SPARC and a lentivirus carrying short-hairpin

The estrogen-integrin αvβ3 crosstalk in breast cancer: Modulating PD-L1 and immune evasion.

The mechanisms governing PD-L1 expression in breast cancer are highly complex and context-dependent. This article proposes a novel "dual-track" regulatory model to resolve the conflicting roles of estrogen in tumor immunity. Genomic signaling through ER-α suppresses PD-L1 transcription, explaining lower PD-L1 levels in ER-α-positive tumors. Conversely, non-genomic signaling mediated by cell-surface integrin αvβ3 drives PD-L1 presentation by triggering downstream PI3K/Akt and MAPK/ERK 1/2 pathways. We synthesize emerging data on how post-translational modulators (RNF31, p66Shc) and metabolic re

Integrin β1 Demarks Precursors of Brain-Residing Antibody-Secreting Cells in Multiple Sclerosis.

BACKGROUND AND OBJECTIVES: Multiple sclerosis (MS) is characterized by CXCR3+ memory B cells that infiltrate the CNS to mature into antibody-secreting cells (ASCs). It remains elusive how to benefit from this disease hallmark as a prognostic tool. We aimed to uncover markers that reflect B-cell entry and maturation in the CNS and, on that basis, explore associations with response to high-efficacy MS therapies. METHODS: Ex vivo single-cell technologies were applied to blood samples and postmortem CNS suspensions from MS and control donors, either treated or untreated. In addition, in vitro ass

Preclinical Evaluation of Stable Integrin αvβ3-Specific [198Au]Gold Nanoparticles for Tumor Therapy.

Objectives: This paper reports the preclinical evaluation of stable tumor-specific gold nanoparticles (AuNPs) activated by neutron irradiation as a therapeutic option for the treatment of cancers characterized by high tumor angiogenesis. Methods: A selection of promising AuNPs with high avidity to αvβ3-expressing glioma (U-87 MG) cells (IC50 = 82-104 nM) were chosen with different surface loading of Arg-Gly-Asp (RGD) peptides as tumor targeting vectors for integrin αvβ3, a target which is overexpressed in tissues displaying high tumor angiogenesis. Three different [198Au]AuNPs were evaluated a

A genetic modifier links integrin α5 to the phenotypic variation in fibronectin 1a mutant zebrafish.

Phenotypic variation is often observed in individuals with the same mutation. However, the mechanisms that contribute to this variation remain largely unknown. Fibronectin mutants in both mouse and zebrafish fail to form a functional cardiovascular system, although the penetrance and expressivity of this phenotype vary depending on the genetic background. Here we investigate the variation of the zebrafish natter phenotype, which is caused by a nonsense mutation in fibronectin 1a (fn1a). natter/fn1a mutants exhibit incompletely penetrant cardia bifida, a phenotype caused by the failure of cardi

Characterization of Sex-Based Differences in Integrin-Mediated Endothelial Cell Adhesion to Bioactive Hydrogels.

Endothelialization promotes thromboresistance in blood-contacting devices, but biomaterial designs often overlook sex differences in the endothelialization processes. In this study, we elucidated sex differences in endothelial cell-material interactions through an investigation of the integrin-ligand interplay with biomaterial substrates and the corollary effects on cell adhesion and spreading. First, integrin expression of human coronary artery endothelial cells (HCECs) was characterized for age-matched donors (3 male, 3 female). Sex-based differences in integrin expression were identified, w
