Taurine-magnesium coordination compound, a potential anti-arrhythmic complex, improves aconitine-induced arrhythmias through regulation of multiple ion channels.

Taurine-magnesium coordination compound (TMCC) exhibits antiarrhythmic effects in cesium-chloride-and ouabain-induced arrhythmias; however, the mechanism underlying these effects on arrhythmia remains poorly understood. Here, we investigated the effects of TMCC on aconitine-induced arrhythmia in vivo and the electrophysiological effects of this compound in rat ventricular myocytes in vitro. Aconitine was used to induce arrhythmias in rats, and the dosages required to produce ventricular premature contraction (VPC), ventricular tachycardia (VT), ventricular fibrillation (VF), and cardiac arrest

Molecular basis of the interaction between gating modifier spider toxins and the voltage sensor of voltage-gated ion channels.

Voltage-sensor domains (VSDs) are modular transmembrane domains of voltage-gated ion channels that respond to changes in membrane potential by undergoing conformational changes that are coupled to gating of the ion-conducting pore. Most spider-venom peptides function as gating modifiers by binding to the VSDs of voltage-gated channels and trapping them in a closed or open state. To understand the molecular basis underlying this mode of action, we used nuclear magnetic resonance to delineate the atomic details of the interaction between the VSD of the voltage-gated potassium channel KvAP and th…

Effects of midazolam, pentobarbital and ketamine on the mRNA expression of ion channels in a model organism Daphnia pulex.

BACKGROUND: Over the last few decades intensive studies have been carried out on the molecular targets mediating general anesthesia as well as the effects of general anesthetics. The γ-aminobutyric acid type A receptor (GABAAR) has been indicated as the primary target of general anaesthetics such as propofol, etomidate and isoflurane, and sedating drugs including benzodiazepines and barbiturates. The GABAAR is also involved in drug tolerance and dependence. However, the involvement of other ion channels is possible. METHODS: Using reverse transcription and quantitative PCR techniques, we syst…

Modulators of ion channels activated by hypotonic swelling in cardiomyocytes: new perspectives for pharmacological treatment of life-threatening arrhythmias.

This review highlights the most important research data related to membrane potential and current changes in cardiomyocytes during hypotonic stress. Relative decrease in osmolarity in extracellular compartment (due to accumulation of metabolic products in the cells) during acute episodes of ischemia in the heart muscle leads to cell swelling caused by water entering the cells. Such condition starts regulatory volume decrease (RDV) in cardiomyocytes--a process involving activation of various ion channels. It seems to be the crucial proarrhythmic mechanism in ischemic heart, probably very often …

Early copper-induced leakage of K(+) from Arabidopsis seedlings is mediated by ion channels and coupled to citrate efflux.

Copper tolerance among Arabidopsis ecotypes is inversely correlated with long-term K(+) leakage and positively correlated with short-term K(+) leakage (A. Murphy, L. Taiz [1997] New Phytol 136: 211-222). To probe the mechanism of the early phase of K(+) efflux, we tested various channel blockers on copper and peroxide-induced K(+) efflux from seedling roots. The K(+) channel blockers tetraethyl ammonium chloride and 4-aminopyridine (4-AP) both inhibited short-term copper-induced K(+) efflux. In contrast, peroxide-induced K(+) efflux was insensitive to both tetraethyl ammonium chloride and 4-AP

[Interactions of intravenous anesthetics with human CNS ion channels. Electrophysiologic studies with a new type of voltage clamp technique].

UNLABELLED: Despite a widespread approach to explain the molecular mechanisms of anaesthetic agents, the complex state of "general anaesthesia" is still not completely understood. Voltage-activated sodium channels from human brain cortex served as a model membrane protein to investigate anaesthetic drug-protein interactions using a novel electrophysiological voltage-clamp technique. Sodium channels are already well-characterized important integral membrane proteins responsible for the generation of the fast-propagated action potential and thus are vital components for neuronal signal integrati

Calcium-activated potassium channels in resting and activated human T lymphocytes. Expression levels, calcium dependence, ion selectivity, and pharmacology.

Ca(2+)-activated K+[K(Ca)] channels in resting and activated human peripheral blood T lymphocytes were characterized using simultaneous patch-clamp recording and fura-2 monitoring of cytosolic Ca2+ concentration, [Ca2+]i. Whole-cell experiments, using EGTA-buffered pipette solutions to raise [Ca2+]i to 1 microM, revealed a 25-fold increase in the number of conducting K(Ca) channels per cell, from an average of 20 in resting T cells to > 500 channels per cell in T cell blasts after mitogenic activation. The opening of K(Ca) channels in both whole-cell and inside-out patch experiments was highly

Structure-activity relationship of philanthotoxins--II. Effects on the glutamate gated ion channels of the locust muscle fibre membrane.

1. PTX-4.3.3, the synthetic product of the wasp toxin, delta-philanthotoxin, blocks the open, glutamate gated, ion channels of the locust muscle. 2. This block is accompanied by the appearance of double exponential distribution of the closed times frequency, and a decrease of the inverse relationship between the open and closed states. 3. The analogues with the shortened polyamine chain, PTX-4.3 and PTX-3.3, are less potent, trifluoromethyl-PTX-4.3.3 is a more potent analogue. 4. Yet, they still behave like PTX-4.3.3 as open ion-channel blockers. 5. Another analogue, without the aromatic nucle…

Ion channel mechanisms of rat tail artery contraction-relaxation by menthol involving, respectively, TRPM8 activation and L-type Ca2+ channel inhibition.

Transient receptor potential melastatin 8 (TRPM8) is the principal cold and menthol receptor channel. Characterized primarily for its cold-sensing role in sensory neurons, it is expressed and functional in several nonneuronal tissues, including vasculature. We previously demonstrated that menthol causes variable mechanical responses (vasoconstriction, vasodilatation, or biphasic reactions) in isolated arteries, depending on vascular tone. Here we aimed to dissect the specific ion channel mechanisms and corresponding Ca2+ signaling pathways underlying such complex responses to menthol and other

The role of mechanosensitive ion channel Piezo1 in the intestinal barrier function changes and related gastrointestinal disease.

Intestinal barrier integrity is fundamental to the maintenance of body homeostasis. The mechanosensitive ion channel Piezo1, acting as a pivotal hub for mechanical signal transduction, converts mechanical stimuli into biochemical signals and plays a central role in the regulation of the intestinal barrier. This review systematically delineates the basic structure and kinetic properties of Piezo1 and comprehensively elaborates on its macro-regulatory functions across the multidimensional network of the intestinal mechanical, chemical, biological, and immune barriers. Furthermore, it investigate

Ion-Channel-Mediated Drug Repurposing Opportunities Validated by Single-Cell Perturbation in Colorectal Cancer.

Colorectal cancer (CRC) remains a leading cause of cancer mortality, yet no systematic effort has linked druggable CRC driver genes to downstream ion channel effectors. We integrated differential expression analysis, weighted gene co-expression network analysis (WGCNA), and protein-protein interaction (PPI) network pharmacology to identify CRC hub genes and their ion channel connections, validated by dual single-cell perturbation approaches: variational graph autoencoder-based virtual knockout (VGAE-KO) and experimental HCT116 CRISPRi Perturb-seq (6 genes, 8445 cells). WGCNA identified 100 hub

The Ion channel, CFTR, assembles with HIPPO pathway proteins TAZ and YAP in polycystic kidney disease.

Adult-onset polycystic kidney disease (ADPKD) leads to a decline in renal function. Cyst expansion is critically dependent on increased proliferation(1). Because proliferation is so central to cyst formation, thwarting these processes is critical for improving the health of patients with ADPKD. We have shown that the CFTR modulator VX-809 reduces cyst formation by creating an absorptive phenotype and inhibiting proliferation. We utilized the pkd1R3277/R3277C (RC) mutant mice and cell lines to study this phenomenon. We show that CFTR closely associates with the HIPPO pathway proteins TAZ and YA

The mechanosensitive ion channel Piezo1 promotes colitis by modulating oxidative stress and ferroptosis-related markers.

Piezo1 plays a crucial role in the gastrointestinal tract. However, whether Piezo1 induces intestinal dysfunction and mitochondrial damage in dextran sulfate sodium (DSS)-induced colitis, and the underlying mechanisms involved, remains unclear. We aimed to investigate whether Piezo1 triggers oxidative stress, alters ferroptosis-associated markers, and induces intestinal epithelial injury in human intestinal epithelial cells (HIECs) and in the colonic tissues of mice with DSS-induced colitis. We established a DSS-induced colitis mouse model by administering 1.5% DSS in drinking water and perfor

Analgesic effects of bulleyaconitine A: new advances in research from ion channel targets to clinical translation.

Chronic pain represents a significant global health concern, posing a severe threat to human wellbeing and affecting up to 20% of the adult population. Bulleyaconitine A (BLA), a diterpenoid alkaloid derived from plants of the Aconitum genus with in the Ranunculaceae family, demonstrates remarkable analgesic properties with a low potential for addiction, thus broad clinical application prospects compared to opioids. Extensive research has elucidated multiple pharmacological mechanisms underlying the analgesic effects of BLA, including state-dependent blockade of voltage-gated sodium channels,

Schwann Cell TRPA1, a Proalgesic Ion Channel, Mediates Neuroinflammation and Fibromyalgia-Associated Behaviours in Mice.

Transient receptor potential ankyrin 1 (TRPA1) is an ion channel that integrates the somatosensory system and is specialised in detecting thermal, mechanical, and chemical stimuli. It acts as both a sensor and an amplifier of reactive oxygen and nitrogen species, carbonylic species and lipid peroxidation products, which are overproduced in several painful conditions, including fibromyalgia. Studies have linked TRPA1 to heightened sensitivity to mechanical and cold pain in fibromyalgia patients. In a preclinical mouse model of fibromyalgia induced by reserpine administration, activated Schwann

Structure of a pH-sensitive pentameric ligand-gated ion channel from the Sarcoptes scabies mite.

Scabies is a skin infestation caused by the mite Sarcoptes scabiei and represents a substantial global health burden exacerbated by emerging resistance to ivermectin. An anionic pentameric ligand-gated ion channel from the mite, SsCl, shows pH-sensitivity and is significantly modulated by ivermectin. Here, we use cryo-EM and electrophysiology to explore the pH-sensing mechanisms of SsCl and the impact of ivermectin on channel activity. Structures of SsCl were resolved in closed (pH 6.5) and desensitized (pH 9) states, alongside ivermectin-bound conformations. The desensitized structure adopts

A duo of redox-sensitive pore-loop cysteines controls the activity of the neural ion channel TRPM3.

Transient Receptor Potential Melastatin 3 (TRPM3) is a non-selective, Ca2+-permeable ion channel that plays a pivotal role in peripheral thermosensation and nociception. Moreover, gain-of-function variants in TRPM3 underlie a spectrum of neurodevelopmental and epileptic disorders in humans, indicating an important role of TRPM3 in the central nervous system. Oxidative stress contributes to various neurological disorders of both the central and peripheral nervous system, but it is unknown whether TRPM3 activity is altered by the cellular redox state. Here, we report a direct, bidirectional modi

Selective blockade of acid-sensing ion channel 1a can provide substantial hippocampal neuroprotection.

BACKGROUND: Acid-sensing ion channel 1a (ASIC1a) is the only member of the ASIC family where Ca2+ osmosis has been reported, and it is highly expressed in neurons of the central nervous system. This study aimed to investigate whether ASIC1a is trafficked to the plasma membrane and regulated by the Rho/ROCK and PI3K signaling pathways in temporal lobe epilepsy (TLE). In addition, further research is required to determine whether selective ASIC1a blockade is a viable therapeutic strategy for TLE. METHODS: The localization and expression levels of ASIC1 and mRNA levels of ASIC1a were detected wh

Optochemical modulation of corneal cold nerve terminal impulse activity with a photochromic ion channel blocker.

BACKGROUND AND PURPOSE: The functional organization of corneal cold nerve endings, critical structures in maintaining the ocular surface, remains poorly understood. Here, the photoisomerizable small-molecule diethylamine-azobenzene-quaternary ammonium (DENAQ) was used to photomodulate activity of cold-sensing nerve terminals in control and chronic tear-deficient corneas. Furthermore, DENAQ was used for in vivo photochemical regulation of the thermally induced blink reflex. EXPERIMENTAL APPROACH: Extracellular nerve terminal impulse activity was recorded on cold terminals in excised corneas of
