Estimating the burden of mucormycosis infections in France (2005-2007) through a capture-recapture method on laboratory and administrative data.

BACKGROUND: Mucormycoses are rare but severe fungal infections whose incidence is increasing, particularly in immunosuppressed and diabetic patients. Following a retrospective study on the characteristics and outcomes of cases who were identified through two sources of information, we carried out a capture-recapture method to estimate the actual burden of the disease in France, 2005-2007. METHODS: An administrative dataset from the national hospital discharge system and a laboratory dataset from the National Reference Centre for Mycoses and Antifungals were combined to identify patients from

The Laboratory Risk Indicator for Necrotizing Fasciitis score for discernment of necrotizing fasciitis originated from Vibrio vulnificus infections.

BACKGROUND: The Laboratory Risk Indicator for Necrotizing Fasciitis (LRINEC) score has been verified as a useful diagnostic tool for detecting necrotizing fasciitis (NF). Its application, however, is mainly for NF types I and II. The practical relevance of the LRINEC score for Vibro vulnificus-related skin and soft tissue infection (SSTI) was hardly ever investigated. The aim of this study was to assess the applicability of the LRINEC scoring system and to identify NF-predicting factors in patients with V. vulnificus-caused SSTI. METHODS: A retrospective study was conducted, enrolling 125 con

Laboratory diagnosis of Pneumocystis carinii infections by PCR directed to genes encoding for mitochondrial 5S and 28S ribosomal RNA.

PCR with 5S mitochondrial ribosomal RNA (5S) target is a sensitive and specific assay for the detection of Pneumocystis carinii in clinical specimens from the respiratory tract. We developed an oligonucleotide probe directed to a 200 bp amplicon generated by fungal-specific universal primers that anneals with sequences specific for P. carinii in the 28S ribosomal RNA gene (28S). Of 50 archived bronchoalveolar lavage 1(BAL) specimens, 46 of 50 samples (92% agreement) gave the same result (23 positive, 23 negative) by PCR directed to the 5S and 28S assays. Results of calcofluor white staining of

Fungal infections in acute leukemia patients during selective decontamination of the digestive tract--a clinical, laboratory and pathological study.

Selective decontamination (SD) is used for prevention of bacterial as well as fungal infection in acute leukemia patients during remission induction therapy. We analyzed 138 treatment periods of 108 patients. The intensified chemotherapy resulted in severe granulocytopenia below 0.1 X 10(9) l for over 25.2 days. In 19 patients there was a suspect of major fungal infection, for which they were given antimycotics (in 15 cases amphotericin B and 5-fluocytosin). Fourteen of them died. Major fungal infections were documented in 5 cases. In 18% of the deceased we found major fungal infection (7 case

Experimental infections with pathogenic free-living amebae in laboratory primate hosts: I (A) A study on susceptibility to Naegleria fowleri.

Studies were conducted on 27 Old World monkeys to determine their susceptibility to pathogenic strains (HB-1 and C-66) of Naegleria fowleri by intranasal, intravenous, or intrathecal inoculation of trophozoites. No clinically detectable disease resulted from either intranasal or intravenous inoculation, but 11 of 18 monkeys inoculated intrathecally succumed to acutely fatal meningoencephalitis, while the other 7 survived with no obvious permanent brain damage. Pathogenicity of N. fowleri appeared to be influenced by the strain virulence, growth phase, and cultural condition of the amebae, as w

The Role of the Medical Laboratory in Identifying 'Super-Spreaders': Correlation Between Viral Load Kinetics and Human-to-Human Infectivity.

Nipah virus (NiV) is a high-consequence zoonotic henipavirus characterised by severe encephalitis and respiratory distress, driving recurrent outbreaks with case fatality rates between 40% and 75% in India and Bangladesh. Within these outbreaks, 'super-spreading' dynamics play a critical role, where a small subset of individuals contributes to a disproportionately large share of secondary infections. To evaluate the relationship between human infectivity and viral load kinetics in Nipah virus infection and to define the role of the medical laboratory in identifying potential super-spreaders, a

From Laboratory to Clinic: Translational Medicine Paradigm of Polymyxin B Nanopreparations for Overcoming Drug-Resistant Bacterial Infections.

The global prevalence of multidrug-resistant (MDR) Gram-negative bacterial infections has posed a severe threat to public health security. As the "last line of defense" antibiotic against such infections, polymyxin B is severely limited in clinical application by dose-dependent nephrotoxicity (25-50% incidence), neurotoxicity, and the global spread of mcr-mediated resistance. Nanotechnology overcomes these limitations through three key mechanisms: (1) targeted enrichment at infection sites to reduce systemic exposure; (2) controlled drug release to minimize peak concentration-related toxicity;

Demonstrated laboratory effectiveness of an anthelmintic medicated feed for the treatment of Oxyspirura petrowi infections in northern bobwhite (Colinus virginianus).

In May 2024, a medicated anthelmintic feed was approved by the U.S. Food and Drug Administration to treat parasitic infection in wild quail following evidence linking parasites to northern bobwhite (Colinus virginianus) population impacts. During the approval process, it was confirmed that the feed significantly reduced parasite burdens in wild bobwhite populations after 3 weeks of treatment. However, no studies were conducted on the drug's efficacy in individual bobwhite in a controlled laboratory setting. In this study, we infected pen-reared bobwhite with Oxyspirura petrowi, an eyeworm comm
