Eosinophil peroxidase levels in hearts and lungs of mice infected with Toxocara canis.

Toxocara canis infection of abnormal hosts results in a condition in which infective larvae migrate through the soft tissues of the body, exclusive of the skin. This condition is known as visceral larva migrans (VLM) and causes a syndrome characterized by hepatosplenomegaly, hyperglobulinemia, hypereosinophilia, and transient pulmonary infiltrates. Because of the known association between hypereosinophilia and eosinophilic heart disease, we have been studying the hearts of mice infected with T. canis for evidence of myocardial damage and have previously described a severe eosinophilic myocardi

Proteomic analysis of extracellular vesicles and extracellular vesicle-depleted excretory-secretory products of Toxocara canis and Toxocara cati larval cultures.

Toxocara canis and Toxocara cati are parasitic nematodes in the order Ascaridida, which inhabit the small intestines of dogs and cats, respectively, as adults. Although often nonpathogenic as adults, nematodes within this genus are capable of causing widespread disease throughout the host while in a larval stage, during which time larvae migrate throughout the body in a process termed larva migrans. Larvae are also capable of surviving within host tissues in an encysted arrested stage, without immune clearance by the host. The ability of larvae to survive within host tissues during migration a

High Prevalence and Regional Heterogeneity of Canine Ancylostoma spp. in Ecuador: A Systematic Review and Meta-Analysis and Its Potential One Health Implications.

Hookworms are zoonotic helminths that cause intestinal diseases in pets and humans, as well as cutaneous larva migrans in the latter; therefore, they are of importance to animal and public health. Stray and domestic dogs play a significant role in the transmission of hookworm disease. This study examines the prevalence of Ancylostoma spp. infections in dogs in Ecuador through a systematic review and meta-analysis. Although based exclusively on canine data, the findings are discussed in the context of available evidence on zoonotic transmission and potential One Health implications. A total of

Retinal Granuloma Progression After Scleral Buckling in Ocular Toxocariasis: A Case Report.

BACKGROUND Toxocariasis has a wide geographic distribution, with high prevalence in tropical and subtropical regions. Ocular larva migrans is one of the most common manifestations in China and classically presents with peripheral granuloma, posterior granuloma, and chronic endophthalmitis. This report describes an atypical case of ocular toxocariasis in an adolescent male patient, initially misdiagnosed as chronic rhegmatogenous retinal detachment. CASE REPORT A 17-year-old male patient presented to our hospital with a 4-month history of progressive vision loss in his right eye, initially misd

Toxocariasis presenting with peripheral eosinophilia and eosinophilic pleural effusion: A case report.

Human toxocariasis is a parasitic infection caused by Toxocara species and is important in the differential diagnosis of patients presenting with unexplained eosinophilia. A case of toxocariasis with an eosinophilic pleural effusion is presented. A 67-year-old Japanese man presented with progressive dyspnea. Laboratory investigations showed marked peripheral eosinophilia and a markedly elevated serum immunoglobulin E level. Chest imaging demonstrated bilateral ground-glass opacities and a left-sided pleural effusion. Analysis of the pleural fluid showed eosinophil predominance without evidence

The Early Bird Catches the Parasite: Potential Invertebrate Pathway for Baylisascaris procyonis to Infect Songbirds.

Baylisascaris procyonis larva migrans is a debilitating and potentially lethal zoonotic, parasitic condition that can infect a large variety of paratenic hosts, including birds and mammals and even humans. The typical pathway to develop baylisascariasis is to ingest B. procyonis eggs from contaminated feces or through contaminated soil where contaminated fecal matter has decayed. I hypothesized that earthworms (Lumbricus terrestris) living in soils contaminated with B. procyonis eggs from decayed feces of its definitive host, raccoons (Procyon lotor), would incidentally ingest B. procyonis egg

Enzyme-linked immunosorbent assay (ELISA) using recombinant Fasciola cathepsin L1 for the diagnosis of human fasciolosis caused by Fasciola hepatica/gigantica hybrid type.

Recombinant Fasciola cathepsin L-1 (rCatL1) was evaluated in enzyme-linked immunosorbent assay (ELISA) for the serodiagnosis of human fasciolosis in Japan. Quality characteristics of the test were accessed by receiver operating characteristic (ROC) analysis, with sera from fasciolosis patients (n = 10), patients with no evidence of parasitic infections (n = 29), and patients with other helminth infections (n = 119). Both the sensitivity and specificity of the test achieved 100% with the control samples. To test the performance of the assay in an authentic situation, 311 serum samples, which ha

Field application of a novel multiplex qPCR assay reveals the occurrence of the zoonotic hookworm Ancylostoma braziliense in Nigerian dogs.

A number of gastrointestinal parasites have been reported to infect dogs in Nigeria, some of which have zoonotic potential. Of these, hookworms are the most prevalent, with both Ancylostoma caninum and Uncinaria stenocephala reported in the country. In this study, we subjected 203 hookworm microscopy-positive samples of the 885 individual faecal samples collected from dogs in Nigeria to a recently developed multiplex qPCR for the detection and characterisation of canine hookworm species. The qPCR demonstrated a diagnostic sensitivity of 98% (95% CI 95-99.4) allowing the detection of A. caninum

Immune response to chronic Toxocara canis infection in a mice model.

AIMS: The zoonotic nematode Toxocara canis causes larva migrans syndrome that induces an immune response characterized by the production of antibodies and eosinophilia. A Th2 polarization has been associated with the infection, but there are still details of the cellular and humoral immune response that need to be described. Thus, the aim of this study was to describe the systemic host immune response to T canis chronic infection in a mouse model. METHODS AND RESULTS: BALB/c mice were inoculated once with 500 T canis embryonated eggs, per os. After 49 days, the amounts of larval found in brai

Robust Phenotypic Activation of Eosinophils during Experimental Toxocara canis Infection.

Eosinophils are multifunctional cells that have cytotoxic proinflammatory activities and stimulate CD4+ T-cells in experimental models of allergy and parasitic infections. Eosinophils, when exposed to antigens, are activated, expressing the CD38/CD69 molecules and exhibited increased expression of major histocompatibility complex (MHC-II), CD80 and CD86, suggesting they play a role upon Toxocara canis antigen stimulation. In the present study, we evaluated the profile of eosinophils using conventional and image flow cytometry upon experimental T. canis infection. T. canis antigens induced a ro

Eosinophilia prevalence and related factors in travel and immigrants of the network +REDIVI.

The population movements during the last decades have resulted in a progressively increasing interest in certain infectious diseases. Eosinophilia is a common finding in immigrants and travellers. One of the most common causes of eosinophilia is helminth infection, and some intestinal protozoa. The aim of this paper is to describe the epidemiological characteristics of cases with eosinophilia and its association with the presence of parasites in the REDIVI data network. This is a multicentre prospective observational study that includes patients diagnosed with eosinophilia registered in the co

Oral ivermectin for the treatment and prophylaxis of scabies in prison.

INTRODUCTION: Oral ivermectin has been widely used to treat various human diseases, such as filariasis, myiasis, larva migrans, strongyloidiasis and scabies (both the common and crusted forms). However, there are only a small number of papers on the effects of this drug for the control of scabies in infested environments. The current study shows the results obtained with the collective treatment of inmates of a public jail. MATERIALS AND METHODS: A total of 123 inmates living in a restricted and contaminated environment were evaluated clinically by experienced dermatologists for the assessmen

Multiple large pyogenic liver abscesses of the tropics: a new entity.

BACKGROUND: Pyogenic liver abscesses most commonly occur in males in -he sixth decade, are usually associated with biliary tract disease, malignancy and immunosuppression and the mortality rate is high. We describe another form of pyogenic abscess occurring in females which, if treated aggressively, carries a much better prognosis. METHODS: Between 1986 and 1993 we treated 8 patients with multiple pyogenic liver abscesses. Diagnosis was established by ultrasound and CT scan followed by needle aspiration to confirm pus. Amoebic aetiology was excluded by a serology, poor response to metronidazo

Improved method for recovering ascarid and other helminth eggs from soil associated with epizootics and during survey studies.

Two noncentrifugal and 5 centrifugal flotation methods for the recovery of infective Baylisascaris procyonis eggs from contaminated soil were evaluated after an epizootic of fatal cerebrospinal nematodiasis in bobwhites. All involved the use of inexpensive, readily available materials and routine procedures. In all but 1 of the methods, sizes of the soil samples were 30 g; in 1 centrifugal method, the sample size was 1 g. The centrifugal methods included various combinations of (i) anionic detergent (Tween) pretreatment of the soil, (ii) 3 X washing of the sediment, (iii) NaNO3 or ZnSO4 flotat

Protein phosphorylation networks in Baylisascaris procyonis revealed by phosphoproteomic analysis.

BACKGROUND: Baylisascaris procyonis is an intestinal ascarid worm that parasitizes in raccoons and causes fatal neural, visceral, and ocular larva migrans in humans. Phosphorylated proteins and protein kinases have been studied as vaccine and drug target candidates against parasitic infections. However, no data are available on protein phosphorylation in the raccoon roundworm. METHODS: In this study, the entire proteome of adult B. procyonis was enzymatically digested. Then, phosphopeptides were enriched using immobilized metal affinity chromatography (IMAC) and analyzed by liquid chromatogra

Optimal ELISA antigen for the diagnosis of Ascaris suum infection in humans.

Ascarid nematodes, Ascaris suum, Toxocara canis and Toxocara cati, are the most important causative species of larva migrans syndrome (LMS) in humans. Although the diagnosis of ascarid LMS is generally based on serological tests, specific serological tests for A. suum infection have not been fully developed. In the present study, the sensitivity and specificity of three A. suum antigen preparations, i.e., the somatic adult worm antigen (As-SWAP), larval excretory-secretory (ES) antigens derived from infective L3 (AsiL3-ES) and larval ES from tissue migratory L3 (AsmL3-ES), were evaluated for t
