Development of a recombinant baculovirus expressing an insect-selective neurotoxin: potential for pest control.

Recombinant nuclear polyhedrosis viruses (NPVs) expressing insect-selective toxins, hormones, or enzymes could enhance their insecticidal properties. We have constructed a recombinant, polyhedrin-positive Autographa californica NPV (AcNPV) that is orally infectious and expresses an insect-selective toxin (AaIT), isolated from the scorpion Androctonus australis, under the control of the p10 promoter. Bioassays with the recombinant baculovirus on 2nd instar larvae of Heliothis virescens demonstrated a significant decrease in the time to kill (LT50 88.0 hours) compared to wild-type AcNPV (LT50 12

Characterizing virulence differences in a parasitoid wasp through comparative transcriptomic and proteomic.

BACKGROUND: Two strains of the endoparasitoid Cotesia typhae (Hymenoptera: Braconidae) present a differential parasitism success on the host, Sesamia nonagrioides (Lepidoptera: Noctuidae). One is virulent on both permissive and resistant host populations, and the other only on the permissive host. This interaction provides a very interesting frame for studying virulence factors. Here, we used a combination of comparative transcriptomic and proteomic analyses to unravel the molecular basis underlying virulence differences between the strains. RESULTS: First, we report that virulence genes are

Significance of the tropical fire ant Solenopsis geminata (hymenoptera: formicidae) as part of the natural enemy complex responsible for successful biological control of many tropical irrigated rice pests.

The tropical fire ant Solenopsis geminata (Fabricius) often nests very abundantly in the earthen banks (bunds) around irrigated rice fields in the tropics. Where some farmers habitually drain fields to the mud for about 3-4 days, the ants can quickly spread up to about 20 m into the fields where they collect food, including pest prey such as the eggs and young of the apple snail Pomacea caniculata (Lamarck) and insects such as lepidopterous larvae and hoppers, notably Nilaparvata lugens (Stäl) the brown planthopper (Bph) and green leafhoppers Nephotettix spp. Even in drained fields, the activi

Revealing fitness and virulence determinants of hypervirulent Klebsiella pneumoniae during infection in Galleria mellonella using a transposon library.

Klebsiella pneumoniae infections represent a significant public health concern. Despite their clinical relevance, the genetic determinants underlying bacterial fitness and virulence remain incompletely characterized. In this study, we systematically identified genes involved in host adaptation by generating a transposon mutant library and integrating a Galleria mellonella infection model with transposon sequencing (Tn-seq) technology. This approach yielded a comprehensive dataset of adaptation-deficient genes in the hypervirulent K. pneumoniae strain ATCC 43816. Using homologous recombination,

Designing of eucalyptol nanoemulsion drug delivery system loaded with amphotericin B for Candida albicans biofilm control: in vitro and in vivo assessment using the Galleria mellonella model.

OBJECTIVE: (s): Candida species can form biofilm in various clinical settings, making them challenging to treat due to their resistance to antifungal drugs. The present study aimed to evaluate the effects of eucalyptol/amphotericin B nanoemulsion (NEA) on Candida albicans (C. albicans) biofilm. MATERIALS AND METHODS: In this study, NEA was synthesized and its capacity to combat biofilms generated by four clinical isolates and a reference strain of C. albicans was investigated according to CLSI M27-A3 guidelines. The NEA was prepared using eucalyptol oil, Tween 80, and ethanol, and characteriz

Correlating Genotype and Phenotype in the Asexual Yeast Candida orthopsilosis Implicates ZCF29 in Sensitivity to Caffeine.

Candida orthopsilosis is diploid asexual yeast that causes human disease. Most C. orthopsilosis isolates arose from at least four separate hybridizations between related, but not identical, parents. Here, we used population genomics data to correlate genotypic and phenotypic variation in 28 C. orthopsilosis isolates. We used cosine similarity scores to identify 65 variants with potential high-impact (deleterious effects) that correlated with specific phenotypes. Of these, 19 were Single Nucleotide Polymorphisms (SNPs) that changed stop or start codons, or splice sites. One variant resulted in

NeuroBactrus, a novel, highly effective, and environmentally friendly recombinant baculovirus insecticide.

A novel recombinant baculovirus, NeuroBactrus, was constructed to develop an improved baculovirus insecticide with additional beneficial properties, such as a higher insecticidal activity and improved recovery, compared to wild-type baculovirus. For the construction of NeuroBactrus, the Bacillus thuringiensis crystal protein gene (here termed cry1-5) was introduced into the Autographa californica nucleopolyhedrovirus (AcMNPV) genome by fusion of the polyhedrin-cry1-5-polyhedrin genes under the control of the polyhedrin promoter. In the opposite direction, an insect-specific neurotoxin gene, Aa

Skin-piercing blood-sucking moths I: ecological and ethological studies on Calpe eustrigata (Lepid., noctuidae).

The Noctuid Calpe [Calyptral] eustrigata Hmps. was reported as a skin-piercing blood-sucking moth for the first time in Malaya (Bänziger, 1968) and is so far the only lepidopteran proved to suck blood by means of a piercing act. A few field observations and the description of the piercing behaviour of caged moths were given. Apart from a taxonomic study of the genus Calpe (Berio, 1956), a single record (Büttiker, 1969) and some notes on the moth's proboscis and possible evolutionary pathway (Bänziger, 1970, 1971, 1972) to our knowledge no other data have been published on the moth after its de

Rapid evolution and genetic basis of spinetoram resistance in Phthorimaea absoluta: evidence from six years of monitoring and laboratory selection.

BACKGROUND: Phthorimaea absoluta (Meyrick) (Lepidoptera: Gelechiidae) is one of the most destructive pests of tomato worldwide and has repeatedly evolved resistance to insecticides. Spinosyn insecticides, particularly spinetoram, are widely used for its control due to their high efficacy and favorable selectivity. This study investigated temporal changes in susceptibility to spinetoram in Brazilian populations of Phthorimaea absoluta over a 6-year monitoring period and characterized the genetic basis, metabolic contribution, and cross-resistance patterns of a resistant population. RESULTS: Di

Differential gene expression patterns between the head and thorax of Gynaephora aureata are associated with high-altitude adaptation.

Grassland caterpillars (Lepidoptera: Erebidae: Gynaephora) are important pests in alpine meadows of the Qinghai-Tibetan Plateau (QTP). These pests have morphological, behavioral, and genetic adaptations for survival in high-altitude environments. However, mechanisms underlying high-altitude adaptation in QTP Gynaephora species remain largely unknown. Here, we performed a comparative analysis of the head and thorax transcriptomes of G. aureata to explore the genetic basis of high-altitude adaptation. We detected 8,736 significantly differentially expressed genes (sDEGs) between the head and tho

Unusual response characteristics of pheromone-specific olfactory receptor neurons in the Asian corn borer moth, Ostrinia furnacalis.

Male moth pheromone-detecting receptor neurons are known to be highly specific and very sensitive. We investigated physiological and behavioral responses to female sex pheromone components in male Ostrinia furnacalis moths (Lepidoptera: Crambidae). Using recordings from a cut-sensillum technique, trichoid sensilla could be grouped into four physiological types (1-4), according to the response of receptor neurons to the two major pheromone components, (E)-12- and (Z)-12-tetradecenyl acetate (E12- and Z12-14:OAc). These types could subsequently be characterized as four subtypes (A-D) depending o

The fall armyworm strain associated with most rice, millet, and pasture infestations in the Western Hemisphere is rare or absent in Ghana and Togo.

The moth pest fall armyworm, Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) is now present throughout much of the Eastern Hemisphere where it poses a significant economic threat to a number of crops. Native to the Western Hemisphere, fall armyworm is one of the primary pests of corn in the Americas and periodically causes significant economic damage to sorghum, millet, cotton, rice, and forage grasses. This broad host range is in part the result of two populations historically designated as host strains (C-strain and R-strain) that differ in their host plant preferences. Reports o

Herbivory Protection via Volatile Organic Compounds Is Influenced by Maize Genotype, Not Bacillus altitudinis-Enriched Bacterial Communities.

Belowground, plants interact with beneficial soil microbes such as plant growth-promoting rhizobacteria (PGPR). PGPR are rhizosphere bacteria that colonize roots and elicit beneficial effects in plants such as improved plant growth, pathogen resistance, abiotic stress tolerance, and herbivore protection. Treatment of plants with PGPR has been shown to trigger the emission of volatile organic compounds (VOCs). Volatile emissions can also be triggered by herbivory, termed herbivore-induced plant volatiles (HIPV), with important ramifications for chemical-mediated plant and insect interactions. M

Apple volatiles synergize the response of codling moth to pear ester.

Codling moth, Cydia pomonella L. (Lepidoptera: Tortricidae), is a major cosmopolitan pest of apple and other pome fruits. Ethyl (E,Z)-2,4-decadienoate (pear ester) has been identified as a host-derived kairomone for female and male codling moths. However, pear ester has not performed similarly in different fruit production areas in terms of the relative magnitude of moth catch, especially the proportion of females caught. Our work was undertaken to identify host volatiles from apples, and to investigate whether these volatiles can be used to enhance the efficacy of host kairomone pear ester fo

Transposon insertion causes cadherin mis-splicing and confers resistance to Bt cotton in pink bollworm from China.

Transgenic crops producing insecticidal proteins from Bacillus thuringiensis (Bt) are cultivated extensively, but rapid evolution of resistance by pests reduces their efficacy. We report a 3,370-bp insertion in a cadherin gene associated with resistance to Bt toxin Cry1Ac in the pink bollworm (Pectinophora gossypiella), a devastating global cotton pest. We found the allele (r15) harboring this insertion in a field population from China. The insertion is a miniature inverted repeat transposable element (MITE) that contains two additional transposons and produces two mis-spliced transcript varia

Serological immunoassay for detection of hepatitis E virus on the basis of genotype 3 open reading frame 2 recombinant proteins produced in Trichoplusia ni larvae.

Hepatitis E virus (HEV) is a major cause of acute hepatitis in humans, and strains of genotypes 1 and 2 are endemic in many regions with suboptimal sanitary conditions. In many industrialized countries, HEV strains of genotype 3 are highly endemic in swine, and an increased number of autochthonous infections with HEV genotype 3 strains have been reported lately. Serological studies of HEV infection are often conducted with commercial assays based on peptides and recombinant proteins of HEV genotype 1 and 2 strains. For some patients with proven HEV genotype 3 infections, these assays failed to

Efficacy of oritavancin alone and in combination against vancomycin-susceptible and -resistant enterococci in an in-vivo Galleria mellonella survival model.

OBJECTIVE: The optimal therapy for serious enterococcal infections, especially vancomycin-resistant enterococci (VRE), remains unclear, although combination therapy is often recommended. Oritavancin has demonstrated in-vitro activity against VRE, but data evaluating oritavancin in combination with other agents and in in-vivo systems are lacking. The objective of this study was to evaluate the efficacy of oritavancin alone and in combination with ceftriaxone, daptomycin, gentamicin, linezolid and rifampin against vancomycin-susceptible enterococci and VRE in an in-vivo Galleria mellonella survi

Coxiella burnetii utilizes both glutamate and glucose during infection with glucose uptake mediated by multiple transporters.

Coxiella burnetii is a Gram-negative bacterium which causes Q fever, a complex and life-threatening infection with both acute and chronic presentations. C. burnetii invades a variety of host cell types and replicates within a unique vacuole derived from the host cell lysosome. In order to understand how C. burnetii survives within this intracellular niche, we have investigated the carbon metabolism of both intracellular and axenically cultivated bacteria. Both bacterial populations were shown to assimilate exogenous [13C]glucose or [13C]glutamate, with concomitant labeling of intermediates in

Vitamin E Increases Antimicrobial Sensitivity by Inhibiting Bacterial Lipocalin Antibiotic Binding.

Burkholderia cenocepacia is an opportunistic Gram-negative bacterium that causes serious respiratory infections in patients with cystic fibrosis. Recently, we discovered that B. cenocepacia produces the extracellular bacterial lipocalin protein BcnA upon exposure to sublethal concentrations of bactericidal antibiotics. BcnA captures a range of antibiotics outside bacterial cells, providing a global extracellular mechanism of antimicrobial resistance. In this study, we investigated water-soluble and liposoluble forms of vitamin E as inhibitors of antibiotic binding by BcnA. Our results demonstr

Identification of multiple small heat-shock protein genes in Plutella xylostella (L.) and their expression profiles in response to abiotic stresses.

We identify and characterize 14 small heat-shock protein (sHSP) genes from the diamondback moth (DBM), Plutella xylostella (L.), a destructive pest. Phylogenetic analyses indicate that, except for sHSP18.8 and sHSP19.22, the other 12 DBM sHSPs belong to five known insect sHSP groups. Developmental expression analysis revealed that most sHSPs peaked in the pupal and adult stages. The transcripts of sHSPs display tissue specificity with two exhibiting constitutive expression in four tested tissues. Expression of sHSP18.8 in fourth instar larvae is not induced by the tested abiotic stressors, and
