Altered lipidome and antioxidative activity of small, dense HDL in normolipidemic rheumatoid arthritis: relevance of inflammation.

OBJECTIVE: High-density lipoprotein (HDL) particles exert potent antiatherogenic activities, including antioxidative actions, which are relevant to attenuation of atherosclerosis progression. Such activities are enriched in small, dense HDL and can be compromised under conditions of chronic inflammation like rheumatoid arthritis (RA). However, structure-function relationships of HDL largely remain indeterminate. METHODS: The relationships between HDL structure and function were evaluated in normolipidemic patients with active RA (DAS28 > 3.2; n = 12) and in normolipidemic age-matched controls

Brain lipidomics identifies mitochondrial redox dysfunction and metabolic trade-offs associated with Parkinson's disease-like pathology induced by Nanoplastics exposure.

Growing nanoplastics exposure raises concern for neurotoxicity, particularly given recent evidence of plastic accumulation within human brain tissue a highly lipid enriched organ, yet effects on brain lipid metabolism remain poorly understood. Here, we employed high-resolution untargeted lipidomics to map brain lipid perturbations in Drosophila melanogaster chronically exposed to polystyrene nanoplastics (PS- NPs). PS-NPs accumulated in fly brains and induced dose-dependent remodeling of mitochondrial membrane lipids, notably cardiolipins and phosphatidylethanolamines, accompanied by increased…

Decoding lipid signatures and aroma characteristics of four Chinese traditional dark teas via integrative lipidomics, volatilomics, and quantitative descriptive analysis.

Dark tea is a microbially post-fermented tea whose aroma formation is closely associated with lipid degradation. In this study, integrated lipidomics and volatilomics were applied to characterize four Chinese dark teas: Anhua dark tea (AHDT), Qingzhuan dark tea (QZDT), Liupao dark tea (LPDT), and Pu-erh dark tea (PEDT). Lipidomic profiling identified 486 lipid species, including 186 differential lipids among the four tea varieties. Glycerophospholipids and glycoglycerolipids were relatively enriched in AHDT, fatty acyls in PEDT, and acylglycerolipids in QZDT and LPDT. Quantification of fatty a…

Lipidomics uncovers metabolic manifestations related to liver steatosis and low-grade systemic inflammation in diet-treated hereditary fructose intolerance patients.

BACKGROUND AND AIMS: Hereditary Fructose Intolerance (HFI), a rare autosomal recessive metabolic disorder, has historically been considered benign when treated with a lifelong fructose-, sucrose and sorbitol-restricted diet. However, adverse metabolic manifestations have recently been reported in treated HFI patients. As serum metabolomics offers a valuable tool for assessing metabolic manifestations underlying inherited metabolic disorders, we aim to compare the serum lipidomic profile of HFI-treated patients with age-, gender-, and body mass index-matched healthy controls. METHODS: Long-ter

Metabolomics and lipidomics predictor of survival in hepatocellular carcinoma patients receiving tyrosine kinase inhibitor and immune checkpoint inhibitor combination therapy.

Although tyrosine kinase inhibitor and immune checkpoint inhibitor (TKI-ICI) combination therapy has emerged as a promising treatment for hepatocellular carcinoma (HCC), reliable biomarkers for predicting long-term survival remain underexplored. Here, we conducted metabolomics and lipidomics profiling in baseline plasma samples from 58 patients with HCC who received TKI-ICI therapy in a prospective phase II trial. Prognostic features were identified using an integrated machine learning framework combining random forest survival analysis, LASSO regression, and Cox modeling. Untargeted metabolom…

Dual fragmentation via collision-induced and oxygen attachment dissociations using water and its radicals for C=C position-resolved lipidomics.

Oxygen attachment dissociation (OAD) is a tandem mass spectrometry (MS/MS) technique for annotating the positions of double bonds (C=C) in complex lipids. Although OAD has been used for untargeted lipidomics, its availability has been limited to the positive ion mode, requiring the independent use of a collision-induced dissociation (CID) method. In this study, we demonstrated the OAD MS/MS technique in the negative-ion mode for profiling phosphatidylserines, phosphatidylglycerols, phosphatidylinositols, and sulfatides, where the fragmentation mechanism remained consistent with that in the pos…

Lipidomics-Based Analysis of the Regulatory Effects of Phytosterol Esters on Lactation Performance and Lipid Metabolism in Tarim Bactrian camels.

Plantsterol esters (PSEs) exert beneficial effects on animal product quality, indicating their potential as a nutritional intervention strategy. This study investigated the effects of dietary PSE supplementation on lactation performance and lipid metabolism in Tarim Bactrian camels (Camelus bactrianus) to establish a scientific basis for its application in their husbandry. Thirty-two mid-lactation female camels were randomly allocated to four groups (n = 8): CON (basal diet), L (200 mg/kg PSE), M (400 mg/kg PSE), and H (800 mg/kg PSE). Since lactation performance is closely linked to metabolic

Parallel Metabolomics and Lipidomics of a PSMA/GCPII Deficient Mouse Model Reveal Alteration of NAAG Levels and Brain Lipid Composition.

Glutamate carboxypeptidase II (GCPII, also known as PSMA or FOLH1) is responsible for the cleavage of N-acetyl-aspartyl-glutamate (NAAG) to N-acetyl-aspartate and glutamate in the central nervous system and facilitates the intestinal absorption of folate by processing dietary folyl-poly-γ-glutamate in the small intestine. The physiological function of GCPII in other organs like kidneys is still not known. GCPII inhibitors are neuroprotective in various conditions (e.g., ischemic brain injury) in vivo; however, their utilization as potential drug candidates has not been investigated in regard t

New insights into the role of lipids in aroma formation during black tea processing revealed by integrated lipidomics and volatolomics.

Lipids are important tea aroma precursors. Due to the complexity of black tea processing involving both enzymatic and thermal reactions, the role of lipids in black tea aroma formation remained unclear. Herein, the dynamic changes of lipids and volatiles during black tea processing were simultaneously analyzed by lipidomics and volatolomics using ultra-high-performance liquid chromatography coupled to Q-Exactive Orbitrap mass spectrometry (UHPLC-Q-Exactive) and gas chromatography-tandem mass spectrometry (GC-MS/MS). The lipidomics method was validated in linearity, reproducibility, and recover…

Gut microbiota in a mouse model of obesity and peripheral neuropathy associated with plasma and nerve lipidomics and nerve transcriptomics.

BACKGROUND: Peripheral neuropathy (PN) is a common complication in obesity, prediabetes, and type 2 diabetes, though its pathogenesis remains incompletely understood. In a murine high-fat diet (HFD) obesity model of PN, dietary reversal (HFD-R) to a low-fat standard diet (SD) restores nerve function and the nerve lipidome to normal. As the gut microbiome represents a potential link between dietary fat intake and nerve health, the current study assessed shifts in microbiome community structure by 16S rRNA profiling during the paradigm of dietary reversal (HFD-R) in various gut niches. Dietary f

Lipidomics combined with transcriptomic and mass spectrometry imaging analysis of the Asiatic toad (Bufo gargarizans) during metamorphosis and bufadienolide accumulation.

BACKGROUND: To adapt to life on land, Asiatic toads (Bufo gargarizans) must remodel their bodies and refine their chemical defenses in water. The full scope of the mechanisms underlying these processes has yet to be revealed. Bufadienolides (BDs) are chemical defense substances secreted by toads when they are in danger, and they have high medicinal value in treating heart failure, cancer, and hepatitis. However, the artificial breeding of toads to increase BDs has been unsuccessful due to the high mortality of toad larvae during metamorphosis. METHOD: Toad larvae at different growth stages we

Discrimination of urinary exosomes from microvesicles by lipidomics using thin layer liquid chromatography (TLC) coupled with MALDI-TOF mass spectrometry.

Источник пометил публикацию как отозванную

Urinary extracellular vesicles (EVs), including microvesicles and exosomes, play several important roles in cell biology and serve as potential biomarkers in various kidney diseases. Although they have differential biophysical properties, specific biomarkers are required to discriminate these EVs during isolation/purification. The present study aimed to define differential lipidome profiles of urinary microvesicles vs. exosomes. Urine samples collected from eight healthy individuals were pooled and underwent lipid extraction using 2:1(v/v) chloroform/methanol. The recovered lipids were resolve
