Dynamics of scFv-targeted VAP2 correlating with IL-16, MIF and IL-1Ra in ANCA-associated vasculitis.

BACKGROUND AND HYPOTHESIS: Using a mouse model of MPA with microvascular lesion with a clone (VasSF) of recombinant single chain fragments of the variable region of human IgG, we previously showed that vasculitis-associated apolipoprotein A2 (VAP2) may be a therapeutic target for vasculitis. The present study estimated the target molecules for VasSF and the association between VAP2 and cytokine levels in patient sera in terms of microvascular lesion severity. METHODS: Sera and clinical information were collected from patients with microscopic polyangiitis and granulomatosis with polyangiitis

Tumor microenvironment macrophage inhibitory factor directs the accumulation of interleukin-17-producing tumor-infiltrating lymphocytes and predicts favorable survival in nasopharyngeal carcinoma patients.

The accumulation of an intratumoral CD4(+) interleukin-17-producing subset (Th17) of tumor-infiltrating lymphocytes (TILs) is a general characteristic in many cancers. The relationship between the percentage of Th17 cells and clinical prognosis differs among cancers. The mechanism responsible for the increasing percentage of such cells in NPC is still unknown, as is their biological function. Here, our data showed an increase of Th17 cells in tumor tissues relative to their numbers in normal nasopharynx tissues or in the matched peripheral blood of NPC patients. Th17 cells in tumor tissue prod

Concentration-dependent activity of mometasone furoate and dexamethasone on blood eosinophils isolated from atopic children: modulation of Mac-1 expression and chemotaxis.

Treatment of asthma with corticosteroids results in downregulation of eosinophilic airway inflammation. We evaluated in vitro the activity of an "inhaled" corticosteroid, mometasone furoate (MF), and of a "systemic" corticosteroid, dexamethasone (DEX), on eosinophil functions, i.e. adhesion molecule expression and cell chemotaxis. Partially purified blood eosinophils were obtained from 18 asthmatic subjects sensitized to house dust mites. The expression of the macrophage antigen (Mac)-1 (CD11b/CD18) was measured by specific monoclonal antibody (mAb) staining and flow cytometry analysis at base

Study of Lawsonia intracellularis interactions with cell culture mitochondria.

Lawsonia intracellularis is an important obligate intracellular bacterium. It localizes in close proximity to mitochondria within host cells. Mitochondria regulate diverse physiological processes in host cells, thereby significantly affecting the intracellular proliferation of bacteria. However, how L. intracellularis interacts with mitochondria to promote its own proliferation remains unclear. In this study, the mitochondrial morphology of McCoy cells infected with L. intracellularis was examined by indirect immunofluorescence assay (IFA), and L. intracellularis infection was found to induce

Thioredoxin improves contact dermatitis through an anti-inflammatory mechanism different from glucocorticoids.

Thioredoxin (TRX), a redox-regulatory protein of 12 kDa, plays an essential role in modulating oxidative stress and mediating inflammatory processes. In this study, we compared and analyzed the anti-inflammatory effects of topically applied recombinant human TRX (rhTRX), hydrocortisone, and their combination on murine models of contact dermatitis (CD). Topical application of rhTRX, hydrocortisone, and their synergistic combination notably ameliorated ear edema, reduced neutrophilic infiltration within the ear tissues and suppressed the production of cytokines. We explored the distinct anti-inf

Single-Cell and Machine Learning Analyses Identify a PFKFB3-Centered Regulatory Network and Potential Salidroside Interaction in Coronary Heart Disease.

Coronary heart disease (CHD) is a leading cause of morbidity and mortality, driven by metabolic remodeling, vascular inflammation, and perivascular adipose tissue (PVAT) dysfunction. We integrated bulk transcriptomic datasets to develop a machine learning-based diagnostic model, evaluated 113 algorithms, and identified a seven-gene signature (PYGL, PTGS2, PFKFB3, MMP9, CYP1B1, CXCR1, ABCB1) with robust predictive performance. Single-cell RNA sequencing (scRNA-seq) of coronary PVAT revealed substantial cellular heterogeneity and prioritized PFKFB3 as a hub linking glycolytic activity to nuclear

Why Do Some Therapies Work While Others Fail? Mechanisms of Response and Emerging Treatments in axial Spondyloarthritis: 2026 SPARTAN Annual Meeting Proceedings.

PURPOSE OF REVIEW: This review summarizes recent advances in the pathogenesis of axial spondyloarthritis (axSpA), examines why TNF, IL-17, and JAK inhibition is effective whereas IL-23 blockade has not demonstrated clinical efficacy, and highlights emerging therapeutic targets. RECENT FINDINGS: TNF, IL-17 A, and IL-17 F are key drivers of inflammation in axSpA and may also contribute to pathological new bone formation. HLA-B27 can present peptides to CD8+ T cells, potentially expanding pathogenic T-cell populations, including TRBV9+ (V9 T cell receptor beta chain) cells, that produce TNF and

Brazilian Red Propolis and Its Active Constituent 7-O-methylvestitol Impair Early and Late Stages of Toxoplasma gondii Infection in Human Placental Models.

Toxoplasma gondii is a globally distributed protozoan parasite and a major cause of congenital infections, particularly in South America. Current therapies for congenital toxoplasmosis are limited by toxicity, long treatment regimens, and suboptimal efficacy, highlighting the urgent need for safer and more effective alternatives. In this study, we evaluated the antiparasitic effects of crude ethanolic extract of Brazilian Red Propolis (BRP) and its isolated compounds, focusing on 7-O-methylvestitol, in human trophoblast (BeWo) cells and third-trimester placental explants. Both BRP and 7-O-meth

Migration Inhibitory Factor in Conditioned Medium from Human Umbilical Cord Blood-Derived Mesenchymal Stromal Cells Stimulates Hair Growth.

Conventional therapeutic applications of mesenchymal stromal cells (MSCs) focus on cell replacement and differentiation; however, increasing evidence suggests that most of their therapeutic effects are carried out by their various secretions. This study investigated the application of conditioned medium (CM) from human umbilical cord blood-derived MSCs (hUCB-MSCs) to improve hair growth and developed a method to reliably produce this optimized CM. Primed MSC-derived CM (P-CM) with combinations of TGF-β1 and LiCl was optimized by comparing its effects on the cell viability of dermal papilla cel

Characterization of Schistosoma mansoni antigen-reactive T cell clones that form granulomas in vitro.

Granuloma formation and modulation in schistosomiasis are a consequence of discrete subpopulations of T lymphocytes and the mediators they produce. In the present study, T cell clones reactive to soluble egg antigen (SEA) were developed to analyze the roles of T cells in Schistosoma mansoni egg-induced granuloma formation. In an in vitro granuloma assay, 1 X 10(5) T cells specifically augmented the response of 2 X 10(6) normal spleen cells to SEA-coupled but not purified protein derivative-coupled polyacrylamide beads. In vitro granulomatous responses by individual clones were correlated with

Interaction between 'sensitized lymphocytes' and antigen in vitro. 3. Release of soluble mediators by particulate antigen.

Guinea-pigs immunized with sheep red cells (SRC) in Freund's complete adjuvant (FCA) developed delayed type hypersensitivity to a SRC urea extract 7 days after sensitization and showed combined Arthus and delayed reactivity from the 14th day on. Animals immunized by an intracardiac injection of SRC in saline and skin tested 4–5 days later with SRC urea extract, responded with an erythematous and indurated lesion reaching its peak 24 hr after testing and resembling a typical delayed hypersensitivity reaction. Lymphocytes from lymph nodes, peritoneal exudate and peripheral blood of guinea-pigs i

MIF and CD74 as Emerging Biomarkers for Immune Checkpoint Blockade Therapy.

Immune checkpoint blockade (ICB) therapy is used to treat a wide range of cancers; however, some patients are at risk of developing treatment resistance and/or immune-related adverse events (irAEs). Thus, there is a great need for the identification of reliable predictive biomarkers for response and toxicity. The cytokine MIF (macrophage migration inhibitory factor) and its cognate receptor CD74 are intimately connected with cancer progression and have previously been proposed as prognostic biomarkers for patient outcome in various cancers, including solid tumors such as malignant melanoma. Exploring T Cell and NK Cell Involvement in Ankylosing Spondylitis Through Single-Cell Sequencing.

To uncover the complex immune mechanisms driving inflammation in ankylosing spondylitis and lay the groundwork for identifying new therapeutic targets and innovative approaches, we conducted 10× single-cell sequencing on bone marrow cell samples collected from the vertebrae of three AS patients and three non-AS patients. Using single-cell sequencing data, we analysed the expression of differentially expressed genes (DEGs) by comparing AS patients with non-AS patients. Key genes among the related DEGs were identified through protein-protein interaction networks and hub gene screening and furthe

Circulating and Synovial Monocytes in Arthritis and Ex-Vivo Model to Evaluate Therapeutic Modulation of Synovial Monocytes.

Monocytes are innate immune cells that play a dual role in protection of host against pathogens and initiation and perpetuation of inflammatory disorders including joint diseases. During inflammation, monocytes migrate from peripheral blood to tissues via chemokine receptors where they produce inflammatory factors. Monocytes are classified into three subsets, namely: classical, intermediate and non-classical, each subset has particular function. Synovium of patients with inflammatory joint diseases, such as rheumatoid arthritis and psoriatic arthritis as well as osteoarthritis, is enriched by

Galleria mellonella proteomic response to infection with Streptococcus pyogenes.

Galleria mellonella is considered a suitable alternative infection model for studying microbial infections. Unlike conventional animal models, invertebrate models are readily available and not restricted by ethical concerns. Husbandry, maintenance, and handling of G. mellonella larvae are relatively simple. Furthermore, the insect innate immune system, consisting of a humoral and a cellular branch, shares similarities with the mammalian immune system. Streptococcus pyogenes is a strictly human pathogen responsible for a range of clinical manifestations, from superficial uncomplicated infection

Caffeic acid reduces Toxoplasma gondii proliferation in human extravillous trophoblast cells (HTR8/SVneo) through induction of pro-inflammatory cytokines and a death process suggestive of apoptosis.

BACKGROUND: Toxoplasmosis, caused by the protozoan Toxoplasma gondii, is a public health problem, especially in congenital infections. Current treatments, with spiramycin or pyrimethamine and sulfadiazine, are highly toxic, prompting the search for effective and safe natural alternatives. OBJECTIVE: This study evaluated the in vitro effect of caffeic acid (CA) on human extravillous trophoblast cells (HTR8/SVneo) infected by T. gondii. METHODS: The microbicidal and metabolic activity of CA against free tachyzoites was analyzed, in addition to its immunological and anti-Toxoplasma action in in

MIF/CD74 axis in hepatic stellate cells mediates HBV-related liver fibrosis.

Chronic hepatitis B virus (HBV) infection is a major risk factor for liver fibrosis, cirrhosis, and hepatocellular carcinoma (HCC). Despite advances in understanding HBV-related liver diseases, effective therapeutic strategies remain limited. Macrophage migration inhibitory factor (MIF) has been implicated in various inflammatory and fibrotic conditions, but its role in HBV-induced liver fibrosis has not been fully explored. This study investigates the involvement of MIF in liver fibrosis and evaluates its potential as a therapeutic target. We found that MIF expression was significantly elevat
