Genetic and molecular signatures highlight diverse pathways linking obesity to type 2 diabetes.

Obesity is a major risk factor for type 2 diabetes, a disease affecting approximately 10% of the global population. Obesity is a heterogeneous condition, with different components exerting distinct, and sometimes opposing, effects on type 2 diabetes risk. Here, we aim to identify molecular mechanisms through which distinct components of obesity influence risk for type 2 diabetes by integrating multi-omics data. We identify SNPs associated with body mass index in males and females and cluster them based on their Mendelian randomisation estimates on type 2 diabetes risk. This analysis reveals fo

Molecular insights into nagashima-type palmoplantar keratoderma: SERPINB7 mutation spectrum and mechanistic perspectives.

Nagashima-type palmoplantar keratoderma (NPPK) is a common inherited palmoplantar keratoderma predominantly affecting East Asian populations and caused by biallelic loss-of-function variants in the SERPINB7 gene. Clinically, NPPK is characterized by diffuse, non-progressive hyperkeratoderma of the palms and soles with erythema extending beyond the palmoplantar margins, frequently accompanied by hyperhidrosis and malodor. Although the disease course is generally benign, these features may lead to a considerable psychosocial burden. Advances in next-generation sequencing have expanded the mutati

Association between primary aldosteronism and type 2 diabetes and identification of potential molecular mechanisms.

BACKGROUND: Primary aldosteronism (PA) is a common cause of secondary hypertension and is associated with an increased risk of cardiovascular and metabolic complications. Emerging evidence suggests a close relationship between PA and type 2 diabetes mellitus (T2DM), with a higher prevalence of impaired glucose metabolism observed in PA patients. However, the underlying molecular mechanisms linking PA and T2DM remain unclear. PURPOSE: This study aimed to investigate the causal relationship between PA and T2DM and to explore the underlying molecular mechanisms and potential therapeutic targets.

Memory Impairments: Type, Causes, and Molecular Players-Memory Dysfunction Across Neurologic Insults.

Viral infections of the central nervous system produce memory impairment through mechanisms that extend beyond acute neuronal injury. Herpes simplex virus type 1, human immunodeficiency virus, varicella zoster virus, cytomegalovirus, Epstein-Barr virus, influenza, SARS-CoV-2, West Nile virus, and Zika virus each enter or engage the brain through distinct routes, yet converge on four shared molecular pathways that selectively damage hippocampal circuits: mitochondria-associated membrane (MAM) dysfunction, chronic neuroinflammation, blood-brain barrier (BBB) disruption, and impaired CREB-BDNF si

Exploration of autophagy-associated genes and potential molecular mechanisms in type 1 diabetes and osteoporosis.

The co-occurrence of osteoporosis (OP) and type 1 diabetes mellitus (T1DM) represents a clinically significant comorbidity pattern, characterized by skeletal fragility and insulin deficiency. While epidemiological links exist, their shared molecular mechanisms remain undefined. This study investigates autophagy-a critical cellular degradation pathway-as a potential mechanistic bridge between OP and T1DM pathogenesis. We integrated multi-omics approaches using GEO datasets to identify autophagy- associated hub genes. Machine learning (LASSO/Random Forest) prioritized CPNE1 and FRAT2, validated

Molecular signatures of alveolar type II cell differentiation in acute respiratory distress syndrome.

BACKGROUND: Acute respiratory distress syndrome (ARDS) is a life-threatening lung condition with high morbidity and mortality. METHODS: We analyzed publicly available single-cell transcriptomic and microarray datasets from murine ARDS models to characterize AT2 cell differentiation trajectories following lipopolysaccharide (LPS)-induced injury. Computational mapping of cell "trajectories" revealed distinct gene expression signatures associated with divergent repair outcomes. These findings were validated in bronchoalveolar lavage fluid (BALF) samples from ARDS patients and in an LPS-induced A

Pediatric-Type Follicular Lymphoma of the Conjunctiva: Shared Histologic and Molecular Features With the Nodal Counterpart.

Pediatric-type follicular lymphoma (PTFL) is a rare and indolent B-cell lymphoma that largely affects children and young adults. Although typically nodal, rare similar lesions have been reported in the conjunctiva, but differential diagnosis with follicular hyperplasia and other low-grade B-cell lymphomas is challenging. We identified 22 B-cell-derived conjunctival lesions in patients aged 7 to 33 years. All biopsies were characterized using morphologic, immunophenotypic, and molecular criteria. Cases showed a marked male predominance (20 males and 2 females). Final diagnoses included PTFL (n=

Molecular Epidemiology and Evolutionary Dynamics of Human Influenza Type-A Viruses in Africa: A Systematic Review.

Genomic characterization of circulating influenza type-A viruses (IAVs) directs the selection of appropriate vaccine formulations and early detection of potentially pandemic virus strains. However, longitudinal data on the genomic evolution and transmission of IAVs in Africa are scarce, limiting Africa's benefits from potential influenza control strategies. We searched seven databases: African Journals Online, Embase, Global Health, Google Scholar, PubMed, Scopus, and Web of Science according to the PRISMA guidelines for studies that sequenced and/or genomically characterized Africa IAVs.
