Lack of motor recovery after prolonged denervation of the neuromuscular junction is not due to regenerative failure.

Motor axons in peripheral nerves have the capacity to regenerate after injury. However, full functional motor recovery rarely occurs clinically, and this depends on the nature and location of the injury. Recent preclinical findings suggest that there may be a time after nerve injury where, while regrowth to the muscle successfully occurs, there is nevertheless a failure to re-establish motor function, suggesting a possible critical period for synapse reformation. We have now examined the temporal and anatomical determinants for the re-establishment of motor function after prolonged neuromuscul

Ultrastructural evidence of arterial denervation following experimental subarachnoid hemorrhage.

Loss of catecholamine histofluorescence, increased sensitivity to norepinephrine, and changes in alpha1 receptor binding have led to the proposal that denervation hypersensitivity may play a role in cerebrovascular spasm. Because the significance of these alterations has remained unclear, the present study was undertaken to determine whether there was direct ultrastructural evidence of arterial denervation following experimental subarachnoid hemorrhage. Under general anesthesia, adult cats were subjected to pre-pontine injection of blood or serum (5 to 7 ml) via a transclival approach. The ani

"Prespondylosis" and some pain syndromes following denervation supersensitivity.

Pain is determined by the neurologic properties of receptor organs, neurons, and their interconnections. These may become supersensitive or hyperreactive following denervation (Cannon's Law). A common cause of denervation in the peripheral nervous system is neuropathy or radiculopathy as a sequel to spondylosis. Spondylosis in its early stage may be "asymptomatic" or painless and hency unsuspected, because small-diameter pain fibers may not initially be involved despite the attenuation of the other component fibers of the nerve. The term "prespondylosis" is introduced here to describe this pre

Envenoming bites by kraits: the biological basis of treatment-resistant neuromuscular paralysis.

Beta-bungarotoxin, a neurotoxic phospholipase A2 is a major fraction of the venom of kraits. The toxin was inoculated into one hind limb of young adult rats. The inoculated hind limb was paralysed within 3 h, and remained paralysed for 2 days. The paralysis was associated with the loss of synaptic vesicles from motor nerve terminal boutons, a decline in immunoreactivity of synaptophysin, SNAP-25 and syntaxin, a loss of muscle mass and the upregulation of NaV(1.5) mRNA and protein. Between 3 and 6 h after the inoculation of toxin, some nerve terminal boutons exhibited clear signs of degeneratio

Three-dimensional computer reconstruction of the neuromuscular junction distribution in the human posterior cricoarytenoid muscle.

Recent studies have indicated that in a denervated muscle highly specific interactions occur between regenerating axons and the basal lamina of the myofiber precisely at the original synaptic sites. Since these findings suggest that synapse regeneration is facilitated at the original synaptic sites, a knowledge of the distribution of neuromuscular junctions in the human posterior cricoarytenoid (PCA) muscle may guide reinnervation procedures in this muscle. We, therefore, have used histochemical and computer graphics techniques to reconstruct the three-dimensional distribution of neuromuscular

Innervation and properties of the rat FDSBQ muscle: an animal model to evaluate voluntary muscle strength after incomplete spinal cord injury.

Muscles innervated from spinal segments close to the site of a human spinal cord injury are often under voluntary control but are weak because they are partially paralyzed and partially denervated. Our objective was to develop an animal model of this clinical condition to evaluate strategies to improve voluntary muscle strength. To do so, we examined the spinal and peripheral innervation of the flexor digitorum superficialis brevis quinti (FDSBQ) muscle of the rat foot, characterized the muscle and motor unit properties, and located the FDSBQ motoneurons. Retrograde labeled motoneurons were in

Immaturity of the neuromuscular junction in spinal muscular atrophy mouse models.

Spinal muscular atrophy (SMA) is caused by deficiency of the survival motor neuron (SMN) protein and is classically defined by degeneration of lower motor neurons. Extensive evidence from mouse models and human tissue demonstrates that dysfunction at the neuromuscular junction (NMJ) emerges early and precedes overt denervation. Here, we review structural, molecular, and functional studies showing that SMA NMJs fail to complete key postnatal maturation programmes that normally scale presynaptic release capacity to muscle growth and increasing functional demand. SMA motor terminals retain multip

Sectional neuroanatomy of the lower limb I: lower back and hip.

This series of two articles is structured to provide anatomically accurate functional schematics of the motor and sensory innervation of the lower back, hip, and lower limb. This first paper provides radiographically oriented schematic axial sections of the lower back and hip in which the muscles are appropriately color-coded to match the peripheral nerves. A companion color-coded summary table allows prediction of unique patterns of denervation from 25 lesion sites. These are divided into three categories (roots T12 to S4, four plexal quadrants, and 11 sectional levels). Correlation between a

A non-permanent tonic pupil in rheumatoid arteritis.

A 76-year-old male with a severely deforming rheumatoid arthritis, eosinophilia, polymyositis, and episcleritis developed a transient tonic pupil. The episcleritis, and a muscle biopsy revealing an occlusive arteritis with eosinophilia, suggest that a wide-spread rheumatoid arteritis caused a reversible ischemic insult to the ciliary ganglion and thus created a transient denervation of the pupil.

An Analysis of Differentially Expressed Coding and Long Non-Coding RNAs in Multiple Models of Skeletal Muscle Atrophy.

The loss of skeletal muscle mass (muscle atrophy or wasting) caused by aging, diseases, and injury decreases quality of life, survival rates, and healthy life expectancy in humans. Although long non-coding RNAs (lncRNAs) have been implicated in skeletal muscle formation and differentiation, their precise roles in muscle atrophy remain unclear. In this study, we used RNA-sequencing (RNA-Seq) to examine changes in the expression of lncRNAs in four muscle atrophy conditions (denervation, casting, fasting, and cancer cachexia) in mice. We successfully identified 33 annotated lncRNAs and 18 novel l

Asymmetric muscle activity on (18)F-FDG PET/CT.

A 51-year-old man referred to our department to undergo an F-FDG PET/CT to detect primary tumor 7 days after diagnosis of one left brain metastasis on MRI in the aftermath of acute right-sided hemiparesis. F-FDG PET/CT was performed without sufficient fasting period. It showed abnormal uptake in the left apical lung nodule, suspicious for primary tumor. Moreover, F-FDG PET/CT showed extensive skeletal muscle accumulation which was more significant as expected but concerning only the left side of the body. So, acute right-sided hemiparesis was responsible for no muscle uptake, probably explaine

Focal myositis associated with S-1 radiculopathy: report of two cases.

Two cases are described of pseudotumoral calf hypertrophy after laminectomy for a compressive S-1 radiculopathy. The serum creatine kinase (CK) level was normal or mildly elevated. T2-weighted magnetic resonance imaging (MRI) showed calf enlargement, with an increased signal of the medial head of the gastrocnemius muscle. Electromyography revealed fibrillation potentials and positive sharp waves, but no complex repetitive discharges in the affected gastrocnemius muscle, with motor unit potentials having mixed neurogenic and myopathic features. Muscle biopsy revealed a focal myositis associated

The physiological response of the detrusor muscle to experimental bladder outflow obstruction in the pig.

An animal model of bladder outflow obstruction in the pig was used to study the physiological response of the detrusor muscle to obstruction. Bladder muscle strips from obstructed pigs showed a significant increase in sensitivity to acetylcholine and other pharmacological agents compared with controls. In contrast, the response to intramural nerve stimulation was significantly reduced in obstruction, with both cholinergic and non-cholinergic neurotransmission being affected. These findings are consistent with partial denervation of the bladder as a result of obstruction, with the subsequent de

[Surgical therapy of spasmodic torticollis. Selective peripheral denervation].

The authors report on their experience with 19 patients treated with a new operative procedure for spasmodic torticollis--microneurosurgical selective denervation of the hyperactive muscles. In comparison with earlier surgical approaches, the procedure, described by the Canadian neurosurgeon Claude Bertrand, is associated with a high success rate and an absence of the sequelae not infrequently seen with other procedures (stereotactic thalamotomy, intradural divisioning of the anterior roots C1-C3, etc.). The procedure is thus indicated whenever spasmodic torticollis still persists two years af

Cellular activation patterns of CD10+ fibro-adipogenic progenitors across acquired disease states in human skeletal muscle biopsies.

Background: Fibro-adipogenic progenitors (FAP) are muscle resident mesenchymal stem cells pivotal for regulation of myofiber repair. Experimental results show in addition involvement in a range of other pathological conditions and potential for pharmacological intervention. FAP histopathology in human muscle biopsies is largely unknown, but has potential to inform translational research. Methods: CD10+ FAPs in 32 archival muscle biopsies from 8 groups (normal, dermatomyositis, inclusion body myositis (IBM), anti-synthetase syndrome, immune-mediated necrotizing myopathy (IMNM), denervation, typ

Characterization of a phenol-based model for denervation of the abdominal aorta and its implications for aortic remodeling.

OBJECTIVE: Sympathetic innervation plays a pivotal role in regulating cardiovascular health, and its dysregulation is implicated in a wide spectrum of cardiovascular diseases. This study seeks to evaluate the impact of denervation of the abdominal aorta on its morphology and wall homeostasis. METHODS: Male and female Sprague-Dawley rats (N = 12), aged 3 months, underwent midline laparotomy for infrarenal aorta exposure. Chemical denervation was induced via a one-time topical application of 10% phenol (n = 6), whereas sham controls received phosphate-buffered saline (n = 6). Animals were allow

Limb Muscle Reinnervation with the Nerve-Muscle-Endplate Grafting Technique: An Anatomical Feasibility Study.

BACKGROUND: Peroneal nerve injuries results in tibialis anterior (TA) muscle paralysis. TA paralysis could cause "foot drop," a disabling condition that can make walking difficult. As current treatment methods result in poor functional recovery, novel treatment approaches need to be studied. The aim of this study was to explore anatomical feasibility of limb reinnervation with our recently developed nerve-muscle-endplate grafting (NMEG) in the native motor zone (NMZ). METHODS: As the NMEG-NMZ technique involves in nerves and motor endplates (MEPs), the nerve supply patterns and locations of t

Differences between foreign and original innervation of skeletal muscle in the frog.

1. In order to investigate the characteristics of synapses formed by a foreign nerve in skeletal muscle of the frog, the hypoglossal nerve in Rana pipiens was implanted under the cutaneus pectoris (c.p.) muscle which was denervated 1 month later.2. Within 1 week of crushing the c.p. nerve, hypoglossal nerve fibres could be seen extending towards the denervated end-plates and the muscle contracted in response to stimulation of the implanted nerve.3. The synapses formed by the foreign nerve differed from those of the original nerve in several ways. The mean quantal content (m) of end-plate poten
