Histological and immunohistochemical evaluation of polyherbal formulations on inflammatory, apoptotic, and angiogenic pathways in streptozotocin-induced diabetic cardiac injury.

This study investigated whether polyherbal formulations (PHF1 and PHF2) have cardioprotective, anti-inflammatory, and anti-apoptotic effects when compared to metformin (MFM) in rats with diabetes induced by streptozotocin (STZ). The rats were divided into five groups: control, STZ, STZ + MFM, STZ + PHF1, and STZ + PHF2. Cardiac tissues underwent histological and immunohistochemical analyses to evaluate tissue-level molecular alterations induced by STZ and the regulatory impact of polyherbal formulations. The STZ-treated group exhibited statistically significant elevations in oxidative stress,

Endothelial Hydrogels Improve Microvascular Regeneration and Perfusion in Tracheal Scaffolds.

OBJECTIVE: Vascularization remains a major barrier to the clinical translation of airway replacement. We previously demonstrated that engineered tracheal scaffolds support neovascularization, although restoration of microvascular perfusion is delayed in part due to a reduction in capillary branching. Endothelial hydrogels (EH) have demonstrated the potential to enhance vascular regeneration in several domains of tissue engineering, yet their application in airway replacement has not been fully evaluated. In this study, we assessed the impact of EH on microvascular regeneration and perfusion in

A novel phosphorylated chitin-hydroxyapatite composite promotes angiogenesis and osteogenesis for bone defect repair.

Current bone repair materials face limitations including donor site morbidity, immune rejection, and insufficient osteogenic capacity. To overcome these challenges, tissue engineering has introduced bioactive composites that better mimic the natural bone microenvironment. Here, we developed a novel composite by integrating phosphorylated chitin, which is an excellent biocompatible and bioactive organic polymer, with biogenic hydroxyapatite, a mineral phase closely resembling native bone. The composite was systematically characterized for its physicochemical properties. In vitro studies demonst

An Experimental Study on 3D-Printed Gyroid-Shaped TC4 Porous Scaffolds Guiding Angiogenesis and Osteogenesis in Bone Defect Areas.

To investigate the ability of novel Gyroid-shaped titanium alloy (TC4) porous bioscaffolds to induce angiogenesis and osteogenesis in bone defect areas. This study employed selective laser melting (SLM) technology to fabricate Gyroid shaped and Cube-shaped TC4 porous bioscaffolds, using the commonly used cube shape as a control. The unit cell size was 4 mm, with a wall thickness or rod diameter of 300 μm and a porosity of approximately 80%. These scaffolds were implanted into rabbit mandibular defect sites (10 mm × 7 mm × 5 mm) to evaluate the angiogenic and osteogenic potential of the Gyroid-

Microfluidic pulp platform with vascular integration to evaluate biofunctional material.

Biofunctional materials are increasingly used to preserve tooth vitality by promoting dental pulp-mediated hard tissue formation. However, existing evaluation platforms, such as conventional in vitro assays or microfluidic systems, fail to replicate the complex histological and physiological characteristics of dental pulp. This study introduces a 4D biofunctional material-to-pulp (4D BFP) platform that recapitulates pulp physiology, integrating three key features of native pulp tissue: layered histoarchitecture, microcirculatory dynamics, and three-dimensional multicellular organization. This

VEGF-E Attenuates Injury After Ischemic Stroke by Promoting Reparative Revascularization.

The angiogenic response after stroke correlates with mild injury and an improved recovery. Stimulation of post-stroke angiogenesis using vascular endothelial growth factor (VEGF)-A is associated with an increased risk of vascular destabilization, leading to life-threatening complications. The non-mammalian VEGF-A homolog, VEGF-E, stimulates stable cutaneous vascularization and promotes wound healing. Herein, we posit that VEGF-E represents a potential mediator of reparative revascularization after ischemic stroke. C57BL6/J wildtype mice were subjected to experimental stroke, and VEGF-E or VEGF

Boosting the angiogenesis potential of self-assembled mesenchymal stem cell spheroids by size mediated physiological hypoxia for vascularized pulp regeneration.

Hypoxia is a pivotal factor in enhancing the vascularization potential of both two-dimensional (2D) cultured cells and three-dimensional (3D) cellular spheroids. Nevertheless, spheroids that closely mimic the in vivo microenvironment often experience excessive hypoxia, leading to the necrotic core and the release of toxic byproducts, ultimately impeding the regenerative process. To balance cell vitality and pro-angiogenic properties of cellular spheroids, this study investigates size-dependent hypoxia in stem cell spheroids utilizing an oxygen transfer finite element model. Subsequently, we de

Self-Organized Vascularized Microtissue Based on the Human Microvascular Unit for Tissue Repair and Regeneration.

Insufficient blood circulation in injured regions is a critical challenge that hinders the application of engineered tissue in the regeneration of tissue defects or losses. Here, we developed a three-dimensional culture system that enables human microvascular units (MVUs) to expand and self-organize into vascularized microtissues containing well-formed microvascular structures and stem-cell-derived functional stroma. Moreover, the vascularized microtissue is customizable in both shape and size. The functionality of vascularized microtissues was evaluated in various tissue defect models includi

A functional cardiac patch with "gas and ion" dual-effect intervention for reconstructing blood microcirculation in myocardial infarction repair.

Postinfarction revascularization is critical for repairing the infarcted myocardium and for stopping disease progression. Considering the limitations of surgical intervention, engineered cardiac patches (ECPs) are more effective in establishing rich blood supply networks. For efficacy, ECPs should promote the formation of more mature blood vessels to improve microcirculatory dysfunction and mitigate hypoxia-induced apoptosis. Developing collateral circulation between infarcted myocardium and ECPs for restoring blood perfusion remains a challenge. Here, an ion-conductive composite ECPs (GMA@OSM

Dual-function MgSrCeAl-LDH nanosheets enable both ROS clearance and enhanced angiogenesis for myocardial ischemia/reperfusion injury therapy.

Myocardial ischemia/reperfusion (I/R) injury represents a major global health concern, driven by excessive reactive oxygen species (ROS), which closely correlates with subsequent myocardial infarction and fibrosis. Combining ROS scavenging with ischemic myocardium revascularization holds therapeutic promise. However, developing multifunctional therapeutic agents that can simultaneously scavenge ROS and promote angiogenesis remains unreported. Herein, we propose MgSrCeAl-layered double hydroxide (LDH) nanosheets as a multifunctional ROS scavenger and angiogenesis promoter for efficient myocardi

Direct in vivo reprogramming to relieve tissue ischemia via induced vasculogenesis.

There is an ongoing need for innovative cell and regenerative medicine therapies to promote vascular repair and regeneration to meet the growing global health concern of ischemic cardiovascular diseases. Direct reprogramming of somatic cells into an induced pluripotent state caused scientists to reconsider prior theories of cell differentiation and develop approaches to directly reprogram cells in vivo into other states or fates to adaptively replace cells lost to aging or disease. While direct in vivo reprogramming of fibroblast to cardiomyocytes, neurons, or pancreatic beta islet cell states

Three-Dimensional Bioprinting of Astrocytes and Endothelial Cells to Direct Retinal Axon Growth and Vascularization.

Retinal organoids (ROs) are currently used to study retinal development and diseases but cannot model glaucoma because they fail to form a nerve fiber layer (NFL) and optic nerve (ON). Utilizing three-dimensional bioprinting, ON head astrocytes (ONHAs) and vascular endothelial cells, both of which contribute to NFL development in vivo but are absent in ROs, were positioned at the center of scaffolds seeded with retinal ganglion cells (RGCs). In experiments using ONHAs isolated from developing retinas, polarization of RGC neurite growth increased by 43% while ONHA from adult retinas or astrocyt

A VEGF gene therapy approach for the treatment of patients with coronary artery disease and refractory angina: assessment of clinical development.

INTRODUCTION: Vascular endothelial growth factor (VEGF) gene therapy is a novel treatment strategy for refractory angina (RA) that works by promoting myocardial neoangiogenesis and collateral circulation formation. XC001 (encoberminogene rezmadenovec) is a novel, replication-deficient, non-integrating recombinant adenovirus vector formally referred to as AdVEGF-All6A+ engineered to produce three isoforms of VEGF A (121, 165, and 189) that are proven to induce neoangiogenesis and constructed specifically to increase the expression of VEGF 189 and 165 to improve safety because of its heparan sul

Blood vessel wall shear stress determines regions of liposome accumulation in angiogenic vasculature.

Nanoparticles used for drug delivery often require intravenous administration exposing them to fluid forces within the vasculature, yet the impact of blood flow on nanoparticle delivery remains incompletely understood. Here, we utilized transgenic zebrafish embryos to investigate the relationship between the accumulation of fluorescently labeled PEGylated liposomes and various hemodynamic factors (such as flow velocity, wall shear stress (WSS), and flow pattern) across a wide range of angiogenic blood vessels. We reconstructed 3D models of vascular structures from confocal images and used comp

ECM-mimetic glucomannan hydrogel promotes pressure ulcer healing by scavenging ROS, promoting angiogenesis and regulating macrophages.

Pressure ulcers (PUs) have emerged as a significant burden on both individuals and society. Effective treatment of PUs is a significant clinical challenge due to the compromised tissue microenvironment characterized by extracellular matrix (ECM) depletion, increased levels of reactive oxygen species (ROS), excessive inflammation and impaired angiogenesis. To this end, we have developed a glucomannan hydrogel (GM-Pgel) that mimics the skin's extracellular matrix to accelerate wound healing by regulating chronic inflammation in the PUs. This hydrogel not only faithfully replicates the components
