Role of the autonomic nervous system in the mediation of LHA electrical stimulation-induced effects on insulinemia and glycemia.

The short-term effects of bilateral electrical stimulation of the lateral component of the lateral hypothalamic/medial forebrain bundle area (LHA) on peripheral plasma levels of glucose (G), immunoreactive insulin (IRI) and glucagon (IRG) were measured in food-derived, anesthetized rats in the presence of continuous low i.v. glucose infusion. In normal rats LHA stimulation induced a rapid but moderate rise in IRI in the presence of a simultaneous rapid rise in G. At the end of the 5 min stimulation period the glucose-induced IRI response was clearly attenuated. In rats receiving a continuous p

Acute Inhibition of Inflammation Mediated by Sympathetic Nerves: The Inflammatory Reflex.

In this review, we will try to convince the readers that the immune system is controlled by an endogenous neural reflex, termed inflammatory reflex, that inhibits the acute immune response during the course of a systemic immune challenge. We will analyse here the contribution of different sympathetic nerves as possible efferent arms of the inflammatory reflex. We will discuss the evidence that demonstrates that neither the splenic sympathetic nerves nor the hepatic sympathetic nerves are necessary for the endogenous neural reflex inhibition of inflammation. We will discuss the contribution of

Identification of small-molecule inhibitors of Zika virus infection and induced neural cell death via a drug repurposing screen.

In response to the current global health emergency posed by the Zika virus (ZIKV) outbreak and its link to microcephaly and other neurological conditions, we performed a drug repurposing screen of ∼6,000 compounds that included approved drugs, clinical trial drug candidates and pharmacologically active compounds; we identified compounds that either inhibit ZIKV infection or suppress infection-induced caspase-3 activity in different neural cells. A pan-caspase inhibitor, emricasan, inhibited ZIKV-induced increases in caspase-3 activity and protected human cortical neural progenitors in both mon

Mediolateral neural patterning during gastrulation precedes dorsoventral patterning in chick and frog.

INTRODUCTION: dorsoventral polarity of the neural tube is based on molecular patterning established during neurulation and is suggested to be induced by fine-tuned interpretation of opposing morphogen gradients in the neural tube: a ventral-to-dorsal gradient is formed by shh secreted from floor plate and notochord whereas a dorsal-to-ventral gradient is created by BMP and Wnt originating from the roof plate. Recent results revealed an unexpected early induction of Shh in the chick prospective floor plate prior to notochord formation. Hence, we addressed the questions as to whether key compone

Norepinephrine transporter inhibition prevents tilt-induced pre-syncope.

OBJECTIVES: We tested the hypothesis that pharmacological norepinephrine reuptake transporter (NET) inhibition delays the onset of head-up tilt-induced presyncope in healthy subjects. BACKGROUND: Treatment of neurally mediated syncope is unsatisfactory. In a previous study in a small number of healthy subjects, pharmacologic NET inhibition delayed the onset of head-up tilt-induced pre-syncope. METHODS: We combined data sets from 3 substudies comprising 51 healthy subjects without a history of syncope. In a double-blind, randomized, cross-over fashion, subjects underwent 2 head-up tilt tests,

Response Dissociation in Hierarchical Cortical Circuits: a Unique Feature of Autism Spectrum Disorder.

A prominent hypothesis regarding the pathophysiology of autism is that an increase in the balance between neural excitation and inhibition results in an increase in neural responses. However, previous reports of population-level response magnitude in individuals with autism have been inconsistent. Critically, network interactions have not been considered in previous neuroimaging studies of excitation and inhibition imbalance in autism. In particular, a defining characteristic of cortical organization is its hierarchical and interactive structure; sensory and cognitive systems are comprised of

Epithelial involution and basement membrane loss during early rhombencephalic tectoria lamina development.

Extracellular material molecules play a key role in the regulation of morphogenesis and differentiation of a large number of organs including the central nervous system. However, the role of the neural basement membrane in the growth of different parts of the neural tube has yet to been delineated. Here, the structural and compositional modifications of the basement membrane (BM) of rhombencephalic tectoria lamina anlage (RTLA) have been examined during the process of RTLA epithelial attenuation. Between stages 10 to 11-the presumptive RTLA epithelium showed a structure, thickness and cell-pro

Molecular Control of Neural Stem Cell Quiescence in the Adult Zebrafish Telencephalon: Established Mechanisms and an Integrative Hypothesis.

The zebrafish telencephalon is an excellent model for adult neurogenesis under physiological and regenerative conditions, as many pathways regulating neural stem cell behavior are conserved with mammalian neurogenic niches. Resident radial glial cells (RGCs) serve as neural stem cells in the adult brain and are readily accessible in vivo, because forebrain eversion repositions the ventricular zone to the dorsal surface. Recent studies have revealed substantial heterogeneity among RGCs, including activation-primed cells and deeply quiescent states during homeostasis. Here, we first review the e

A Molecular Analysis of Neural Olfactory Placode Differentiation in Human Pluripotent Stem Cells.

During embryonic development, the olfactory sensory neurons (OSNs) and the gonadotropic-releasing hormone neurons (GNRHNs) migrate from the early nasal cavity, known as the olfactory placode, to the brain. Defects in the development of OSNs and GNRHNs result in neurodevelopmental disorders such as anosmia and congenital hypogonadotropic hypogonadism, respectively. Treatments do not restore the defective neurons in these disorders, and as a result, patients have a diminished sense of smell or a gonadotropin hormone deficiency. Human pluripotent stem cells (hPSCs) can produce any cell type in th

Changes in clinical pain in fibromyalgia patients correlate with changes in brain activation in the cingulate cortex in a response inhibition task.

OBJECTIVE: The primary symptom of fibromyalgia is chronic, widespread pain; however, patients report additional symptoms including decreased concentration and memory. Performance-based deficits are seen mainly in tests of working memory and executive functioning. It has been hypothesized that pain interferes with cognitive performance; however, the neural correlates of this interference are still a matter of debate. In a previous, cross-sectional study, we reported that fibromyalgia patients (as compared with healthy controls) showed a decreased blood oxygen level dependent (BOLD) response rel

Abnormal dorsal premotor-motor inhibition in writer's cramp.

The authors hypothesized that a deficient premotor-motor inhibitory network contributes to the unwanted involuntary movements in dystonia. The authors studied nine controls and nine patients with writer's cramp (WC). Dorsal premotor-motor cortical inhibition (dPMI) was tested by applying conditioning transcranial magnetic stimulation (TMS) to the dorsal premotor cortex and then a test pulse to the ipsilateral motor cortex at an interval of 6 ms. The authors used an H-reflex in flexor carpi radialis paired with TMS over the premotor cortex to assess for spinal cord excitability change. Finally,

Chronic sustained hypoxia enhances both evoked EPSCs and norepinephrine inhibition of glutamatergic afferent inputs in the nucleus of the solitary tract.

The nucleus of the solitary tract (NTS) receives inputs from both arterial chemoreceptors and central noradrenergic neural structures activated during hypoxia. We investigated norepinephrine (NE) modulation of chemoreceptor afferent integration after a chronic exposure to sustained hypoxia (CSH) (7-8 d at 10% FIO(2)). Whole-cell recordings of NTS second-order neurons identified by DiA (1,1'-dilinoleyl-3,3,3',3'-tetra-methylindocarbocyanine, 4-chlorobenzenesulphonate) labeling of carotid bodies were obtained in a brain slice. Electrical stimulation of the solitary tract was used to evoke EPSCs.

Non-Invasive Brain Stimulation of the Ventromedial Prefrontal Cortex Improves Behavioral Inhibition by Enhancing the Processing Depth and Anticipation of Outcomes in a Gambling Task.

Disordered gambling and other behavioral addictions are characterized by a lack of behavioral inhibition, such that patients repeatedly make disadvantageous decisions and fail to disengage from maladaptive behavior. Previous research has shown that behavioral addictions are associated with altered ventromedial prefrontal cortex (vmPFC) activity, making it an interesting target for neuromodulation. Using a gambling paradigm containing positive and negative expected value trials, we investigated decision-making and feedback processing while stimulating the vmPFC via transcranial direct current s

Phenothiourea sensitizes zebrafish cranial neural crest and extraocular muscle development to changes in retinoic acid and IGF signaling.

1-Phenyl 2-thiourea (PTU) is a tyrosinase inhibitor commonly used to block pigmentation and aid visualization of zebrafish development. At the standard concentration of 0.003% (200 µM), PTU inhibits melanogenesis and reportedly has minimal other effects on zebrafish embryogenesis. We found that 0.003% PTU altered retinoic acid and insulin-like growth factor (IGF) regulation of neural crest and mesodermal components of craniofacial development. Reduction of retinoic acid synthesis by the pan-aldehyde dehydrogenase inhibitor diethylbenzaldehyde, only when combined with 0.003% PTU, resulted in ex

Comprehensive profiling of brain dynamics during anesthesia across phylogeny.

The behavioral effects of anesthetics are highly conserved across species, hinting at shared and fundamental underlying mechanisms. Here we compile a dataset of multiscale neural activity during wakefulness and anesthesia, encompassing human, macaque, marmoset, mouse, zebrafish and nematode. Applying massive feature extraction, we characterize local neural dynamics across >6,000 time-series features. This reveals a conserved dynamical profile of anesthesia across species, characterized by shorter intrinsic timescales of neural activity and dampened inter-regional synchrony. Deep-brain stimulat

Adnp2 promotes neural induction by antagonizing the ved and vent homeobox genes.

In vertebrates, neural induction requires the inhibition of BMP signals by dorsal organizer-derived antagonists. However, the underlying mechanism is not completely understood. Here, using zebrafish as a model, we show that ADNP2 orthologs adnp2a and adnp2b are required for proper neural induction in gastrulating embryos. Deficiency of adnp2 causes deficits in neuroectoderm specification, showing reduced expression of both anterior and posterior neural markers. Adnp2 functions as a transcriptional repressor, and directly occupies and suppresses the BMP-related ved/vent/vox homeobox genes, ther

Enhancing inhibition-induced plasticity in tinnitus--spectral energy contrasts in tailor-made notched music matter.

Chronic tinnitus seems to be caused by reduced inhibition among frequency selective neurons in the auditory cortex. One possibility to reduce tinnitus perception is to induce inhibition onto over-activated neurons representing the tinnitus frequency via tailor-made notched music (TMNM). Since lateral inhibition is modifiable by spectral energy contrasts, the question arises if the effects of inhibition-induced plasticity can be enhanced by introducing increased spectral energy contrasts (ISEC) in TMNM. Eighteen participants suffering from chronic tonal tinnitus, pseudo randomly assigned to eit

Specific recruitment properties of spinal reflex of thigh muscle in sprinter.

Thigh muscles activation pattern plays a key role in high-performance sprint running. We investigated spinal neural mechanisms underlying sprinters' specific thigh muscle control by examining spinal reflex recruitment properties and reciprocal inhibition of the thigh muscles in sprinters. Ten high-level national sprinters and ten controls participated in this study. Transcutaneous spinal cord stimulation (tSCS) was used to evoke spinal reflexes in biceps femoris (BF) and soleus (SOL) muscles. To assess the recruitment properties of thigh muscles, in Experiment 1, tSCS was applied at varying in

Laminin potentiates differentiation of PCC4uva embryonal carcinoma into neurons.

The embryonal carcinoma PCC4uva differentiates into neurons in response to treatment with retinoic acid and dbcAMP. We used this in vitro model system to study the effects of laminin on early neural differentiation. Laminin substrata markedly potentiate neural differentiation of retinoic acid and dbcAMP-treated cultures. Only laminin induced more rapid neural cell body clustering, neurite growth and neurite fasciculation as compared to type IV collagen, type I collagen, and fibronectin substrata. Exogenous laminin substrata promoted greater cell attachment, cellular spreading and growth to con

Optoelectronic artificial synapse for lateral inhibition-enhanced retinal biomimicry.

Optoelectronic synaptic devices enable in-sensor processing of enhanced edge detection and contrast resolution in complex visual scenes due to their excellent capability to emulate the functions of visual neurons, such as light perception and image processing, while lateral inhibition synaptic plasticity refines spatial selectivity and extends the dynamic range by suppressing redundant signals and amplifying subtle variations in input intensity. The incorporation of lateral inhibition into a single optoelectronic synaptic device will offer a cost-effective and energy-efficient route for direct
