The prognosis of traumatized permanent anterior teeth with fracture of the enamel and dentin.

The purpose of this study was to examine, in the light of a 2-year follow-up, the diagnostic and prognostic value of vitality tests in teeth which had suffered fracture of the enamel and dentin without pulp exposure, to assess the period in which late pathologic changes may occur, and to determine the recommended time intervals for follow-up examinations. Eighty-four childred, 6 to 14 years of age, with 123 traumatized teeth were included in the study. Of these teeth, 87 percent were "vital" at the initial examination, and most of these remained vital throughout the 2 years. Most pathologic ch

Wnt1's Differential Effects on Craniofacial Bone and Tooth Development.

The development of craniofacial bones and teeth relies heavily on the Wnt signaling pathway, yet the specific mechanisms and Wnt variants involved remain under continual investigation. Using publicly available single-cell sequencing data from the mouse incisor, we reveal Wnt1 expression across dental structures and investigate its role using a Col1a1-dependent Wnt1 transgenic mouse model. Inducing Wnt1 early on affects craniofacial bone without disturbing tooth development, but prolonged embryonic induction leads to postnatal mortality with osteopetrosis-like bone overgrowth and malformed teet

Biomechanical stress regulates mammalian tooth replacement via the integrin β1-RUNX2-Wnt pathway.

Renewal of integumentary organs occurs cyclically throughout an organism's lifetime, but the mechanism that initiates each cycle remains largely unknown. In a miniature pig model of tooth development that resembles tooth development in humans, the permanent tooth did not begin transitioning from the resting to the initiation stage until the deciduous tooth began to erupt. This eruption released the accumulated mechanical stress inside the mandible. Mechanical stress prevented permanent tooth development by regulating expression and activity of the integrin β1-ERK1-RUNX2 axis in the surrounding

The epidemiological and clinical features of odontogenic infective endocarditis.

Antibiotic prophylaxis (AP) of infective endocarditis (IE) in dental practice is a controversial topic. We evaluated the characteristics of the odontogenic IE and assessed the practice and sources of information pertaining to the topic utilized by the Croatian dentists. We conducted a retrospective review of consecutive medical charts of adult patients with IE, admitted to the University Hospital for Infectious Diseases in Zagreb, Croatia, between January 2007 and December 2017. In addition, a cross-sectional, self-reporting questionnaire survey was conducted with participation of 348 Croatian

DNA methylation is critical for tooth agenesis: implications for sporadic non-syndromic anodontia and hypodontia.

Hypodontia is caused by interactions among genetic, epigenetic, and environmental factors during tooth development, but the actual mechanism is unknown. DNA methylation now appears to play a significant role in abnormal developments, flawed phenotypes, and acquired diseases. Methylated DNA immunoprecipitation (MeDIP) has been developed as a new method of scanning large-scale DNA-methylation profiles within particular regions or in the entire genome. Here, we performed a genome-wide scan of paired DNA samples obtained from 4 patients lacking two mandibular incisors and 4 healthy controls with n

Pre-eruptive intra-coronal resorption: controversies and treatment options.

Pre-eruptive intra-coronal resorption (PIER) is a defect located in the dentin of an unerupted tooth, just beneath the dentin-enamel junction, with a prevalence of 0.5-2% of the teeth. The depth of the lesion is variable and may also reach the pulp. In the past, these lesions were confused with caries, and were therefore called "hidden" or "pre-eruptive caries". These defects are usually detected incidentally in routine dental radiographs. It has been proven that in the pre-eruptive stage the lesions contain soft tissue and inflammatory cells. The present report describes the clinical manageme

The dental age in the child with coeliac disease.

AIM: The coeliac disease deprives affected individual of the main nutritive factors, which are essential to promote body development: coeliac children are often subject to weight loss and have a lower somatic growth rate compared to healthy children. In addition, it is proven that prolonged malnutrition can have irreversible effects on dental eruption: teeth development also suffers a delay or it is slowed down in coeliac children. The purpose of this study was to evaluate and quantify the persistence of a delay in the dental age in children with coeliac disease and the presence of a possible

Dentoalveolar changes after unilateral extractions of mandibular first molars and their influence on third molar development and position.

AIM: To investigate the spontaneous tooth position changes after unilateral extraction of mandibular first molars and the influence on third molar position. METHODS: Panoramic radiographs of 111 individuals (mean age 19 years 8 months) in whom one mandibular first molar was extracted at least 5 years prior. Comparison of all measurements of the control and the affected side was performed by paired Student t test. RESULTS: The mandibular second molars tipped mesially, whereas the premolars, canines, and incisors moved distally toward the extraction space. Vertical alveolar resorption was sign

Mouse models of tooth abnormalities.

Tooth number is abnormal in about 20% of the human population. The most common defect is agenesis of the third molars, followed by loss of the lateral incisors and loss of the second premolars. Tooth loss appears as both a feature of multi-organ syndromes and as a non-syndromic isolated character. Apart from tooth number, abnormalities are also observed in tooth size, shape, and structure. Many of the genes that underlie dental defects have been identified, and several mouse models have been created to allow functional studies to understand, in greater detail, the role of particular genes in t

Radiographic follow-up of impacted third molars from age 20 to 32 years.

Nineteen patients (13 male, six female) with 34 impacted third molars, 21 in the mandible and 13 in the maxilla were radiographically followed from age 20 to 32. All were examined clinically and panoramic radiographs were taken at baseline and at the end of the study. Radiographic analysis included resorption of teeth, enlargement of the follicle, development of the root, change in inclination of the third molar, state of impaction, relative depth of the third molar in bone and relation to the ramus of the mandible and to the second molar tooth. In the mandible, the mean change in inclination

The novel gene locus for agenesis of permanent teeth (He-Zhao deficiency) maps to chromosome 10q11.2.

He-Zhao deficiency has been recently characterized with a distinct form of agenesis of permanent teeth that is different from other previously reported disorders of tooth agenesis. This inherited abnormality suggests that some gene(s) associated with the development of permanent teeth may mutate. In this study, we map the gene locus to chromosome 10q11.2. The DNA pooling method combined with two-point and multi-point linkage analysis has been successfully applied. The maximum LOD (Zmax) scores for two-point and multi-point analyses are 13.29 (on marker D10S196) at recombination fraction (theta

Dental development in Apert syndrome.

OBJECTIVE: Apert syndrome has been extensively studied and described. However, an area that has not been studied is the dental development of these individuals. The purpose of this study was to evaluate the development of the dentition and compare it with that of unaffected children. There appears to be clinical observations indicating delayed eruption of the permanent teeth in the Apert child. METHODS: This retrospective study examined all Apert syndrome patients from four craniofacial centers who had a panoramic radiograph taken before the age of 16 years. Thirty-six individuals, 19 boys an

Growth factors in bones and teeth.

Human bone contains an abundance of polypeptide growth factors. These growth factors stimulate the proliferation and activity of bone cells and can stimulate bone formation. Data from this laboratory and others suggest that bone growth factors may act to couple bone formation to resorption to maintain bone mass during remodeling. Research is underway to study these growth factors in bones and teeth, and their possible roles in both the pathogenesis and the treatments of osteoporosis and dental diseases.

Effects of bone marrow transplantation on impacted dental germs in osteopetrotic op/op rats.

The osteopetrotic op/op rats suffered from a complete failure of tooth eruption in association with a reduced bone resorption. An unresorbed occlusal bone overlaid the germs which remained embedded in bone. The process of prefunctional eruption in limited growing teeth depends on the resorption of the alveolar bone covering the germs. The aim of this study was to investigate the effects of hematopoïetic grafts in newborn op/op rats on the process of prefunctional eruption. Fifteen op/op treated rats were used for radiographic, microradiographic and histologic examinations. After transfusion of

Some unusual cases of apexification subsequent to trauma.

Three cases are reported in which apexification occurred without the benefit of endodontic treatment. These reports illustrate that immature teeth which are avulsed and replanted, dislocated and repositioned, or traumatized with pulp necrosis supervening, may progress to form complete apices without intracanal therapy. The value of using calcium hydroxide is not in question, but may not be as essential an aid to apical closure as was previously thought. In one case, the appearance of isolated apixes forming, deep in the bone after loss of very immature lateral incisors poses the question of wh

In vivo bioengineered tooth formation using decellularized tooth bud extracellular matrix scaffolds.

The use of dental implants to replace lost or damaged teeth has become increasingly widespread due to their reported high survival and success rates. In reality, the long-term survival of dental implants remains a health concern, based on their short-term predicted survival of ~15 years, significant potential for jawbone resorption, and risk of peri-implantitis. The ability to create functional bioengineered teeth, composed of living tissues with properties similar to those of natural teeth, would be a significant improvement over currently used synthetic titanium implants. To address this pos

Effects of Porphyromonas gingivalis lipopolysaccharide on the behavior of human dental pulp stem cells in vitro and their inflammation-related transcriptomics profile.

OBJECTIVE: The inflammatory microenvironment in pulpitis and periapical periodontitis critically impairs dental pulp stem cells (DPSCs) functionality, yet the underlying molecular mechanisms remain poorly defined. This study aimed to investigate the effects of Porphyromonas gingivalis lipopolysaccharide (P. gingivalis LPS) on human DPSCs behavior and elucidate the transcriptomic changes underlying LPS-induced inflammation. DESIGN: Human DPSCs were exposed to 1 µg/mL LPS for 24 hours to establish an inflammatory model, with subsequent evaluation of proliferation (CCK-8), migration (Transwell),
