Autophagy dysfunction in iPSCs-derived neurons and midbrain organoids carrying a SNCA triplication.

Parkinson's disease (PD), characterized by α-Synuclein aggregation and dopaminergic neuronal loss, has no current cure. Autophagy is critical for α-Synuclein clearance, yet its real-time dynamics remain challenging to assess in human-relevant systems. Here, we used live-cell imaging to assess autophagy within human neuronal cultures and midbrain organoids (hMOs) derived from induced pluripotent stem cells (iPSCs) of PD patients carrying a triplication of the α-Synuclein gene (3xSNCA). Using the LC3-Rosella dual-fluorescent reporter, we quantified autolysosomes dynamics in real time. In 3xSNCA …

On-Demand Generation of Patient-Derived Organoids for Precision Oncology: Progress and Prospects.

Precision oncology has become a focus of both clinical practice and basic research. With the rapid evolution of organoid culture techniques and the integration of biomedical engineering approaches, patient-derived organoid (PDO) systems are playing an increasingly pivotal role in precision cancer therapy. By recapitulating patient-specific tumor characteristics, these platforms demonstrate considerable potential for clinical translation. Despite remarkable progress, the impact of PDO models on real-world therapeutic decision-making remains limited. Most existing studies have focused on preclin

Integrative characterization of human hepatocellular carcinoma-derived organoids and their HBV-associated biology.

BACKGROUND: Hepatocellular carcinoma (HCC) is one of the most aggressive malignancies and is characterized by a high degree of heterogeneity. However, HCC treatment remains difficult, highlighting the need to better define molecular subgroups of HCC and understand their potential relationships with therapeutic response. METHODS: In this study, we established 20 patient-derived organoids and 12 patient-derived cell lines from 20 individuals diagnosed with HCC. These models were comprehensively characterized using whole-exome sequencing, RNA sequencing, drug screening, and hepatitis B virus (HB…

ErbB receptor stimulation is required for mouse Colon adenoma organoids to form crypts.

When cultured with Wnt, R-spondin, EGF, Noggin, myofibroblast conditioned medium and Matrigel, crypts from normal mouse colon mucosa form crypt-producing organoids and can be passaged every seven days. Under the same culture and passage conditions, crypts isolated from colon adenomas derived from Apcmin/+ mice typically grow as spheroidal cysts and do not produce crypts. The adenoma organoids require EGF, but not Wnt, R-spondin or Noggin for continuous passaging. However, when mouse colon adenoma spheroids are grown for more than 10 days in the presence of EGF, crypt formation occurs. EGF, ERE…

Patient-derived cancer organoids to tailor personalized treatment strategies in upper gastrointestinal malignancies.

Upper gastrointestinal malignancies bear a high morbidity and mortality burden. Curative treatment requires a multimodal approach, subjecting patients to preoperative chemotherapy or chemoradiation to downstage the tumor before resection. Various preoperative regimens exist but there is no tailor-made approach based on the tumor sensitivity for the individual patient. This predisposes a considerable subset of patients to inadequate treatment and highlights the need for personalized medicine, for which the potential of patient-derived cancer organoids (PDCOs) was investigated. PDCOs were establ…

Mosaic human cortical organoids model mTOR-related focal cortical dysplasia through DEPDC5 deletion.

Focal cortical dysplasia type II (FCDII), a major cause of paediatric drug-resistant focal epilepsy, results from brain somatic variants in mTOR pathway genes, including germline and somatic second-hit loss-of-function variants in the mTOR repressor DEPDC5. Here, we present a proof-of-concept model of DEPDC5 two-hit inactivation mosaicism using patient-derived human cortical organoids (hCOs). Mosaic hCOs displayed increased mTOR activity that was rescued by the mTOR inhibitor rapamycin. Mosaic hCOs also exhibited dysmorphic-like neurons and enhanced neuronal excitability, recapitulating key FC…

Organoids in translation: a bench-to-bedside framework for pancreatic cancer precision medicine.

INTRODUCTION: Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal malignancies with a 5-year survival rate of < 13%. Standard treatments such as FOLFIRINOX or gemcitabine/nab-paclitaxel yield modest response rates, underscoring the urgent need for precision oncology approaches. Patient-derived organoids (PDOs) preserve the genomic, phenotypic, and histopathological features of the source tumor and offer a promising platform for drug screening, biomarker development, and personalized therapy. However, a systematic evaluation of their translational capacities is lacking. METHODS: …

Butyrate blocks cell cycle progression in colorectal cancer organoids partially through HDAC2 inhibition.

INTRODUCTION: Butyric acid is a major gut microbiota metabolite that exhibits many biological functions, including the suppression of colorectal cancer (CRC) growth. However, previous studies have mainly been conducted using cell lines, which do not recapitulate the genuine three-dimensional spatial characteristics of CRC. In this study, we explored the therapeutic outcome and precise molecular targets of butyrate with sodium butyrate (NaB) using tumor organoids of CRC. METHODS: Firstly, we examined the influence of butyrate on CRC subcutaneous allografts. And then the biological effects of b…

High-content screening of organoids reveals the mechanisms of human pancreas acinar specification.

Organoids derived from pluripotent stem cells have emerged as powerful models to study human development. To investigate signaling pathways regulating human pancreas differentiation and morphogenesis, we developed a high-content, image-based screen and quantitative multivariate analysis pipelines robust to heterogeneity to extract single-cell and organoid features using pancreatic progenitor organoids. Here, we identified 54 compounds affecting cell identity and/or morphological landscape. Focusing on one family of compounds, we found that glycogen synthase kinase 3α/β (GSK3A/B) inhibition via…

Bovine endometrial organoids: a new tool to study conceptus-maternal interactions in mammals†.

We developed a hormonally responsive organoid model of the bovine endometrium. Bovine glandular epithelial cells isolated from reproductive tracts were cultured in an extracellular matrix hydrogel (Cultrex 2) at 38.5°C, 5% CO2 (n = 3). RNA was extracted and qPCR confirmed the presence of gland markers: leukemia inhibitory factor, mucin-1, insulin-like growth factor binding protein-1, kruppel-like factor-5, and forkhead box protein-A2. Organoids were imaged at specific time-points to monitor growth. Morphologically, organoids were spherical and fast-growing at passages 0 and 1, but this decline…

Comparative Drug Response Profiling in Neuroblastoma Cell Lines and Patient-Derived Tumor Organoids.

High-risk neuroblastoma remains a leading cause of pediatric cancer mortality, and improved preclinical models are needed to guide therapeutic developments. We screened seven high-risk neuroblastoma cell lines and three patient-derived tumor organoids with 528 compounds alongside bone marrow controls, and compared them with external datasets. Unsupervised clustering of drug response clearly separated two-dimensional (2D) and three-dimensional (3D) models, with 3D models showing more coherent responses and overall higher drug sensitivity. Mitotic and DNA-damage pathways emerged as key vulnerabi…

[The application of breast cancer organoids in drug screening and gene function research].

Organoids refer to cell clusters with certain spatial structures formed by human adult stem cells or pluripotent stem cells through three-dimensional culture in vitro, which can be stably passaged. Organoids can simulate real organs in both structure and function, and to the greatest extent reproduce the structure and function of living organs or tissues. They have three-dimensional structural features, retain cellular heterogeneity, and can self-renew and self-organize. In the field of breast cancer research, breast cancer organoid models can faithfully reproduce the heterogeneity, tissue str…
