Jiedu Quyu Ziyin prescription alleviated renal injury in MRL/lpr by inhibiting PPP-mediated oxidative stress through regulating p53/G6PD/NOX pathway.

ETHNOPHARMACOLOGICAL RELEVANCE: Jiedu Quyu Ziyin prescription (JP), derived from classical Chinese medicinal formulations (Shengma Biejia Tang), alleviates systemic lupus erythematosus (SLE) progression by modulating immunological derangements. The underlying mechanism by which JP regulates metabolism to retard SLE progression has not yet been fully elucidated. AIM OF THE STUDY: This study aims to investigate whether JP attenuates renal injury in MRL/lpr lupus-prone mice by suppressing hyperactivation of the pentose phosphate pathway (PPP) and its mediated oxidative stress. MATERIALS AND MET

Dysregulated metabolism underpins Zika-virus-infection-associated impairment in fetal development.

Zika virus (ZIKV) is an Aedes-mosquito-borne flavivirus that causes debilitating congenital and developmental disorders. Improved understanding of ZIKV pathogenesis could assist efforts to fill the therapeutic and vaccine gap. We use several ZIKV strains, including a pair differing by a single phenylalanine-to-leucine substitution (M-F37L) in the membrane (M) protein, coupled with unbiased genomics to demarcate the border between attenuated and pathogenic infection. We identify infection-induced metabolic dysregulation as a minimal set of host alterations that differentiates attenuated from pa

A comparative transcriptome analysis of the novel obligate methanotroph Methylomonas sp. DH-1 reveals key differences in transcriptional responses in C1 and secondary metabolite pathways during growth on methane and methanol.

BACKGROUND: Methanotrophs play an important role in biotechnological applications, with their ability to utilize single carbon (C1) feedstock such as methane and methanol to produce a range of high-value compounds. A newly isolated obligate methanotroph strain, Methylomonas sp. DH-1, became a platform strain for biotechnological applications because it has proven capable of producing chemicals, fuels, and secondary metabolites from methane and methanol. In this study, transcriptome analysis with RNA-seq was used to investigate the transcriptional change of Methylomonas sp. DH-1 on methane and

Nuclear magnetic resonance studies of sugar metabolism in the human infant lens.

We investigated sugar metabolism in lenses isolated from infants with stage-5 retinopathy of prematurity (ROP). The lenses were incubated in 13C-labeled glucose (35.5 mM) or galactose (30 mM) for 24 h at 37.5 degrees C. Six to 8 lenses were pooled for each experiment. 13C-nuclear magnetic resonance spectroscopy was then performed on perchloric acid extracts of these lenses. Active aldose metabolism was indicated by the production of lactate. However, neither sorbitol nor dulcitol was detected. On the other hand, an experimental aldose reductase inhibitor, sorbinil, caused a decrease in the hex

Region-specific gene expression in the epididymis of Yak.

Immature spermatozoa undergo series of events in the epididymis to acquire motility and fertilizing ability. These events are a direct result of exposure to, and interaction with, the luminal environment created by the epididymal epithelium. The three conventional regions of the epididymis namely; caput, corpus and cauda have been identified to play specific roles in the epididymal maturation process of the spermatozoa; their respective roles have been associated with specific gene expression patterns that account for the composition of the luminal fluid that bathe the spermatozoa as they tran

EMT-Derived Alterations in Glutamine Metabolism Sensitize Mesenchymal Breast Cells to mTOR Inhibition.

Epithelial-to-mesenchymal transition (EMT) is a fundamental developmental process with strong implications in cancer progression. Understanding the metabolic alterations associated with EMT may open new avenues of treatment and prevention. Here we used 13C carbon analogs of glucose and glutamine to examine differences in their utilization within central carbon and lipid metabolism following EMT in breast epithelial cell lines. We found that there are inherent differences in metabolic profiles before and after EMT. We observed EMT-dependent re-routing of the TCA-cycle, characterized by increase

Are both hemolysis and impaired myocyte integrity associated with glucose-6-phosphate dehydrogenase deficiency?

Glucose-6-phosphate dehydrogenase (G6PD) is a potent antioxidant enzyme. G6PD deficiency, predominant among males, is a neonatal "neurotoxic" risk, characterized by episodic hemolysis that is sometimes infection triggered. Clinical risk among heterozygous G6PD-deficient females is often under-recognized. Recent reports of rhabdomyolysis among G6PD-deficient individuals are reviewed here. In one case, a male athlete with G6PD deficiency (<10% activity) experienced multiple, acute episodes of rhabdomyolysis, painful muscle symptomatology, injury and hemolysis after strenuous exertion. Both his m

RuBP availability contributes to the regulation of RUBISCO activity in Synechocystis.

RIBULOSE-1,5-BISPHOSPHATE CARBOXYLASE/OXYGENASE (RUBISCO) and the Calvin-Benson-Bassham (CBB) cycle assimilate most of the inorganic carbon on earth. Conserved CP12 proteins control diurnal C-assimilation of the CBB cycle. Their function originated in cyanobacteria that deactivate GLYCERALDEHYDE 3-PHOSPHATE DEHYDROGENASE (GAPDH) and PHOSPHORIBULOKINASE (PRK) during darkness by redox-regulated complex-formation with CP12. This mechanism indicates metabolic regulation of C-assimilation. We measured C-assimilation in vivo at the day/night transition by dynamic 13CO2 labeling and 1-C-positional mo

Colonic metabolomic and transcriptomic alterations in a mouse model of metabolic syndrome.

Metabolic syndrome (MetS), characterized by abdominal obesity, insulin resistance, dyslipidemia, and hypertension, affects a substantial proportion of the global population and increases the risk for cardiovascular disease, diabetes, and metabolic dysfunction-associated steatotic liver disease (MASLD). Despite its prevalence, there are currently no effective pharmacological therapies targeting MetS, highlighting the need to identify novel etiological mechanisms, particularly within the gastrointestinal (GI) tract. Using a mouse model of MetS and healthy lean controls, we assessed the colonic m

Immunometabolic alterations in post-traumatic stress disorder.

Post-traumatic stress disorder (PTSD) has been linked to various alterations within the immune system, yet the metabolic programming of immune cells remains unexplored. In the current exploratory cross-sectional study, we interrogated immunometabolic function by applying cell-specific metabolic flow cytometry, serum biomarker profiling, and targeted gene expression analysis in peripheral blood mononuclear cells from patients with PTSD (n = 34) compared with healthy controls (n = 32). PTSD was associated with elevated serum IL-6 (p = 0.005) and higher glycolysis- and oxidative pentose phosphate

Integrative proteomics and metabolomics analysis of the mechanism of pancreatic β-cell dysfunction in aged mice.

OBJECTIVES: The prevalence of type 2 diabetes mellitus has increased worldwide and is higher among older individuals. Exploring the mechanisms underlying pancreatic β-cell dysfunction may help elucidate the pathogenesis of age-related diabetes. METHODS: Islet function-related parameters were measured in four young and four aged mice. Endogenous proteins and metabolites in the pancreas were detected using liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based proteomics and metabolomics, and integrated data analysis was performed. RESULTS: Compared with young mice, aged mice presente

Mechanistic insights into the efficient degradation and detoxification of chloramphenicol by Clostridium pasteurianum DSM 525.

Chloramphenicol (CAP), a chlorinated nitroaromatic antibiotic, posed significant ecological and human health risks due to its persistence and toxicity. Although microbial degradation represented a promising remediation strategy, its practical application was often constrained by slow degradation kinetics and incomplete biotransformation. In this study, rapid and efficient CAP degradation by the Gram-positive anaerobe Clostridium pasteurianum DSM 525 was demonstrated. The strain achieved 83.6 % removal of 400 mg L-1 CAP within 8 h, and the degradation kinetics followed a first-order model (R2 ≥

Integrative Multi-Omics Analysis Reveals Transcriptomic and Metabolic Remodeling Associated with Enhanced Peanut Nodulation Under Arbuscular Mycorrhizal Fungal Inoculation and Calcium Application.

Peanut (Arachis hypogaea L.) yield depends on biological nitrogen fixation, but the molecular mechanisms underlying the combined effects of arbuscular mycorrhizal fungi (AMF) and calcium fertilizer on nodulation remain unclear. Here, we used integrated transcriptomic and metabolomic analyses to investigate potential mechanisms in peanut roots. Compared with the non-inoculated control, AMF inoculation alone was associated with a 22.1% higher nodule number per plant. The combined application of AMF and CaO showed a 35.9% higher nodulation than AMF alone, and a 30.9% higher AMF colonization rate

Decoding the core microbiota during liquor fermentation of sauce-aroma baijiu: Advances in the characteristics of core microbial communities and their regulatory strategies.

Sauce-aroma Baijiu, a traditional Chinese solid-state fermented liquor, owes its unique quality to the synergistic metabolism of core microbiota (bacteria, yeasts, molds) during stacking and cellar fermentation. However, previous studies have predominantly focused on high-temperature Daqu, leaving the core microbiota in fermented grains insufficiently characterized, particularly regarding their succession dynamics, functional attributes, and targeted regulatory approaches. Additionally, the complex interspecific interactions and environmental adaptation mechanisms of core microbiota remain unc

Fenofibrate ameliorates salt-sensitive hypertension by improving renal metabolic homeostasis.

Salt-sensitive hypertension (SSH) presents a formidable clinical challenge, given its intricate pathophysiology and a scarcity of targeted treatment options. The present study investigates the therapeutic potential and underlying mechanisms of fenofibrate, a peroxisome proliferator-activated receptor α (PPARα) activator, in SSH. Clinical observations indicate that SSH in male patients is often accompanied by dyslipidemia, with a positive correlation observed between blood pressure and lipid profiles-particularly triglycerides. In male Dahl salt-sensitive (SS) rats, a four-week oral administrat

Autonomous Redox Rhythms and Their Coupling to the Circadian Clock.

Circadian rhythms have traditionally been considered to be outputs of genetic transcription-translation feedback loop (TTFL) oscillators. However, the discovery of circadian peroxiredoxin oxidation rhythms in anucleate red blood cells suggests that cells also possess an autonomous circadian redox rhythm. The conservation of this rhythm across all lineages of life, as well as in TTFL-defective backgrounds in multiple organisms, further indicates its importance. Rather than functioning as an isolated system, the redox rhythm is bidirectionally coupled to the genetic circadian clock. Perturbation

Integrated Physiological and Transcriptomic Analyses of Saccharomyces cerevisiae Under Syringaldehyde Stress.

Syringaldehyde is a major lignin-derived inhibitor in lignocellulosic hydrolysates that affects growth, metabolism, and ethanol fermentation in Saccharomyces cerevisiae; however, the mechanisms underlying its toxicity and cellular adaptation remain poorly understood. In this study, fermentation analysis, physiological characterization, and transcriptomic profiling were integrated to investigate the response of S. cerevisiae to syringaldehyde stress. Syringaldehyde inhibited yeast growth and ethanol fermentation in a concentration-dependent manner. At 1.4 g/L, it caused only a minor decrease in
