[Association between anti-phospholipid antibodies and thrombotic complications in systemic lupus erythematosus].

A number of previous studies have shown that anti-phospholipid(aPL) antibodies(Abs) do not bind primarily to the negatively-charged phospholipid itself but rather to complexes of the phospholipid and plasma proteins, and that the most common antigenic targets are beta 2-glycoprotein I recognized by anticardiolipin Abs and prothrombin recognized by most lupus anticoagulants. However, resent studies suggest that other phospholipid-binding proteins, particularly protein C, protein S, and annexin V, may be important targets as well. To clarify the association between the various types of aPL Abs a

Preparation by spray drying of amphotericin B-phospholipid composite particles and their anticellular activity.

As a potent drug carrier for systemic fungal infections, amphotericin B(AmB)-phospholipid composite particles (APCPs) were prepared by the spray drying method. AmB and egg phosphatidylcholine, co-dissolved in methanol (0.0425-0.45 mg AmB/ml, 0.17-1.8 mg lipid/ml), was nebulized at 2 ml/min. The aerosol produced was carried by air at 1000 ml/min to the inner tubes of a serially connected distilling column system, of which the outer tubes were supplied with circulating water of 95 degrees C. The particles, by scanning electron microphotography, are spherical and submicronsized. Upon hydration of

Effects of oxidative stress on phospholipid signaling in rat cultured astrocytes and brain slices.

Although reactive oxygen species (ROS) are conventionally viewed as toxic by-products of cellular metabolism, a growing body of evidence suggests that they may act as signaling molecules. We have studied the effects of hydrogen peroxide (H(2)O(2))-induced oxidative stress on phospholipid signaling in cultured rat cortical astrocytes. H(2)O(2) stimulated the formation of phosphatidic acid and the accumulation of phosphatidylbutanol, a product of the phospholipase D (PLD)-catalyzed transphosphatidylation reaction. The effect of exogenous H(2)O(2) on the PLD response was mimicked by menadione-ind

Effects of tirilazad mesylate on vasospasm and phospholipid hydroperoxides in a primate model of subarachnoid hemorrhage.

BACKGROUND AND PURPOSE: Tirilazad mesylate has been used in the attempt to prevent cerebral vasospasm after subarachnoid hemorrhage (SAH), although the actual targets of this agent in vivo have thus far been controversial. Chemiluminescence/high-performance liquid chromatography provided a new method for direct measurements of phosphatidylcholine hydroperoxide (PCOOH) and phosphatidylethanolamine hydroperoxide (PEOOH) in vivo and showed that phosphatidylcholine is the lipid class most susceptible to lipid peroxidation. In the present study we measured those levels in a primate model of SAH for

Deletion of central alpha-helices in human apolipoprotein A-I: effect on phospholipid association.

In order to better understand the structure-function properties of apolipoprotein (apo) A-I, we have constructed and expressed three apoA-I … at the amphipathic alpha-helices within residues 100-186 are directly involved in interactions with phospholipids. The helical region 100-121 appears to be more important to the stabilization of the lipid-apoprotein complex formed whereas helices within residues 122-186 appear to be critical to the initial rates of association of the apoprotein with DMPC. These …

Intrahepatic biliary cholesterol and phospholipid transport in humans: effect of obesity and cholesterol cholelithiasis.

The mode of transport of biliary lipids within the hepatocyte and the role of the bile canalicular membrane (BCM) in biliary lipid secretion are not well understood. We hypothesized that biliary cholesterol and phospholipid are co-transported across the hepatocyte in vesicular form from the endoplasmic reticulum to the bile across the BCM. We obtained wedge liver biopsies and fasting gallbladder bile from 15 cholesterol gallstone patients and 10 control subjects. BCM, basolateral membrane (BLM), and many microsomal vesicular fractions were isolated by centrifugation. One of the vesicular fract

The effect of Ambroxol on bleomycin-induced changes in phospholipid composition of rat lung surfactant.

The influence of Ambroxol on bleomycin-induced changes in lung surfactant phospholipid composition in rats was examined. Bleomycin alone (intratracheally 10 mg/kg, single dose) produces a significant fall in phosphatidylcholine (especially in disaturated species) and phosphatidylglicerol content in both lavage and postlavaged lung tissues surfactants. Ambroxol (8 days, 2 mg/kg s.c. per day) normalizes these changes. Thus, Ambroxol has an salutary effect on bleomycin-induced changes in lung surfactant phospholipids.

The effects of pH on the interaction of anthrax toxin lethal and edema factors with phospholipid vesicles.

Bacillus anthracis secretes three distinct proteins which interact in binary combinations to produce two toxins. The two effector moieties, edema factor (EF) and lethal factor (LF), interact competitively with the cell receptor-binding moiety, protective antigen (PA), to produce biologically distinct effects. The passage of the toxins through an acidified endosomal compartment is an essential step in the intoxication process, and it has been shown that low pH triggers the insertion of the activated form of PA, PA63, into model lipid bilayers. In this study, we have examined the effects of pH o

Biodistribution, metabolism, and excretion of radioiodinated phospholipid ether analogs in tumor-bearing rats.

The phospholipid ether analog, [125I]-1-O-[12-(m-iodophenyl)dodecyl]propanediol-3-phosphocholine (NM-295) was synthesized and evaluated for its ability to visualize tumors. Preliminary studies were performed in rats bearing the Walker 256 carcinosarcoma. Most of the radioactivity was cleared from the animals during the first 24 hours. However, the tumor showed a decreased rate of clearance of radioactivity when compared with non-target tissue. This difference in the clearance rate allowed for excellent images of the tumor at 24 hours. Scintigraphic images compared favorably with other radioiod

Anti-phospholipid antibodies bind to platelet microparticles in idiopathic (autoimmune) thrombocytopenic purpura.

We recently reported that IgM antibody-related microparticles exist in some patients with idiopathic thrombocytopenic purpura (ITP) [14]. In this study, we investigated the relationship between antiphospholipid (cardiolipin and phosphatidylinositol) antibodies and microparticles in 56 ITP patients. We used an ELISA to detect anti-phospholipid antibodies. IgG antibodies against cardiolipin and phosphatidylinositol were detected in 13 and 12 patients, respectively. The titers of IgG antibodies against these phospholipids did not correlate with the platelet-associated IgG level or the platelet co…

Phospholipid metabolism and intracellular Ca2+ homeostasis in cultured rat hepatocytes intoxicated with cyanide.

The killing of cultured hepatocytes by 1 mM sodium cyanide was reduced by 100 microM chlorpromazine or cytochalasin B (25 micrograms/ml) or by lowering the pH of the culture medium to 6.0. In each case, ATP was depleted despite the decreased number of dead cells. The cell killing by cyanide was accompanied by an accelerated release of 3H-labeled arachidonate from phospholipids. Depletion of ATP by oligomycin did not accelerate phospholipid degradation or kill the hepatocytes. Chlorpromazine, cytochalasin B, and extracellular acidosis reduced the rate of phospholipid degradation in control cell…

Channels formed in phospholipid bilayer membranes by diphtheria, tetanus, botulinum and anthrax toxin.

Diphtheria, tetanus, botulinum, and anthrax toxin are multipartate toxins, one of the domains of which is (or is presumed to be) an enzyme. Cell intoxication requires that the enzymatic portion gain access to the cytosol via endocytosis into an acidic vesicle compartment of the cell. Translocation of the enzyme across the vesicular membrane is dependent on the low pH of the vesicle and involves another domain of the toxin; for each of these toxins, that domain is capable of forming channels in phospholipid bilayer membranes. These channels are large (greater than 12 A diameter) and voltage-gat

Anthrax toxin: channel-forming activity of protective antigen in planar phospholipid bilayers.

The three separate proteins that make up anthrax toxin--protective antigen (PA), edema factor (EF), and lethal factor (LF)--act in binary combinations to produce two distinct reactions in experimental animals: edema (PA + EF) and death (PA + LF). PA is believed to interact with a membrane receptor, and after proteolytic processing, to mediate endocytosis and subsequent translocation of EF or LF into the cytosol. PA can be separated, after mild trypsinolysis, into two fragments, PA65 (65 kDa) and PA20 (20 kDa). We demonstrate that trypsin-cleaved PA is capable of forming cation-selective channe

Presence of phospholipidic lamellar bodies on the mucosa of rabbit eustachian tube. Ultrastructural aspects.

Electron microscopy of the rabbit eustachian tube showed the presence of osmiophilic lamellar bodies in the cytoplasm of secretory cells and of osmiophilic free lamellar membranes in the lumen of tubal glands or above the epithelium. Similar structures have been reported in other parts of the respiratory system and in the pulmonary alveoli. The functional significance of these morphological observations is suggestive of a surface-tension-lowering effect on the mucosal lining layer of the eustachian tube and of a possible relationship to the phospholipid surfactant of the lung.

Proton/hydroxide conductance through phospholipid bilayer membranes: effects of phytanic acid.

Mechanisms of proton/hydroxide conductance (GH/OH) were investigated in planar (Mueller-Rudin) bilayer membranes made from decane solutions of phospholipids or phospholipids plus phytanic acid (a 20-carbon, branched chain fatty acid). At neutral pH, membranes made from diphytanoylphosphatidylcholine or bacterial phosphatidylethanolamine had GH/OH values in the range of (2-5) X 10(-9) S X cm-2, corresponding to H+/OH- 'net' permeabilities of about (0.4-1.0) X 10(-5) cm X s-1. GH/OH was inhibited by serum albumin, phloretin, glycerol and low pH, but was increased by chlorodecane and voltage grea…

The use of phospholipid liposomes for lithium administration. Polydipsic effect and tissue distribution of lithium.

Administration of lithium entrapped in phospholipid liposomes increased the lithium-induced polydipsia, but did not accelerate the onset of this effect. It also resulted in a larger accumulation of lithium in the liver, kidney and spleen, but not in the brain. The time course of polydipsia suggests that it depends on the intracellular lithium concentration. However, the rate of development of this effect depends on some additional factor.
