Optimization of Exosome Isolation from Vitis vinifera Cell Cultures.

Plant extracellular vesicles (exosomes) are considered promising nanostructures for biomedical and biotechnological applications; however, methods for their isolation from plant cell cultures remain insufficiently optimized. The aim of this study was to evaluate the efficiency of exosome isolation from callus and suspension cultures of Vitis vinifera and to investigate the effects of culture age and extraction method on the yield of extracellular vesicles. It was demonstrated that suspension culture provides a higher exosome yield compared to callus tissue. Isolated particles were characterize

Nitric Oxide: Its Generation and Interactions with Other Reactive Signaling Compounds.

Nitric oxide (NO) is an immensely important signaling molecule in animals and plants. It is involved in plant reproduction, development, key physiological responses such as stomatal closure, and cell death. One of the controversies of NO metabolism in plants is the identification of enzymatic sources. Although there is little doubt that nitrate reductase (NR) is involved, the identification of a nitric oxide synthase (NOS)-like enzyme remains elusive, and it is becoming increasingly clear that such a protein does not exist in higher plants, even though homologues have been found in algae. Down

Salicylic acid regulates Plasmodesmata closure during innate immune responses in Arabidopsis.

In plants, mounting an effective innate immune strategy against microbial pathogens involves triggering local cell death within infected cells as well as boosting the immunity of the uninfected neighboring and systemically located cells. Although not much is known about this, it is evident that well-coordinated cell-cell signaling is critical in this process to confine infection to local tissue while allowing for the spread of systemic immune signals throughout the whole plant. In support of this notion, direct cell-to-cell communication was recently found to play a crucial role in plant defen

RGD-functionalized Wolffia globosa-derived Exosome-like Nanoparticles for Targeted Doxorubicin Delivery in Breast Cancer Therapy.

INTRODUCTION: Plant-derived exosome-like nanoparticles (ELNs) have attracted considerable attention as biocompatible drug delivery vehicles. Wolffia globosa (Kai-pum), a native Thai aquatic plant, represents a novel and underexplored source of ELNs. This study describes the isolation, characterization, and functional evaluation of W. globosa-derived exosome-like nanoparticles (WGENs) that are surface-modified with the integrin-targeting Arg-Gly-Asp (RGD) peptide and loaded with doxorubicin (Dox) for targeted breast cancer therapy. METHODS: WGENs were isolated by differential ultracentrifugati

Mobilization of lipids and fortification of cell wall and cuticle are important in host defense against Hessian fly.

BACKGROUND: Wheat - Hessian fly interaction follows a typical gene-for-gene model. Hessian fly larvae die in wheat plants carrying an effective resistance gene, or thrive in susceptible plants that carry no effective resistance gene. RESULTS: Gene sets affected by Hessian fly attack in resistant plants were found to be very different from those in susceptible plants. Differential expression of gene sets was associated with differential accumulation of intermediates in defense pathways. Our results indicated that resources were rapidly mobilized in resistant plants for defense, including exten

Multiple Pathways Revealing the CeO2 Nanoparticle-Biostimulant-Based "Stress Training" Strategy for Enhanced Medicago sativa L. Antiviral Capability.

"Nanophytovirology" is a potential management approach to combat plant viral diseases. Herein, the impact of foliar application of CeO2 nanoparticles (CeO2-NPs) on the growth, plant cell ultrastructure, and physiology of alfalfa mosaic virus (AMV)-infected alfalfa seedlings was explored at different concentrations and application strategies. In a solar greenhouse, 20 mL of CeO2-NP suspensions of 50, 100, 200, and 500 mg/L was sprayed onto the plant surface every other day for 9 days. As a result, the total volume applied per pot was 100 mL. It showed that the beneficial effects of CeO2-NPs on

TurboID-based proteomic profiling reveals proxitome of the IRT1 metal transporter and new insight into IRT1 endocytosis.

Iron Regulated Transporter1 (IRT1) is the major root iron transporter responsible for broad-spectrum metal absorption in Arabidopsis thaliana root epidermal cells. Non-iron metal substrates of IRT1 were recently shown to regulate IRT1 cell surface levels by endocytosis and vacuolar degradation. TurboID-based proximity labeling was recently developed to detect protein-protein interactions and thus shed light on the intricate regulation of proteins in living cells. Although TurboID-based proximity labeling technology has been successfully established in mammals, its application in plant systems

Synchronous Induction and Microscopic Analysis of Hyphal Growth Using Laboratory Strain AB33.

In nature, hyphal growth of Ustilago maydis is strictly coupled to plant infection and initiated after mating of two genetically compatible yeast cells on the plant surface. Compatibility is defined by mating type loci: the biallelic a locus mediates recognition and fusion through a pheromone/receptor system, whereas the multiallelic b locus, encoding transcription factor subunits bEast (bE) and bWest (bW), establishes and maintains the dikaryotic hypha controlled via the assembly of compatible subunits into active heterodimers after plasmogamy. The laboratory strain AB33 enables a controlled

Development of host strains and vector system for an efficient genetic transformation of filamentous fungi.

An ability to synthesize extracellular enzymes degrading a wide spectrum of plant and algae polymeric substrates makes many fungi relevant for biotechnology. The terrestrial thermophilic and marine fungal isolates capable of plant and algae degradation have been tested for antibiotic resistance for their possible use in a new genetic transformation system. Plasmids encoding the hygromycin B phosphotransferase (hph) under the control of the cauliflower mosaic virus 35S promoter, the trpC gene promoter of Aspergillus nidulans, and the Aureobasidium pullulans TEF gene promoter were delivered into

Contribution of glutathione to the control of cellular redox homeostasis under toxic metal and metalloid stress.

The accumulation of toxic metals and metalloids, such as cadmium (Cd), mercury (Hg), or arsenic (As), as a consequence of various anthropogenic activities, poses a serious threat to the environment and human health. The ability of plants to take up mineral nutrients from the soil can be exploited to develop phytoremediation technologies able to alleviate the negative impact of toxic elements in terrestrial ecosystems. However, we must select plant species or populations capable of tolerating exposure to hazardous elements. The tolerance of plant cells to toxic elements is highly dependent on g…

Soybean homologs of MPK4 negatively regulate defense responses and positively regulate growth and development.

Mitogen-activated protein kinase (MAPK) cascades play important roles in disease resistance in model plant species such as Arabidopsis (Arabidopsis thaliana) and tobacco (Nicotiana tabacum). However, the importance of MAPK signaling pathways in the disease resistance of crops is still largely uninvestigated. To better understand the role of MAPK signaling pathways in disease resistance in soybean (Glycine max), 13, nine, and 10 genes encoding distinct MAPKs, MAPKKs, and MAPKKKs, respectively, were silenced using virus-induced gene silencing mediated by Bean pod mottle virus. Among the plants s

Plant water transport and aquaporins in oxygen-deprived environments.

Oxygen deprivation commonly affects plants exposed to flooding and soil compaction. The resulting root hypoxia has an immediate effect on plant water relations and upsets water balance. Hypoxia inhibits root water transport and triggers stomatal closure. The processes contributing to the inhibition of root hydraulic conductivity and conductance (hydraulic conductivity of the whole root system) are complex and involve changes in root morphology and the functions of aquaporins. Aquaporins (AQPs) comprise a group of membrane intrinsic proteins that are responsible for the transport of water, as w

Metabolism meets transport: a new perspective on auxin homeostasis at subcellular level.

This review highlights recent advances revealing auxin homeostasis as an integrated network of metabolism and transport, emphasizing subcell … t knowledge on IAA metabolism and places it into the broader framework of auxin distribution within plant cells. The spatial distribution of auxin is tightly linked to its metabolic regulation, as localized auxin accumulation underlies many developmental and signaling processes. Subcellular compartmentalization of auxin into organelles such as the endoplasmic …

Ascorbic acid in plants: Biosynthetic regulation and biotechnology strategies for enhancing human health.

Ascorbic acid (AsA) is a crucial water-soluble antioxidant in plant cells, playing a central role not only in plant growth, development, and stress responses but also serving as an essential nutrient for human health. Since the human body cannot synthesize AsA independently, its intake primarily relies on fresh fruits and vegetables. This paper reviews, from a plant physiological perspective, how AsA accumulation is finely regulated by multiple processes including biosynthesis, transport, degradation, and regeneration cycles. It discusses how environmental signals trigger a series of changes i…

Insights into plant regeneration: cellular pathways and DNA methylation dynamics.

Plants, known for their immobility, employ various mechanisms against stress and damage. A prominent feature is the formation of callus tissue-a cellular growth phenomenon that remains insufficiently explored, despite its distinctive cellular plasticity compared to vertebrates. Callus formation involves dedifferentiated cells, with a subset attaining pluripotency. Calluses exhibit an extraordinary capacity to reinitiate cellular division and undergo structural transformations, generating de novo shoots and roots, thereby developing into regenerated plants-a testament to the heightened developm

Plant cystine-knot peptides: pharmacological perspectives.

Cystine-knot miniproteins are a class of 30-50 amino acid long peptides widespread in eukaryotic organisms. Due to their very peculiar three-dimensional structure, they exhibit high resistance to heat and peptidase attack. The cystine-knot peptides are well represented in several plant species including medicinal herbs and crops. The pharmacological interest in plant cystine-knot peptides derives from their broad biological activities, mainly cytotoxic, antimicrobial and peptidase inhibitory and in the possibility to engineer them to incorporate pharmacophoric information for oral delivery or

Thymus vulgaris (thyme) inhibits proliferation, adhesion, migration, and invasion of human colorectal cancer cells.

Colorectal cancer (CRC) remains one of the most common malignancies and a leading cause of cancer-related deaths. Its prognosis remains poor for patients with several grades of this disease. This underscores the need for alternative modalities, such as herbal medicines, to treat this disease. A commonly used plant that appears to be of high medicinal value is Thymus vulgaris L. However, the effects of this plant on the malignant behavior of human CRC cells remains poorly investigated. This study was undertaken to determine the anticancer efficacy of T. vulgaris extract (TVE) in CRC cells. Our
