Infection Dynamics and Host Responses to Two IPNV Isolates in Liver of Atlantic Salmon (Salmo salar).

The infectious pancreatic necrosis virus (IPNV) used to be one of the largest loss factors in Atlantic salmon farming. Since 2009, marker-assisted selection for resistance to IPN, targeting a single major quantitative trait locus (QTL), has led to a ten-fold decrease in the number of IPN outbreaks in Norway. However, some IPN-related problems remain, due to isolates of the virus which seem to bypass the resistance mechanism of the QTL. We comparatively characterized a classical isolate affected by the IPN-QTL (cIPNV) and an isolate that circumvents the QTL-based protection (rIPNV). Using both

Pathogenic genes associated with immune-related genes in graves' disease: a multi-omics Mendelian randomization analysis.

Immune-related genes (IRGs) have been implicated in autoimmune diseases, but their causal role in GD remains unclear. We conducted a multi-omics Mendelian randomization (MR) analysis integrating genome-wide association study (GWAS) data from GD patients with blood-based methylation quantitative trait loci(mQTLs), expression QTLs(eQTLs), and protein QTLs(pQTLs). SMR analysis was performed to assess the causal associations between IRGs and GD, followed by HEIDI tests to screen for pleiotropy. Colocalization analysis identified shared genetic determinants. We further validated the findings in Fin…

Genetic analysis of stress hormone levels in hair of healthy nursery pigs and their relationships with backtest responses.

This study explored the genetics of the levels of stress hormones (cortisol, cortisone, DHEA, and DHEA-S) in hair of 863 clinically healthy Yorkshire × Landrace male pigs at ∼40 days of age and evaluated their potential as biomarkers of innate stress response by estimating genetic correlations with responses to a 30 s backtest performed at ∼27 days of age. Backtest responses included the number and intensity of vocalizations (VN and VI) and struggles (SN and SI). With pigs genotyped using a 50 K single nucleotide polymorphism (SNP) panel that was imputed to 650 K SNPs, heritability estimates f

Mendelian randomization reveals causal circulatory microRNAs in prostate cancer pathogenesis and prognosis.

Prostate cancer (PCa) lacks precise diagnostic tools, necessitating noninvasive biomarkers. MicroRNAs (miRNAs) show promise due to fluid stability and oncogenic regulation. Using 2-sample Mendelian randomization (MR), we analyzed plasma miRNA expression quantitative trait loci (eQTLs) from discovery and validation cohorts. Cis-acting eQTLs served as instrumental variables, with causal effects assessed via inverse-variance weighted (IVW) and MR-Egger regression. Functional and survival analyses leveraged The Cancer Genome Atlas (TCGA)-PRAD data and bioinformatics tools. Five miRNAs (hsa-miR-127…

Genome-wide association meta-analysis identifies 29 new acne susceptibility loci.

Acne vulgaris is a highly heritable skin disorder that primarily impacts facial skin. Severely inflamed lesions may leave permanent scars that have been associated with long-term psychosocial consequences. Here, we perform a GWAS meta-analysis comprising 20,165 individuals with acne from nine independent European ancestry cohorts. We identify 29 novel genome-wide significant loci and replicate 14 of the 17 previously identified risk loci, bringing the total number of reported acne risk loci to 46. Using fine-mapping and eQTL colocalisation approaches, we identify putative causal genes at sever

Pleiotropy and epistasis within and between signaling pathways defines the genetic architecture of fungal virulence.

Cryptococcal disease is estimated to affect nearly a quarter of a million people annually. Environmental isolates of Cryptococcus deneoformans, which make up 15 to 30% of clinical infections in temperate climates such as Europe, vary in their pathogenicity, ranging from benign to hyper-virulent. Key traits that contribute to virulence, such as the production of the pigment melanin, an extracellular polysaccharide capsule, and the ability to grow at human body temperature have been identified, yet little is known about the genetic basis of variation in such traits. Here we investigate the genet

Hearing loss and tinnitus: association studies for complex-hearing disorders in mouse and man.

Genome-wide association studies (GWAS) provide an unbiased first look at genetic loci involved in aging and noise-induced sensorineural hearing loss and tinnitus. The hearing phenotype, whether audiogram-based or self-report, is regressed against genotyped information at representative single nucleotide polymorphisms (SNPs) across the genome. Findings include the fact that both hearing loss and tinnitus are polygenic disorders, with up to thousands of genes, each of effect size of < 0.02. Smaller human GWAS' were able to use objective measures and identified a few loci; however, hundreds of th

The DNA methylome of inflammatory bowel disease (IBD) reflects intrinsic and extrinsic factors in intestinal mucosal cells.

Abnormal DNA methylation has been described in human inflammatory conditions of the gastrointestinal tract, such as inflammatory bowel disease (IBD). As other complex diseases, IBD results from the balance between genetic predisposition and environmental exposures. As such, DNA methylation may be the consequence (and potential effector) of both, genetic susceptibility variants and/or environmental signals such as cytokine exposure. We attempted to discern between these two non-excluding possibilities by performing a combined analysis of published DNA methylation data in intestinal mucosal cell
