Development of a mycolic acid-graphene quantum dot probe as a potential tuberculosis biosensor.

The development of amine-functionalized graphene quantum dots (GQDs) linked to mycolic acids (MAs) as a potential fluorescent biosensor to detect tuberculosis (TB) biomarkers is described. GQDs have attractive properties: high fluorescence, excellent biocompatibility, good water solubility, and low toxicity. MAs are lipids that are found in the cell wall of Mycobacterium tuberculosis that are antigenic, however, they are soluble only in chloroform and hexane. Chloroform-soluble MAs were covalently linked to synthesized water-soluble GQDs using an amide connection to create a potential fluoresc…

Quantum dot clusters as self-assembled antennae with phycocyanine and phycobilisomes as energy acceptors.

In this study, we investigated an experimental and Monte-Carlo computational characterization of self-assembled antennae built using CdTe colloidal quantum dots (QDs). These clusters provide efficient excitation of phycocyanine (PC) or phycobilisomes (PBSs). PBSs are light-harvesting complexes (LHCs) of cyanobacteria, made of several PC units, organized in disks and rods. Each PC contains three separate cofactors. Therefore, we analyzed variations in multi-donor and multi-acceptor systems. The self-assembled QD clusters were formed mostly by electrostatic interactions, possibly due to the intr…

Toxicity of different types of quantum dots to mammalian cells in vitro: An update review.

Currently, there are a great quantity type of quantum dots (QDs) that has been developed by researchers. Depending on the core material, they can be roughly divided into cadmium, silver, indium, carbon and silicon QDs. And studies on the toxicity of QDs are also increasing rapidly, but in vivo tests in model animals fail to reach a consistent conclusion. Therefore, we review the literatures dealing with the cytotoxicity of QDs in mammalian cells in vitro. After a short summary of the application characteristics of five types of QDs, the fate of QDs in cells will be discussed, ranging from the …

Quantum dot encapsulated nanocolloidal bioconjugates function as bioprobes for in vitro intracellular imaging.

Bioimaging probes incorporating quantum dots (QDs) are important for identifying organelles and monitoring their movement/location in living cells. Organelle specificity can be accomplished by functionalizing probe surfaces with chemical groups that can react with antibodies capable of targeting specific organelle-protein epitopes. Here, such a bioprobe is generated by encapsulating ZnS-capped CdSe QDs within polystyrene (PS) nanocolloids via Pickering miniemulsion using laponite nanoclay platelets as solid-stabilizers. The surfaces of these platelets are modified with aminopropyltriethoxysila…

The role of NLRP3 inflammasome activation in the neuroinflammatory responses to Ag2Se quantum dots in microglia.

Silver selenide quantum dots (Ag2Se QDs) provide bright prospects for the application of QDs in the field of biomedicine because they contain low-toxic compounds and show great advantages in the imaging of deep tissues and tiny vascular structures. However, the biosafety of these novel QDs has not been thoroughly evaluated, especially in one main target for toxicity-the central nervous system (CNS). Our previous studies have suggested severe inflammatory responses to cadmium-containing QDs in the hippocampus, which gives us a hint regarding the risk assessment of Ag2Se QDs. In this study, micr…

Quantum dot induced acute changes in lung mechanics are mouse strain dependent.

INTRODUCTION: Concerns have been raised regarding occupational exposure to engineered nanomaterials (ENMs). Potential impacts on lung function from inhalation exposures are of concern as the lung is a sensitive ENM target in animals. Epidemiological data suggest that occupational exposure to ENMs may impact respiratory and cardiovascular health. Quantum dots (QDs) are ENMs with outstanding semiconductor and fluorescent properties with uses in biomedicine and electronics. QDs are known to induce inflammation and cytotoxicity in rodents and high dose exposures impact lung function 2 weeks after …

Surface biofunctionalized CdS and ZnS quantum dot nanoconjugates for nanomedicine and oncology: to be or not to be nanotoxic?

Herein, for the first time, we demonstrated that novel biofunctionalized semiconductor nanomaterials made of Cd-containing fluorescent quantum dot nanoconjugates with the surface capped by an aminopolysaccharide are not biologically safe for clinical applications. Conversely, the ZnS-based nanoconjugates proved to be noncytotoxic, considering all the parameters investigated. The results of in vitro cytotoxicity were remarkably dependent on the chemical composition of quantum dot (CdS or ZnS), the nature of the cell (human cancerous and embryonic types), and the concentration and time period of

Nucleo-mitochondrial interaction of yeast in response to cadmium sulfide quantum dot exposure.

Cell sensitivity to quantum dots (QDs) has been attributed to a cascade triggered by oxidative stress leading to apoptosis. The role and function of mitochondria in animal cells are well understood but little information is available on the complex genetic networks that regulate nucleo-mitochondrial interaction. The effect of CdS QD exposure in yeast Saccharomyces cerevisiae was assessed under conditions of limited lethality (<10%), using cell physiological and morphological endpoints. Whole-genomic array analysis and the screening of a deletion mutant library were also carried out. The result…

Quantum Dot and Polymer Composite Cross-Reactive Array for Chemical Vapor Detection.

A cross-reactive chemical sensing array was made from CdSe Quantum Dots (QDs) and five different organic polymers by inkjet printing to create segmented fluorescent composite regions on quartz substrates. The sensor array was challenged with exposures from two sets of analytes, including one set of 14 different functionalized benzenes and one set of 14 compounds related to security concerns, including the explosives trinitrotoluene (TNT) and ammonium nitrate. The array was broadly responsive to analytes with different chemical functionalities due to the multiple sensing mechanisms that altered…

A time-dependent DFT study of the absorption and fluorescence properties of graphene quantum dots.

Absorption and fluorescence spectra of graphene quantum dots (GQDs) have been computed by using time-dependent density functional theory (TDDFT). Different functionals, including PBE, TPSSh, B3LYP, PBE0, CAM-B3LYP, and LC-ωPBE, have been tested and B3LYP/6-31G(d) has been proven to be the most accurate method for our work. The bulk solvent effects of toluene and dichloromethane have been assessed by using the polarizable continuum model (PCM). The absorption wavelength of GQDs in solvents is red-shifted compared with that in the gas phase. Edge functionalization effects analysis shows that a s…

Genetic algorithm-guided discovery of additive combinations that direct quantum dot assembly.

The use of combinations of organic additives to control crystallization, as occurs in biomineralization, is rarely investigated due to the vast potential reaction space. It is demonstrated here that combinatorial approaches led by genetic algorithm heuristics can enable identification of active additive combinations, and four key organic molecules are rapidly identified, which generate highly fluorescent CdS quantum dot superstructures.

Fabrication of the optical fiber pH sensor based on CdSe/ZnS quantum dot.

During the past decade, there has been a lot of research on the application of quantum dots (QDs) in photonics and biotechnology. QDs are semiconductor nanocrystals which show many peculiar optical and electrical properties due to their quantum confinement effects. In this work, the correlation of the fluorescence intensity of QDs and the pH under ambient temperature was studied. An optical fiber pH sensor was prepared using CdSe/ZnS carboxyl quantum dots as sensing media. QDs with different sizes and emission colors (green, yellow, and red) were immobilized on the surface of the optical fiber…

CdTe quantum dot as a fluorescence probe for vitamin B(12) in dosage form.

We here report the CdTe quantum dot (CdTe QDs)-based sensor for probing vitamin B12 derivatives in aqueous solution. In this paper, simple and sensitive fluorescence quenching measurements has been employed. The Stern-Volmer constant (KSV), quenching rate constant (kq) and binding constant (K) were rationalized from fluorescence quenching measurement. Furthermore, the fluorescence resonance energy transfer (FRET) mechanism was discussed. This method was applicable over the concentration ranging from 1 to 14μg/mL (VB12) with correlation coefficient of 0.993. The limit of detection (LOD) of VB12

Improved thermal cycling durability and PCR compatibility of polymer coated quantum dot.

Quantum dots have experienced rapid development in imaging, labeling and sensing in medicine and life science. To be suitable for polymerase chain reaction (PCR) assay, we have tested QD thermal cycling durability and compatibility, which have not been addressed in previous reports. In this study, we synthesized CdSe/ZnS QDs with a surface modification with high-MW amphiphilic copolymers and observed that Mg²⁺ ions in the PCR reaction could induce the QDs to precipitate and reduce their fluorescence signal significantly after thermal cycling. To overcome this problem, we used mPEG2000 to conju…

Cysteamine CdS quantum dots decorated with Fe3+ as a fluorescence sensor for the detection of PPi.

A new sensitive and selective fluorescence sensor for the detection of pyrophosphate (PPi) in aqueous media based on the Fe(3+) decorated cysteamine CdS QDs ([Cys-CdS QDs]-Fe(3+)) was proposed. The presence of PPi can induce the fluorescence quenching of [Cys-CdS QDs]-Fe(3+) due to the high formation constants between the phosphate group and Fe(3+). Because the complex between Fe(3+) and PPi acts as an efficient quencher, the concentration of PPi can be evaluated by tracking the degree of fluorescence quenching. The fabricated sensor was optimized to obtain the best sensor selectivity and sens

Dynamic energy budget approach to modeling mechanisms of CdSe quantum dot toxicity.

A mechanistic model of bacterial growth based on dynamic energy budget (DEB) theory is utilized to investigate mechanisms of toxicity of CdSe quantum dots (QDs). The model of QD toxicity is developed by extending a previously published DEB model of cadmium ion toxicity to include a separate model of QD toxic action. The extension allows for testing whether toxicity from QD exposure can be explained fully by dissolved cadmium exposure only, or if the separate effects of QDs need to be taken into account as well. Two major classes of QD toxicity mechanisms are considered: acclimation expressed t…

Degradation products from consumer nanocomposites: a case study on quantum dot lighting.

Most nanomaterials enter the natural environment as nanoenabled products, which are typically composites with primary nanoparticles bound on substrates or embedded in liquid or solid matrices. The environmental risks associated with these products are expected to differ from those associated with the as-produced particles. This article presents a case study on the end-of-life emission of a commercial prototype polymer/quantum-dot (QD) composite used in solid-state lighting for homes. We report the extent of cadmium release upon exposure to a series of environmental and biological simulant flui

Evaluation of all-inorganic CdSe quantum dot thin films for optoelectronic applications.

Exchanging the original organic ligands of colloidal CdSe quantum dots (QDs) with inorganic metal chalcogenide SnS(4) ligands resulted in absorption peak redshifts and complete photoluminescence (PL) quenching in QD solids. The SnS(4)-capped QDs, meanwhile, were able to retain strong excitonic absorption. After the ligand exchange, the ITO/QDs/Al structure showed much higher electrical conductivity and reduced space-charge limited current. Its photocurrent spectral response increased by over two orders of magnitude and closely resembled the absorption spectrum of the QDs. However, it was found…

Transparent conducting films of CdSe(ZnS) core(shell) quantum dot xerogels.

A method of fabricating sol-gel quantum dot (QD) films is demonstrated, and their optical, structural and electrical properties are evaluated. The CdSe(ZnS) xerogel films remain quantum confined, yet are highly conductive (10(-3) S cm(-1)). This approach provides a pathway for the exploitation of QD gels in optoelectronic applications.

Selective and absolute quantification of endogenous hypochlorous acid with quantum-dot conjugated microbeads.

Endogenous hypochlorous acid (HOCl) secreted by leukocytes plays a critical role in both the immune defense of mammalians and the pathogenesis of various diseases intimately related to inflammation. We report the first selective and absolute quantification of endogenous HOCl produced by leukocytes in vitro and in vivo with a novel quantum dot-based sensor. An activated human neutrophil secreted 6.5 ± 0.9 × 10(8) HOCl molecules into its phagosome, and kinetic measurement for the secretions showed that the extracellular generation of HOCl was temporally retarded, but the quantity eventually atta
