An improved pseudotargeted GC-MS/MS-based metabolomics method and its application in radiation-induced hepatic injury in a rat model.

The liver is the pivotal metabolic organ primarily responsible for metabolic activities, detoxification and regulation of carbohydrate, protein, amino acid, and lipid metabolism. However, very little is known about the complicated pathophysiologic mechanisms of liver injury result from ionizing radiation exposure. Therefore, a pseudotargeted metabolomics approach based on gas chromatography-tandem mass spectrometry with selected reaction monitoring (GC-MS-SRM) was developed to study metabolic alterations of liver tissues in radiation-induced hepatic injury. The pseudotargeted GC-MS-SRM method

Topical Deferoxamine Alleviates Skin Injury and Normalizes Atomic Force Microscopy Patterns Following Radiation in a Murine Breast Reconstruction Model.

BACKGROUND: Breast cancer is most commonly managed with a combination of tumor ablation, radiation, and/or chemotherapy. Despite the oncologic benefit of these treatments, the detrimental effect of radiation on surrounding tissue challenges the attainment of ideal breast reconstruction outcomes. The purpose of this study was to determine the ability of topical deferoxamine (DFO) to reduce cutaneous ulceration and collagen disorganization following radiotherapy in a murine model of expander-based breast reconstruction. METHODS: Female Sprague-Dawley rats (n = 15) were divided into 3 groups: co

Heparin-binding EGF-like growth factor (HB-EGF) protects the intestines from radiation therapy-induced intestinal injury.

PURPOSE: Radiation therapy (RT) often induces enteritis by inhibiting proliferation and inducing apoptosis. Heparin-binding EGF-like growth factor (HB-EGF) has been shown to protect the intestine in several animal injury models. The objective of this study was to examine whether HB-EGF affects RT-induced intestinal injury. METHODS: HB-EGF or PBS was administered intraperitoneally to mice daily for 3 days, followed by total body irradiation (TBI). Three days after TBI, intestinal segments were harvested, and BrdU immunohistochemistry was performed to identify proliferating crypts (n=25). Four

Radiation injury of the parotid glands during treatment for head and neck cancer: assessment using dynamic contrast-enhanced MR imaging.

The parotid gland is an important organ at risk of complications of radiotherapy for head and neck cancer. In this study, we examined the potential of dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) for assessment of radiation injury to the parotid glands. DCE-MRI was performed before and 3 months after radiotherapy in patients treated for head and neck cancer. DCE-MRI was analyzed using the pharmacokinetic model proposed by Tofts and Kermode to produce three DCE parameters: k(trans), v(e) and v(p). These parameters were correlated with the dose of radiation delivered to the par

In Situ-Forming Bioresponsive Liposome-Anchored Hydrogel Restores Redox-Immune Homeostasis in Mucosal Macrophages for Radiation-Induced Rectal Injury Repair.

Radiation-induced rectal injury (RRI) is a prevalent complication of pelvic radiotherapy that severely affects the life quality of patients, but its effective treatment remains a major challenge in the clinic. Herein, we identify that the redox-immune dyshomeostasis of mucosal macrophages as a major pathological driver of RRI-associated acute inflammation and chronic fibrosis and developed an in situ forming reactive oxygen species (ROS)-responsive liposome-anchored hydrogel (Lip@H151@TK-PF127 Gel) for effective RRI repair. For this purpose, liposomes were further grafted with mannose moieties

Gut Microbiota-Derived Tyrosol Alleviates Radiation-Induced Intestinal Injury via Targeting SCD1-MUFA Axis to Suppress ER Stress.

Radiation-induced intestinal injury (RIII) represents a major, clinically recalcitrant complication of radiotherapy, with current protective options remaining extremely limited. In this study, we identify tyrosol, a gut-derived phenolic metabolite enriched in the feces of irradiated mice, as a potent radioprotective agent. It reduced intestinal epithelial cell death and improved survival in lethally irradiated mice by preserving mucosal barrier and villus-crypt architecture, and downregulating pro-inflammatory cytokines. Mechanistically, we for the first time reveal that tyrosol directly targe

Exosomes derived from bone marrow mesenchymal stem cells facilitate repair of radiation-Induced skin injury by attenuating inflammation and apoptosis.

Radiation-induced skin injury (RISI) refers to injury to the skin resulting from exposure to ionizing radiation, for which current treatment options are limited. Exosomes derived from bone marrow mesenchymal stem cells (BMSCs-Exos) have demonstrated significant potential in tissue repair. This study evaluated the healing efficacy of BMSCs‑Exos in a rat model of RISI. BMSCs-Exos were isolated and delivered via subcutaneous injection into the RISI. Our findings revealed that BMSCs-Exos reduced the wound area and lowered the radiation injury score, thereby indicating their capacity to facilitate

LAT1-mediated delivery of engineered R13A-MOTS-c attenuates radiation-induced lung injury via Nrf2 activation and mitochondrial protection.

MOTS-c exhibits substantial antioxidant and anti-inflammatory properties, yet its therapeutic potential is constrained by poor membrane permeability due to its high polarity. To overcome this limitation, we engineered R13A-MOTS-c by substituting the polar arginine at position 13 with alanine in the wild-type peptide (Met-Arg-Trp-Gln-Glu-Met-Gly-Tyr-Ile-Phe-Tyr-Pro-Arg-Lys-Leu-Arg). This modification increased the peptide's hydrophobicity index from -0.938 to -0.544, measurably improving its cellular uptake. Functional uptake assays, including competition with canonical LAT1 substrates (leucine

Mitigation of Radiation-induced Acute Hematopoietic System and Intestine Injury by Resveratrol-loaded Polymeric Nanoparticles after Whole Body Irradiation in Mice.

INTRODUCTION: Radiation-induced damage to the hematopoietic and gastrointestinal systems, especially the intestine, is a major concern for individuals exposed to whole-body radiation during an accident. Resveratrol has shown potential in mitigating radiation-induced toxicity, but its efficacy may be limited by its low bioavailability. In this study, we aimed to evaluate the effectiveness of resveratrol-loaded polymeric-based nanocapsules in mitigating radiation-induced injury in the hematopoietic system and intestine after whole-body exposure to radiation. METHODS: Sixty male mice were random

Maresin-1 alleviates lipid peroxidation-induced ferroptosis after radiation-induced brain injury in mice through the RORα/NRF2 pathway.

Ferroptosis plays a critical role in radiation-induced brain injury (RIBI). The role of Maresin-1, which has anti-inflammatory and antiferroptotic properties, in RIBI is still unclear. This study aimed to explore the effects and mechanisms of Maresin-1 on ferroptosis after RIBI in mice. A mouse model of RIBI was constructed through whole-brain irradiation. Short-term neurological functions were evaluated by the modified Garcia score and the beam balance score, and long-term neurological functions were evaluated by the Morris water maze and the rotarod test. Sex Differences in Urinary Metabolite Profiles between Survivors and Non-Survivors of Radiation-induced Lung Injury in the C57L/J Murine Model.

Novel biodosimetry assays are needed to categorize both acute ionizing radiation injury and delayed effects of radiation exposure, such as radiation-induced lung injury (RILI) -associated mortality. In this study, we utilized the C57L/J mouse model, a well-established system for replicating the clinical pathology of RILI. Lung injury was induced using a combination of neutron total-body irradiation (TBI) (30% of total dose +7% of total dose concomitant gamma rays) and whole-thoracic X-irradiation (WTI) boost for the balance of the required dose at total doses of 9, 9.5, 10 and 10.5 Gy.
