Vision at the limits: Absolute threshold, visual function, and outcomes in clinical trials.

The study of individual differences in perception at absolute threshold has a rich history, with much of the seminal work being driven by the need to identify those with superior abilities in times of war. Although the popularity of such testing waned in the latter half of the 20th century, interest in measures of visual function at the absolute limit of vision is increasing, partly in response to emerging treatments for retinal diseases, such as gene therapy and cellular therapies, that demand "new" functional measures to assess treatment outcomes. Conventional clinical, or clinical research,

Cones in ageing and harsh environments: the neural economy hypothesis.

PURPOSE: Cones are at great risk in a wide variety of retinal diseases, especially when there is a harsh microenvironment and retinal pigment epithelium is damaged. We provide established and new methods for assessing cones and retinal pigment epithelium, together with new results. We investigated conditions under which cones can be imaged and could guide light, despite the proximity of less than ideal retinal pigment epithelium. RECENT FINDINGS: We used a variety of imaging methods to detect and localise damage to the retinal pigment epithelium. As age-related macular degeneration is a parti

Spatial receptive fields in the retina and dorsal lateral geniculate nucleus of mice lacking rods and cones.

In advanced retinal degeneration loss of rods and cones leaves melanopsin-expressing intrinsically photosensitive retinal ganglion cells (ipRGCs) as the only source of visual information. ipRGCs drive non-image-forming responses (e.g., circadian photoentrainment) under such conditions but, despite projecting to the primary visual thalamus [dorsal lateral geniculate nucleus (dLGN)], do not support form vision. We wished to determine what precludes ipRGCs supporting spatial discrimination after photoreceptor loss, using a mouse model (rd/rd cl) lacking rods and cones. Using multielectrode arrays

Neuronal apoptosis, axon damage and synapse loss occur synchronously in acute ocular hypertension.

Retinal ganglion cells (RGCs) apoptosis and their axon degeneration are pivotal features in glaucoma. Previous studies suggest that the process of RGCs soma degeneration is distinct from axon degeneration and that both of them lead to vision loss but separately. However, since a normal visual function relies on the integrity of axon, synapse and soma in the retina, a comprehensive understanding of the changes of these neuron components in glaucoma is desired. Therefore, in an acute ocular hypertension (AOH) model in mice, we systematically evaluated retinal neuron soma, axon and synapse altera

Disrupted cholesterol metabolism promotes age-related photoreceptor neurodegeneration.

Photoreceptors have high intrinsic metabolic demand and are exquisitely sensitive to metabolic perturbation. In addition, they shed a large portion of their outer segment lipid membranes in a circadian manner, increasing the metabolic burden on the outer retina associated with the resynthesis of cell membranes and disposal of the cellular cargo. Here, we demonstrate that deletion of both ABCA1 and ABCG1 in rod photoreceptors leads to age-related accumulation of cholesterol metabolites in the outer retina, photoreceptor dysfunction, degeneration of rod outer segments, and ultimately blindness.

Signal processing in a simple vertebrate photoreceptor system: the teleost pineal organ.

The integrating circuitry and efferent pathways for neural signals evoked in the photosensory pineal organ by changes in ambient illumination have been investigated by a multidisciplinary approach. Intrapineal efferent neurons were identified by means of retrograde filling with horseradish peroxidase (HRP). In addition to several types of neurons, photoreceptor cells that emitted axons to the brain via the pineal tract were observed. The presence of several populations of local interneurons (putatively cholinergic, GABAergic and substance P-containing) and possible afferent (putatively noradre

Keeping the balance.

How we see our environment is the result of a multi-level, parallel processing effort by the central nervous system. This computation is initiated within the retina at the very first synapse in the visual pathway - the photoreceptor ribbon synapse. Two recent studies shed light on the critical role of balanced calcium channel activity in maturation of this highly specialized synapse. (1, 2.)

From pineal photoreception to optogenetics: functional colour opponency based on a pineal bistable opsin.

Most animals sense light through opsins, which are photosensitive G protein-coupled receptors that utilize light information to regulate various physiological functions. In vertebrates, a representative example of such functions is vision, which is based on photoreception by visual opsins expressed in rod and cone photoreceptor cells of the retina. Accumulating photobiological evidence has gradually revealed the significance of the molecular properties of these opsins, as well as those of signal transduction-related molecules such as transducin, rhodopsin kinases, and arrestins, and their coll

Macular impairment in mitochondrial diseases: a potential biomarker of disease severity.

The high-energy demands of the retina are thought to contribute to its particular vulnerability to mitochondrial dysfunction. Photoreceptors are the cells with the higher oxygen consumption within the retina, and among these, the cones contain more mitochondria and have a higher energy demand than rods. A cohort of twenty-two patients with genetically-defined mitochondrial diseases (MDs) were enrolled to determine if the macula is functionally and anatomically impaired in these metabolic disorders. Visual acuity and fERG amplitude of patients with primary mitochondrial dysfunction were reduced

The Retinol-Binding Protein Receptor 2 (Rbpr2) Is Required for Photoreceptor Survival and Visual Function in the Zebrafish.

Vitamin A/retinol (ROL) and its metabolites (retinoids) play critical roles in eye development and photoreception. Short-term dietary vitamin A deficiency (VAD) manifests clinically as night blindness, while prolonged VAD is known to cause retinal pigment epithelium (RPE) and photoreceptor degeneration. Therefore, sustained uptake of dietary vitamin A, for ocular retinoid production, is essential for photoreceptor health and visual function. The mechanisms influencing the uptake, storage, and supply of dietary vitamin A, for ocular retinoid production, however, are not fully understood. We inv

Single-cell transcriptomic analysis of ferroptosis-associated cell populations in the progression of diabetic retinopathy.

BACKGROUND: Diabetic retinopathy (DR), a common microvascular complication of diabetes mellitus, has been associated with ferroptosis-related pathological mechanisms. Evidence indicates that ferroptosis may contribute to retinal injury and disease progression. The present analysis aimed to characterize ferroptosis-associated cell subsets and their roles in the progression of DR. METHOD: Single-cell RNA sequencing data derived from diabetic rat models of DR were obtained from the Gene Expression Omnibus database. A total of 464 ferroptosis-related genes were used to calculate ferroptosis score

Wide-field cellular-resolution retinal imaging using deformable mirror-based sensorless adaptive optics time-domain full-field OCT.

Adaptive optics (AO) enables cellular-resolution retinal imaging by correcting ocular aberrations, but its widespread clinical adoption remains limited by the narrow field of view (FOV) imposed by the isoplanatic patch of the eye. In this study, we present a deformable mirror (DM)-based sensorless AO time-domain full-field OCT (FFOCT) system that overcomes these limitations by leveraging the inherent robustness of FFOCT to ocular aberrations under spatially incoherent illumination. Using both phantom eye simulations and in vivo experiments, we demonstrate that correction of only three to five

Design, Synthesis, and In Vitro Evaluation of Novel 8-Amino-Quinoline Combined with Natural Antioxidant Acids.

Overproduction of reactive oxygen species (ROS) and alterations in metallostasis are common and related hallmarks in several neurodegenerative diseases (NDDs). Nature-based derivatives always represent an attractive tool in MTDL drug design, especially against ROS in NDDs. On this notion, we designed a new series of 8-quinoline-N-substituted derivatives with a natural antioxidant portion (i.e., lipoic, caffeic, and ferulic acids). These compounds were shown to chelate copper, a metal involved in ROS-induced degeneration, and scavenger oxygen radicals in DPPH assay. Then, selected compounds 4 a

Electroretinographic abnormalities in parents of patients with Leber congenital amaurosis who have heterozygous GUCY2D mutations.

BACKGROUND: Leber congenital amaurosis (LCA) is an infrequently encountered congenital form of retinitis pigmentosa with marked genetic and clinical heterogeneity. Thus far, 10 genes have been identified in this disorder since 1996. In the future, LCA may become treatable by gene and/or pharmacological intervention, and these therapies will likely be gene specific, giving major significance to rapid gene identification and gene-phenotype studies. OBJECTIVE: To test the hypothesis that parents of patients with LCA have identifiable electroretinographic and psychophysical changes. SUBJECTS, MAT
