Studies have demonstrated that adult male fish exposed to estrogenic endocrine-disrupting compounds (EDCs) exhibit reduced sperm motility and fertility. However, the effects of prepubertal estrogenic EDC exposure on testicular development and subsequent reproductive capacity remain poorly understood. In this study, prepubertal male (XY) tilapia at 30 days after hatching (dah) were exposed to 17β-estradiol (E2) for 60 days, covering the prepubertal period, to investigate its impact on testicular development and fertility at sexual maturity. Morphological and histological analyses revealed that

BACKGROUND: Male fertility has declined globally. Diabetes and aging are major contributors to the reduced sperm production and testicular dysfunction. Elevated glucagon signaling leads to diabetic complications, yet its impact on male reproductive function remains unclear.
METHODS: Streptozotocin-induced type 1 diabetic mice, naturally aged mice, and healthy young male mice were treated with a glucagon receptor (GCGR) monoclonal antibody or control. Sperm quality and testicular outcomes, including histology, oxidative stress, and apoptosis, were assessed. We then generated global Gcgr knocko

Sperm production starts from puberty in the seminiferous tubules providing testosterone production by the Leydig cells that takes place in the interstice of the testicles. Normal spermatogenesis depends on specific signalling from the hypothalamic-pituitary-gonadal axis. GnRH, FSH and LH are the main hormones involved in the production and maturation of spermatozoa. Exogenous administration of androgens influences the hypothalamic- pituitary-gonadal axis with negative feedback that may lead to a partial or complete cessation of spermatogenesis by decreasing FSH and LH. Despite the fact that ma

This paper presents data on the cellular localization of the testis-expressed mouse Cyrn gene product, cyritestin. This cysteine-rich protein is a member of a family including various rodent and primate proteins and snake venom proteins of the metalloproteinase and disintegrin types. By using antibodies raised against recombinant proteins generated in bacteria and against synthetic peptides we show that (i) Cyrn mRNA is present in germ cells 4 days prior to translation; (ii) cyritestin protein is localized in the acrosomal region of spermatids and spermatozoa; and (iii) cyritestin has an appar

The effects of estradiol benzoate administration (50 micrograms/day/rat for 15 days) to healthy intact male albino rats of Holtzman strain were investigated. Estradiol benzoate (E2B) treatment caused significant reduction in androgen-target tissues, viz., testis, epididymis, vas deferens, seminal vesicle, and ventral prostate. This led to alterations in the milieu of the epididymis and vas deferens rendering them hostile for sperm maturation, motility, metabolism, and survival as is evident by reduced sperm motility, alterations in their morphology, and the resulting loss of fertility. The red…

In vitro production of functional gametes can revolutionize reproduction by reducing generation intervals and accelerating genetic breeding in aquaculture, especially in fish with relatively long generations. Nevertheless, functional sperm production from in vitro-cultured spermatogonia remains a challenge in most aquaculture fish. In this study, we isolated and characterized premeiotic spermatogonia from marine four-eyed sleepers ( Bostrychus sinensis), which are prone to ovotesticular or sterile testicular development, and induced the differentiation of the spermatogonia into flagellated spe

DNA damage in the male germ line cells is correlated with male infertility. Nuclear DNA damage in mature sperm includes single strand nicks and double strand breaks that can arise as a consequence of errors in chromatin rearrangement during spermiogenesis, abortive apoptosis, and oxidative stress. Methods of assisted reproduction may be connected with a risk of the using of sperm with impaired DNA integrity that can lead to spontaneous abortion or delivery of a child with congenital abnormalities.

The epididymis is an essential organ for sperm maturation and reproductive health. The epididymal epithelium consists of intricately connected cell types that are distinct not only in molecular and morphological features but also in physiological properties. These differences reflect their diverse functions, which together establish the necessary microenvironment for the post-testicular sperm development in the epididymal lumen. The understanding of the biophysical properties of the epididymal epithelial cells is critical for revealing their functions in sperm and reproductive health, under bo…

Carbamazepine (CBZ) is an anti-epileptic drug that acts on Leydig cells, affecting steroidogenesis and causes fetal malformation. The aim of this study was to investigate the effects of CBZ on male sexual maturation and other male parameters. Rat dams were treated with CBZ during pregnancy and breastfeeding. The anogenital distance (AGD) and the anogenital index (AGI) were obtained. Testicular descent and preputial separation were also evaluated. The offspring was euthanized at PND 41 and 63. The accessory glands were weighed and the testes were collected for histopathological, morphometric an

Several studies have demonstrated interesting results considering the implication of three growth factors (GFs), namely nerve growth factor (NGF), erythropoietin (EPO), and the insulin-like growth factor-I (IGF-1) in the physiology of male reproductive functions. This review provides insights into the effects of NGF, EPO, and IGF-1 on the male reproductive system, emphasizing mainly their effects on sperm motility and vitality. In the male reproductive system, the expression pattern of the NGF system varies according to the species and testicular development, playing a crucial role in morphoge

PRSS37, a putative trypsin-like serine protease, is highly conserved during mammalian evolution as revealed by multiple sequence alignment. Mice deficient for Prss37 gene exhibit male infertility, but their mating behavior, spermatogenesis, sperm morphology, and motility remain unaffected, similar to a situation called unexplained male infertility (UMI) in men (human being). Here, we demonstrated that PRSS37 is restrictively expressed in human testis, where it is mainly located in the elongating and elongated spermatids during spermiogenesis as shown by immunohistochemical analysis of normal h

Ureaplasma urealyticum (UU) is one of the most commonly occurring pathogens associated with genital tract infections in infertile males, but the impact of seminal UU infection in semen on intrauterine insemination (IUI) outcomes is poorly understood. We collected data from 245 infertile couples who underwent IUI at The First Affiliated Hospital of USTC (Hefei, China) between January 2021 and January 2023. The subjects were classified into two groups according to their UU infection status: the UU-positive group and the UU-negative group. We compared semen parameters, pregnancy outcomes, and neo

It is generally assumed that men with congenital bilateral absence of the vas deferens (CBAVD) have azoospermia because of obstruction and that sperm production is normal. This study examines spermatogenesis in men with CBAVD to assess the validity of this assumption. We identified all men with CBAVD who had undergone either a diagnostic or therapeutic fertility procedure. Procedures included diagnostic biopsy, testis fine needle aspiration (FNA) mapping, microscopic epididymal sperm aspiration (MESA), and testis sperm extraction (TESE). Among 33 CBAVD men, 18 underwent testis biopsy, 27 had M

Knowledge of the capabilities and limitations of the reproductive capacity of bulls is vital in maximizing reproductive efficiencies. Bull semen collection guidelines established by researchers and industry personnel to maximize the sperm harvest from bulls have been evolving for more than 60 years. Today, a mature artificial insemination industry employs those strategies to meet demands. These efficient management schemes exploit the reproductive potential of each sire while minimizing the associated risk of injury to bulls and reduce associated production costs. Personnel employed by a semen

Three sexually mature goshawks reared in captivity and imprinted on humans to express reproductive behavior according to the cooperative method were studied for three consecutive breeding seasons to assess the quality of their sperm. The following parameters were analyzed: ejaculate volume and sperm concentration, motility, viability, and morphology. Ejaculate volume, sperm concentration and motility fluctuated along the reproductive season, revealing the greatest quality of the reproductive material at full springtime (i.e., April). Motility of the sperm collected in March strongly reduced wi

Impaired sperm motility is a leading cause of male infertility. Studies indicated that FK506, an immunosuppressive drug, resulted in male mouse infertility or an overall decline in the fertilization capacity of male renal transplant recipients. However, the underlying mechanism is not fully elucidated. Here, we reported that the disruption of FK506 binding protein 12.6 (FKBP12.6) significantly alleviated FK506-induced male infertility in mice by restoring sperm motility and mitochondrial functions in immature sperm. Mechanically, we identified that the FK506-FKBP12.6 complex preferentially bou
