Revitalising Brewers' Spent Grains and Enriching With Biogenic Compounds Through the Fermentation of Fructophilic Lactic Acid Bacteria and Yeasts.

The large output of spent grains from the brewing industry presents environmental concerns but also offers promising nutritional and functional potential for valorization by researchers and industrial stakeholders. In this perspective, we investigated how non-conventional starters like Fructobacillus fructosus PL22 and Wickerhamomyces anomalus GY1 can drive the fermentation of brewer's spent grain (BSG), a solid by-product of the brewing industry, to enrich its portfolio of bioactive compounds. While sugar reduction was comparable between started- and unstarted-BSG, the effect of the fermentat

Integrated transcriptomics and metabolomics reveal key metabolic pathway responses in Pistia stratiotes under Cd stress.

Cadmium (Cd) can interfere with plant gene expression, change the content of metabolites and affect plant growth. In this study, untargeted metabolomics (LC-MS) and RNA-Seq sequencing were performed on root tissues of Pistia stratiotes exposed to Cd stress. The results showed that cadmium stress affected the accumulation and transport of cadmium in plants and increased the content of soluble sugar, the activities of ascorbate peroxidase (APX), and peroxidase (POD) by 34.89%, 41.45%, and 6.71% on average, and decreased the activity of superoxide dismutase (SOD) by 51.51% on average. At the same

Screening of Dietary Ingredients against the Honey Bee Parasite Nosema ceranae.

Nosema ceranae is a major pathogen in the beekeeping sector, responsible for nosemosis. This disease is hard to manage since its symptomatology is masked until a strong collapse of the colony population occurs. Conversely, no medicaments are available in the market to counteract nosemosis, and only a few feed additives, with claimed antifungal action, are available. New solutions are strongly required, especially based on natural methods alternative to veterinary drugs that might develop resistance or strongly pollute honey bees and the environment. This study aims at investigating the nosemos

Physico-Chemical Conversion of Lignocellulose: Inhibitor Effects and Detoxification Strategies: A Mini Review.

A pretreatment of lignocellulosic biomass to produce biofuels, polymers, and other chemicals plays a vital role in the biochemical conversion process toward disrupting the closely associated structures of the cellulose-hemicellulose-lignin molecules. Various pretreatment steps alter the chemical/physical structure of lignocellulosic materials by solubilizing hemicellulose and/or lignin, decreasing the particle sizes of substrate and the crystalline portions of cellulose, and increasing the surface area of biomass. These modifications enhance the hydrolysis of cellulose by increasing accessibil

Description of gamma radiation-resistant Geodermatophilus dictyosporus sp. nov. to accommodate the not validly named Geodermatophilus obscurus subsp. dictyosporus (Luedemann, 1968).

A gamma radiation-resistant, Gram reaction-positive, aerobic and chemoorganotrophic actinobacterium, initially designated Geodermatophilus obscurus subsp. dictyosporus G-5(T), was not validly named at the time of initial publication (1968). G-5(T) formed black-colored colonies on GYM agar. The optimal growth range was 25-35 °C, at pH 6.5-9.5 and in the absence of NaCl. Chemotaxonomic and molecular characteristics of the isolate matched those described for members of the genus Geodermatophilus. The DNA G + C content of the strain was 75.3 mol%. The peptidoglycan contained meso-diaminopimelic ac

Regulation of sialic acid O-acetylation in human colon mucosa.

The expression of O-acetylated sialic acids in human colonic mucins is developmentally regulated, and a reduction of O-acetylation has been found to be associated with the early stages of colorectal cancer. Despite this, however, little is known about the enzymatic process of sialic acid O-acetylation in human colonic mucosa. Recently, we have reported on a human colon sialate-7(9)-O-acetyltransferase capable of incorporating acetyl groups into sialic acids at the nucleotide-sugar level [Shen et al., Biol. Chem. 383 (2002), 307-317]. In this report, we show that the CMP-N-acetyl-neuraminic aci

Structural and Functional Characterization of a Novel Broad-Spectrum Oligosaccharide Uptake Channel OmpV25 in Vibrio campbellii.

Vibrio campbellii (formerly Vibrio harveyi) is a marine pathogen that causes vibriosis, a major disease affecting global aquaculture. This bacterium inhabits chitin-rich environments and uses chito-oligosaccharides as a main nutrient source. Here, we identify the novel outer membrane protein OmpV25 as a porin mediating broad-spectrum oligosaccharide uptake in V. campbellii. OmpV25 was heterologously expressed in Escherichia coli BL21 (DE3) Omp8, purified by nickel affinity and size exclusion chromatography, and crystallized. The resulting 2.70 Å crystal structure revealed the 25-kDa OmpV25 to

Mining and functional characterization of NADPH-cytochrome P450 reductases of the DNJ biosynthetic pathway in mulberry leaves.

BACKGROUND: 1-Deoxynojirimycin (DNJ), the main active ingredient in mulberry leaves, with wide applications in the medicine and food industries due to its significant functions in lowering blood sugar, and lipids, and combating viral infections. Cytochrome P450 is a key enzyme for DNJ biosynthesis, its activity depends on the electron supply of NADPH-cytochrome P450 reductases (CPRs). However, the gene for MaCPRs in mulberry leaves remains unknown. RESULTS: In this study, we successfully cloned and functionally characterized two key genes, MaCPR1 and MaCPR2, based on the transcriptional profi

Gut microbiota derived short-chain fatty acids in physiology and pathology: An update.

Short-chain fatty acids (SCFAs) are essential molecules produced by gut bacteria that fuel intestinal cells and may also influence overall health. An imbalance of SCFAs can result in various acute and chronic diseases, including diabetes, obesity and colorectal cancer (CRC). This review delves into the multifaceted roles of SCFAs, including a brief discussion on their source and various gut-residing bacteria. Primary techniques used for detection of SCFAs, including gas chromatography, high-performance gas chromatography, nuclear magnetic resonance and capillary electrophoresis are also discus

Expanding the Role of Sub-Exploited DOE-High Energy Extraction and Metabolomic Profiling towards Agro-Byproduct Valorization: The Case of Carotenoid-Rich Apricot Pulp.

Traditional extraction remains the method-of-choice for phytochemical analyses. However, the absence of an integrated analytical platform, focusing on customized, validated extraction steps, generates tendentious and non-reproducible data regarding the phytochemical profile. Such a platform would also support the exploration and exploitation of plant byproducts, which are a valuable source of bioactive metabolites. This study deals with the incorporation of (a) the currently sub-exploited high energy extraction methods (ultrasound (UAE)- and microwave-assisted extraction (MAE)), (b) experiment

Probing the metabolic network in bloodstream-form Trypanosoma brucei using untargeted metabolomics with stable isotope labelled glucose.

Metabolomics coupled with heavy-atom isotope-labelled glucose has been used to probe the metabolic pathways active in cultured bloodstream form trypomastigotes of Trypanosoma brucei, a parasite responsible for human African trypanosomiasis. Glucose enters many branches of metabolism beyond glycolysis, which has been widely held to be the sole route of glucose metabolism. Whilst pyruvate is the major end-product of glucose catabolism, its transamination product, alanine, is also produced in significant quantities. The oxidative branch of the pentose phosphate pathway is operative, although the

Lack of mutagenicity and clastogenicity of PNAEmu-NLS targeted to a regulatory sequence of the translocated c-myc oncogene in Burkitt's lymphoma.

Peptide nucleic acids (PNAs) are synthetic homolog of nucleic acids in which the phosphate-sugar polynucleotide backbone is replaced by a flexible polyamide. They bind complementary polynucleotide sequences with higher affinity and specificity than their natural counterparts. PNAs linked to the appropriate nuclear localization signal (NLS) peptide have been used to selectively down-regulate the expression of several genes in viable cells. For example in Burkitt's lymphoma (BL) cells the c-myc oncogene is translocated in proximity to the Emu enhancer of the Ig gene locus and upregulated. PNAs c

Antiviral activity of Flos verbasci infusion against influenza and Herpes simplex viruses.

The lyophilized infusion from flowers of Verbascum thapsiforme Schrad. (FVI) showed antiviral activity in in vitro studies against Fowl plague virus, several influenza A strains, influenza B strain as well as Herpes simplex virus. Influenza viruses titer decreased by 1-3 log units, while of H. simplex virus by 2.3 log. FVI has shown virucidal activity on H. simplex virus at 300 micrograms/ml, but did not inactivate influenza viruses. Phytochemical investigations of FVI have shown the presence of flavonoids, iridoids, phenolic acids, saponins, amino acids and free sugars.

Inhibitory effects of 2-deoxytetronic acid, a putative endogenous satiety factor, on paraventricular neurons of rats, in vitro.

The endogenous sugar acid, 2-deoxytetronic acid (2-DTA) was investigated for its effects on activity of paraventricular nucleus (PVN) neurons in slice preparations from rats. 2-DTA dose-responsively inhibited the spontaneous activity of 23 of the 53 PVN neurons tested at concentrations of 10(-5) to 10(-2) M, and 2 were excited. The inhibitory responses were not due to osmotic changes by 2-DTA administration. The high concentration of 2-DTA needed to produce responses suggests that it acts as substrate for oxidative metabolism rather than as a neurotransmitter or neuromodulator in the suppressi

Blending Carbohydrate and Quinine-Based Polymers Imparts Colloidal Stability, Improved Performance, and Cell Specificity for mRNA Delivery.

Quinine-based polymers have shown promise for effective delivery of nucleic acids; however, quinine's hydrophobicity has often resulted in uncontrolled aggregation (>1000 nm) of complexes. PEGylation is often used to improve the colloidal stability of nucleic acid delivery vehicles; however, this frequently results in reduced cellular internalization. Herein, we describe the synthesis of two diblock quinine polymers that utilize the carbohydrates glucose and galactose in place of PEG as the neutral hydrophilic block to deliver mRNA. These diblock stabilizers were blended with five hydrophobic

Nutrition in inflammatory bowel diseases.

The relationship between nutrition and inflammatory bowel disease (IBD) is a complex one, the evaluation and correction of nutritional deficits being integral part of therapy in these patients. The diet that consists in excessive consumption of meat, sugar, fats (with a higher 0 6 /o) 3 polyunsaturated fatty acids ratio) are factors involved in the epidemiology and pathogenesis of IBD. Malnutrition is present among most IBD patients, being the result of multiple mechanisms from digestive symptoms to inflammatory process, intestinal resection and administration of medications. Oral diet is high

Some cultural and physiological aspects of methane-utilizing bacteria.

A number of different methane-utilizing bacteria are described and compared with isolates of other investigators. The strains can be divided into three groups based on pigmentation, cell morphology and internal membrane structures. The oxidation of hydrocarbons, alcohols, aldehydes, fatty acids, methyl ethers and sugar phosphates by these bacteria was studied. There was much similarity between strains within the same group. Differences between groups as regards oxidative properties could be detected, but these were mainly quantitative and could not be used as taxonomical criteria. In addition,
