Aflatoxin B1-induced toxicity, oxido-inflammatory damage, and apoptosis in male rat reproductive circuitry were abrogated by co-treating with the xanthophyll-lutein and zeaxanthin.

Reproductive dysfunction is a recognised adverse effect of exposure to aflatoxin B1 (AFB1) in humans and animals. Despite the widely acknowledged health risks, exposure to AFB1 remains unavoidable. Conversely, lutein (LUT) and zeaxanthin (ZEA) are plants' potent antioxidants and anti-inflammatory agents, exhibiting promising potential for modulating inflammatory and apoptotic signalling pathways. This study aimed to investigate the effects of co-treatment with LUT/ZEA on reproductive function in rats intoxicated with AFB1. The study utilised male Wistar rats (n = 20 and n = 4 per cohort). The

Assessment of the Impact of Humic Acids on Intestinal Microbiota, Gut Integrity, Ileum Morphometry, and Cellular Immunity of Turkey Poults Fed an Aflatoxin B1-Contaminated Diet.

A recent study published data on the growth performance, relative weights of the organs of the gastrointestinal tract, liver histology, serum biochemistry, and hematological parameters for turkey poults fed an experimental diet contaminated with aflatoxin B1 (AFB1) and humic acids (HA) extracted from vermicompost. The negative effects of AFB1 (250 ng AFB1/g of feed) were significantly reduced by HA supplementation (0.25% w/w), suggesting that HA might be utilized to ameliorate the negative impact of AFB1 from contaminated diets. The present study shows the results of the remaining variables, a

Stannous Chloride-Modified Glass Substrates for Biomolecule Immobilization: Development of Label-Free Interferometric Sensor Chips for Highly Sensitive Detection of Aflatoxin B1 in Corn.

This study presents the development of stannous chloride (SnCl2)-modified glass substrates for biomolecule immobilization and their application in fabricating sensor chips for label-free interferometric biosensors. The glass modification process was optimized, identifying a 5% SnCl2 concentration, a 45 min reaction time, and a 150 °C drying temperature as conditions for efficient protein immobilization. Based on the SnCl2-modified glass substrates and label-free spectral-phase interferometry, a biosensor was developed for the detection of aflatoxin B1 (AFB1)-a highly toxic and carcinogenic con

Assessing the Validity of Normalizing Aflatoxin B1-Lysine Albumin Adduct Biomarker Measurements to Total Serum Albumin Concentration across Multiple Human Population Studies.

The assessment of aflatoxin B1 (AFB1) exposure using isotope-dilution liquid chromatography-mass spectrometry (LCMS) of AFB1-lysine adducts in human serum albumin (HSA) has proven to be a highly productive strategy for the biomonitoring of AFB1 exposure. To compare samples across different individuals and settings, the conventional practice has involved the normalization of raw AFB1-lysine adduct concentrations (e.g., pg/mL serum or plasma) to the total circulating HSA concentration (e.g., pg/mg HSA). It is hypothesized that this practice corrects for technical error, between-person variance i

Aflatoxin B1 exposure triggers hepatic lipotoxicity via p53 and perilipin 2 interaction-mediated mitochondria-lipid droplet contacts: An in vitro and in vivo assessment.

Aflatoxin B1 (AFB1) is one of the most toxic mycotoxins widely found in food contaminants, and its target organ is the liver. It poses a major food security and public health threat worldwide. However, the lipotoxicity mechanism of AFB1 exposure-induced liver injury remains unclear and requires further elucidation. Herein, we investigated the potential hepatic lipotoxicity of AFB1 exposure using in vitro and in vivo models to assess the public health hazards of high dietary AFB1 exposure. We demonstrated that low-dose of AFB1 (1.25 μM for 48 h, about one-fifth of the IC50 in HepG2 and HepaRG c

Increased risk of hepatocellular carcinoma in male hepatitis B surface antigen carriers with chronic hepatitis who have detectable urinary aflatoxin metabolite M1.

We followed 145 men with chronic hepatitis B virus (HBV) hepatitis for 10 years to determine whether exposure to aflatoxin, or concomitant exposure to hepatitis C virus (HCV), or family history of hepatocellular carcinoma (HCC) increased the risk of developing HCC. We collected 8 monthly urine samples before beginning follow-up and pooled them to detect aflatoxin metabolite M1 (AFM1). AFM1 was detected in 78 (54%) of the subjects. The risk of HCC was increased 3.3-fold (with a 95% confidence interval of 1.2-8.7) in those with detectable AFM1 (above 3.6 ng/L). This relative risk was adjusted fo

Modulation by dietary copper of aflatoxin B1-induced activity of DNA repair enzymes poly (ADP-ribose) polymerase, DNA polymerase beta and DNA ligase.

The activity of some nuclear enzymes associated with DNA repair was examined following aflatoxin B1 administration in rats maintained on different levels of dietary copper. Induction of poly(ADP-ribose) polymerase, DNA polymerase beta and DNA ligase was found to be significantly higher in copper-deficient rats. Copper supplementation, even at marginal doses, was able to bring down the induction to the level observed in normal rats. The results emphasize the protective role of copper against the DNA damaging effects of aflatoxin B1.

[Diet imbalances and aflatoxicosis].

Generally we know influences of diet's balances or imbalances upon toxicants. Some actions appear with aflatoxins as shown by studies on rat, pig, calf, poultry, turkey and duckling. According to species considered, oral administration of aflatoxins reduces food consummation, Nitrogen balance, respiratory coefficient. Therefore those modifications may be induced by a decrease of foodstuffs ingestion. It has been underlined that hypoproteic diet increases intoxication and diet rich in lipotropic factors decreases it. The greatest number of experiments carried out are concerning vitamins A and D…

The Effects of Lycopene on the Growth Performance, Antioxidant Capacity, and Liver Health of Hybrid Groupers (♀ Epinephelus fuscoguttatus × ♂ E. lanceolatus) Under the Influence of Aflatoxin B1.

The continued rise in fishmeal prices and the scarcity of fishery resources have led to an increase in the use of plant-based protein sources in compound feed, making mold contamination in compound feed a frequent occurrence. Aflatoxin B1 (AFB1) contamination in aquaculture feed can seriously harm fish growth and liver health; it is extremely toxic, posing an even greater risk to carnivorous fish such as grouper. Lycopene, a natural carotenoid, has antioxidant and anti-inflammatory physiological functions and may act as a functional feed additive to mitigate damage induced by mycotoxins. This

β-Nicotinamide mononucleotide reduces aflatoxin B1 induced liver injury via the gut microbiota-bile acid-farnesoid X receptor (FXR/NR1H4) axis.

BACKGROUND: Aflatoxin B1 (AFB1) is a potent hepatotoxin that poses significant health risks to both livestock and humans. β-Nicotinamide mononucleotide (NMN) has potential therapeutic benefits for liver diseases. However, its mechanism against AFB1-induced liver injury remains unclear. OBJECTIVE: This study investigates whether NMN supplementation reduces AFB1-induced liver injury through the modulation of the gut-liver axis and elucidates the underlying molecular mechanism. METHODS: Mice were exposed to AFB1 (0.75 mg·kg-1, p.o.) for 2 weeks to induce liver injury, with or without NMN (300 m

Protective effects of ganoderic acid C2 from Ganoderma lucidum against aflatoxin B1-induced splenorenal injury: Inhibition of NOX4-mediated oxidative stress and inflammation.

Aflatoxin B1 (AFB1) is a mycotoxin known to induce splenorenal toxicity through oxidative stress and chronic inflammation, yet effective protective strategies remain limited. Ganoderic Acid C2 (GA-C2), a triterpenoid isolated from Ganoderma lucidum, possesses antioxidant and anti-inflammatory potential, but its role against AFB1-induced toxicity has not been fully clarified. In this study, seventy-two male BALB/c mice were treated with AFB1 (0.75 mg/(kg·day)) alone or in combination with GA-C2 (5, 10, 15 mg/(kg·day)) or the NADPH oxidase 4 (NOX4) inhibitor GKT137831 (30, 60 mg/(kg·day)) for 30
