A Porcine Model of Intervertebral Disc Injury Recapitulates Human Discogenic Pain via Notochordal Cell Loss and Pain-inducing Nucleus Pulposus Cell Emergence.

Lower back pain (LBP) is one of the most common causes of disability, with up to 40% of LBP cases attributed to intervertebral disc (IVD) degeneration. While small animal models are widely used to study IVD and LBP, the small size of their IVDs limits translational and biological relevance. Large animal models more accurately emulate human disease; however, methods of measuring LBP are not well established. Pigs were also considered unfit for LBP research, due to notochordal cells (NCs) persistence through life, unlike humans. We developed a comprehensive porcine model with quantitative measur

Veterinary Student Skills Learned at an Access to Care Clinic: Beyond Medicine and Surgery.

Incorporating curriculum to effectively help veterinary students learn how to provide accessible quality care to all pet owners is needed. The primary aims of this study are to explore how a 2-week rotation at a veterinary medical service-learning clinic (Wisconsin Companion Animal Resources, Education, and Social Services [WisCARES]) improves (a) comfort in working with clients from diverse race and low socioeconomic (SES) backgrounds and (2) confidence in leading cases, communication skills, and providing a spectrum of care options. Students were surveyed at five time points pre-rotation: mi

Advances in cholera research: from molecular biology to public health initiatives.

The aquatic bacterium Vibrio cholerae is the etiological agent of the diarrheal disease cholera, which has plagued the world for centuries. This pathogen has been the subject of studies in a vast array of fields, from molecular biology to animal models for virulence activity to epidemiological disease transmission modeling. V. cholerae genetics and the activity of virulence genes determine the pathogenic potential of different strains, as well as provide a model for genomic evolution in the natural environment. While animal models for V. cholerae infection have been used for decades, recent ad

Sensitivity of the S1 neuronal calcium network to insulin and Bay-K 8644 in vivo: Relationship to gait, motivation, and aging processes.

Neuronal hippocampal Ca2+ dysregulation is a critical component of cognitive decline in brain aging and Alzheimer's disease and is suggested to impact communication and excitability through the activation of a larger after hyperpolarization. However, few studies have tested for the presence of Ca2+ dysregulation in vivo, how it manifests, and whether it impacts network function across hundreds of neurons. Here, we tested for neuronal Ca2+ network dysregulation in vivo in the primary somatosensory cortex (S1) of anesthetized young and aged male Fisher 344 rats using single-cell resolution techn

Integrated approaches to howler monkey (Alouatta spp.) medicine in professional care and conservation.

Howler monkeys (Alouatta spp.) are threatened by anthropogenic pressures such as habitat fragmentation and deforestation, while conservation efforts are challenging to coordinate as natural geographic distribution ranges are the largest of any New World primate. On a One Health front, howler monkeys represent a great model to investigate the infectious disease dynamics between wild primates and humans as several infectious diseases affecting howlers have a demonstrated zoonotic potential. Howler monkey populations in professional care offer a window to investigate susceptibility to diseases in

If you build it, they will learn: A review of models in veterinary surgical education.

Surgical skills are learned through deliberate practice, and veterinary educators are increasingly turning to models for teaching and assessing surgical skills. This review article sought to compile and review the literature specific to veterinary surgical skills models, and to discuss the themes of fidelity, educational outcomes, and validity evidence. Several literature searches using broad terms such as "veterinary surgery model," "veterinary surgical model," and "veterinary surgical simulator" were performed using PubMed, CAB abstracts, and Google scholar. All articles describing a model c

Endocrine disrupting polyhalogenated organic pollutants interfere with thyroid hormone signalling in the developing brain.

Persistent polyhalogenated organic pollutants are present worldwide and accumulate along the food chain. They interfere with human and animal health and are particularly harmful for pre- and perinatal neurodevelopment. The mechanisms behind the observed effects vary depending on the specific compound investigated. Co-planar polychlorinated biphenyls (PCBs) can act via the arylhydrocarbon receptor while many ortho-substituted PCBs disrupt intracellular Ca(2+) homeostasis. A common mechanism for a wide variety of PCBs is interference with thyroid hormone (TH) signalling in developing brain, by c

Behavioral alterations in rats prenatally exposed to valproic acid: animal model of autism.

Autism is a severe behavioral disorder characterized by pervasive impairments in social interactions, deficits in verbal and nonverbal communication, and stereotyped, repetitive patterns of behaviors and interests. Recently, a new rodent model of autism was created by exposure of rat fetuses to valproic acid (VPA) on the 12.5th day of gestation (VPA rats). The model has striking anatomical, pathological, and etiological similarities to human data; however, it has not been characterized behaviorally. In order to determine if VPA rats present behavioral aberrations observed in autism, their beha

Evidence of transmission and dissemination of diverse blaNDM-5-producing Escherichia coli clones between refugee and host communities and their environment: a multicenter cross-sectional study.

UNLABELLED: The global spread of carbapenemase-producing Escherichia coli (CP-Ec) poses a significant public health threat, with particularly severe consequences for vulnerable populations in resource-limited settings. To address this, we conducted in-depth genetic analyses and examined the relatedness of CP-Ec isolates recovered from hospitalized patients, refugees, animals, water, and environmental sources within refugee camps and in marginalized host communities in Lebanon. Nineteen putative CP-Ec isolates, identified by MALDI-TOF MS and designated as community isolates, harbored either NDM

Are we ready for the next anthrax outbreak? Lessons from a simulation exercise in a rural-based district in Uganda.

Anthrax is a bacterial zoonotic disease caused by Bacillus anthracis. We qualitatively examined facilitators and barriers to responding to a potential anthrax outbreak using the capability, opportunity, motivation behaviour model (COM-B model) in the high-risk rural district of Namisindwa, in Eastern Uganda. We chose the COM-B model because it provides a systematic approach for selecting evidence-based techniques and approaches for promoting the behavioural prompt response to anthrax outbreaks. Unpacking these facilitators and barriers enables the leaders and community members to understand ex

Neurobehavioral sex-related differences in Nf1+/- mice: female show a "camouflaging"-type behavior.

BACKGROUND: Neurofibromatosis type 1 (NF1) is an inherited neurocutaneous disorder associated with neurodevelopmental disorders including autism spectrum disorder (ASD). This condition has been associated with an increase of gamma-aminobutyric acid (GABA) neurotransmission and, consequently, an excitation/inhibition imbalance associated with autistic-like behavior in both human and animal models. Here, we explored the influence of biological sex in the GABAergic system and behavioral alterations induced by the Nf1+/- mutation in a murine model. METHODS: Juvenile male and female Nf1+/- mice an

Surveillance and Control of African Swine Fever in the Early Phase of the COVID-19 Pandemic, March-May 2020: A Multi-Country E-Survey.

Stringent COVID-19 public health and social measures (PHSMs) have challenged the work of animal health professionals, especially in the early phase of the pandemic. We aimed to qualitatively describe how COVID-19 PHSMs have affected the surveillance and control of African swine fever (ASF) in Europe, assess how professionals engaged in these activities perceived the impact of the COVID-19 crisis, and identify potential areas of improvement. An online questionnaire was proposed via email between 9 December 2020 and 22 January 2021 to professionals engaged in ASF-related activities in Europe and

Increased efficacy of combining prebiotic and postbiotic in mouse models relevant to autism and depression.

The Microbiota-Gut-Brain axis (MGBA) is a bidirectional communication pathway between gut bacteria and the central nervous system (CNS) (including the intestine) that exerts a profound influence on neural development, neuroinflammation, activation of stress response and neurotransmission, in addition to modulating complex behaviours, such as sociability and anxiety. Several MGBA modulating approaches are possible, such as probiotic administration. A reasonable pharmacological approach would also be the contemporarily administration of both prebiotics and postbiotics. To test this hypothesis, w

Consumers' understanding and concerns about bovine spongiform encephalopathy (BSE): comparison among Canadian, American, and Japanese consumers.

In spite of much analysis of the impact of bovine spongiform encephalopathy (BSE) on consumer perceptions and meat purchases, there has been little explicit analysis of the level of BSE knowledge. In this study the role of knowledge about BSE was examined in Canada, the United States, and Japan. In addition, the level of knowledge was linked to human health concerns regarding BSE and whether there is agreement with paying a premium for beef with BSE animal tests. From a public policy perspective, understanding whether higher or lower knowledge is linked to public concern and desire for market

The meat and plant protein blend alternative: A systematic review of factors influencing hybrid meat consumption.

Because hybrid meat products blend characteristics of both meat and plant-based foods, they may represent an ambiguous or intermediate category that does not fit neatly within existing food classifications. Often referred to within industry as balanced proteins, these products combine animal-derived ingredients with alternative, non-animal ingredients or protein sources. Despite growing commercial and policy interest, consumer research on hybrid meat remains fragmented and conceptually underdeveloped. Using the theories, theoretical constructs, context, and methodologies (TCCM) framework, this

Analysis of human-oriented facial signals of the domestic dog using eye tracker technology.

INTRODUCTION: Understanding canine facial signals is crucial for responsible dog ownership and animal welfare, yet how humans visually process these signals in breeds with extreme morphology remains under-researched. METHODS: This study examined visual attention patterns using eye-tracking when humans viewed brachycephalic versus normocephalic dog faces. Forty-four undergraduate students viewed photographs of Boston Terriers (brachycephalic) and Jack Russell Terriers (normocephalic) depicting four emotional contexts: positive (Called by Name, Play) and negative (Separation, Stranger Threat).

ASPIS Academy: A blueprint for building early-career researcher networks to advance NAM-based risk assessment.

Chemical risk assessment is moving away from traditional animal-based testing toward New Approach Methodologies (NAMs) that are more human-relevant, efficient, and aligned with the replacement, reduction, and refinement of animal use. This transition depends not only on new methods and technologies, but also on the researchers who will develop, apply, and promote them. Early-career researchers (ECRs) are central to this effort, yet they often work within fragmented professional networks and have limited opportunities for mentoring, visibility, and leadership. The ASPIS Academy (AA) was establi

Systematic review of changes in extracellular vesicles associated with obstructive sleep apnoea: implications for diagnosis and treatment.

Obstructive sleep apnoea (OSA) is a major public health concern, strongly linked to cardiovascular disease and cancer. Extracellular vesicles (EVs) have emerged as key mediators in intercellular communication, oxidative stress and inflammation, carrying molecules that can influence OSA pathophysiology. However, their role in OSA pathophysiology remains underexplored. This systematic review consolidates current research on EVs in OSA, focusing on their cargo, surface proteins and impact on oxidative stress, inflammation, cancer progression and cardiovascular dysfunction. Customizable artificial simulator for developing, planning, and training personnel on neurophysiology and surgical procedures in non-human primates.

BACKGROUND: Neuroscience researchers often surgically implant hardware into model organisms to measure and manipulate neural activity. Designing and optimizing these procedures in non-human primates often requires sedated or euthanized animals. Artificial tissue technologies can reduce animal use in this process, but existing simulators do not include all relevant tissues and do not facilitate iterative design processes. NEW METHOD: We created a comprehensive, customizable, and modular surgical simulator for neuroscience research. Our simulator incorporates artificial skull, brain, and soft t
