BACKGROUND: Shiga toxin-producing Escherichia coli O157:H7 is a foodborne pathogen that causes severe human diseases including hemolytic uremic syndrome (HUS). The virulence factor that mediates HUS, Shiga toxin (Stx), is encoded within the genome of a lambdoid prophage. Although draft sequences are publicly available for a large number of E. coli O157:H7 strains, the high sequence similarity of stx-converting bacteriophages with other lambdoid prophages poses challenges to accurately assess the organization and plasticity among stx-converting phages due to assembly difficulties.

Type 1 von Willebrand disease (VWD) is a common inherited disorder characterized by mild to moderate bleeding and reduced levels of von Willebrand factor (VWF). An animal model for human type 1 VWD, the RIIIS/J mouse strain, exhibits a prolonged bleeding time and reduced plasma VWF levels. We have previously mapped the defect in RIIIS/J to distal mouse Chr 11, distinct from the Vwf locus on Chr 6. This locus, Mvwf, was localized to an approximately 0.5-cM interval, tightly linked to Gip, distal to Ngfr, and proximal to Hoxb. We have now used these genetic markers to construct a contig of yeast

Avian pathogenic Escherichia coli (APEC) infections are associated with major economic losses, cause perihepatitis, pericarditis, septicemia and even systemic infections in the poultry industry. APEC infections have usually been controlled by antibiotics, resulting in an increased prevalence of antibiotic-resistant E. coli. Concerns have been increased that transfer of antibiotic-resistant APEC via the food chain may cause risks for extra-intestinal infection of humans related to zoonotic transfer and increased difficulties in the treatment of human infections caused APEC-related E. coli types

The rapid expansion of cervid farming raises concerns about antimicrobial resistance (AMR) dissemination, yet its impact on the Cervidae gut microbiome remains poorly characterized. We integrated 89 newly sequenced fecal metagenomes with 599 publicly available datasets, comprising 285 metagenomes from farmed cervids and 370 from wild cervids, to construct a catalog of 15,494 non-redundant metagenome-assembled genomes (MAGs) representing 2,401 species. Our analysis demonstrates that farming profoundly reshapes the gut microbiome's functional composition. Specifically, farmed cervids exhibited s

OBJECTIVE: Multidrug-resistant (MDR) Gram-negative bacteria, particularly Klebsiella pneumoniae, represent a significant threat to global health. Given the shortage of new antibiotics, bacteriophages (phages) offer a promising alternative. Understanding bacteria-phage interactions and resistance mechanisms is vital for optimising phage therapy. We aimed to investigate the resistance mechanism of an MDR clinical isolate, K. pneumoniae UCSD1 (designated as KpUCSD1) against a newly isolated phage ΦKpUCSD1.
METHODS: A phage-resistant mutant KpUCSD1R was collected at 24 h following treatment of Kp

INTRODUCTION: The cervicovaginal microbiome is a key determinant of reproductive health. Its multi-kingdom structure and ecological interactions remain insufficiently characterized across diverse populations. This study aimed to define the composition and cross-kingdom dynamics of the cervical microbiome in women without HPV infection and with normal cytology in a Kazakhstani population.
METHODS: In this cross-sectional study, cervical samples from 92 reproductive-age women were analyzed using whole-genome metagenomic sequencing to characterize bacterial, viral, fungal, and archaeal communiti

Acinetobacter baumannii is a bacterial pathogen that is commonly associated with hospital-acquired illnesses. Antimicrobial drug resistance in A. baumannii includes several penicillin classes, first and second-generation cephalosporins, cephamycins, most aminoglycosides, chloramphenicol, and tetracyclines. The recent rise in multidrug-resistant A. baumannii strains has resulted in an increase in pneumoniae associated with ventilators, urinary tract infections associated with the catheter, and bloodstream infections, all of which have increased complications in treatment, cost of treatment, and

The factors contributing to the survival of enveloped viruses (e.g., influenza and SARS-CoV-2) on fomite surfaces are of societal interest. The bacteriophage Phi6 is an enveloped viral surrogate commonly used to study viability. To investigate how viability changes during the evaporation of droplets on polypropylene, we conducted experiments using a fixed initial Phi6 concentration while systematically varying the culture concentration and composition (by amendment with 2% fetal bovine serum (FBS), 0.08 wt % BSA, or 0.5 wt % SDS). The results were consistent with the well-founded relative humi

A predator-prey model is used to investigate the interactions between phages and bacteria by considering the lytic and lysogenic life cycles of phages and the prophage induction. We provide answers to the following conflictual research questions: (1) what are conditions under which the presence of phages can purify a bacterial infected environment? (2) Can the presence of phages triggers virulent bacterial outbreaks? We derive the basic offspring number $\mathcal N_0$ that serves as a threshold and the bifurcation parameter to study the dynamics and bifurcation of the system. The model exhibit

Solar radiation-driven inactivation of bacteria, virus and protozoan pathogen models was quantified in simulated drinking water at a temperate latitude (34 degrees S). The water was seeded with Enterococcus faecalis, Clostridium sporogenes spores, and P22 bacteriophage, each at ca 1x10(5) mL(-1), and exposed to natural sunlight in 30-L reaction vessels. Water temperature ranged from 17 to 39 degrees C during the experiments lasting up to 6h. Dark controls showed little inactivation and so it was concluded that the inactivation observed was primarily driven by non-thermal processes. The optimis

The classical T-even bacteriophages recognize host cells with their long tail fibers. Gene products 35, 36, and 37 constitute the distal moiety of these fibers. The free ends of the tail fibers, which are formed by the CO2H terminus of gene product 37, possess the host range determinants. It was found that 4 out of 10 different strains of Escherichia coli K-12 contained regions of chromosomal DNA which hybridized with a probe consisting of genes 35, 36, and 37 of the T-even phage K3. From one strain this homologous DNA, which was associated with an EcoRI fragment of about 5 kilobases, was clon

OBJECTIVES: In Staphylococcus aureus, WTA glycosylation participates in the interaction with the immune system and bacteriophage adsorption. Indeed, virulent phages with podovirus morphology and some myoviruses require β-glycosylation (catalized by TarS), and are hindered by α-glycosylation (carried out by TarM) of WTA. In contrast, other myophages, like phiIPLA-RODI and phage K, exhibit a broad-host range and infect strains without either glycosylation according to the spot test. Here, we set out to determine the impact of glycosylation type on susceptibility to the Kayvirus phiIPLA-RODI.

INTRODUCTION: Various systemic and enteric diseases have been attributed to C. perfringens, including healthcare-associated infections and intoxications. Humans and animals are thus exposed to serious health risks from these bacteria. Infections caused by these antibiotic-resistant bacteria are becoming an increasing concern. Bacteriophages (phages), which possess specific lytic activity, offer a viable alternative for the treatment of Clostridium perfringens.
METHODS: In this study, a novel lytic C. perfringens phage designed as CpP_VarHS was isolated from wastewater and targeted the host C.

The emergence of multidrug-resistant Klebsiella pneumoniae, including carbapenem-resistant K. pneumoniae (CRKP), as one of the most common and notable superbugs, has long been a major threat to public health. As natural predators of bacteria, bacteriophages (or phages) can induce the lysis of bacterial cells. Herein, we report the isolation and characterization of two phages and their efficacy in the control of CRKP. Using the sequence type 11 (ST11) CRKP strain THR60 and its related strain THR60r as the host bacteria, phages GZ7 and GZ9 were isolated from hospital sewage, respectively. GZ7 is

There are at least six well-established pathotypes of Escherichia coli (E. coli), including extraintestinal E. coli (ExPEC), enteroinvasive E. coli (EIEC), enteropathogenic E. coli (EPEC), enteroaggregative E. coli (EAEC), enterotoxigenic E. coli (ETEC), and Shiga toxin-producing E. coli (STEC). Here, we reviewed the role hybrid STEC pathotypes have played in causing outbreaks of gastrointestinal infectious disease (GIID) from the epidemics of dysentery at the end of the 19th century, to recent foodborne outbreaks and outbreaks of haemolytic uraemic syndrome (HUS). We assessed genomic and epid

Salmonella Typhimurium is a highly lethal pathogenic bacterium in weaned piglets, causing significant treatment costs and economic losses in the swine industry. Additionally, due to its ability to induce zoonotic diseases, resulting in harm to humans through the transmission of the pathogen from pork, it presents a serious public health issue. Bacteriophages (phages), viruses that infect specific bacterial strains, have been proposed as an alternative to antibiotics for controlling pathogenic bacteria. In this study, we isolated SLAM_phiST1N3, a phage infecting a multidrug-resistant (MDR) S. T

Amino acid substitutions conferring resistance of herpes simplex virus 1 (HSV-1) and human cytomegalovirus (HCMV) to foscarnet (PFA) are located in the genes UL30 and UL54, respectively, encoding the DNA polymerase (pol). In this study, we analyzed the impact of substitutions located in helix K and region II that are involved in the conformational changes of the DNA pol. Theoretical substitutions were identified by sequences alignment of the helix K and region II of human herpesviruses (susceptible to PFA) and bacteriophages (resistant to PFA) and introduced in viral genomes by recombinant phe

The rise of vancomycin-resistant Enterococcus spp. (VRE) has led to treatment challenges in hospital settings worldwide. Hospital wastewater (HW) might disseminate this threat to the aquatic environment. Thus, this study elucidates the VRE resistance quotient (RQ) of different environmental matrixes in wastewater and compares genomic determinants of VRE strains recovered from HW to water resources. Presumptive Enterococcus spp. and VRE were quantified and isolated using standard microbiological procedures. Fourteen VRE genomes were then sequenced using an Illumina HiSeq X™ Ten platform. Subseq
