Transplantation of neural stem cells: cellular & gene therapy for hypoxic-ischemic brain injury.

We have tracked the response of host and transplanted neural progenitors or stem cells to hypoxic-ischemic (HI) brain injury, and explored the therapeutic potential of neural stem cells (NSCs) injected into mice brains subjected to focal HI injury. Such cells may integrace appropriately into the degenerating central nervous system (CNS), and showed robust engraftment and foreign gene expression within the region of HI inury. They appeared to have migrated preferentially to the site of ischemia, experienced limited proliferation, and differentiated into neural cells lost to injury, trying to re

[Electrophysiological brain-stem diagnosis in patients with a pronounced obstructive sleep apnea syndrome].

BACKGROUND AND OBJECTIVE: The pathophysiological basis of obstructive sleep apnoea syndrome (OSAS) is a loss of tone of the oropharyngeal mu … uscular tone, one can demonstrate an increased incidence of subclinical, functional disorder in the brain stem as well as any pointers to a central localization of abnormalities in OSAS. PATIENTS AND METHODS: 19 patients with severe OSAS were examined for the presence of subclinical brain-stem lesions by means of acoustic evoked potentials, blinking reflex, …

[Reliability of brain stem audiometry in specific learning disorders (disorders of sensory integration)].

Diagnostic audiological tests performed in ten children from 1985 to 1991 showed remarkable differences between the results of behavioral audiometry (free field or pure tone audiometry) and those of auditory evoked response audiometry. Auditory brainstem response (ABR) was absent in seven children with sensory neural hearing loss. Neuropsychological evaluation was performed on four children, neuropsychological observation was performed on two children, pediatric examination was performed on three children and one child showed signs of neuropediatric disorder. The results of neuropsychological

Magnetic resonance imaging and brain-stem auditory evoked potentials in neuro-Behçet's disease.

We studied central nervous system lesions in patients with neuro-Behcet's disease using magnetic resonance imaging (MRI) of the brain and recording of brain-stem auditory evoked potentials (BAEPs). MRI revealed abnormal findings in seven of eight patients. MRI studies demonstrated extensive regions with high intensity signal in the brain stem and/or basal ganglia on T2-weighted images obtained during the acute stage of the disease in three patients. One of these patients had a strongly gadolinum-enhanced round lesion in the lower pons. In four of the other five patients with chronic disease, b…

Brain stem compression secondary to adipose graft prolapse after translabyrinthine craniotomy: case report.

Three cases of fat graft prolapse into the cerebellopontine angle with clinical deterioration are presented. These patients had undergone translabyrinthine craniotomy for cerebellopontine angle tumors and had autologous abdominal fat strips placed to obliterate the dural defect and the mastoid cavity. Two patients suffered significant morbidity, and one patient died. Clinical presentation, computed tomographic scan findings, methods of treatment, and avoidance of this complication are discussed.

The long-term outcome after surgical treatment of dorsally exophytic brain-stem gliomas.

Dorsally exophytic brain-stem gliomas represent a distinctive subgroup of pediatric brain-stem neoplasms that are amenable to radical excisi … peration, the tumors filled the fourth ventricle, fungating out of a broad-based area of the dorsal brain stem. The exophytic tumor was resected, but no attempt was made to remove tumor from the brain stem. Histological examination showed that 16 of the tumors were grade I or II astrocytomas, one was a ganglioglioma, and one was an otherwise benign-appearing …

Spinal cord and brain stem anomalies in scoliosis. MR screening of 26 cases.

The spinal cord and brain were examined with magnetic resonance (MR) in 26 patients with idiopathic scoliosis to study the prevalence of spinal cord and brainstem anomalies. Two patients had small centrally located spinal cord syrinxes, one at C6-C8 and the other at T6-T8, without association with any brainstem or cerebellar deformity. The lowest position of the cerebellar tonsils was 0.5 cm below the foramen magnum, which was also seen in 2 other patients. Scoliosis associated with syringomyelia may be more common than previously thought, and may be wrongly classified as idiopathic. A neuroge

Trismus caused by paradoxical activity of jaw-closing muscles in brain stem tuberculoma.

An ipsilateral trismus of jaw-closing muscles during jaw opening is reported in a patient with brain stem tuberculoma. This condition is only superficially simulated by spasticity or tetanus. The decisively differentiating clinical and electromyographic features and the possible pathophysiologic mechanism underlying such trismus are discussed.

Brain stem auditory evoked potentials in patients with the lateral medullary (Wallenberg's) syndrome.

The purpose of this study is to investigate the diagnostic value of using brain stem auditory evoked potentials (BSEP) in patients with the lateral medullary (Wallenberg's) syndrome. The observed changes included: prolongation of the interpeak latency intervals from wave I to V and III to V; prolongation of the interaural latency difference for wave V; splitting or absence of wave III, and the presence of a broad partly fused wave III-V complex. The present study showed that BSEP can be used to confirm the diagnosis of this syndrome and ascertain the site of the lesion.

Effects of substance P on nociceptive and non-nociceptive trigeminal brain stem neurons.

As little information is available on the chemistry of synaptic transmission in trigeminal brain stem nuclei, an iontophoretic study was done on the effects of glutamate and substance P on single neurons in trigeminal nuclei oralis and caudalis in cats anesthetized with chloralose and paralyzed. The neurons were additionally studied for their responses to natural noxious and innocuous cutaneous and intraoral stimuli as well as to bipolar stimulation of the ipsilateral and contralateral canine tooth pulps, the exposed infraorbital and superior laryngeal nerves and forepaw. Glutamate excited all…

[Reticular formation nuclei and bioelectric activity in patients with ischemic brain stem strokes].

The study pertains to new approaches in the study of corrlations between the EEG data and the degree of lesions in the nuclei of the reticular formation in ischemic strokes due to occlusive processes in the vertebrobasillar system. In all cases, because of disorders of vital functions, the authors convened artificial lung ventilation which permitted to prolong the life of the patient and to study the dynamics of the EEG. In 10 lethal cases the brain stem was studied on a series of continuous preparations to determine the degree of lesions in the nuclei of the reticular formation in the pons Va…

Somatosensory cerebral evoked potentials after vascular lesions of the brain-stem and diencephalon.

Clinical and electrophysiological observations are described in 7 patients with clinically well-identified vascular lesions of the brain-stem or diencephalon. In the patients of Group A with a thalamic syndrome, the somatosensory cerebral evoked potentials had a reduced voltage and increased latency on the affected side. No significant anomalies were recorded in the patients of Group B with a Wallenberg or Weber syndrome. In patients of group C with a locked-in syndrome, the cerebral evoked potentials presented marked bilateral anomalies which provided interesting data about the extension of t

Cell Biology Meets Cell Metabolism: Energy Production Is Similar in Stem Cells and in Cancer Stem Cells in Brain and Bone Marrow.

Energy production by means of ATP synthesis in cancer cells has been investigated frequently as a potential therapeutic target in this century. Both (an)aerobic glycolysis and oxidative phosphorylation (OXPHOS) have been studied. Here, we review recent literature on energy production in glioblastoma stem cells (GSCs) and leukemic stem cells (LSCs) versus their normal counterparts, neural stem cells (NSCs) and hematopoietic stem cells (HSCs), respectively. These two cancer stem cell types were compared because their niches in glioblastoma tumors and in bone marrow are similar. In this study, it

Targeted delivery of neural stem cells to the brain using MRI-guided focused ultrasound to disrupt the blood-brain barrier.

Stem cell therapy is a promising strategy to treat neurodegenerative diseases, traumatic brain injury, and stroke. For stem cells to progress towards clinical use, the risks associated with invasive intracranial surgery used to deliver the cells to the brain, needs to be reduced. Here, we show that MRI-guided focused ultrasound (MRIgFUS) is a novel method for non-invasive delivery of stem cells from the blood to the brain by opening the blood brain barrier (BBB) in specific brain regions. We used MRI guidance to target the ultrasound beam thereby delivering the iron-labeled, green fluorescent
