Diseases involving the Golgi calcium pump.

Secretory-pathway Ca2(+)-transport ATPases (SPCA) provide the Golgi apparatus with Ca2+ and Mn2+ needed for the normal functioning of this organelle. Loss of one functional copy of the human SPCA1 gene (ATP2C1) causes Hailey-Hailey disease, a rare skin disorder characterized by recurrent blisters and erosions in the flexural areas. Here, we will review the properties and functional role of the SPCAs. The relationship between Hailey-Hailey disease and its defective gene (ATP2C1) will be adressed as well.

Calcium, vitamin D and involutional osteoporosis.

PURPOSE OF REVIEW: Previous studies suggest that combined calcium and vitamin D supplementation decreases the risk of fractures in older people, particularly those living in care homes, but trials of vitamin D alone in fracture prevention have generated inconsistent results. This review examines the physiological functions of calcium and vitamin D, and the contrasting views of what constitutes an adequate dietary calcium intake and vitamin D sufficiency in adults, and highlights the results of recent large studies of calcium and vitamin D supplementation. RECENT FINDINGS: The RECORD study sho…

Recent advances in physiological calcium homeostasis.

A constant extracellular Ca2+ concentration is required for numerous physiological functions at tissue and cellular levels. This suggests th … ovided new insights into the molecular mechanisms of Ca2+ transport in cells. The G-protein coupled calcium-sensing receptor, responsible for the exquisite ability of the parathyroid gland to respond to small changes in serum Ca2+ concentration was discovered about a decade ago. Research has focussed on the molecular mechanisms determining the serum level …

Calcium-channel modulators for cardiovascular disease.

It is generally accepted that hypertension doubles the risk of cardiovascular disease, of which coronary heart disease is the most common and lethal. Hypertension is a predisposing factor for the development of stroke, peripheral arterial disease, heart failure and end-state renal disease. Atherosclerosis-causing coronary heart disease is related to the severity of hypertension. Inhibition of calcium entry reduces the active tone of vascular smooth muscle and produces vasodilatation. This pharmacological action has been the basis for the use of calcium-channel blockers (CCBs) for the managemen…

[Periorbital swelling caused by carbasalate calcium].

A 61-year-old man presented with visual problems due to a marked two-sided periorbital swelling that had started three years earlier. There was no obvious underlying cause and he had persistent rhinoconjunctival symptoms. At the time the swelling started he was already suffering from xanthelasmata palpebrarum and chronic obstructive pulmonary disease. He had been taking simvastatin and salmeterol-fluticason for a year and carbasalate calcium for two years. There was marked periorbital swelling accompanied by xanthelasmata palpebrarum and slight chemosis and erythema of the eyelids. After exclu…

Vocal fold augmentation with calcium hydroxylapatite.

OBJECTIVES: Voice disorders affect more than 3% of the general population. Vocal fold atrophy is a part of the normal aging process, with up to 60% of 60-year-old individuals displaying evidence of glottal insufficiency. A safe, effective, and durable substance for injection augmentation of the vocal folds is not currently available. The purpose of this investigation was to describe our preliminary experience with calcium hydroxylapatite (CaHA) for vocal fold augmentation. METHODOLOGY: All patients undergoing injection augmentation of the vocal folds with CaHA between January 1, 2002 and June…

Perinatal calcium metabolism: physiology and pathophysiology.

Disturbances in mineral homeostasis are common in the neonatal period, especially in premature infants and infants who are hospitalised in an intensive care unit. In many cases these disturbances are thought to be exaggerated responses to the normal physiological transition from the intrauterine environment to neonatal independence. By contrast, some disturbances in calcium homeostasis are the result of genetic defects, which in many instances can now be identified at the molecular level. In other cases hypocalcaemia or hypercalcaemia may result from pathological intrauterine conditions, birth…

[Calcium pyrophosphate dihydrate-crystal induced arthropathy].

Calcium pyrophosphate dihydrate (CPPD) crystal induced arthropathy (CPPD-CA; systemic articular chondrocalcinosis) characterized by deposition of CPPD-crystals in fibro- and hyaline cartilage, joint capsule and periarticular tissues is associated with a variable clinical spectrum of inflammatory degenerative and occasionally destructive joint and vertebral manifestations including neurologic complications and rarely tophaceous-tumoral tissue calcifications. Microcrystal formation in the extracellular matrix of cartilage and tendons is based on genetic and acquired dysregulation of chondrocyte …

Calcium current subtypes in GnRH neurons.

Calcium plays roles in excitability, rhythm generation, and neurosecretion. Identifying channel subtypes that regulate calcium influx is thus important to understanding rhythmic GnRH secretion, which is a prerequisite for reproduction. Whole-cell voltage-clamp recordings were made from short-term dissociated GnRH adult ovariectomized (OVX) mice (n = 21) to identify channel subtypes that carry calcium current using selective channel blockers and voltage characteristics. Low-voltage activated (LVA) currents were not observed in 42 GnRH neurons tested, although most non-GnRH neurons (4/6) display…

Inhibitors of articular calcium crystal formation.

The coexistence of calcium phosphate and calcium pyrophosphate crystals in osteoarthritis is a well-described phenomenon. In addition to deposition in articular cartilage, the crystals induce numerous changes to biochemical parameters in the surrounding fluid. This survey discusses crystal-inhibitor interactions whereby defining the molecular structure of crystals assists in an optimal inhibitor design. Crystal growth studies in the presence and absence of a test compound can generate data on crystal face changes. Together with knowledge of the inhibitors' molecular structure, computer modelin…

Calcium pyrophosphate dihydrate crystal deposition disease.

Calcium pyrophosphate dihydrate (CPPD) crystal deposition disease is characterized by the accumulation of pyrophosphate dihydrate crystals in articular and periarticular tissues. Various terms have been utilized to describe this arthropathy, which has led to some confusion. CPPD crystal deposition disease is among many conditions that may result in crystal deposition within cartilage. Chondrocalcinosis is a pathologic and radiographic term denoting calcification of cartilage within joints including both hyaline articular cartilage and fibrocartilage. Pseudogout is a clinical term applied to an…

[Calcium-sensing receptors: physiology and pathology].

The near constancy of extracellular calcium concentration is required for the numerous physiological functions of extra- and intracellular calcium. This implies that any change in extracellular calcium concentration must be detected in order to allow the appropriate correction by the homeostatic systems. The identification and cloning of a calcium-sensing receptor (CaR), which is expressed in the plasma membrane of parathyroid cells as well as many other cell types, has been a major advance in the understanding of the mechanisms involved in the control of extracellular calcium concentration. I…

Calcium requirements of physically active people.

Dietary calcium and physical activity have been independently, but inconsistently, associated with the development of increased peak bone mass and reduced bone loss later in life. An examination of the literature points to important effects of dietary calcium on bone health. During the development of peak bone mass, calcium intakes of <1 g/d are associated with lower bone mineral density. At intakes approaching calcium requirements, physical activity is a more important predictor of bone mineral density than is calcium intake. In studies of postmenopausal women, calcium intakes of 1 g (25 mmol…

Intestinal calcium absorption from mineral water.

Little is known about the bioavailability of calcium in water and various beverages. Some mineral waters contain large amounts of calcium that could compensate for insufficient consumption of dairy products. The fractional intestinal absorption of calcium (FCA) was measured in 12 healthy adult volunteers, using a trace dose of radiocalcium and 200 mg of calcium carrier, part of which was calcium contained in mineral water. Measurements were performed in fasting subjects consuming a standard breakfast. In all the subjects, three mineral waters with a calcium concentration of 10.4, 78, and 467 m…

Defective calcium transport in vitiliginous melanocytes.

Melanocytes were successfully established from involved and uninvolved skin of a patient with acute acrofacial vitiligo. Cells from involved epidermis showed a fivefold decrease in the rate of radiolabelled 45Ca uptake compared with uninvolved and control cells. These results are similar to previous findings in keratinocytes from involved skin in patients with vitiligo. Since 6-biopterin is cytotoxic to melanocytes and calcium controls the redox status of the 6-biopterin/(6R)5, 6, 7, 8-tetrahydrobiopterin equilibrium via the thioredoxin reductase/thioredoxin system, these results underline the…

[Renal protective effects of calcium antagonists].

Calcium antagonists (C-A) induce different, renal hemodynamic alterations. They reduce afferent arteriolar resistance and do not change glomerular filtration rate. C-A prevent acute renal failure, especially after kidney transplantation. Calcium antagonists decrease nephrotoxicity of cyclosporine and radiocontrast media. C-A are recommended in the therapy of renovascular hypertension.

Calcium channel blockers for anxiety disorders?

The role of calcium in the etiology of anxiety has been proposed for several decades. Calcium channel blockers profoundly influence calcium metabolism and the transport of calcium. Even though the evidence for the role of calcium remains weak, drugs affecting calcium might be useful in the treatment of anxiety disorders. One of these compounds, verapamil, has been used to treat mood disorders. Calcium channel blockers have also been tried in other indications such as premenstrual syndrome, irritable bowel syndrome, schizophrenia, tardive dyskinesia, and Tourette's syndrome. However, the number…
