High rate of chromosomal abnormalities in HTLV-I-infected T-cell colonies derived from prodromal phase of adult T-cell leukemia: a study of IL-2-stimulated colony formation in methylcellulose.

To detect chromosomal abnormalities in prodromal phase of adult T-cell leukemia (ATL), we established a clonal culture method for human T-lymphotropic virus type I (HTLV-I) infected T-cells in methylcellulose containing recombinant human interleukin 2 (rhIL-2). We tried to analyze chromosomes of 187 colonies (4, 23, 69, 74, and 17, from HTLV-I-uninfected normal T-cells, HTLV-I-Infected normal T-cells, HTLV-I carriers, smoldering ATL, and chronic ATL, respectively), using chromosomal banding methods. In the prodromal group, 53% of colonies (84/160) (36/69, 37/74, 11/17 in HTLV-I carriers, smold

Colonized Silence: Confronting the Colonial Link in Rural Alaska Native Survivors' Non-Disclosure of Child Sexual Abuse.

Though there is evidence linking the history of colonialism and oppression of Indigenous people to the high rates of rape and child sexual abuse experienced by this population today, it is less understood how colonial processes, past and present, condition the decision to disclose or report victimization. Drawing on a survivorship storytelling study of rural Alaska Native survivors of child sexual abuse, this paper underscores the importance of centering colonialism in understanding the culture of silence surrounding sexual victimization among Indigenous people. Results show that reasons for n…

Small colony variants and cefiderocol resistance in clinical Escherichia coli: an in vitro mechanistic study.

BACKGROUND: The emergence of cefiderocol resistance in Escherichia coli poses a threat to this last line antibiotic against carbapenem-resistant infections. This study aimed to induce and characterize the mechanisms of cefiderocol resistance in vitro. METHODS: Cefiderocol resistance was induced in four clinical E. coli strains (carrying bla NDM-5, bla KPC-2, bla OXA-48, bla IMP-4) and ATCC 25922 via serial passaging in increasing drug concentrations. Resistant strains were subjected to whole-genome sequencing (WGS) and RNA sequencing (RNA-Seq) to identify genetic determinants. Phenotypic chan

The Role of Granulocyte Colony-Stimulating Factor in Endometrial Preparation for Embryo Implantation in In Vitro Fertilization.

Granulocyte colony-stimulating factor (G-CSF) has been suggested as a supplementary approach for endometrial preparation in IVF. Clinical results continue to be inconsistent. This narrative review synthesises molecular and clinical information to elucidate the function of G-CSF in modifying endometrial receptivity and to identify patient categories most likely to benefit. A thorough assessment was conducted on published research on G-CSF administration in women with treatment-resistant thin endometrium, recurrent implantation failure, and unselected IVF populations. The research demonstrates t

Skeletal and body composition effects of orchiectomy in a colony of diet-induced obese cynomolgus macaques.

Obesity is a leading cause of secondary hypogonadism or low testosterone. Conversely, hypogonadism frequently leads to excess body fat/obesity demonstrating a potentially reciprocal relationship between the two. This study describes the impact of bilateral orchiectomy (ORX) in a colony of diet-induced obese (DIO) male Mauritius cynomolgus macaques (DIO-CM) maintained on a Western Style Diet (WSD). Body composition analysis via DXA was performed prior and up to 15 m postsurgery to determine the long-term changes in hypogonadism. As expected, ORX significantly reduced testosterone, inhibin B, an

Protective effect of granulocyte colony stimulating factor on cyclophosphamide-induced prostate injury in rats.

Cyclophosphamide (CPA) is a commonly used chemotherapeutic agent whose metabolites readily accumulate in male reproductive organs such as the testes and epididymis, exhibiting reproductive toxicity. Granulocyte colony-stimulating factor (G-CSF) possesses anti-inflammatory and antioxidant effects, but its protective mechanism against CPA-induced prostate injury remains unclear. This study aimed to investigate the protective effect of G-CSF against CPA-induced prostate injury in rats. Thirty SPF male Sprague-Dawley (SD) rats were randomly divided into a control group (C group), a CPA model group

Semaglutide-associated agranulocytosis requiring hospitalisation and granulocyte colony-stimulating factor: a case report.

Semaglutide, a glucagon-like peptide-1 receptor agonist (GLP-1RA), is widely used in obesity and type 2 diabetes. Although generally well tolerated, severe haematological adverse reactions remain poorly characterised. A 48-year-old woman treated with subcutaneous semaglutide for obesity developed febrile agranulocytosis with an absolute neutrophil count of 0.0×10⁹/L after several months of therapy. Baseline blood counts had been normal prior to treatment. Bone marrow aspirate and biopsy showed preserved cellularity with granulocytic maturation arrest at the promyelocyte/myelocyte stage. Semagl

Spatial constraints determine the spread of plasmid-encoded antibiotic resistance between bacterial colonies.

Plasmid transfer among bacteria is an important driver of the spread of antibiotic resistance. Surface-associated bacterial biomass is a hotspot for plasmid transfer due to the dense spatial packing of cells, but this biomass is often sparse (composed of discrete bacterial colonies). Compared to plasmid dynamics within a single colony, the determinants of plasmid transfer between discrete colonies are less understood. Yet, colonies routinely physically collide with each other as they grow and expand across surfaces. Here, we experimentally demonstrate that collisions between colonies of Stutze

Frequent Queen Replacement and Presence of Unrelated Queens in Colonies of a Functionally Monogynous Ant Species.

In eusocial insects, social parasitism-the exploitation of the host's brood care behavior for survival and reproduction-can occur either within or between species. Parasitic queens invade host nests and aggressively replace the resident queen. While the adoption of conspecific queens is a common feature of species with multiqueen colonies (polygyny), the origin of parasitic founding is not fully understood. Functionally monogynous ants, in which nestmate queens establish social and reproductive hierarchies through biting and antennal boxing, may provide a link between peaceful adoption and soc

Granulocyte colony-stimulating factor (G-CSF)-induced aortitis in a woman with breast cancer.

Granulocyte colony-stimulating factor (G-CSF) is used alongside cytotoxic chemotherapy regimens to reduce the severity and duration of neutropenia and decrease infection risk. Here, we add to the growing body of evidence that aortitis can be considered a recognised side effect of G-CSF therapy.A woman in her 60s with breast carcinoma treated with G-CSF cover alongside adjuvant chemotherapy presented with intermittent fever and epigastric pain. There were no localising signs of infection. Results demonstrated raised inflammatory markers. The patient did not respond to antibiotics, and imaging s

Neuroprotective effects of granulocyte colony-stimulating factor against tramadol-induced cerebellar neurotoxicity.

BACKGROUND: Tramadol (TRM) is a centrally acting synthetic opioid and serotonin/norepinephrine reuptake inhibitor. Despite being a potent painkiller, long-term use can induce permanent neurotoxicity. Granulocyte colony-stimulating factor (G-CSF) is a cytokine that helps to mobilize stem cells and facilitate their integration over injured neurons. AIM: This work aims to study the histopathological, biochemical, and molecular alterations in the cerebellar cortex induced by TRM in comparison to the postulated protective effect of G-CSF versus TRM withdrawal. METHODS: 32 adult male albino rats w

Granulocyte Colony-Stimulating-Factor-Producing Bladder Carcinoma: A Case Report and Literature Review.

Granulocyte colony-stimulating factor (G-CSF)-producing bladder carcinoma has an aggressive clinical course. We report a case of G-CSF-producing bladder carcinoma in a 78-year-old Japanese man who had a bladder tumor with a diameter of 38 mm. Transurethral resection of the bladder tumor was performed. Pathological examination revealed a high-grade muscle-invasive urothelial carcinoma (pT2). The patient had three courses of neoadjuvant chemotherapy with a combination of gemcitabine and carboplatin and thereafter underwent robot-assisted radical cystectomy. The surgically resected bladder specim

Colonial harm in new packaging: Indigenous critiques of the tobacco industry's 'harm reduction' rhetoric.

Over the last 75 years, we have learned that commercial tobacco use causes widespread disease and death. However, the commercial Tobacco and Nicotine Industry continues to promote, market, and sell tobacco and nicotine products to protect and expand profit. This reflects their legal obligation to act in shareholders' best interests. While the Tobacco and Nicotine Industry heavily promotes alternative products such as electronic cigarettes and nicotine pouches, for now, these represent a relatively small share of profits compared with combustible cigarette sales. The continued reliance on and e

Distinct osteoclastogenic potential of granulocyte-macrophage colony-stimulating factor-induced monocyte subsets.

BACKGROUND: Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by synovial inflammation and progressive bone destruction. Although osteoclasts mediate bone resorption in RA, recent evidence suggests that inflammatory osteoclasts differ from physiological osteoclasts in various aspects, including their progenitor origins. This study aimed to compare the osteoclastogenic potential of monocyte-derived dendritic cells (moDCs) and other granulocyte-macrophage colony-stimulating factor (GM-CSF)-induced cells to identify potential progenitor populations involved in inflammatory b

Generating Natural Neural Response in Visual Neuroprosthetics Using the Artificial Bee Colony Algorithm.

A significant challenge that hampered visual prosthetic systems is its ability to produce a meaningful perception of vision. It is hypothesised that mimicking the natural spiking pattern of the visual cortex would lead to a more natural perception of vision. In this study, an optimisation algorithm, namely the Artificial Bee Colony (ABC) algorithm, is utilised to automatically generate a stimulus waveform to produce a response that best matches a target neural pattern. An in silico model of realistic retinal ganglion cells (RGC) was used to simulate neural responses in a visual prosthesis and
