Changes in p53 and cyclin D1 protein levels and cell proliferation in different stages of human esophageal and gastric-cardia carcinogenesis.

The objective of this study was to quantify the changes in p53 and cyclin D1 protein levels in different stages of human esophageal and gastric cardia carcinogenesis in a high-risk population in Henan, China. Immunoreactivity of p53, cyclin D1 and proliferating-cell nuclear antigen (PCNA) was observed in the cell nuclei of esophageal and gastric cardia biopsies. The number of p53-immunostaining-positive cells was low in normal epithelia, slightly increased in basal-cell hyperplasia (BCH), markedly increased in dysplasia (DYS) (10-fold), and further increased in squamous-cell carcinoma (SCC) (4

Raman spectroscopy reveals a novel mechanism by which cyclin Y promotes breast cancer progression by regulating lipid metabolism.

Raman spectroscopy is a technique for disease diagnosis and treatment evaluation through directly measuring the molecular features of cellular metabolites at the single-cell level. Nevertheless, the application of Raman spectroscopy in studying the mechanisms of breast cancer remains limited. Cyclins are critical regulators of cell proliferative activity that are often dysregulated in cancer. Although cyclin Y (CCNY), as a key cyclin, is increasingly recognized as an oncogene in various cancers, including lung, renal, hepatocellular, ovarian and laryngeal cancers, CCNY, which is essential for …

Cyclin D1 and the resistance landscape of HIF2 inhibition in clear-cell renal cell carcinoma: Insights from a mini-review.

Clear-cell renal cell carcinoma (ccRCC) is primarily driven by VHL inactivation and subsequent stabilization of hypoxia-inducible factor 2α (HIF2α). The recent approval of the HIF2 inhibitor belzutifan represents a landmark advance, yet both primary and acquired resistance remain major barriers. In this mini-review, we summarize emerging insights into the molecular basis of HIF2 inhibitor response and resistance, with a particular focus on Cyclin D1 as a key downstream effector linking HIF2 signaling to cell-cycle progression. Persistent Cyclin D1 expression, as well as functional compensation

Deciphering the role of Ki67: prognostic insights in advanced breast cancer therapy with Cyclin-dependent kinase 4/6 inhibitors.

BACKGROUND: Cyclin-dependent kinase 4/6 inhibitors (CDK4/6i) are the standard of care in advanced breast cancer. While Ki67 is prognostic in early-stage disease, including adjuvant CDK4/6i, its role in metastatic setting is unclear. This study assessed Ki67’s prognostic value in advanced breast cancer treated with CDK4/6i. METHODS: We conducted a retrospective analysis of patients with advanced breast cancer receiving CDK4/6i at our cancer center between 2018 and 2023. RESULTS: The study included 368 patients with Ki67 results. The median age was 63 years (IQR 52–70.5). 283 patients (76.9%) re

Cyclin E1 Expression Predicts Poorer Outcome in HER2-positive Breast Cancer with Residual Disease after Neoadjuvant Chemotherapy.

BACKGROUND: Among HER2-positive breast cancer patients, those who do not achieve pathological complete response (non-pCR) after neoadjuvant chemotherapy (NAC) generally have poorer outcomes. Although additional postoperative therapies have been developed, it remains unclear which patients truly require treatment intensification. This study aimed to identify prognostic factors in non-pCR patients. METHODS: Of 283 patients with HER2-positive invasive breast cancer treated with NAC, including anti-HER2 therapy between 2006 and 2023, 174 were classified as non-pCR. Of these, 37 patients developed

Impact of COX-2 inhibition by DuP-697 on cyclin B1/Cdc2 dynamics in human medullary thyroid carcinoma cells.

BACKGROUND: Cyclooxygenase-2 (COX-2) and its product prostaglandin E₂ (PGE₂) are key mediators of parafollicular cell proliferation and calcitonin secretion in medullary thyroid carcinoma (MTC). Although COX-2 inhibitors exhibit anti-proliferative effects, the molecular mechanisms underlying their influence on cell-cycle regulation remain unclear. OBJECTIVE: This study explored the effects of the selective COX-2 inhibitor DuP-697 on proteins associated with G₂/M regulatory signaling in human medullary thyroid carcinoma TT cells. METHODS: TT cells were treated with DuP-697 (40-160 nM) for 3-7

Inhibition of cyclin-dependent kinase 8 modulates chondrocyte transition to hypertrophy and provides in vivo protection against spontaneous osteoarthritis.

Chondrocyte hypertrophy is a key hallmark of osteoarthritis (OA) that drives a multitude of whole-joint disease processes. There are no approved disease-modifying pharmaceuticals in clinical use, emphasising the urgent need for innovative therapeutics. Screening of a focussed chemical library using a novel in vitro platform discovered that inhibition of CDK8/19 mediator kinases modulated chondrocyte behaviours with potential to limit OA hallmarks. Transcriptomic analysis of various chondrogenic cell populations revealed that CDK8/19-inhibitor treatment upregulated pro-anabolic/anti-catabolic m

Absent Cyclin D1 Expression in Myeloid Sarcomas Distinguishes From Malignant Histiocytic Neoplasms: When Morphologic Ambiguity is Deceptive.

Myeloid sarcoma (MS) is a tumor mass involving any extramedullary site composed of myeloid-lineage blasts with myeloid, myelomonocytic, or monocytic differentiation. Distinguishing MS from malignant histiocytic neoplasms (MHNs) is diagnostically challenging due to morphologic and immunophenotypic overlap, particularly for MS with monocytic differentiation, which can mimic MHNs, especially the subtypes of histiocytic sarcoma and interdigitating dendritic cell sarcoma, leading to potential misdiagnosis and inappropriate management. Cyclin D1 overexpression is implicated in MHNs, but its utility

Cyclin-dependent kinase 4 and 6 inhibitors and the breast cancer immune ecosystem: immune remodeling, resistance, and therapeutic reprogramming.

Cyclin-dependent kinase 4 and 6 inhibitors (CDK4/6 inhibitors) combined with endocrine therapy have become a therapeutic backbone for hormone receptor-positive, human epidermal growth factor receptor 2-negative breast cancer, yet durable disease control is frequently limited by intrinsic and acquired resistance. Canonical tumor-cell mechanisms, including retinoblastoma-pathway escape, cyclin E-cyclin-dependent kinase 2 (CDK2) activation, endocrine adaptation, and phosphoinositide 3-kinase (PI3K)-AKT-mechanistic target of rapamycin (mTOR) signaling, explain only part of this failure because the

Functional insight into cyclin-dependent kinase (CDK)7 via chemical inhibition of the priority fungal pathogen Cryptococcus neoformans.

UNLABELLED: Cyclin-dependent kinase (CDK)7 regulates the cell cycle and transcription in higher eukaryotes, but its function and the antifungal potential of CDK7 inhibitors remain unexplored in fungal pathogens. CDK7 inhibitors were identified to probe CDK7 function in the priority fungal pathogen, Cryptococcus neoformans (Cn). The CDK7 ortholog in Cn was tagged with mNeonGreen and shown to form a nuclear-localized, CDK-activating kinase (CAK) complex with cyclin H and assembly factor Mat1. The pulled-down CAK complex was demonstrated to be active and was used to identify several human CDK7 in

Metastatic Mammary-Like Adenocarcinoma of the Vulva Responding to Cyclin-Dependent Kinase (CDK) Inhibitor: A Case Report and Literature Review.

Mammary-like adenocarcinoma originating in the vulva is extremely rare, even rarer when metastasized at diagnosis. It may arise from the primitive mammary ridge or mammary-like glands of the skin in the anogenital region. We report a case of a 47-year-old woman with diffuse bone metastasis and a vulvar lesion compatible with mammary-type adenocarcinoma, with no evidence of primary breast cancer. Systemic treatment followed protocols for hormone receptor (HR)-positive, human epidermal growth factor receptor 2 (HER2)-negative advanced breast cancer, using an aromatase inhibitor and ribociclib, r

Gallstone Ileus in a 25-Year-Old Female With Cyclin-Dependent Kinase-Like 5 Deficiency Disorder: A Case Report.

Gallstone ileus is an uncommon complication of cholelithiasis and the delayed diagnosis may be associated with increased risk of mortality. When gallstones block the cystic duct, they can lead to cholecystitis. If a fistula forms between the inflamed gallbladder and the adjacent intestine, the gallstones may pass into the intestinal tract and cause obstruction in the intestine. We report a case of 25-year-old female with developmental and epileptic encephalopathy who was intraoperatively diagnosed with gallstone ileus during surgery for small bowel obstruction of unknown origin. The patient ha

Cyclin-dependent kinase 4/6 inhibitors and cardiotoxic events in breast cancer: A pharmacovigilance study based on the FAERS database.

Recent trials have highlighted the cardiotoxicity of ribociclib, a CDK4/6 inhibitor, particularly its association with QT prolongation. However, studies on the link between CDK4/6 inhibitors and cardiotoxic events show inconsistent results, and the factors influencing these events and related drug interactions remain underexplored. To address these uncertainties, our study utilizes the FDA adverse event reporting system database (Q1 2015 to Q1 2024) to examine the cardiotoxic events of CDK4/6 inhibitors in breast cancer patients. We employed a comprehensive analytical framework, applying dispr
