Unveiling Clinostomum spp. In farmed nile tilapia from upper Egypt: morphological clues and molecular evidence.

BACKGROUND: Clinostomum species (spp.) are significant fish-borne zoonotic helminths worldwide, causing yellow grub disease which diminishes the quality, marketability, and productivity of infected fish stocks. Despite their veterinary and public health relevance, detailed molecular investigations of Clinostomum spp. infecting Nile tilapia in Egypt remain limited, especially in Upper Egypt. A total of 400 Nile tilapias were collected from fish farms in Sohag Governorate, Upper Egypt, and initially screened for encysted metacercariae through visual inspection and subsequent light microscopic ex

Redescription of the Larval Trematode Taxon Monilicaecum ventricosum (Hemiuroidea: Didymozoidae) from Type Host Cololabis saira, with Inference of the Definitive Host Based on DNA Analysis.

The larval trematode taxon Monilicaecum ventricosum Yamaguti, 1942 (Didymozoidae) was redescribed from its type host, the Pacific saury Cololabis saira (Brevoort, 1856), collected from the North Pacific off Eastern Hokkaido, Japan. Because didymozoid larvae and adults differ markedly in morphology, linking these two stages using morphological data alone has traditionally been difficult. Consequently, the monotypic genus Monilicaecum Yamaguti, 1942 is currently regarded as a collective group, similar to genera established solely on larval forms of other trematodes. However, recent advances in m

The faecal egg count reduction test: Will identification of larvae to species improve its utility?

In the faecal egg count reduction test, visual identification of larvae cultured from faeces enables the egg counts to be apportioned to species/genera, resulting in a more accurate test. However, morphology cannot reliably differentiate some species meaning that, in some cases, efficacy can only be estimated at the genus or species-complex level. We investigated the benefits of identifying larvae to species using DNA to determine how often this would alter the diagnosis of resistance and whether increasing the number of larvae identified would alter the repeatability of an efficacy estimate.

First report of Saccocoelioides nanii (Digenea: Haploporidae) infecting an extremophile host, Jenynsia sulfurica (Cyprinodontiformes: Anablepidae).

A species of Saccocoelioides (Haploporidae) is reported from the intestine of the livebearer fish Jenynsia sulfurica (Anablepidae), which inhabits an extremophile environment in northwestern Argentina. This host species lives in a sulfide-rich thermal stream characterized by several extreme environmental conditions, including high temperatures, low oxygen levels, moderate pH, and elevated total dissolved solids. Morphological and molecular analyses indicate that the parasite under study is consistent with Saccocoelioides nanii. Morphological comparisons revealed minor variations in body size a

Species-specific detection of Schistosoma japonicum using the 'SNAILS' DNA-based biosensor.

The neglected tropical disease schistosomiasis continues to be a global health concern, especially in low- and middle-income countries, with at least 250 million people infected worldwide and a further 779 million at risk of infection. Schistosoma japonicum, which is found in parts of South Asia, causes intestinal schistosomiasis in humans, as well as infecting up to forty other mammalian species. Therefore, novel diagnostics that can detect S. japonicum are desirable. In this study, we have further developed and refined the 'SNAILS' DNA-based biosensor technology for the detection of schistos

Extensive Countrywide Molecular Identification and High Genetic Diversity of Haemonchus spp. in Domestic Ruminants in Greece.

The gastrointestinal nematode parasite Haemonchus spp. is one of the most pathogenic parasites of ruminants, due to its blood-sucking activity, which causes large economic losses in the ruminant industry. The latest epizootiological data recorded an increase in the infection, not only in Greece but also in other countries, mainly attributed to climatic changes. The study of the population structure and the investigation of the phylogenetic relationships of Haemonchus spp. are essential for the understanding of its biology and epizootiology to implement appropriate control and prevention strate

The variety of abomasal nematode communities of captive and free-roaming populations of European bison, Bison bonasus (L.): a morphometric and molecular approach.

Most studies concerning parasitic infections in European bison have been performed on free-ranging animals: comparatively little is known about the abomasal nematodes of captive wisents, which are widely used in reintroduction programmes. The aim of the study was to determine the infection level and species composition of abomasal nematodes in captive European bison in enclosures (including zoos) and breeding centres compared to free-ranging individuals. It also includes a morphological analysis of the parasites based on figures and measurement data. Altogether, 11 species of nematodes were de

Laboratory and field validation of the recombinase polymerase amplification assay targeting the Schistosoma mansoni mitochondrial minisatellite region (SmMIT-RPA) for snail xenomonitoring for schistosomiasis.

Improvements in diagnostics for schistosomiasis in both humans and snail hosts are priorities to be able to reach the World Health Organization (WHO) goal of eliminating the disease as a public health problem by 2030. In this context, molecular isothermal amplification tests, such as Recombinase Polymerase Amplification (RPA), are promising for use in endemic areas at the point-of-need for their accuracy, robustness, simplicity, and time-effectiveness. The developed recombinase polymerase amplification assay targeting the Schistosoma mansoni mitochondrial minisatellite region (SmMIT-RPA) was u

Echinococcus granulosus Sensu Stricto and Echinococcus multilocularis in a Gray Wolf (Canis lupus) in Turkey: Further Evidence for Increased Risk of Alveolar Echinococcosis in Urban Areas.

OBJECTIVE: The aim of this study was to identify Echinococcus species by morphological and molecular means. METHODS: A dead gray wolf (Canis lupus) was found near Erzurum province and brought to the parasitology laboratory. Sedimentation and counting technique (SCT) and polymerase chain reaction (PCR) analysis were conducted. RESULTS: The SCT implications indicated that the wolf had a substantial worm burden (62,720 and 49,280 parasites) due to a co-infection of E. granulosus s.l. and E. multilocularis. Genus/species-specific PCR was used to analyze DNA extracted from adult worms and confirm

Gray foxes (Urocyon cinereoargenteus) as natural definitive hosts of Paragonimus mexicanus.

Paragonimus mexicanus is a trematode that causes pulmonary and extrapulmonary infections in humans, characterized by chest pain, dyspnea, fever, and weight loss. The detection of Paragonimus spp. is primarily achieved through the microscopic observation of eggs in feces, sputum, and pleural fluid. Paragonimus mexicanus has been found in various wild animals, including dogs, cats, raccoons, and opossums. Although the reservoirs of P. mexicanus in Costa Rica are unknown, this study analyzed fecal samples from gray foxes (Urocyon cinereoargenteus) using microscopic and molecular methods. In the m

A Molecular Phylogenetic Study of the Eye Fluke Philophthalmus lacrymosus (Trematoda: Philophthalmidae) Found in Larus dominicanus (Aves: Laridae) from Brazil.

PURPOSE: Species of the genus Philophthalmus are eye flukes with a complex taxonomy, which began to be improved with the help of molecular data only recently. However, most described species have never been placed into a phylogenetic context. In this study, eye flukes previously found on kelp gulls, Larus dominicanus, from Brazil and identified as Philophthalmus lacrymosus were subjected to molecular analysis. METHODS: For the molecular analyses, we analyzed parasites found in six infected gulls (one worm per bird) collected from different municipalities of the state of Santa Catarina, Brazil

Molecular evidence of new freshwater turtle blood flukes (Digenea: Spirorchiidae) in the intermediate snail host Biomphalaria occidentalis Paraense, 1981 in an urban aquatic ecosystem in Brazil.

In this study, two potentially new species of turtle blood flukes (TBFs) (Digenea: Spirorchiidae) have been recorded from South Brazil. The spirorchiid parasites infect the vascular system of turtles, thereby compromising their health. The life cycle of these parasites is not well studied. The larval stage of cercaria is found in intermediate gastropod hosts, with some species presenting similar morphological characteristics, which can result in misinterpretations when using only morphological taxonomy for species identification. In this study, we recorded a single morphotype belonging to the

A new species of Auriculostoma (Trematoda: Allocreadiidae) from the intestine of Brycon guatemalensis (Characiformes: Bryconidae) from the Usumacinta River Basin, Mexico, based on morphology and 28S rDNA sequences, with a key to species of the genus.

We describe a new species of Auriculostoma Scholz, Aguirre-Macedo & Choudhury, 2004 based on several sources of information including morphology (light and scanning electron microscopy [SEM]), sequences of two nuclear genes, host association, and geographical distribution. Morphologically, the new species most closely resembles Auriculostoma astyanace Scholz, Aguirre-Macedo & Choudhury, 2004, but differs by having deeply lobated testes and cirrus-sac extending posteriorly to seminal receptacle level. Auriculostoma lobata n. sp. can be readily distinguished from all the other congeners by the c

Avian Schistosomes from the South American Endemic Gastropod Genus Chilina (Pulmonata: Chilinidae), with a Brief Review of South American Schistosome Species.

Our current knowledge of avian schistosomes from South America is scarce in all respects, including species and generic diversity, their life cycles, patterns of host use, potential to cause dermatitis outbreaks, and evolutionary affinities. As a step towards addressing this shortcoming, the goal of this study was to provide discrete reference points relating to snail hosts, locality records, morphological attributes, sequence for nuclear 28S and ITS, and partial mitochondrial cox1 genes, and phylogenetic relationships for schistosome cercariae recovered from different species of Chilina, whic

A diagnostic study of Echinococcus multilocularis in red foxes (Vulpes vulpes) from Great Britain.

Alveolar echinococcosis is caused by a parasitic tapeworm Echinococcus multilocularis and is a serious disease with high fatality in humans. The definitive primary host is the red fox (Vulpes vulpes) but domestic animals (dogs and to a lesser extent cats) as well as several genera of rodents can also be infected with the parasite. There is, to date, no evidence of indigenous cases of E. multilocularis in Great Britain (GB) but in most of continental Europe the parasite is considered to be endemic and/or slowly spreading. All pet dogs entering the United Kingdom (UK) under the pet travel scheme

Comparison of conventional versus real-time PCR detection of Brugia malayi DNA from dried blood spots from school children in a low endemic area.

Microscopic detection of active phase of lymphatic filariasis is indicated by the presence of microfilaria in whole blood. This method is not sensitive and requires relatively large amount of blood sample. PCR allows very sensitive detection of the parasite DNA using a smaller amount of blood; and the use of dried blood spots facilitates sample transportation. Nevertheless, limited studies have been reported on PCR using dried blood spot for detection of Brugia malayi. In this study, we investigated the effects of concentrating whole blood genomic DNA sample and the amplification methods [conv

[Screening of mimic antigen epitopes of Echinococcus granulosus by phage-displayed peptide library].

The 7-mer phage random peptide libraries was screened with the specific anti-EgB polyclonal antibody. Sixty clones picked randomly were deduced by DNA sequencing and their amino acid sequences were compared with EgB. After 5 rounds of panning, the phage recovery rate increased from 4.15 x 10(-5) to 4.30 x 10(-2). 45 out of 60 selected clones were ELISA positive by anti-EgB polyclonal antibody with a positive rate of 75%, 45 positive clones were sequenced and the predicted amino acid sequences showed low homology to EgB. The reactivity of selected peptide and sera from patients of anaphylactic

Molecular epidemiology of Schistosoma mansoni: a robust, high-throughput method to assess multiple microsatellite markers from individual miracidia.

Schistosomiasis is one of the major unconquered infectious diseases afflicting people of developing countries, particularly in Africa. A deeper understanding of the epidemiology of schistosomes is complicated by the intravascular location of adult worms which makes them routinely unavailable for study. Their progeny, miracidia, which are hatched from eggs that are passed in feces, are available and can provide valuable insights about human infections, but they are small in size, hindering robust molecular analyses. Here we present a new high-throughput technique to assess the genotypes at 21 p

Pool screen polymerase chain reaction for estimating the prevalence of Onchocerca volvulus infection in Simulium damnosum sensu lato: results of a field trial in an area subject to successful vector control.

Detection of infective parasites in the vector population can be an early indicator of recrudescence in areas freed of new cases of onchocerciasis. However, dissection of vector black flies is inefficient in areas subject to effective control. Recently, a polymerase chain reaction (PCR)-based assay has been used to detect a single Onchocerca volvulus-infected black fly in pools containing large numbers of uninfected flies. This method had not been validated on wild-caught black flies in an area subject to effective vector control. Here, we report a method of restricting the pool screen PCR ass
