Effects of potassium bromide disk formation on the infrared spectra of dried model proteins.

The most common method of sample preparation for Fourier transform infrared spectroscopic analysis of proteins in the solid state is compression after mixing with potassium bromide (KBr). Recently, questions have arisen as to whether proteins are conformationally altered by this process. In this study, the amide I Fourier transform infrared spectra of two model proteins, lysozyme and alpha-chymotrypsinogen, were measured before and after compression in KBr, and the effects of moisture exposure and the ratio of KBr to protein were examined. Contrary to earlier reports, compaction of the KBr/pro

Replantation of an avulsed incisor after prolonged dry storage: a case report.

Management of tooth avulsion in the permanent dentition often presents a challenge. Definitive treatment planning and consultation with specialists is seldom possible at the time of emergency treatment. Replantation of the avulsed tooth can restore esthetic appearance and occlusal function shortly after the injury. This article describes the management of a child with an avulsed maxillary permanent incisor that had been air-dried for about 18 hours. The replanted incisor retained its esthetic appearance and functionality 2 years after replantation, yet the long-term prognosis is not good becau

Revision myringoplasty with solvent-dehydrated human dura mater (Tutoplast).

OBJECTIVES: Several grafting materials have been used in the reconstruction of the perforations of the tympanic membrane. The use of dehydrated dura homografts as a substitute for the tympanic membrane has not been fully documented. On the other hand, revision surgery is more critical in terms of patients' expectations. The aim of this study is to determine the tissue stiffness, the compliance, and the wound healing rate of dura homografts in revision surgery of perforations of the tympanic membrane. STUDY DESIGN AND SETTING: The charts of 45 patients who have received dura grafts because of

Multifaceted approach to the impact of drying techniques on Lycium barbarum polysaccharide yield, quality, and bioactive retention: insights from HPLC monosaccharide quantification.

Lycium barbarum (goji berry, LB) is a nutrient-dense fruit that requires drying for preservation, but this process can degrade its bioactive compounds. This study compares the effects of hot-air drying (HAD), infrared drying (IRD), and pulsed vacuum drying (PVD) on drying kinetics, microstructure, and polysaccharide yield, with HPLC used for monosaccharide quantification and FT-IR for structural analysis. PVD significantly reduced drying time to 11 h, representing a 50.0% reduction compared to HAD (22 h), a 47.6% reduction compared to IRD (21 h), and a 42.1% reduction compared to HAD+ (19 h),

Optimizing post-printing drying conditions for point-of-care manufacturing of SSE-3D printed medicines.

Post-printing drying conditions could affect the quality of oral use printlets elaborated by semi-solid extrusion (SSE) 3D printing, yet their systematic characterization as Critical Process Parameters (CPPs) remains limited. This study evaluated the influence of temperature (20-40°C) and relative humidity (20-80%) on acetazolamide printlets drying process using a full factorial design (n = 27). Response variables included weight stabilization time (the time required for the batch to complete drying, operationally defined as achieving constant weight), disintegration time, dissolution profile,

Low-temperature vacuum evaporation as a novel dehydration process for the long-term preservation of transplantable human corneal tissue.

Globally there is a shortage of available donor corneas with only 1 cornea available for every 70 needed. A large limitation to corneal transplant surgery is access to quality donor tissue due to inadequate eye donation services and infrastructure in many countries, compounded by the fact that there are few available long-term storage solutions for effectively preserving spare donor corneas collected in countries with a surplus. In this study, we describe a novel technology termed low-temperature vacuum evaporation (LTVE) that can effectively dry-preserve surplus donor corneal tissue, allowing

Stabilization of HSV-2 viral vaccine candidate by spray drying.

This work demonstrates that an HSV-2 candidate vaccine can be thermostabilized by spray drying to reduce cold chain demands. This work is also to optimize the process responses by varying spray dry parameters for pre-screened suitable excipients; and to determine the validity of current prescreening techniques. Vaccine activity losses were measured by in vitro plaque forming assay with Vero cell line. An accelerated storage condition of 45 °C for 10 days was used to determine spray dried sample stability. Prescreening studies demonstrated that trehalose and sucrose were superior to other teste

Optimization of Spray Drying Conditions for Yield, Particle Size and Biological Activity of Thermally Stable Viral Vectors.

PURPOSE: This work examines the relevance of viral activity in the optimization of spray drying process parameters for the development of thermally stable vaccine powders. In some instances, the actual active pharmaceutical ingredient (API) is not included in the process optimization as it is deemed too costly to use until the final selection of operating conditions, however, that approach is inappropriate for highly labile biopharmaceutics. We investigate the effects of spray drying parameters on i) yield, ii) particle size and iii) viral vector activity of a mannitol/dextran encapsulated rec

Particle formation of budesonide from alcohol-modified subcritical water solutions.

Recently, subcritical water (SBCW: water that has been heated to a temperature between 100°C and 200°C at pressures of up to 70bar) has been used to dissolve several hydrophobic pharmaceutical compounds (Carr et al., 2010a). Furthermore, a number of active pharmaceutical ingredients (APIs) have been rapidly precipitated from SBCW solutions (Carr et al., 2010b,c). It is possible to alter the precipitate morphology by altering the processing variables; including the SBCW-API solution injection temperature and adding impurities (such as pharmaceutical excipients, e.g. lactose) to the precipitatio
