Isolation, Genomic Characterization, and Pathogenicity of a Novel Duck Orbivirus Genetically Similar to Corriparta Virus in China.

Corriparta virus (CORV), an arbovirus within the Orbivirus genus, exhibits a broad vertebrate host range but limited pathogenicity. In this study, we report the first isolation and characterization of a novel orbivirus genetically related to CORV, temporarily designed as novel duck orbivirus (NDORV), from Beijing ducks in Henan province, China, in 2024. Genomic characterization revealed that NDORV possesses a 10-segment double-stranded RNA (dsRNA) genome, consistent with the structural hallmarks of the Orbivirus genus, with a high genetic similarity to Parry's Lagoon virus (PLV) and CORV. To e…

First establishment of a Duck Model for in vivo and in vitro studies of West nile virus (Kunjin subtype).

We present a method for constructing a Kunjin virus (KUNV) infectious clone capable of stable proliferation in bacterial systems. This KUNV infectious clone successfully produces infectious viral particles within host cells. In vitro, the recombinant virus demonstrates the ability to replicate in primary duck cells and induces 100 % mortality in duck embryos. In vivo, the recombinant virus exhibits virulence in our established duckling model, leading to clinical signs such as hepatic haemorrhage, splenomegaly, and pancreatic haemorrhage. Furthermore, the recombinant rKUNV induced a mortality r

Pioneering discovery: US3-phosphorylated sites on VP22 orchestrate duck plague virus release and pathogenicity.

Duck plague virus (DPV), a member of the Alphaherpesvirinae subfamily, causes severe infectious disease in waterfowl and Anseriformes birds. Although the VP22 protein (encoded by the UL49 gene) is known to play a pivotal role in secondary viral envelopment, its post-translational modifications and regulatory mechanisms during the viral life cycle remain poorly understood. In this study, we found that the conserved serine/threonine kinase US3 of DPV phosphorylates VP22. We identified eight serine residues (Ser37, Ser66, Ser68, Ser82, Ser114, Ser198, Ser226, and Ser227) as phosphorylation target

A targeted mutation in ICP8 attenuates duck plague virus by disrupting replication and proposes a vaccine strategy.

Controlling duck plague (DP) is essential for waterfowl health and industry productivity. The herpesviral single-stranded DNA-binding protein ICP8 is essential for replication compartment assembly, yet its exploitation for vaccine development in avian herpesviruses is limited. In this study, we generated two ICP8 mutants (H825A and F829A/P830A) in duck plague virus (DPV) via BAC-based homologous recombination. Using structural modeling, in vitro replication kinetics, co-immunoprecipitation, and in vivo duckling models, we assessed impacts on replication complex assembly, virulence attenuation,

Multiscale 3D genome organization underlies duck fatty liver with no adipose inflammation or serious injury.

Non-alcoholic fatty liver disease (NAFLD) is the most common form of chronic liver disease. Little is known about how gene expression and chromatin structure are regulated in NAFLD due to lack of suitable model. Ducks naturally develop fatty liver similar to serious human non-alcoholic fatty liver (NAFL) without adipose inflammation and liver fibrosis, thus serves as a good model for investigating molecular mechanisms of adipose metabolism and anti-inflammation. Here, we constructed a NAFLD model without adipose inflammation and liver fibrosis in ducks. By performing dynamic pathological and t…

2A2 protein of DHAV-1 induces duck embryo fibroblasts gasdermin E-mediated pyroptosis.

The duck hepatitis A virus type 1 (DHAV-1) causes rapid death in ducklings by triggering a severe cytokine storm. Pyroptosis is an inflammatory form of programmed cell death that is directly related to an increase in pro-inflammatory cytokine levels. Only a few studies have explored the mechanisms underlying pyroptosis in virus-infected avian cells. In this study, we established an avian infection model in vitro by infecting duck embryo fibroblasts (DEFs) with the virulent DHAV-1 LY0801 strain. DHAV-1 infection induced pyroptosis in the DEFs by activating gasdermin E (GSDME) protein via caspas

A study of duck detection using deep neural network based on RetinaNet model in smart farming.

In a duck cage, ducks are placed in various states. In particular, if a duck is overturned and falls or dies, it will adversely affect the growing environment. In order to prevent the foregoing, it was necessary to continuously manage the cage for duck growth. This study proposes a method using an object detection algorithm to improve the foregoing. Object detection refers to the work to perform classification and localization of all objects present in the image when an input image is given. To use an object detection algorithm in a duck cage, data to be used for learning should be made and th…

Characteristics of the a sequence of the duck Plague virus genome and specific cleavage of the viral genome based on the a sequence.

During the replication process, the herpesvirus genome forms the head-to-tail linked concatemeric genome, which is then cleaved and packaged into the capsid. The cleavage and packing process is carried out by the terminase complex, which specifically recognizes and cleaves the concatemeric genome. This process is governed by a cis-acting sequence in the genome, named the a sequence. The a sequence and genome cleavage have been described in some herpesviruses, but it remains unclear in duck plague virus. In this study, we analysed the location, composition, and conservation of a sequence in the

Antimicrobial Resistance Genes and Clonal Relationships of Duck-Derived Salmonella in Shandong Province, China in 2023.

Salmonella is a major threat to both human and animal health. However, the diversity and antibiotic resistance of animal-derived Salmonella and their association with human infections remain largely unexplored. In this study, Salmonella strains were isolated, identified, and sequenced from dead embryos and cloacal swab samples obtained from 278 large-scale duck farms in 11 cities in Shandong Province. The results show that a total of 57 Salmonella strains were isolated, with the dominant sequence types (ST) being ST17 (15/57) and ST19 (9/57), while the dominant serotypes were S. Indiana (15/57

Research Note: Genetic characterization of novel duck reoviruses from Shandong Province, China in 2022.

Since 2005, novel duck reoviruses have been outbreaks in duck breeding areas such as central China and South China. In recent years, the incidence rate of this disease is still increasing, bringing serious economic losses to waterfowl breeding industry. This study isolated 3 novel duck reoviruses (NDRV-SDLS, NDRV-SDWF, and NDRV-SDYC) from sick ducks in 3 local duck farms in Shandong Province. The study aimed to investigate the characteristics of these viruses. The virus is inoculated into duck embryo fibroblasts, where the virus replicates to produce syncytium and dies within 3 to 5 d. The vir…

Identification of novel B cell epitopes in Fiber-2 protein of duck adenovirus 3 and their application.

Duck adenovirus 3 (DAdV-3), a newly emerged duck adenovirus, has resulted in significant economic losses to the duck industry across China since 2014. However, little is known about the B cell epitopes in major antigen of DAdV-3 and the serological approach for detection of DAdV-3 is not available. In this study, four monoclonal antibodies (mAbs) specific to Fiber-2 protein of DAdV-3 were first generated and designated as 2G10, 3D9, 5E6, and 6B12. Indirect immunofluorescence assay (IFA) showed that all of the mAbs reacted with the Fiber-2. Moreover, mAbs 2G10, 5E6, and 6B12 demonstrated good a

BX795, a kinase inhibitor, inhibit duck plague virus infection via targeting US3 kinase.

Duck plague virus (DPV) is a typical DNA virus of waterfowl, it causes huge economic losses to the duck industry due to the higher mortality and lower egg production. The disease is one of the frequent epidemics and outbreaks on duck farms because present vaccines could not provide complete immunity and there are no specific antiviral drugs available. Therefore, the development of antiviral drugs is urgently needed. In this study, we evaluated the antiviral activity of BX795, a specific kinase inhibitor of 3-phosphoinositide-dependent kinase 1 (PDK1), protein kinase B (AKT) and Tank binding ki

Curcumin attenuates AFB1-induced duck liver injury by inhibiting oxidative stress and lysosomal damage.

Aflatoxin B1 (AFB1), as the most toxic secondary metabolite produced by Aspergillus flavus, is a serious threat to human and animal health. Curcumin, a polyphenol from the plant turmeric, has demonstrated unique anti-damage properties in several studies. But, its ability to alleviate AFB1-induced liver damage in ducks and the underlying mechanisms are not completely elucidated. In this study, we investigated the intervention of curcumin on AFB1-induced hepatotoxicity in ducks. Research data showed that the combination of curcumin and AFB1 alleviated oxidative stress, reduced malondialdehyde (M…

Enzymatic hydrolysis re-endows desalted duck egg white nanogel with outstanding foaming properties.

Heat-induced gel-assisted desalination could efficiently and inexpensively remove salt from salted egg whites. However, it was at the expense of the excellent foaming properties of egg whites, caused by the denaturation and aggregation of proteins during heating treatment. Hence, in this current work, the enzymatic treatment was used to re-endow duck egg white nanogels (DEWN) with outstanding foaming properties. We found that low levels of hydrolysis (DH = 2.27 %) could dramatically improve the foaming capability (FC), reaching >200 %, which also enhanced the foaming stability (FS). As the hyd

Metabolomics reveals changes in metabolite composition of duck eggs under the impact of long-term storage.

BACKGROUND: Eggs are essential food sources as they provide low cost and high nutritional content of animal protein. The preservation period is one of the apparent factors affecting egg quality. Previous studies based on traditional detection techniques demonstrated that storage period would significantly influence egg weight, eggshell weight, albumen height, haugh unit (HU) and albumen viscosity. Herein, we employed non-targeted metabolome technology to reveal the comprehensive changes in metabolite composition in duck eggs under the impacts of storage period. RESULTS: The results showed tha
