BACKGROUND: Hereditary spastic paraplegias (HSPs) are a clinically and genetically heterogeneous group of neurodegenerative diseases. To date, at least 84 distinct loci (SPGs) and 67 causative genes have been identified. Even though the number of known causative genes is constantly increasing, a substantial portion of patients remains without a molecular diagnosis. Variants in the dynein, cytoplasmic 1, heavy chain 1(DYNC1H1)gene have been reported to cause a range of neurogenetic diseases, including spinal muscular atrophy (SMA), Charcot-Marie-Tooth disease (CMT), and cortical malformations.

Cilia-associated human genetic disorders are striking in the diversity of their abnormalities and their complex inheritance. Inactivation of the retrograde ciliary motor by mutations in DYNC2H1 causes skeletal dysplasias that have strongly variable expressivity. Here we define previously unknown genetic relationships between Dync2h1 and other genes required for ciliary trafficking. Mutations in mouse Dync2h1 disrupt cilia structure, block Sonic hedgehog signaling and cause midgestation lethality. Heterozygosity for Ift172, a gene required for anterograde ciliary trafficking, suppresses cilia p

Flagella and cilia are two very similar organelles that "beat" to move cells and to propel fluid over tissues. They are highly conserved, being found in organisms ranging from prokaryotes to plant and animal eukaryotes. In humans, cilia are present in almost every organ, and several human conditions involve dysfunctional cilia; for example, lateralization defects, where the positions of organs are reversed, and primary ciliary dyskinesia, a rare condition where patients suffer from recurrent respiratory infections. In this article, we will discuss how information gained from studies on algae h

STUDY QUESTION: Is the Tcte1 mutation causative for male infertility?
SUMMARY ANSWER: Our collected data underline the complex and devastating effect of the single-gene mutation on the testicular molecular network, leading to male reproductive failure.
WHAT IS KNOWN ALREADY: Recent data have revealed mutations in genes related to axonemal dynein arms as causative for morphology and motility abnormalities in spermatozoa of infertile males, including dysplasia of fibrous sheath (DFS) and multiple morphological abnormalities in the sperm flagella (MMAF). The nexin-dynein regulatory complex (N-D

Human cerebral cortical malformations are associated with progenitor proliferation and neuronal migration abnormalities. Progenitor cells include apical radial glia, intermediate progenitors and basal (or outer) radial glia (bRGs or oRGs). bRGs are few in number in lissencephalic species (e.g. the mouse) but abundant in gyrencephalic brains. The LIS1 gene coding for a dynein regulator, is mutated in human lissencephaly, associated also in some cases with microcephaly. LIS1 was shown to be important during cell division and neuronal migration. Here, we generated bRG-like cells in the mouse embr

BACKGROUND: Asthenozoospermia is one of the most common causes of male infertility, and its genetic etiology is poorly understood. DNAH9 is a core component of outer dynein arms in cilia and flagellum. It was reported that variants of DNAH9 (OMIM: 603330) might cause primary ciliary dyskinesia (PCD). However, variants in DNAH9 lead to nonsyndromic severe asthenozoospermia have yet to be reported.
METHODS: Whole exome sequencing (WES) was performed for two individuals with nonsyndromic severe asthenozoospermia from two non-consanguineous families, and Sanger sequencing was performed to verify

Multiple morphological abnormalities of the sperm flagellum (MMAF) is a severe form of male infertility defined by the presence of a mosaic of anomalies, including short, bent, curled, thick, or absent flagella, resulting from a severe disorganization of the axoneme and of the peri-axonemal structures. Mutations in DNAH1, CFAP43, and CFAP44, three genes encoding axoneme-related proteins, have been described to account for approximately 30% of the MMAF cases reported so far. Here, we searched for pathological copy-number variants in whole-exome sequencing data from a cohort of 78 MMAF-affected

Defects in motile cilia cause a range of disorders, including heterotaxy (HTX), congenital heart disease (CHD), and primary ciliary dyskinesia (PCD). Although these conditions often co-occur, the genetic and mechanistic bases for tissue-specific manifestations remain poorly understood. Here, we identify compound heterozygous variants in DAW1, a dynein arm assembly factor, in a proband with HTX and complex congenital heart disease but no clinical signs of PCD. Whole-genome sequencing revealed a maternally inherited canonical splice-site variant (c.648 + 1G > A) and a paternally inherited missen

Primary ciliary dyskinesia (PCD) is a severe sperm defect, leading to male infertility. PCD affects both respiratory function and sperm motility, as motile cilia and sperm flagella rely on axonemal architecture. Multiple morphological abnormalities of the sperm flagella (MMAF) is a distinct form of asthenoteratozoospermia, characterized by a heterogeneous spectrum of flagellar defects. In recent years, coiled-coil domain-containing ( CCDC ) genes have been shown to play crucial roles in both MMAF and PCD. In this study, a homozygous mutation in CCDC39 , c.1528-2A>G, was identified in a patient

Energy metabolic dysfunction is a major cause of impaired chronic wounds healing, in which disrupted mitochondrial transfer and autophagy imbalance further aggravate the cellular energy crisis. In this study, single-cell RNA sequencing (scRNA-seq) of clinical diabetic wound samples first identified a pivotal role for macrophage-to-fibroblast mitochondrial transfer in wound healing. This finding was further validated using diabetic wound models and histological analyses, highlighting these processes as potential therapeutic targets for alleviating energy metabolic stress. Based on these finding

INTRODUCTION: Mast cells (MCs) mediate immune, allergic, and neuroinflammatory responses by releasing inflammatory mediators upon activation through the immunoglobulin E (IgE) receptor FcεRI or innate stimuli acting through Mas-related G protein coupled receptors (Mrgprs). We previously showed that Rab12 negatively regulates mediator release by recruiting the Rab-interacting lysosomal protein (RILP)-dynein complex to the secretory granules (SGs). Because Rab12 also interacts with the RILP-like proteins RILP-L1 and RILP-L2, we examined whether phosphorylation controls Rab12 distribution among i

BACKGROUND Primary ciliary dyskinesia (PCD) is an uncommon autosomal recessive disease resulting from dysfunction of motile cilia that causes impaired mucociliary clearance and abnormal embryonic left-right axis differentiation. The CCDC39 gene is a known cause of PCD, which is most commonly associated with inner dynein arm defects and microtubular disorganization. Although heterotaxy-related congenital heart defects are well described in PCD, their presence in patients with CCDC39 mutations, particularly those with congenitally corrected transposition of the great arteries (ccTGA), has not be

Defective endosomal sorting and trafficking are increasingly recognised as key drivers of neurodegeneration, including hereditary spastic paraplegia (HSP) and other motor neuron disorders. Early endosomal tubule fission (ETF) is essential for sorting cargoes for recycling and retrograde transport, yet the mechanisms coordinating this process are incompletely defined. Here, we identify the endoplasmic reticulum (ER)-resident protein protrudin-previously shown to promote axonal regeneration after injury-as a key regulator of ETF. Using CRISPR interference in human cells, we show that loss of pro

Primary ciliary dyskinesia (PCD) is a rare autosomal recessive disorder characterized by dysfunction of motile cilia in various organ systems, including the respiratory and reproductive tracts. A key manifestation in males is infertility, primarily attributed to impaired sperm motility. Although sperm vitality may be preserved, immotility or abnormal flagellar function significantly impairs natural conception. While a minority of men with PCD achieve fatherhood without medical intervention, most require assisted reproductive techniques (ART). The genetic etiology of PCD-related male infertilit

Background: Genetic defects in motile cilia cause primary ciliary dyskinesia (PCD), a rare disease with no specific therapeutics. Individuals with PCD often have impaired fertility and laterality defects and universally suffer from upper and lower airway diseases. Chronic rhinosinusitis is a universal feature of PCD, and mucus accumulation and subsequent infections of the sinonasal cavity cause significant morbidity in individuals with PCD. Despite this, there are no approved treatments that specifically target mucus. Objective: The goals of this study were to determine whether computed tomogr

UNLABELLED: DNAAF5 is a dynein motor assembly factor associated with the autosomal heterogenic recessive condition of motile cilia, primary ciliary dyskinesia (PCD). The effects of allele heterozygosity on motile cilia function are unknown. We used CRISPR-Cas9 genome editing in mice to recreate a human missense variant identified in patients with mild PCD and a second, frameshift null deletion in Dnaaf5 . Litters with Dnaaf5 heteroallelic variants showed distinct missense and null gene dosage effects. Homozygosity for the null Dnaaf5 alleles was embryonic lethal. Compound heterozygous animals
