The winged helix transcription factor Fkh10 is required for normal development of the inner ear.

Fkhl0 is a member of the forkhead family of winged helix transcriptional regulators. Genes encoding forkhead proteins are instrumental during embryogenesis in mammals, in particular during development of the nervous system. Here we report that mice with a targeted disruption of the Fkh10 locus exhibit circling behaviour, poor swimming ability and abnormal reaching response-all common findings in mice with vestibular dysfunction. These animals also fail to elicit a Preyer reflex in response to a suprathreshold auditory stimulation, as seen in mice with profound hearing impairment. Histological

A totally implantable drug delivery system for local therapy of the middle and inner ear.

Local therapy of middle and inner ear diseases is being used, but is restricted to cases of ear drum perforation or to repeated invasive intratympanic drug application by the physician. In accordance with the Medical Device Directive (class III), a bone-anchored, totally implantable drug delivery system (TI-DDS) has been developed. It includes a micropump for subcutaneous, patient-controlled activation, a drug reservoir and a septum port. A thin guide-wired catheter leads from the pump outlet to the point of application in the mastoid or middle ear cavities. Local inner ear therapy with suitab

Receptor pharmacological models for inner ear therapies with emphasis on glutamate receptors: a survey.

With the aid of microiontophoretic techniques we evaluated the action of different postsynaptic glutamate receptor subtypes that mediate neurotransmission between the inner hair cell and the afferent neuron. The sensory input is modulated by axodendritic efferents. In the central nervous system, excessive activation of glutamate receptors is thought to be responsible for a wide variety of neurotoxic actions, and calcium is involved in the etiology of glutamate-induced cell damage. Glutamatergic neurotoxicity may form an appropriate pathophysiological model to explain a variety of inner ear dis

Structural diversity in the inner ear of teleost fishes: implications for connections to the Mauthner cell.

A body of literature suggests that the Mauthner cell startle response can be elicited by stimulation of the ear. While we know that there are projections to the M-cell from the ear, the specific endorgan(s) of the ear projecting to the M-cell are not known. Moreover, there are many reasons to question whether there is one pattern of inner ear to M-cell connection or whether the endorgan(s) projection to the M-cell varies in species that have different hearing capabilities of hearing structures. In this paper, we briefly review the structure of fish ears, with an emphasis on structural regional

Hair cell damage in the inner ear of the guinea pig due to noise in a workshop.

Guinea pigs were exposed to noise in various workshops at a shipyard. The equivalent sound level ranged from 87 to 90 dB(A) and the exposure time was 30 days of 8 hours. The numbers of lost outer hair cells were greater than in control animals, thus establishing a relation between the number of lost hair cells and the total noise dose. A comparison between the results obtained here and results obtained in laboratory experiments shows that although many physical sound parameters may be identical, the effect on the hair cells can be quite different. In particular, the total noise dose is not a p

Provocation of nystagmus and deviation by type I allergy in the inner ear of the guinea pig.

Nystagmus, head-deviation, and hearing loss were provoked by Type I allergy induced in guinea pigs with passive sensitization and specific antigen challenge. Mast cell degranulation, edema of connective tissue of the endolymphatic sac and endolymphatic hydrops were histologically observed. These findings indicated that Type I allergy can be induced in the inner ear, and that symptoms analogous to Meniere's disease may be induced by this response.

[Lymphocyte subpopulations and HLA-DR determinations in diseases of the inner ear and Bell's palsy].

Thymus lymphocytes-subpopulations (T-helper T4 and T-suppressorcells T8) were determined in inner ear diseases (e.g. sudden hearing loss, neuronitis vestibularis, Morbus Menière, Bell's Palsy) by specific monoclonal antibodies. T-cell subset ratio (T4/T8) was elevated (less than 3) in about 50% of all patients. DR-typifying was performed because of the well-known control of the immunoregulation through the class II HLA-DR antigens. There was a relative risk of 5.2 in peripheral vestibular lesion (neuronitis vestibularis). The relative risk of autoimmune diseases is found at this level.

[Nimodipine (Bay e 9736) a new therapy concept in diseases of the inner ear?].

In an open study 30 patients with the symptom of tinnitus were treated for 12 weeks with nimodipin (Bay e 9736) 3 X 30 mg/die. 12 patients showed sudden deafness with a history of one day up to more than 20 years. In six cases we observed complete remission, in two cases remission up to 70-80%, in two cases slight improvement. Four patients showed no side reaction to the treatment of nimodipin. Ten patients suffered from so-called degenerative inner ear deafness. 3 (n = 10) showed complete remission, one nearly 90% improvement, another patient showed 60% improvement, one slight improvement, in

Cochlear neuronal populations in developmental defects of the inner ear. Implications for cochlear implantation.

A histological study was made to determine the cochlear neuronal populations of 20 human ears having hearing loss caused by developmental defects. The neuronal populations ranged from 7677 in an ear with Mondini dysplasia to 30 753 in an ear with DiGeorge's syndrome, the norm for young human subjects being 35 000 neurons. The length of the cochlear (spiral) ganglion varied from 7.3 mm to 14.8 mm, the norm for human subjects being 12 mm. The sensorineural hearing losses in all cases were attributable to malformation or degeneration of the sensorineural structures. The hearing loss was moderate

The origin of centrifugal fibers to the inner ear in Caiman crocodilus. A horseradish peroxidase study.

The origin of acoustic and vestibular efferent fibers was investigated in Caiman crocodilus. After injection of horseradish peroxidase (HRP) into the cochlear duct or into the ampullae of the horizontal and anterior semi-circular canals, cells in the medulla oblongata exhibited retrogradely transported HRP reaction product. Efferent vestibular fibers were found in the medial reticular nucleus, at the level of the oliva superior. There were more labeled neurons ipsilateral to the injection site. Efferent acoustic neurons were found close to or inside the rostroventral division of the oliva supe

Unilateral complete aplasia of the inner ear with associated tracheoesophageal fistula: report of a case.

A case of unilateral complete aplasia of the petrous apex and inner ear associated with H-type tracheoesophageal fistula is reported. polytomography, computerized axial tomography, and plain film radiography confirmed the presence of this rare variant of inner ear aplasia. This patient, who has normal facial nerve function, is, to our knowledge, the first reported case of a nonthalidomide, Michel-type aplasia since the original report of Michel more than a century ago. Unilateral complete inner ear aplasia associated with tracheoesophageal fistula has not been previously reported.

Inner ear pathologic features following mumps infection. Report of a case in an adult.

Temporal bone studies in an adult with a moderately severe, bilateral sensorineural hearing loss revealed bilateral cochlear changes 13 years after mumps infection. The organ of Corti was completely absent in the greater part of the superior horizontal basal limbs, with occasional hair cell loss throughout the rest of the cochlea. The outer sulcus cell area was degenerated. The stria vascularis was normal, as was the tectorial membrane, except for small hyaline droplets. The number of nerve fibers was extremely decreased in the spiral bony lamina of the basal turns. Basophilic material, possib

Exploration of biodistribution of mRNA-delivering lipid nanoparticles in the neonatal mouse inner ear.

Hearing loss affects millions worldwide, yet hearing aids and cochlear implants have major limitations. mRNA-based gene therapy could address the underlying molecular causes, but lipid nanoparticle (LNP) delivery in the inner ear remains underexplored and its biodistribution has not been systematically characterized. Here we establish and scale up an mRNA-encapsulated LNP formulation using a design-of-experiments (DoE) approach built on predefined critical quality attributes, and map the route-dependent distribution of reporter mRNA expression in the neonatal mouse cochlea. In HEI-OC1 auditory

Regulatory Networks Driving the Specification, Differentiation, and Diversification of Neurons in the Mouse Inner Ear.

Vestibular and spiral ganglion neurons (VGNs and SGNs) developed in the inner ear, where they extend fibers to innervate the vestibular and cochlear hair cells and project centrally to the vestibular and cochlear nuclei. This review focuses on representative molecular factors that regulate key processes in the development of inner ear neurons, including their specification, differentiation, axon targeting, and functional diversification. A temporal regulatory cascade defines the initial precursors through factors such as Smarca4, Six1, Eya1, followed by Sox2. While Sox2 deletion abolishes hair

A novel minimally invasive stereotaxic technique to target inner ear neurons in the mouse.

Sensorineural hearing loss (SNHL), a leading cause of disability worldwide, arises from damage to hair cells (HCs) or spiral ganglion neurons (SGNs) within the cochlea. Among its etiologies, auditory neuropathy (AN) is characterized by disrupted signal transmission due to SGN damage. Traditional interventions, such as hearing aids and cochlear implants, provide limited benefit in cases of AN, where neuronal damage impairs signal transduction to the brain. Emerging regenerative therapies, including cell replacement and gene delivery, hold potential to restore SGN function, but their application
