L-Arginine Nutrition and Metabolism in Ruminants.

L-Arginine (Arg) plays a central role in the nitrogen metabolism (e.g., syntheses of protein, nitric oxide, polyamines, and creatine), blood flow, nutrient utilization, and health of ruminants. This amino acid is produced by ruminal bacteria and is also synthesized from L-glutamine, L-glutamate, and L-proline via the formation of L-citrulline (Cit) in the enterocytes of young and adult ruminants. In pre-weaning ruminants, most of the Cit formed de novo by the enterocytes is used locally for Arg production. In post-weaning ruminants, the small intestine-derived Cit is converted into Arg primari…

Clinical Features and Pathophysiological Mechanisms of COVID-19-associated Gastrointestinal Manifestations.

AIM AND OBJECTIVE: According to the literature, gastrointestinal (GI) involvement may have a remarkable influence on the course of coronavir … s, and methodological differences. Several studies proved that this novel coronavirus gets into the enterocytes by attaching to angiotensin-converting enzyme 2 receptor. Some of them have shown a direct viral invasion and replication of the virus within enterocytes. Along with detection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in patien …

Celiac Disease: A Transitional Point of View.

Celiac disease (CeD) is a chronic, lifelong, multifactorial, polygenic, and autoimmune disorder, characteristically triggered by exposure to … ll interactions, and production of specific antibodies, resulting in the immune-mediated killing of enterocytes and, macroscopically, in duodenal inflammation. Here, the most relevant correlations between cellular and molecular aspects and clinical manifestations of this complex disease are reviewed, with final considerations on nutritional aspects for diseas …

Flavonoids as drugs at the small intestinal level.

Flavonoids represent a biologically active class of secondary plant compounds. For selected members there is convincing evidence regarding their beneficial effects on human health. Accordingly these compounds are tested as supporting or alternative therapies for a number of diseases such as cancer or type-II diabetes. Subsequent to their ingestion a first site of interference are digestive enzymes and transporters in the small intestine. Through interactions with glucose transporters in the apical membranes of enterocytes flavonoid glycosides and also some aglycones appear to reduce postprandi…

Protein p60 participates in intestinal host invasion by Listeria monocytogenes.

The role of p60 in intestinal invasion by Listeria monocytogenes was assessed after oral infection of mice with the p60 low-expressing mutan … ocytogenes EGD, but not uncoated listeriae were still detectable 24 h p.i. at the apical surface of enterocytes in the intestine. We conclude that p60 contributes to host invasion through the natural port of listerial entry, the intestinal epithelium.

The histologic and molecular alterations during the cold storage of the ascending human colon.

Inclusion of a colon segment in visceral grafts is increasing as this may improve water and electrolyte absorption and ultimately improve recipients' renal function. In contrast with the vast knowledge on the ischemia/reperfusion injury in the small bowel, similar data on the colon is almost inexistent. Using light microscopy, immunofluorescence and Western blot, we assessed several histological and molecular changes following the cold storage (CS) of the ascending colon of 18 human brain-dead organ donors for up to 24 h. CS induced progressive yet limited epithelial detachment and goblet cell

Intestinal goblet cell autoantibody associated enteropathy.

This report describes a case of refractory enteropathy with circulating intestinal goblet cell autoantibodies (IGA). A 19 year old man with hyperthyroidism had suffered from protracted diarrhoea for nearly 10 years. Histological examination showed evidence of collagenous enterocolitis. The diarrhoea did not improve despite fasting under total parenteral nutrition. An immunofluorescence assay demonstrated IGA without anti-enterocyte autoantibodies, the hallmark of autoimmune enteropathy, although other criteria were fulfilled. None of 109 controls, including 55 cases of inflammatory bowel disea

Nonsteroidal-induced benign strictures of the colon: a case report and review of the literature.

Colonic strictures are a rare complication reported to result from chronic use of sustained release formulations of indomethacin and diclofenac. Such strictures often present with associated mucosal ulceration and are thought to result from nonsteroidal antiinflammatory drug-induced alterations in enterocyte homeostasis. Strictures generally occur in the cecum, ascending, and proximal transverse colon with symptoms of occult blood loss, obstruction, changes in bowel habits, and rarely, perforation. The first reported case of a 69-year-old woman who developed recurrent colonic strictures with i

Complementation of Listeria seeligeri with the plcA-prfA genes from L. monocytogenes activates transcription of seeligerolysin and leads to bacterial escape from the phagosome of infected mammalian cells.

Infection experiments have shown that the avirulent species Listeria seeligeri invaded the enterocyte-like cell line Caco-2 with low efficiency but was unable to escape from the phagosome. Introduction of the listeriolysin gene (hly) from L. monocytogenes into L. seeligeri via a recombinant plasmid did not change these characteristics. No measurable transcription of this gene or of the structurally intact chromosomal seeligerolysin gene (lso) was detected. Transformation with a plasmid carrying the bicistronically transcribed plcA-prfA genes from L. monocytogenes resulted in the efficient expr

The sterols from ingested vegetable sources, the phytosterols, are expelled from enterocytes and thus indirectly help our gut in coping with abundant cholesterol in the lumen. Higher plants synthesize many phytosterols, but in marine organisms, we can find other innumerous sterol molecules. The diversity of sterol molecules produced and res …

Marked microcytosis and increased transferrin saturation: Think about variants in SLC11A2 (DMT1).

Congenital microcytic anemias are rare diseases associated with decreased hemoglobin synthesis and red blood cells of low corpuscular volume. DMT1/NRAMP2 is a highly conserved divalent cation transporter encoded by the SLC11A2 gene, expressed at the membrane of various cells. It ensures ferrous iron absorption from the apical membrane of enterocytes, iron recovery from urine by renal tubules, and acidified endosome uptake after transferrin internalization. Pathogenic DMT1 variants have been described in 10 individuals to date, associated with recessive hypochromic anemia and iron overload. Her…

A novel mutation of the GLUT2 gene in a Turkish patient with Fanconi-Bickel syndrome.

Fanconi-Bickel syndrome is a rare inherited disorder of carbohydrate metabolism. The disease is characterized by the association of a massive hepatomegaly due to glycogen accumulation, severe hypophosphatemic rickets and marked growth retardation due to proximal renal tubular dysfunction. Fanconi-Bickel syndrome is a single gene disease and is caused by defects in the facilitative glucose transporter 2 (GLUT2) gene (SLC2A2) on chromosome 3q26.1-26.3, which encodes for the glucose transporter protein 2 expressed in hepatocytes, pancreatic beta-cells, enterocytes, and renal tubular cells. Severa…

[New horizons of gluten sensitivity studies].

Gluten sensitivity may be a cause of gluten-sensitivity celiac disease (GCD). Some gluten-sensitive subjects may have symptoms of GCD, but lack its characteristic changes in the small bowel mucosa (SBM) and a gluten-free diet results in the disappearance of clinical symptoms of GCD. If there is no gluten allergy, the concept "gluten intolerance (GI) unassociated with celiac disease" is applicable in these cases. There is an increase in the prevalence of GCD and GI, which is associated with the use of gluten in food industry to improve the taste and energy density of foods and with the damaging

The role of transforming growth factor (TGF)-β in modulating the immune response and fibrogenesis in the gut.

Transforming growth factor (TGF)-β, a pleiotropic cytokine released by both immune and non-immune cells in the gut, exerts an important tolerogenic action by promoting regulatory T cell differentiation. TGF-β also enhances enterocyte migration and regulates extracellular matrix turnover, thereby playing a crucial role in tissue remodeling in the gut. In this review we describe the mechanisms by which abnormal TGF-β signaling impairs intestinal immune tolerance and tissue repair, thus predisposing to the onset of immune-mediated bowel disorders, such as inflammatory bowel disease and celiac dis

Aggregation and inhibition of rat intestinal alkaline phosphatase by high concentrations of calcium. Reversibility of the processes.

Intestinal alkaline phosphatase (IAP) is an enzyme of the brush border of the enterocyte. The activity of IAP biphasically depends on calcium. Although calcium increases IAP activity, when calcium is higher than 20 mmole/L, IAP activity decreases and the amount of an aggregated form increases. The reversibility of the effect of calcium and the aggregation process are unknown. The isoelectric point of the enzyme was higher in the presence of calcium, but was the same for the enzyme and the aggregated form. The treatment with EGTA after calcium addition did not restore the enzymatic activity but

We demonstrated that the subsequent increased intestinal renewal is mediated by both the Notch and JAK/STAT pathways and is deleterious to Drosophila survival. In conclusion, this work highlights that the ability of Mabs to induce an exacerbated an …
