The effect of H2O2 on lens epithelial cell glutathione.

Investigation of lens epithelial cells indicates that under normal conditions, essentially all of the detectable cellular glutathione is in a reduced state. However, exposure to levels of H2O2 in the range found in the aqueous fluid of cataract patients causes rapid, very large changes in the glutathione redox ratios. Immediately following short-term exposure to 0.15-0.2 mM H2O2, reduced glutathione drops to 19% of its normal level and the remainder of the total glutathione is found in the oxidized form. Within the next few minutes, the redox ratio returns to normal. However, total glutathione…

"Blue-body" epithelial cell inclusions in conjunctivitis.

In 10.4% of conjunctival scrapings received from 1731 patients with conjunctivitis during the years 1970 through 1980, basophilic cytoplasmic inclusions of unknown etiology were found in the epithelial cells when Giemsa stained. These "blue-bodies" differed in morphology from bacteria, chlamydia, and other frequent cytoplasmic inclusions. By electron microscopy, they consisted of a variety of dense bodies and phagolysosomal vacuoles containing complex lipids, indicating injury to the epithelial cells. The presence of these inclusions had a statistically significant association with the use of …

Epithelial cell kinetics in the descending colon of the rat.

Epithelial cell kinetics were investigated in the descending colon of the rat. The number of cells per crypt was found to be approximately 625, with 33 cells per cell column and 19 cell columns per crypt circumference. The growth fraction of the colonic crypt was 0.42, and proliferating cells were situated largely in the lower half of the crypt. The cell cycle time was 50.5 h, with values for the G1, S and G2 phases of 40.0, 7.6 and 2.9 h respectively. Cell migration studies showed that it took 60-72 h for a cell to migrate from the upper border of the proliferative cell compartment in the cry

Tea tree oil action on human corneal epithelial cells biological activity.

PURPOSE: In ophthalmology, tea tree oil is used as an anti-infective and wound-healing compound. It is also effective in combating parasitic eyelid infections. Unfortunately, it has been linked to corneal epithelial defects when it contacts the ocular surface. METHODS AND RESULTS: It was shown that concentrations of TTO exceeding 0.01% may have adverse effects on corneal epithelial cells, reducing cell viability. The IC50 (MTT assay) was 0.055% and the IC50 (NR uptake assay) was 0.1%. Cell death occurred primarily through apoptosis. Flow cytometry revealed that TTO increased the release of re…

Sulforaphane Alleviates Zearalenone-Induced Oxidative Stress in Bovine Mammary Epithelial Cells.

Zearalenone (ZEA) is a common contaminant in crops and animal feed. However, research on the effects of ZEA on animal mammary tissue is relatively limited. Sulforaphane (SFN) is a naturally active compound mainly derived from cruciferous vegetables (such as broccoli), with significant antioxidant and cytoprotective effects. The purpose of this study is the effect of SFN on ZEA-induced toxicity in bovine mammary epithelial cells (MAC-T). By treating MAC-T cells with different concentrations of ZEA and SFN for 24 h, the results showed that different concentrations of ZEA (10, 20, 40, 60, 80, or …

P2Y2 Receptor Controls TLR3-Induced Chemokine Production in Intestinal Epithelial Cells.

Toll-like receptors (TLRs) and P2 receptors are key regulators of innate immunity. During infection, pathogen-associated molecular patterns (PAMPs) activate TLRs, whereas extracellular nucleotides engage P2 receptors and influence inflammatory responses. Here, we show that polyinosinic:polycytidylic acid (poly[I:C])-activated TLR3 and P2Y2 signalling functionally interact in intestinal epithelial cells (IECs) to regulate chemokine production. HT-29 cells were stimulated with PAMPs, including the TLR3 agonist poly(I:C), in the presence or absence of P2 receptor signalling inhibitors. CXCL8/IL-8…

ACE2 Activation in Intestinal Epithelial Cells Prevents Radiation-Induced Intestinal Injury.

AIMS: Radiation-induced intestinal injury (RIII) significantly impairs the quality of life in patients with abdominal/pelvic cancer undergoi … of ISCs both in vitro and in vivo postradiation. Mechanistically, DIZE directly targeted intestinal epithelial cells (IECs), preventing the activation of radiation-induced MAPK (p38/JNK) and NF-κB pathways. This effect was abolished by ACE2 knockdown in a human intestinal epithelial cell line (HIECs) in vitro. Intriguingly, DIZE failed to inhibit endothelial cell …

Microtubule-Targeting Drugs Induce Mesenchymal-Epithelial Transition in Hepatic Stellate Cells.

BACKGROUND AND AIMS: Liver fibrosis is a progressive pathological condition characterized by excessive extracellular matrix accumulation, primarily driven by activated hepatic stellate cells (HSCs). While molecular-based strategies have been extensively explored for HSC deactivation, clinically effective therapies remain elusive. Previous studies suggest that epithelial-like features of quiescent HSCs are lost upon activation, implying that mesenchymal-epithelial transition (MET) may be a potential therapeutic mechanism for reversing HSC activation. METHODS: To identify compounds capable of r

TRPA1 Regulates Fibrosis-Associated Transcriptional Pathways in Human Lung Epithelial Cells.

Transient receptor potential ankyrin 1 (TRPA1) is a cation channel originally identified in lung fibroblasts and extensively studied in sensory neurons, where it is associated with pain and neurogenic inflammation. We and others have recently shown that TRPA1 is also expressed in lung epithelial cells and that its expression is regulated by the cytokine environment. In the present study, we used next-generation sequencing to analyse the role of TRPA1 and the effects of its inhibition on human A549 lung epithelial cell phenotype under inflammatory conditions. Two different TRPA1 inhibitors (HC-…

Optimized primary organotypic culture from murine neonatal tracheal airway epithelial cells.

Neonatal airway development and injury are poorly understood, in part due to challenges of studying extremes of phenotype in human pathological samples and difficulties obtaining relevant comparator samples. Ex vivo model systems are needed to improve understanding of airway development, injury, and repair in the neonatal lung. We optimized a protocol for organotypic culture of primary murine neonatal tracheal epithelial cells (MNTECs). We compared expansion and differentiation properties of MNTECs in five different media conditions, ranging from previously published "lab-made" media to commer…

MUC13 enhances IL-17 signaling pathway activity in nasal epithelial cells.

BACKGROUND: MUC13 is a membrane-bound mucin involved in epithelial protection and barrier maintenance, but its role in nasal epithelial inflammation remains unclear. This study aimed to investigate whether MUC13 regulates inflammatory signaling in primary nasal epithelial cells (NECs). METHODS: Patient-derived primary NECs were transfected with human MUC13 mRNA to establish a dose-dependent overexpression model, and RNA sequencing was performed to characterize MUC13-associated transcriptomic changes. Differential expression analysis, GO/KEGG enrichment analysis, and GSEA were used to identify…

Improved protocol for isolation and characterization of human thymic epithelial cells.

Thymic epithelial cells (TECs) play a vital role in the generation of immunocompetent and immunotolerant T cells. The understanding of TEC biology in model animals such as mice has advanced thanks to the cellular and molecular analyses of purified TEC populations, including cortical TECs (cTECs) and medullary TECs (mTECs). On the other hand, studies of human TECs have fallen behind because of the lack of a well-established method for purifying TEC populations. Here, we show that using 0.5 U/mL liberase to digest thymus tissue into single cells is quicker than using collagenase D or 0.1 U/mL li…

Cuproptosis of bronchial epithelial cells triggers airway remodeling in chronic asthma.

Airway remodeling is a critical cause of chronic asthma that is difficult to control. Cell death contributes to the development and progression of chronic asthma. However, the novel copper-dependent cell death, cuproptosis, and especially cuproptosis of bronchial epithelial cells, its role, and underlying mechanisms in airway remodeling are unclear. Repetitive OVA exposure was used to establish a mouse model of chronic asthma. The human bronchial epithelial cell line BEAS-2B (BEAS-2B cells) was incubated with recombinant human IL-4 (rhIL-4) and TGF-β1 to simulated airway remodeling in vitro. C…

Contact-compression induces inflammatory and remodeling responses in bronchial epithelial cells.

Lung implantable devices, such as stents and valves, are used as treatment for lung cancer and chronic obstructive pulmonary disease (COPD). They apply continuous compressive stress to airway tissue, potentially triggering adverse effects such as chronic inflammation, granulation tissue hyperplasia, and fibrosis at the implant site. To identify the biological responses underlying this process, we developed an in vitro contact-compression model that applies variable compressive stress to bronchial epithelial cells. Confluent layers of bronchial epithelial cells (16HBE) were subjected to compres…

Melatonin alleviates ferroptosis in lens epithelial cells via regulating METTL14 expression.

OBJECTIVES: Ferroptosis in lens epithelial cells (LECs) is increasingly recognized as a key pathological process in cataract formation, driven by oxidative stress, lipid peroxidation, and aberrant RNA modification. This study investigated the role of METTL14 in UVB-induced ferroptosis in LECs and explored the protective mechanism of melatonin (MT). MATERIALS AND METHODS: mRNA and protein levels of METTL14 and ferroptosis-related markers (GPX4, SLC7A11) were quantified. Lipid reactive oxygen species (ROS), malondialdehyde (MDA), and glutathione levels were measured to assess ferroptotic activi…

Thymic alveolar type 2 epithelial mimetic cells revealed by RUNX1 deficiency.

Autoimmune diseases can affect the lungs, and the mechanisms used to prevent autoimmune lung disease are incompletely understood. Recent studies in the thymus have identified unique populations of medullary thymic epithelial cells (mTECs) called mimetic cells that transcriptionally mimic peripheral epithelial populations. These mimetic cells have important functions in the thymus in immune tolerance. Here, we used a mouse line with thymic-specific deletion of Runt-related transcription factor 1 (Runx1) to identify a new mimetic cell akin to alveolar type 2 (AT2) lung epithelial cells. AT2 mTEC…

KDM6A suppresses proliferation and epithelial-mesenchymal transition in prostate cancer cells.

BACKGROUND AND OBJECTIVE: Prostate cancer is one of the most common male malignancies worldwide. KDM6A, a histone H3K27 demethylase, has been implicated in various cancers; however, its specific functions in prostate cancer remain controversial. This study aimed to investigate the regulatory role of KDM6A in castration-resistant prostate cancer (CRPC). METHODS: We analyzed the GEO public database and conducted in vitro and in vivo experiments to examine the function of KDM6A in CRPC. RESULTS: KDM6A expression was downregulated in CRPC compared with primary prostate cancer. KDM6A acted as a t

Overexpression of GDE1 decreases glycerophosphoinositol in intestinal epithelial cells in vivo.

Glycerophosphoinositol (GPI), a product of phospholipases acting on membrane phosphoinositides, is increasingly being recognized as an important bioactive molecule. However, its cellular functions in vivo remain unexplored. Recently, several mammalian glycerophosphodiesterases (GDEs) have shown to be involved in the catabolism of GPI, possibly regulating its intracellular levels. Given the growing interest in the biological significance of GPI in intestinal epithelial cells, this study focused on screening a GDE involved in intracellular GPI metabolism. Our results showed that GDE1 mRNA was hi…
