Tangeretin Targets Macrophage M1 Polarization to Alleviate Liver Injury in a Murine Model of Hepatic Sinusoidal Obstruction Syndrome.

Hepatic sinusoidal obstruction syndrome (HSOS) is a life-threatening liver microvascular disorder characterized by hepatic sinusoidal endothelial cells (HSECs) injury, for which safe and effective pharmacological treatments are currently lacking. Tangeretin (TAN), a natural polymethoxylated flavone, exhibits anti-inflammatory, antioxidant, and hepatoprotective properties. However, its therapeutic potential and mechanisms in HSOS remain unclear. This study investigated the protective role and molecular mechanisms of TAN against monocrotaline (MCT)-induced HSOS. In the murine HSOS model, TAN int

Probiotic-Driven Metabolic Transformation in Dark Tea: Enhanced Bioactives and Gut Microbiota Modulation Underlie Its Alleviation Effect Against Constipation.

Constipation is a widespread gastrointestinal condition with complex causes and limited therapeutic efficacy of current treatments. Probiotics and natural-function foods like dark tea (DT) have emerged as promising alternatives. We investigated the protective effects of dark tea (DT) and Lactiplantibacillus paracasei K34-fermented dark tea (FDT) against constipation induced by loperamide (LOP) in mice. FDT exhibited a superior effect over DT in alleviating constipation, as evidenced by increased fecal water content, reduced defecation time, and accelerated small intestinal transit. FDT also re

The Role of Flavonoids in Alleviating Mammary Gland Inflammation: A Review.

Mammary gland inflammation, clinically expressed as mastitis in dairy ruminants and lactating women, arises from a self-amplifying loop of pathogen sensing, oxidative stress and innate immune activation, and remains a leading driver of antibiotic use, milk-quality loss and morbidity. Mounting pressure to reduce antimicrobial dependence has refocused attention on flavonoids-structurally diverse plant polyphenols with multi-target bioactivity, derived mainly from in vitro and rodent mastitis models. This review integrates contemporary evidence on the six principal flavonoid subclasses within a u

Dihydroxyflavones as potential therapeutic agents against inflammation and renal injury via p38 MAP kinase inhibition: an in silico and experimental study.

Nephrotoxicity poses a significant clinical challenge, contributing to renal complications and increased mortality, thus necessitating novel therapeutics with superior efficacy and safety. Our study addresses this need by evaluating flavone derivatives as potential modulators of p38-MAPK to tackle both inflammation and renal damage, using cell-based assays, cytotoxicity profiling, molecular docking, and ADME-T profiling. Notably, 3,6-dihydroxyflavone (3,6-DHF) and 3,7-dihydroxyflavone (3,7-DHF) demonstrated a significant reduction in KIM-1 mRNA expression in CCl4-induced damaged renal epitheli

A potent icaritin derivative ameliorates Parkinson's disease via GPER-mediated mitophagy restoration and dopaminergic neuroprotection.

Icaritin (ICT), a major prenylated flavone from Epimedium with anti-osteoporosis, anti-cancer, and anti-inflammatory properties, is limited clinically by poor water solubility and low bioavailability. In this study, ICT derivatives were prepared via multi-step chemical reactions and five ICT derivatives (ICTA, ICTB, ICTC, ICTD and ICTE) were successfully synthesized. Compared with ICT, most derivatives exhibited improved solubility and reduced cytotoxicity. Notably, ICTC showed a significant improvement in aqueous solubility (at least 12-fold higher), favorable lower cytotoxicity (at least 2-f

How Gut Microbes Transform Dietary Phytonutrients: Enzymatic Pathways and Human Metabolite Exposure.

Compounds identified in plant foods are often not the species that reach human tissues. Glycosides, tannins and incompletely absorbed aglycones enter the colon, where gut microorganisms hydrolyse, reduce, cleave, demethylate or dehydroxylate them. This critical narrative review examines pathways resolved beyond taxonomic association: daidzein-to-S-equol conversion; cooperative urolithin and enterolignan formation; flavone reduction and C-ring cleavage; stereoselective catechin metabolism; glucosinolate activation; and transformations of resveratrol and hydroxycinnamate-derived metabolites. Comprehensive metabolomic and pharmacological analyses elucidate antioxidant mechanisms and flavor profiles associated with red pericarp color protein in cooked brown rice.

Red pericarp color (OsRc) protein is the key regulator of red seed coat pigmentation in colored rice. However, its detailed role in regulating the cooking properties of brown rice remains unclear. Here, we evaluated the antioxidant properties and analyzed the metabolite profiles of cooked brown rice. The results showed that wild-type (WT) rice exhibited significantly greater antioxidant activity than the two rc mutant rice varieties. Moreover, the total phenolic and flavonoid contents of WT rice were 2.2-fold and 5.0-fold higher than OsRc knockout. Non-volatile metabolomics analysis via Ultra-

Characterization of intestinal microbiota in patients with psoriasis combined with metabolic syndrome.

The co-morbidity of psoriasis (PsO) and metabolic syndrome (MetS) is prevalent. While the mechanisms causing PsO in MetS remain unknown, it is understood that gut microbiota plays a critical role in maintaining immune and metabolic balance. However, the specific mechanisms and whether biomarkers are involved in the pathophysiological process of PSO in MetS patients have yet to be elucidated. This study involved 29 patients with PSO and MetS (PM) and 29 with MetS (M). It analyzed their clinical parameters, microbial composition, function, diagnostic, and predictive performance. The results unve

Antibacterial and Antioxidant Activity of Synthetic Polyoxygenated Flavonoids.

Flavonoids are an abundant class of naturally occurring compounds with broad biological activities, but their limited abundance in nature restricts their use in medicines and food additives. Here we present the synthesis and determination of the antibacterial and antioxidant activities of twenty-two structurally related flavonoids (five of which are new) by scientifically validated methods. Flavanones (FV1-FV11) had low inhibitory activity against the bacterial growth of MRSA 97-7. However, FV2 (C5,7,3',4' = OH) and FV6 (C5,7 = OH; C4' = SCH3) had excellent bacterial growth inhibitory activity

3,5,7-trihydroxyflavone restricts proliferation of androgen-independent human prostate adenocarcinoma cells by inducing ROS-mediated apoptosis and reduces tumour growth.

Flavonoids are among the largest groups of secondary metabolites. Studies suggest that dietary intake of flavonoids reduces the risk of cancer. 3,5,7-trihydroxyflavone (THF) belongs to the flavone class of flavonoids and potentially inhibits the growth of many cancers; however, it is unexplored in prostate cancer. This study reports the antiproliferative potential of THF in prostate cancer cell line via reactive oxygen species (ROS)-mediated cascades and examines the tumour reduction potential in swiss albino mice. The potency of THF was evaluated by employing cytotoxicity assays and wound hea

Eupatilin Ameliorates Hepatic Fibrosis and Hepatic Stellate Cell Activation by Suppressing β-catenin/PAI-1 Pathway.

The activation of hepatic stellate cells (HSCs) has proved to be pivotal in hepatic fibrosis. Therefore, the suppression of HSC activation is an effective anti-fibrotic strategy. Although studies have indicated that eupatilin, a bioactive flavone found in Artemisia argyi, has anti-fibrotic properties, the effect of eupatilin on hepatic fibrosis is currently unclear. In this study, we used the human hepatic stellate cell line LX-2 and the classical CCl4-induced hepatic fibrosis mouse model for in vitro and vivo experiments. We found that eupatilin significantly repressed the levels of the fibro
