Enhancing salicylic acid levels by its exogenous pretreatment to mitigate Fusarium oxysporum-induced biotic stress in Vigna mungo: defense pathways insights.

Fusarium oxysporum disrupts redox homeostasis in Vigna mungo, likely by interfering with salicylic acid signaling, which can be ameliorated by boosting PAL and its related pathways via salicylic acid pretreatment. Fusarium oxysporum, a widespread soil-borne fungus, significantly threatens global crops. This study centers on elucidating the infection strategies employed by F. oxysporum against a new and underexplored host Vigna mungo, a leguminous crop of high agronomic value, and the defense mechanisms that can be activated against the infection, aiming to uncover how these responses can be le…

Integrated analysis of transcriptomics and defense-related phytohormones to discover hub genes conferring maize Gibberella ear rot caused by Fusarium Graminearum.

BACKGROUND: Gibberella ear rot (GER) is one of the most devastating diseases in maize growing areas, which directly reduces grain yield and quality. However, the underlying defense response of maize to pathogens infection is largely unknown. RESULTS: To gain a comprehensive understanding of the defense response in GER resistance, two contrasting inbred lines 'Nov-82' and 'H10' were used to explore transcriptomic profiles and defense-related phytohormonal alterations during Fusarium graminearum infection. Transcriptomic analysis revealed 4,417 and 4,313 differentially expressed genes (DEGs) fr…

Bacillus tequilensis PKDN31 and Bacillus licheniformis PKDL10 -As double headed swords to combat Fusarium oxysporum f. sp. lycopersici induced tomato wilt.

Wilt disease, caused by Fusarium oxysporum. f. sp. lycopersici, is a global threat to tomato production that needs to be addressed seriously. The current research envisages the use of two self-compatible Bacillus strains, Bacillus tequilensis PKDN31 and Bacillus licheniformis PKDL10, in a combinatorial approach. The spent supernatant of liquid cultures from strains PKDN31 and PKDL10 showed in vitro antifungal activity against Fusarium sp. attaining an inhibition percentage of 95.33% and 96.54%, respectively. The bacterial isolates lytic activity against Fusarium oxysporum was evaluated by scan…

Carbendazim-resistance associated β2 -tubulin substitutions increase deoxynivalenol biosynthesis by reducing the interaction between β2 -tubulin and IDH3 in Fusarium graminearum.

Microtubule is a well-known structural protein participating in cell division, motility and vesicle traffic. In this study, we found that β2 -tubulin, one of the microtubule components, plays an important role in regulating secondary metabolite deoxynivalenol (DON) biosynthesis in Fusarium graminearum by interacting with isocitrate dehydrogenase subunit 3 (IDH3). We found IDH3 negatively regulate DON biosynthesis by reducing acetyl-CoA accumulation in F. graminearum and DON biosynthesis was stimulated by exogenous acetyl-CoA. In addition, the expression of IDH3 significantly decreased in the c…

Biotransformation of anabolic compound methasterone with Macrophomina phaseolina, Cunninghamella blakesleeana, and Fusarium lini, and TNF-α inhibitory effect of transformed products.

Microbial transformation of methasterone (1) was investigated with Macrophomina phaseolina, Cunninghamella blakesleeana, and Fusarium lini. Biotransformation of 1 with M. phaseolina yielded metabolite 2, while metabolites 3-7 were obtained from the incubation of 1 with C. blakesleeana. Metabolites 8-13 were obtained through biotransformation with F. lini. All metabolites, except 13, were found to be new. Methasterone (1) and its metabolites 2-6, 9, 10, and 13 were then evaluated for their immunomodulatory effects against TNF-α, NO, and ROS production. Among all tested compounds, metabolite 6 s…

Fungal Peritonitis Due to Fusarium solani Species Complex Sequential Isolates Identified with DNA Sequencing in a Kidney Transplant Recipient in Brazil.

Fungal peritonitis is a rare serious complication most commonly observed in immunocompromised patients under peritoneal dialysis. Nevertheless, this clinical condition is more difficult to treat than bacterial peritonitis. Bacterial peritonitis followed by the use of antibiotics is the main risk factor for developing fungal peritonitis. Candida spp. are more frequently isolated, and the isolation of filamentous fungi is only occasional. Here we describe a case of Fusarium solani species complex peritonitis associated with bacterial peritonitis in a female kidney transplant recipient with previ…

Baseline sensitivity to the SDHI fungicide pydiflumetofen and resistance mechanisms in Fusarium oxysporum, one of the causal agents of soybean root rot in China.

BACKGROUND: Soybean root rot (SRR) is a devastating disease that severely reduces yield and quality in China, with Fusarium oxysporum recognized as one of the most prevalent causal agents. Pydiflumetofen, a new-generation succinate dehydrogenase inhibitor (SDHI) fungicide, exhibits potent efficacy against Fusarium spp. However, the resistance risk and underlying mechanisms of the SRR pathogen to pydiflumetofen remain largely unexplored. RESULTS: Baseline sensitivity was determined using 104 isolates collected from five major soybean-producing provinces. The half-maximal effective concentratio…

A novel endophytic Penicillium sp. JL76001 and its metabolite sclerotiorin: A multi-target and sustainable strategy for biocontrol of tomato Fusarium wilt.

Tomato Fusarium wilt, caused by Fusarium oxysporum f. sp. lycopersici (FOL), is a devastating disease that leads to yield losses of up to 67% in severely affected regions. Current control strategies, such as chemical treatments and resistant cultivars, face challenges including environmental toxicity and pathogen resistance. In this study, we isolated an endophytic fungus, Penicillium sp. JL76001 (JL76001), from the medicinal plant Asarum forbesii, which exhibited strong antagonism against FOL. Plate confrontation assays revealed a 40.48% inhibition of FOL growth, attributed to the secondary m…

An InDel insertion in the promoter of a UDP-ᴅ-glucuronate 4-epimerase 1 gene enhances maize resistance to Fusarium ear rot.

Fusarium ear rot (FER), caused by Fusarium verticillioides, results in substantial yield losses and poses a significant threat to maize production worldwide. However, the genetic basis of FER resistance remains poorly understood. Utilizing QTL-seq and association analysis, we identified a gene encoding UDP-ᴅ-glucuronate 4-epimerase 1 (ZmGAE1). A 141-base pair insertion was revealed as the natural functional variation in the promoter of ZmGAE1, which decreases its expression and enhances resistance to FER. Functional validation confirmed that ZmGAE1 acts as a negative regulator of maize resista…

Effect of in Planta Treatment of 'Cavendish' Banana with Herbicides and Fungicides on the Colonisation and Sporulation by Fusarium oxysporum f.sp. cubense Subtropical Race 4.

Fusarium wilt caused by the soil-borne fungus Fusarium oxysporum f.sp. cubense (Foc) is a significant constraint to banana production worldwide, with the recent expansion of banana growing regions impacted by Foc Tropical Race 4 (TR4). The lack of commercially acceptable Cavendish cultivars with Foc resistance means the only current means of effective control is through strict quarantine and inoculum management. One method of control that is currently advocated includes the removal of infected plants which have been killed using herbicide injections. The aim of this work was to examine the eff…

Fungal biotransformation of chlorogenic and caffeic acids by Fusarium graminearum: New insights in the contribution of phenolic acids to resistance to deoxynivalenol accumulation in cereals.

Fusarium Head Blight and Gibberella Ear Rot, mainly caused by the fungi Fusarium graminearum and Fusarium culmorum, are two of the most devastating diseases of small-grain cereals and maize. In addition to yield loss, these diseases frequently result in contamination of kernels with toxic type B trichothecenes. The potential involvement of chlorogenic acid in cereal resistance to Fusarium Head Blight and Gibberella Ear Rot and to trichothecene accumulation was the focus of this study. The effects of chlorogenic acid and one of its hydrolyzed products, caffeic acid, on fungal growth and type B …

Jasmonic Acid, Abscisic Acid, and Salicylic Acid Are Involved in the Phytoalexin Responses of Rice to Fusarium fujikuroi, a High Gibberellin Producer Pathogen.

Fusarium fujikuroi, the causal agent of bakanae disease, is the main seedborne pathogen on rice. To understand the basis of rice resistance, a quantitative method to simultaneously detect phytohormones and phytoalexins was developed by using HPLC-MS/MS. With this method dynamic profiles and possible interactions of defense-related phytohormones and phytoalexins were investigated on two rice cultivars, inoculated or not with F. fujikuroi. In the resistant cultivar Selenio, the presence of pathogen induced high production of phytoalexins, mainly sakuranetin, and symptoms of bakanae were not obse…

FgFim, a key protein regulating resistance to the fungicide JS399-19, asexual and sexual development, stress responses and virulence in Fusarium graminearum.

Fimbrin is an actin-bundling protein found in intestinal microvilli, hair cell stereocilia and fibroblast filopodia. Its homologue Sac6p has been shown to play a critical role in endocytosis and diverse cellular processes in Saccharomyces cerevisiae. FgFim from the wheat scab pathogenic fungus Fusarium graminearum strain Y2021A, which is highly resistant to the fungicide JS399-19, was identified by screening a mutant library generated by HPH-HSV-tk cassette-mediated integration. The functions of FgFim were evaluated by constructing a deletion mutant of FgFim, designated ΔFgFim-15. The deletion…

The spectrum of Fusarium infection in immunocompromised patients with haematological malignancies and in non-immunocompromised patients: a single institution experience over 10 years.

Fusarium is a newly emerging fungal pathogen associated with significant morbidity and mortality in the immunocompromised host. We have reviewed our hospital's experience with Fusarium between 1985 and 1995. Fusarium species were isolated from 22 specimens, representing 11 patients. Cases were not clustered by time period. The median age of the patients was 36.5 years (range 17-69 years). The sources of the organism were 12 skin lesions from eight patients, seven blood cultures from two patients and one specimen each from a Hickman catheter tip, nail clippings and a bronchoalveolar lavage. Sev…

In Situ Fusarium Mycotoxin Mitigation During Wholegrain Wheat Flour Fermentation with Sourdough Lactic Acid Bacteria and Further Insights into Reduction Mechanisms.

Fusarium mycotoxins are major contaminants of cereal-based products, yet effective clean-label mitigation strategies are still limited. Although lactic acid bacteria (LAB) have been extensively investigated as biological tools for mycotoxin reduction, evidence is largely based on in vitro studies. Here, nineteen LAB strains representative of sourdough microbiota were screened for their ability to reduce deoxynivalenol (DON) in wholegrain wheat dough. Pronounced strain-dependent differences were observed, leading to the selection of Lactiplantibacillus plantarum 1A7, Limosilactobacillus ferment…
