Hypoxia is a key component of the tumor microenvironment and represents a well-documented source of therapeutic failure in clinical oncology. Recent work has provided support for the idea that non-coding RNAs, and in particular, microRNAs, may play important roles in the adaptive response to low oxygen in tumors. Specifically, all published studies agree that the induction of microRNA-210 (miR-210) is a consistent feature of the hypoxic response in both normal and malignant cells. miR-210 is a robust target of hypoxia-inducible factors, and its overexpression has been detected in a variety of

Neprilysin (NEP) is an integral membrane-bound metallopeptidase with a wide spectrum of substrates and physiological functions. It plays an important role in proteolytic processes in the kidney, cardiovascular regulation, immune response, cell proliferation, foetal development etc. It is an important neuropeptidase and amyloid-degrading enzyme which makes NEP a therapeutic target in Alzheimer's disease (AD). Moreover, it plays a preventive role in development of cancer, obesity and type-2 diabetes. Recently a role of NEP in COVID-19 pathogenesis has also been suggested. Despite intensive resea

Switching of cellular energy production from oxidative phosphorylation (OXPHOS) by mitochondria to aerobic glycolysis occurs in many types of tumors. However, the significance of this switching for the development of gastric carcinoma and what connection it may have to Helicobacter pylori infection of the gut, a primary cause of gastric cancer, are poorly understood. Therefore, we investigated the expression of OXPHOS complexes in two types of human gastric carcinomas ("intestinal" and "diffuse"), bacterial gastritis with and without metaplasia, and chemically induced gastritis by using immuno

Eradication of cancer cells while minimizing damage to healthy cells is a primary goal of cancer therapy. Highly selective drugs are urgently needed. Here we demonstrate a new prodrug strategy for selective cancer therapy that utilizes increased histone deacetylase (HDAC) and tumour-associated protease activities produced in malignant cancer cells. By coupling an acetylated lysine group to puromycin, a masked cytotoxic agent is created, which is serially activated by HDAC and an endogenous protease cathepsin L (CTSL) that remove the acetyl group first and then the unacetylated lysine group lib

Environmental exposures during development can alter susceptibility later in life to adult diseases including uterine leiomyoma, a phenomenon termed developmental reprogramming. The goal of this study was to identify genes developmentally reprogrammed by diethylstilbestrol (DES) and aberrantly expressed in leiomyomas. Transcriptional profiling identified 171 genes differentially expressed in leiomyomas relative to normal myometrium, of which 6/18 genes with putative estrogen responsive elements and confirmed to be estrogen-responsive in neonatal uteri were reprogrammed by neonatal DES exposure

The treatment of advanced stage thymomas and thymic carcinomas is a multimodal therapy. New therapeutic targets are currently under investigation, including the epidermal growth factor receptor (EGFR) as well as KIT. A number of studies have shown protumorigenic potential of Cyclooxygenase-2 (COX-2) in a variety of human malignancies, but so far it is unknown whether COX-2 is expressed in primary malignancies of the thymus. Using tissue microarrays, the expression of COX-2, microsomal-PGES-1 and -PGES-2 (mPGES-1 and mPGES-2), as well as EGFR was evaluated in different subtypes of thymoma and t

OBJECTIVES: Kallikrein gene 5 (KLK5; formerly designated as kallikrein-like gene 2, or human stratum corneum tryptic enzyme) is one of the new members of the human kallikrein gene family on chromosome 19q13.4. Although it is expressed at low levels in various tissues, KLK5 expression is highest in the human mammary gland and testis. Previous investigations have established that the expression of KLK5 is estrogen and progestin driven in the BT-474 breast cancer cell line. In this study, we focused on KLK5 expression in normal and cancerous testicular tissue.

Chemotherapy remains a cornerstone of cancer treatment, yet therapeutic resistance continues to limit durable clinical responses. MicroRNAs (miRNAs) are key post-transcriptional regulators of gene expression that orchestrate diverse cellular processes implicated in drug sensitivity and resistance. However, the contribution of miRNA regulatory networks to chemotherapy response has not been systematically characterized using clinical patient cohorts. Here we present DrugMiR, an integrative multi-omics modeling framework that integrates drug response-associated miRNAs with experimentally validate

Hepatocellular carcinoma (HCC) remains a leading cause of cancer-related mortality worldwide, underscoring the need for robust molecular biomarkers to improve prognosis and therapeutic strategies. Although advances have been made in imaging, surgery, as well as systemic therapies, the prognosis of HCC remains poor due to late detection, high recurrence, and molecular heterogeneity, underscoring the significance of identifying robust prognostic biomarkers and therapeutic targets. mRNA-sequencing data from the TCGA-HCC cohort were examined to recognize differentially expressed genes (DEGs) betwe

The greatest cause of death from breast cancer is metastasis, yet little is known about the molecular mechanisms behind this phenomenon. Using four publically accessible datasets, we conducted a thorough transcriptome analysis of 187 samples from seven breast cancer metastatic sites: the brain, bone, lung, liver, lymph nodes, skin, and local-regional skin (skinlr). Of the 12,005 genes that were found to be shared by all samples in this investigation, 604-885 differentially expressed genes (DEGs) were unique to each metastatic location. Pathways including PI3K-Akt signaling, prolactin signaling

The lncRNA NEAT1 is overexpressed in multiple myeloma (MM) plasma cells and plays a key role in MM pathogenesis. NEAT1 is involved in ceRNA network in several cancers; however, data in MM are virtually absent. This study identified a NEAT1-driven ceRNA network involving 96 miRNAs and 40 target genes, selected as concurrently downregulated in NEAT1-KD AMO1 cells and upregulated in NEAT1-overexpressing AMO1 cells (AMO1-OVX). The co-expression of NEAT1 and the targets was validated in MM patients (GSE116294, GSE13591, GSE6477, CoMMpass), and in NEAT1-KD NCI-H929, LP1, and KMS27 cell lines, showin

Assessing the contributions of microRNAs to gene expression regulation and treatment outcomes in malignancies is challenging. Traditional analyses focus on individual microRNAs, overlooking competitive binding among RNA species sharing common microRNAs. We evaluated the impact of microRNA's impact on the survival of patients with mesothelioma in response to treatment with immune checkpoint inhibitors (ICI) within the context of competing endogenous RNA (ceRNA) networks. Using two large public mesothelioma datasets, we constructed ceRNA networks (ceRNETs) and analyzed differentially expressed m

BACKGROUND: Cancers are complex diseases that have heterogeneous genetic drivers and varying clinical outcomes. A critical area of cancer research is organizing patient cohorts into subtypes and associating subtypes with clinical and biological outcomes for more effective prognosis and treatment. Large-scale studies have collected a plethora of omics data across multiple tumor types, providing an extensive dataset for stratifying patient cohorts. Network-based stratification (NBS) approaches have been presented to classify cancer tumors using somatic mutation data. A challenge in cancer strati
