A 2-sample Mendelian randomization study exploring the causal effect of depression on rotator cuff injury.

The aim of this study was to use Mendelian randomization (MR) method to deeply explore the possible causal association between depression and rotator cuff injury. Using pooled data from genome-wide association studies covering large-scale populations, independent genetic loci associated with depression and rotator cuff injuries and showing significant genetic associations in populations of European ancestry were carefully selected as study instrumental variables. The potential causal effect of depression on rotator cuff injury was systematically assessed by implementing 3 MR analysis strategie

Hyperthyroidism drives gout risk: A Mendelian randomization observational study.

The causal interplay between thyroid dysfunction and gout remains controversial because of confounding and reverse causation in observational studies. Using Mendelian randomization (MR), this study investigated the causal effects of hyperthyroidism and hypothyroidism on the risk of gout. Summary statistics from the UK Biobank and FinnGen genome-wide association study databases were analyzed using 2-sample MR. Genetic instruments for thyroid dysfunction were selected under stringent criteria (P < 5 × 10-8, r2 < 0.001). Causal estimates were derived using inverse-variance weighted regression, su

Investigating the genetic causality between periodontitis and intracranial aneurysms: A 2-sample Mendelian randomization study.

Previous studies have suggested an association between periodontitis and intracranial aneurysms (IAs). However, it remains unclear whether the association is causal. This study systematically investigated the potential genetic link between periodontitis and IAs, including the formation and rupture of IAs. This study utilized publicly available genome-wide association study summary statistics data for a 2-sample bidirectional Mendelian randomization (MR) analysis. The main statistical analysis method used was inverse variance weighting. The reliability of the results was verified through sensit

Neuroticism and inflammatory skin diseases: a bidirectional Mendelian randomization study.

Previous observational studies have linked inflammatory skin diseases with mental health issues and neuroticism. However, the specific impact of neuroticism and its subclusters (i.e. worry, depressed affect, and sensitivity to environmental stress and adversity) on these conditions remains underexplored. In this work, we explored causal associations between common inflammatory skin diseases and neuroticism. We conducted a two-sample, bidirectional Mendelian randomization (MR) analysis using data from genome-wide association studies in psoriasis, atopic dermatitis, neuroticism and relevant gene

Evaluating the effect of childhood sunburn on the risk of cutaneous melanoma through Mendelian randomization.

Despite numerous observational studies indicating an increased risk of cutaneous melanoma (CM) due to childhood sunburn, no studies have established a definitive cause-and-effect relationship. Therefore, our objective was to employ a Mendelian randomization (MR) design to explore a possible causal association between childhood sunburn and the risk of CM. To investigate the causal relationship between childhood sunburn and CM, we used large-scale genetic summary-level data from genome-wide association studies (GWAS), including childhood sunburn (n = 346,955) and CM (n = 262,288), building upon

Shared genetic architecture between constipation and aging-related phenotypes: Insights from large-scale genome-wide cross-trait analysis.

While the frequency of constipation increases sharply with age, the genetic links between constipation and aging-related phenotypes remain largely unexplored. So far, research has typically focused only on individual characteristics while neglecting broader genetic connections. In this study, we evaluated the shared genetic architecture of constipation and aging-related phenotypes in order to better understand the causes of aging-related intestinal dysfunction. Using publicly available genome-wide association study summary statistics, we developed a multi-stage analysis pipeline to identify co

Glucose Levels and Granulocyte Colony-Stimulating Factor May Partially Mediate the Association Between Genetic Liability to Diabetes and Carpal Tunnel Syndrome.

BACKGROUND: Carpal tunnel syndrome (CTS) is the most common entrapment neuropathy, imposing substantial clinical and socioeconomic burdens due to chronic pain, functional disability, and reduced quality of life. While diabetes is a well-established risk factor for CTS in observational studies, the causal nature of this association and the mediating roles of glucose levels and inflammatory pathways remain unclear. METHODS: We conducted a two-sample Mendelian randomization study using genetic data primarily from the UK Biobank. We assessed the causal effect of genetic liability to diabetes on C

The dominant findings of a recessive man: from Mendel's kid pea to kidney.

The research of Mendel, born two centuries ago, still has many direct implications for our everyday clinical work. He introduced the terms "dominant" and "recessive" characters and determined their 3:1 ratio in the offspring of heterozygous "hybrid" plants. This distribution allowed calculation of the number of the phenotype-determining "elements," i.e., the alleles, and has been used ever since to prove the monogenic origin of a disorder. The Mendelian inheritance of monogenic kidney disorders is still of great help in distinguishing them from those with multifactorial origin in clinical prac

Causal Effects of Homocysteine, Folate, and Cobalamin on Kidney Function: A Mendelian Randomization Study.

Blood homocysteine level and related vitamin levels are associated with various health outcomes. We aimed to assess causal effects of blood homocysteine, folate, and cobalamin on kidney function in the general population by performing Mendelian randomization (MR) analysis. Genetic instruments for blood homocysteine, folate, and cobalamin levels were introduced from a previous genome-wide association (GWAS) meta-analysis of European individuals. Summary-level MR analysis was performed for the estimated glomerular filtration rate (eGFR) from the CKDGen consortium GWAS that included 567,460 Europ

Cortical Morphology and Susceptibility to Fall-Related Fractures: A Bidirectional Mendelian Randomization Study.

AIM: Injurious falls and low-energy fractures after a simple fall cause substantial morbidity in older adults; observational brain-structure associations are vulnerable to confounding and reverse causation. To test whether predicted regional cortical thickness or surface area causally influences falls and fractures after a simple fall, and to evaluate reverse causality. METHODS: Bidirectional two-sample Mendelian randomization used cortical morphology across 34 regions (> 51 000 Europeans). Outcomes were falls/tendency to fall and simple-fall fractures. Causal association between blood immune cells and chronic hepatitis B: Univariable and multivariable Mendelian randomization.

Current evidence underscores the pivotal role of the immune system in the pathogenesis of chronic hepatitis B (CHB). Nevertheless, the precise causal nexus between immune cells and CHB remains enigmatic, with numerous unresolved queries. Leveraging data from the latest summary-level genome-wide association studies, we examined immune cell genetics in 3757 individuals from Sardinia. CHB data predominantly originated from the Biobank Japan project, encompassing 1394 cases and 211,059 controls. Our investigation entailed a comprehensive univariable Mendelian randomization (MR) analysis to dissect

Relationship between serum vitamin D levels and thyroid- and parathyroid-related diseases: a Mendelian randomisation study.

Previous studies have indicated an association between vitamin D and thyroid- and parathyroid-related diseases. However, it remains unclear whether it is a cause of the disease, a side effect of treatment or a consequence of the disease. The Mendelian randomisation (MR) study strengthens the causal inference by controlling for non-heritable environmental confounders and reverse causation. In this study, a two-sample bidirectional MR analysis was conducted to investigate the causal relationship between serum vitamin D levels and thyroid- and parathyroid-related diseases. Inverse variance weight

Relationships between immune cells, blood metabolites, and intrahepatic cholangiocarcinoma: a Mendelian randomization study with experimental validation.

INTRODUCTION: Increasing studies have found links between immune traits and intrahepatic cholangiocarcinoma (ICC). However, the presence of confounding variables makes it difficult to determine if this correlation shows a real cause-and-effect relationship. METHODS: To ascertain the genetic association between immune cell profiles and the risk of ICC, we employed two-sample Mendelian randomization (MR) to figure out the potential therapeutic targets. A mediation analysis was performed to identify potential blood metabolite mediators, thereby elucidating the underlying biological mechanisms. T
