Abnormal functional lateralization and cooperation in primary angle-closure glaucoma correlate with cell type-specific transcriptional signatures.

BACKGROUND: Primary angle-closure glaucoma (PACG) damages retinal ganglion cells (RGCs) and is associated with neurodegeneration. This study used resting-state functional magnetic resonance imaging (fMRI) to analyze hemispheric lateralization and cooperative functional alterations in PACG. Machine learning assessed the classification efficacy of neuroimaging indicators, while integrated transcriptomics described spatial relationships between neuroimaging changes and genes, neurotransmitters, and cell types. METHODS: Resting-state fMRI data were collected from 101 subjects (44 patients with PA

A Novel Rabbit Model of Angle-Closure Glaucoma Via Ciliary Block and Elevated Trans-Lens Pressure Difference.

PURPOSE: Angle closure is a hallmark of primary angle-closure glaucoma (PACG), but the upstream biomechanical drivers initiating this process remain poorly understood. It has been recently proposed that ciliary block elevates the trans-lens pressure difference, initiating anterior chamber shallowing. However, this mechanism lacks direct in vivo validation. To address this gap, we established and characterized a novel rabbit model of angle-closure glaucoma induced by ciliary block-mediated trans-lens pressure difference. METHODS: Thirty rabbits were divided into three groups: intraocular lens

Topological Organization of the Brain Network in Patients with Primary Angle-closure Glaucoma Through Graph Theory Analysis.

Primary angle-closure glaucoma (PACG) is a sight-threatening eye condition that leads to irreversible blindness. While past neuroimaging research has identified abnormal brain function in PACG patients, the relationship between PACG and alterations in brain functional networks has yet to be explored. This study seeks to examine the influence of PACG on brain networks, aiming to advance knowledge of its neurobiological processes for better diagnostic and therapeutic approaches utilizing graph theory analysis. A cohort of 44 primary angle-closure glaucoma (PACG) patients and 44 healthy controls

Functional investigation suggests CNTNAP5 involvement in glaucomatous neurodegeneration obtained from a GWAS in primary angle closure glaucoma.

Primary angle closure glaucoma (PACG) affects more than 20 million people worldwide, with an increased prevalence in south-east Asia. In a prior haplotype-based Genome Wide Association Study (GWAS), we identified a novel CNTNAP5 genic region, significantly associated with PACG. In the current study, we have extended our perception of CNTNAP5 involvement in glaucomatous neurodegeneration in a zebrafish model, through investigating phenotypic consequences pertinent to retinal degeneration upon knockdown of cntnap5 by translation-blocking morpholinos. While cntnap5 knockdown was successfully vali
