Crosstalk between HIV and hepatitis C virus during co-infection.

An estimated one-third of individuals positive for HIV are also infected with hepatitis C virus (HCV). Chronic infection with HCV can lead to serious liver disease including cirrhosis and hepatocellular carcinoma. Liver-related disease is among the leading causes of death in patients with HIV, and individuals with HIV and HCV co-infection are found to progress more rapidly to serious liver disease than mono-infected individuals. The mechanism by which HIV affects HCV infection in the absence of immunosuppression by HIV is currently unknown. In a recent article published in BMC Immunology, Qu e

HIV-1 infection and the PPARγ-dependent control of adipose tissue physiology.

PPARγ is a ligand-dependent master transcription factor controlling adipocyte differentiation as well as multiple biological processes taking place in other cells present in adipose tissue depots such as macrophages. Recent research indicates that HIV-1 infection-related events may alter adipose tissue biology through several mechanisms involving PPARγ, ranging from direct effects of HIV-1-encoded proteins on adipocytes to the promotion of a proinflammatory environment that interferes with PPARγ actions. This effect of HIV-1 on adipose tissue cells can occur even in the absence of direct infec

The Vaginal Acquisition and Dissemination of HIV-1 Infection in a Novel Transgenic Mouse Model Is Facilitated by Coinfection with Herpes Simplex Virus 2 and Is Inhibited by Microbicide Treatment.

UNLABELLED: Epidemiological studies have demonstrated that herpes simplex virus 2 (HSV-2) infection significantly increases the risk of HIV-1 acquisition, thereby contributing to the expanding HIV-1 epidemic. To investigate whether HSV-2 infection directly facilitates mucosal HIV-1 acquisition, we used our transgenic hCD4/R5/cT1 mouse model which circumvents major entry and transcription blocks preventing murine HIV-1 infection by targeting transgenic expression of human CD4, CCR5, and cyclin T1 genes to CD4(+) T cells and myeloid-committed cells. Productive infection of mucosal leukocytes, pr

No change in key HIV target cell markers following initiation of three progestin-based hormonal contraception methods: findings from the CHIME study.

INTRODUCTION: Depot medroxyprogesterone acetate (DMPA) injectable, etonogestrel subdermal implant (ENG-implant), and levonorgestrel intrauterine device (LNG-IUD) are effective, widely used female hormonal contraceptives (HC). Observational studies, but not a randomized trial, suggest increased risk of HIV acquisition with HC use, particularly DMPA. Sexual acquisition of HIV occurs via CD4+ T cells expressing C-C chemokine receptor type 5 (CCR5), though other immunologic cells play a role. This study examined longitudinal changes in CCR5+ T cells and other immunologic cells in the female genita

HIV-1 capsid stability and reverse transcription are finely balanced to minimize sensing of reverse transcription products via the cGAS-STING pathway.

UNLABELLED: A critical determinant for early post-entry events, the HIV-1 capsid (CA) protein forms the conical core when it rearranges around the dimeric RNA genome and associated viral proteins. Although mutations in CA have been reported to alter innate immune sensing of HIV-1, a direct link between core stability and sensing of HIV-1 nucleic acids has not been established. Herein, we assessed how manipulating the stability of the CA lattice through chemical and genetic approaches affects innate immune recognition of HIV-1. We found that destabilization of the CA lattice resulted in potent

Oral PrEP pause and restart among adolescent girls and young women (AGYW) who initiated PrEP in an HIV prevention program in Kampala, Uganda: a qualitative study.

INTRODUCTION: There is interest in PrEP uptake amongst young people at high risk of HIV. However, there is a significant decline in PrEP starting from three to six months after PrEP initiation, thus affecting PrEP persistence and continuation due to pausing and stopping of PrEP, with a few restarting it. Understanding why AGYW pause PrEP use and what motivates the few to restart is critical for enhancing PrEP adherence and continuation. We explored pausing of PrEP and restarting it amongst AGYW in an HIV prevention program, in Kampala, Uganda, to inform future HIV health education intervention

Effect of Routine Opt-Out HIV Screening on Emergency Department Operational Metrics: Results From the Pragmatic Randomized HIV TESTED Trial.

STUDY OBJECTIVE: To assess the effect of routine opt-out HIV screening on emergency department (ED) patient throughput. METHODS: We performed a multicenter, prospective, pragmatic 3-arm, parallel-group, randomized clinical trial to evaluate the effectiveness of 3 triage-driven, opt-out HIV screening strategies-nontargeted, enhanced targeted, and traditional targeted screening-when fully integrated into usual ED practices. As the 3 screening approaches were performed concurrently, the trial was nested in an equivalent time samples quasi-experiment to allow for the evaluation of HIV screening o

HIV-TB Coinfection among 57 Million Pregnant Women, Obstetric Complications, Alcohol Use, Drug Abuse, and Depression.

OBJECTIVE: HIV and tuberculosis represent diseases of major public health importance worldwide. Very little is known about HIV-TB coinfection among pregnant women, especially from industrialized settings. In this study, we examined the association between TB, HIV, and HIV-TB coinfection among pregnant mothers and obstetric complications, alcohol use, drug abuse, and depression. METHOD: We examined inpatient hospital discharges in the United States from January 1, 2002, through December 31, 2014. We employed multivariable survey logistic regression to generate adjusted estimates for the associ

HIV testing among pregnant wives of migrant men in a rural district of India: urgent call for scale up.

In India, despite the fact that more pregnant women are being tested for HIV under the purview of the Prevention of Parent-to-Child HIV Transmission program, official figures indicate low rates of HIV testing, evidencing missed opportunities for HIV prevention. The present study examined the prevalence of HIV testing and the barriers to testing among pregnant women, whose vulnerability to HIV is enhanced by their spouses' risky behaviors. A cross-sectional study was conducted from November 2010 to January 2011 among 357 women who had given birth in the last two years in a district in Orissa. O

[Exploring the issue of cervical cancer prevention and control in women living with HIV/AIDS].

Shared risk factors mean that women engaging in unprotected sex are at risk of contracting both the human immunodeficiency virus (HIV) and the human papillomavirus (HPV). As HIV suppresses bodily immune systems, an HIV-positive woman with HPV will likely see a faster progression of cancer cells around the cervix. Therefore, it is important to prevent and control cervical cancer in HIV-positive women. Findings from the literature on this topic include: 1) HIV-positive women face a relatively greater risk of: HPV surveillance, contracting high-risk HPV subtypes, contracting more subtypes of HPV,

Pseudotyped human lentiviral vector-mediated gene transfer to airway epithelia in vivo.

We used a replication defective human lentiviral (HIV) vector encoding the lacZ cDNA and pseudotyped with the vesicular stomatitis virus (VSV) glycoprotein (G) to evaluate the utility of this vector system in airway epithelia. In initial studies, apical application of vector to polarized well differentiated human airway epithelial cell cultures produced minimal levels of transgene expression whereas basolateral application of vector enhanced levels of transduction approximately 30-fold. Direct in vivo delivery of HIV vectors to the nasal epithelium and tracheas of mice failed to mediate gene t

Host proteins interacting with integrase of human immunodeficiency virus type 1.

Integrase (IN) is a key enzyme that catalyzes the integration of Human Immunodeficiency Virus (HIV) viral DNA into the human genome. It consists of three domains: the N-terminal domain, the catalytic core domain, and the C-terminal domain. In the cytoplasm, IN, viral DNA, and multiple host factors assemble into a pre-integration complex (PIC). The translocation of PIC from the cytoplasm to the nucleus is a prerequisite for integration. Numerous host proteins modulate IN activity during PIC translocation and the integration process. Interactions between IN and host factors represent promising t

Impact of Hepatitis C Virus Co-infection on Antiretroviral Therapy Outcomes in Adults Living with HIV - China, 2002-2023.

WHAT IS ALREADY KNOWN ABOUT THIS TOPIC?: The impact of hepatitis C virus (HCV) co-infection on antiretroviral therapy (ART) outcomes in people living with human immunodeficiency virus (HIV) remains controversial across studies. WHAT IS ADDED BY THIS REPORT?: Using stratified matching methods, we constructed two cohorts - one with HIV/HCV co-infection and one with HIV mono-infection - to compare ART outcomes. Our analysis revealed that HIV/HCV co-infected individuals faced significantly elevated risks of mortality, virological failure, and attrition relative to their HIV mono-infected counterp

The role of co-infections in mother-to-child transmission of HIV.

In HIV-infected women, co-infections that target the placenta, fetal membranes, genital tract, and breast tissue, as well as systemic maternal and infant infections, have been shown to increase the risk for mother-to-child transmission of HIV (MTCT). Active co-infection stimulates the release of cytokines and inflammatory agents that enhance HIV replication locally or systemically and increase tissue permeability, which weakens natural defenses to MTCT. Many maternal or infant co-infections can affect MTCT of HIV, and particular ones, such as genital tract infection with herpes simplex virus,

Distribution of STD clinic patients along a stages-of-behavioral-change continuum--selected sites, 1993.

Human immunodeficiency virus (HIV) counseling and testing are important components of the public health effort to contain the acquired immunodeficiency syndrome (AIDS) epidemic. Project RESPECT is a multicenter randomized trial being conducted by CDC at five sexually transmitted disease (STD) clinics to evaluate the efficacy of HIV counseling interventions in preventing HIV and other STDs. Because consistent and proper use of latex condoms is one method to substantially reduce the risk for transmitting HIV and other STDs (1), the HIV prevention interventions being evaluated focus primarily on

Use of intravenous immunoglobulin in acquired immune deficiency syndrome.

Patients infected with the human immunodeficiency virus (HIV) may have an antibody deficiency and a deficiency of cellular immunity. Intravenous immunoglobulin (IVIG) preparations may benefit HIV-infected children and adults with recurrent bacterial infections at doses of 200 to 400 mg/kg every 2 to 4 weeks. In addition, IVIG (1 to 2 g/kg) is effective at raising platelet counts to hemostatic levels in HIV-infected patients with idiopathic thrombocytopenic purpura and life-threatening bleeding. Indirect evidence also suggests that IVIG may be effective in preventing Pneumocystis carinii pneumo

Enhancement of Human Immunodeficiency Virus-Specific CD8+ T Cell Responses with TIGIT Blockade Involves Trogocytosis.

Natural killer (NK) and CD8+ T cell function is compromised in human immunodeficiency virus type 1 (HIV-1) infection by increased expression of inhibitory receptors such as TIGIT (T cell immunoreceptor with Ig and ITIM domains). Blocking inhibitory receptors or their ligands with monoclonal antibodies (mAb) has potential to improve antiviral immunity in general and facilitate HIV eradication strategies. We assessed the impact of TIGIT engagement and blockade on cytotoxicity, degranulation, and interferon-gamma (IFN-γ) production by CD8+ T cells from persons living with HIV (PLWH). The effect o

Long-term mucosal T cell activation and homing phenotypes in recipients of an Ad5-vectored HIV vaccine.

BACKGROUND: Vaccine-induced mucosal immune responses may be critical for protection against HIV infection, but may also result in short or long-term changes that enhance susceptibility to infection in some individuals, such as those with baseline seroreactivity to vaccine vector antigens. We examined cellular immune responses in blood and gut mucosal tissue roughly two years following vaccination with placebo or the Step study vaccine MRKAd5/HIV-1. METHODS: Participants vaccinated with either placebo or MRKAd5/HIV-1 during participation in HVTN 071, and HVTN 502/Merck 023 underwent phlebotomy
