A Molecular Window into the Biology and Epidemiology of Pneumocystis spp.

Pneumocystis, a unique atypical fungus with an elusive lifestyle, has had an important medical history. It came to prominence as an opportunistic pathogen that not only can cause life-threatening pneumonia in patients with HIV infection and other immunodeficiencies but also can colonize the lungs of healthy individuals from a very early age. The genus Pneumocystis includes a group of closely related but heterogeneous organisms that have a worldwide distribution, have been detected in multiple mammalian species, are highly host species specific, inhabit the lungs almost exclusively, and have ne

JMT-1: a novel, spherical lytic halotolerant phage isolated from Yuncheng saline lake.

This work described a novel halotolerant phage, JMT-1, with a spherical morphology. JMT-1, which was isolated from a hypersaline lake, could produce clear plaques on Chromohalobacter sp. LY7-3. The purified virions are spherical, have no visible tail, and are about 30-50nm in diameter. JMT-1 has a wide host range, and this study showed that the phage can infect at least five halophilic bacteria. The proteins of JMT-1 were analyzed using sodium dodecyl sulfate polyacrylamide gel electrophoresis, and six proteins were detected. Results show that JMT-1 is a bacteriophage with a linear double-stra

A new species of Dactylogyrus (Monogenea: Dactylogyridae) parasitic on an endangered freshwater fish, Rhodeus atremius atremius, endemic to Japan.

A new dactylogyrid monogenean Dactylogyrus bicorniculus sp. nov. is described from the gills of the kazetoge bitterling, Rhodeus atremius atremius (Jordan and Thompson, 1914), an endemic species in Japan, from Saga Prefecture, northern Kyūshū. D. bicorniculus sp. nov. resembles Dactylogyrus bicornis Malevitskaja, 1941 and Dactylogyrus lophogonus Zhang and Ji, 1980 because they have two common features, a large V-shaped ventral bar and well-developed second marginal hooks. However, the new species is distinguished from these congeners by a shorter penis and an accessory piece. A phylogenetic an

Prevalence and intensity of Streblidae in bats from a Neotropical savanna region in Brazil.

Bats of the family Phyllostomidae are common hosts to streblids known as bat flies. Here, we discuss the component community, prevalence and intensity of infection with species of Streblidae on an assemblage of phyllostomid bats in the Cafuringa Environmental Protection Area (APA Cafuringa) in the core area of the Cerrado in Central Brazil. A total of 1 841 streblid individuals of 24 species occurred on 752 bats of 14 species. Ten species of streblids infected Glossophaga soricina (Pallas), whereas seven or fewer streblid species infected the other bat species. Nine bat fly species presented a

Galactomannan detection for invasive aspergillosis in immunocompromised patients.

BACKGROUND: Invasive aspergillosis is the most common life-threatening opportunistic invasive mycosis in immunocompromised patients. A test for invasive aspergillosis should neither be too invasive nor too great a burden for the already weakened patient. The serum galactomannan enzyme-linked immunosorbent assay (ELISA) seems to have the potential to meet both requirements. OBJECTIVES: To obtain summary estimates of the diagnostic accuracy of galactomannan detection in serum for the diagnosis of invasive aspergillosis. SEARCH METHODS: We searched MEDLINE, EMBASE and Web of Science with both M

The N-terminus of vaccinia virus host range protein C7L is essential for function.

Vaccinia virus (VACV), a member of the Poxviridae family of large double-stranded DNA viruses, is being used as a smallpox vaccine as well as an expression vector for immunization against other infectious diseases and cancer. The host range of wild type VACV is very broad among mammalian cells. C7L is a host range gene identified in VACV and is well conserved in mammalian poxviruses except for parapoxviruses and molluscum contagiosum virus. The molecular mechanisms by which the C7L gene exerts host range function are not well understood. The C7L protein does not have any known conserved domain

Experimental infection of goats with Sarcoptes scabiei var. ovis.

Goats were experimentally infected with Sarcoptes scabiei var. ovis from sheep, proving that this mite is not completely host specific. More severe lesions were produced on goats when the mites were applied to lacerated than when applied to scarified or non-scarified areas. Lesions were more pronounced on moistened areas than dry areas, and lesions produced on dry scarified areas resembled those produced on non-scarified moistened ones. Skin scrapings from the experimental lesions in the goats contained numerous mites of all stages, proving that the sheep mites had become well established and

Functional Divergence of the Paralog Salmonella Effector Proteins SopD and SopD2 and Their Contributions to Infection.

Salmonella enterica is a leading cause of bacterial food-borne illness in humans and is responsible for millions of cases annually. A critical strategy for the survival of this pathogen is the translocation of bacterial virulence factors termed effectors into host cells, which primarily function via protein-protein interactions with host proteins. The Salmonella genome encodes several paralogous effectors believed to have arisen from duplication events throughout the course of evolution. These paralogs can share structural similarities and enzymatic activities but have also demonstrated diverg

Computational identification of physicochemical signatures for host tropism of influenza A virus.

Avian influenza viruses from migratory birds have managed to cross host species barriers and infected various hosts like human and swine. Epidemics and pandemics might occur when influenza viruses are adapted to humans, causing deaths and enormous economic loss. Receptor-binding specificity of the virus is one of the key factors for the transmission of influenza viruses across species. The determination of host tropism and understanding of molecular properties would help identify the mechanism why zoonotic influenza viruses can cross species barrier and infect humans. In this study, we have co

The interplay between host and viral factors in shaping the outcome of cytomegalovirus infection.

Cytomegalovirus (CMV) remains a major human pathogen causing significant morbidity and mortality in immunosuppressed or immunoimmature individuals. Although significant advances have been made in dissecting out certain features of the host response to human CMV (HCMV) infection, the strict species specificity of CMVs means that most aspects of antiviral immunity are best assessed in animal models. The mouse model of murine CMV (MCMV) infection is an important tool for analysis of in vivo features of host-virus interactions and responses to antiviral drugs that are difficult to assess in humans

Survival of Borrelia burgdorferi Strain B31 in Human Serum Is Not Dependent on C4BP Binding to the Bacterial Surface.

Lyme disease is a vector-borne illness caused by spirochetes belonging to the Borrelia burgdorferi species group. These bacteria employ several mechanisms to survive within the vertebrate host, including evasion of the complement system. In this study, we examine the protection against human serum killing by the binding of host complement regulators C4b-binding protein (C4BP) and factor H (FH) to the bacterial surface of B. burgdorferi. Via serum depletion of isolated complement regulators, we found that the absence of C4BP did not alter the survival of B. burgdorferi strain B31; however, the

Viral immunity: it takes two to tango.

Viral infections were a major cause of mortality until the use of vaccination and antiviral drugs became widespread. During the last 10 years, immunologists have made tremendous progress in dissecting mechanisms that contribute to the success of vaccines, in understanding the molecular and cellular basis of innate immunity, and in recognizing the complexity of immunological memory. Virologists have also made great progress in elucidating the molecular mechanisms of how viruses evade, manipulate, and interact with host cellular processes. In spite of these advances, most of the basic mechanisms

Assessing Host-Virus Codivergence for Close Relatives of Merkel Cell Polyomavirus Infecting African Great Apes.

UNLABELLED: It has long been hypothesized that polyomaviruses (PyV; family Polyomaviridae) codiverged with their animal hosts. In contrast, recent analyses suggested that codivergence may only marginally influence the evolution of PyV. We reassess this question by focusing on a single lineage of PyV infecting hominine hosts, the Merkel cell polyomavirus (MCPyV) lineage. By characterizing the genetic diversity of these viruses in seven African great ape taxa, we show that they exhibit very strong host specificity. Reconciliation analyses identify more codivergence than noncodivergence events. I…

Statin-induced chronic cholesterol depletion inhibits Leishmania donovani infection: Relevance of optimum host membrane cholesterol.

Leishmania are obligate intracellular protozoan parasites that invade and survive within host macrophages leading to leishmaniasis, a major cause of mortality and morbidity worldwide, particularly among economically weaker sections in tropical and subtropical regions. Visceral leishmaniasis is a potent disease caused by Leishmania donovani. The detailed mechanism of internalization of Leishmania is poorly understood. A basic step in the entry of Leishmania involves interaction of the parasite with the host plasma membrane. In this work, we have explored the effect of chronic metabolic choleste
