Uncovering Proteomic and Biochemical Alterations in Plasma from Lesch-Nyhan Disease Patients.

Lesch-Nyhan disease (LND) is an ultra-rare X-linked inborn error of metabolism caused by complete or partial deficiency of hypoxanthine-guanine phosphoribosyltransferase (HPRT), a key enzyme in the purine salvage pathway. This defect leads to uric acid overproduction and a broad spectrum of neurological and behavioral manifestations, whose severity depends on the degree of residual enzymatic activity. Although emerging evidence implicates HPRT deficiency in widespread cellular dysfunctions, particularly within midbrain dopaminergic neurons, the molecular mechanisms underlying the neurobehavior

Unleashing the Power of Induced Pluripotent stem Cells in in vitro Modelling of Lesch-Nyhan Disease.

Lesch-Nyhan disease (LND) is a monogenic rare neurodevelopmental disorder caused by a deficiency in hypoxanthine-guanine phosphoribosyltransferase (HPRT), the key enzyme of the purines salvage pathway. Beyond its well-documented metabolic consequences, HPRT deficiency leads to a distinctive neurobehavioral syndrome characterized by motor disabilities, cognitive deficits, and self-injurious behavior. Although various cell and animal models have been developed to investigate LND pathology, none have adequately elucidated the underlying mechanisms of its neurological alterations. Recent advances

Pleiotropic effects of gold(I) mixed-ligand complexes of 9-deazahypoxanthine on transcriptional activity of receptors for steroid hormones, nuclear receptors and xenoreceptors in human hepatocytes and cell lines.

Development of metal-based compounds is an important research avenue in anti-cancer and anti-inflammatory drug discovery. Here we examined the effects of three gold (I) mixed-ligand complexes with the general formula [Au(Ln)(PPh3)] (1, 2, 3) involving triphenylphosphine (PPh3) and a deprotonated form of O-substituted derivatives of 9-deazahypoxanthine (Ln) on the transcriptional activity of aryl hydrocarbon receptor (AhR), androgen receptor (AR), glucocorticoid receptor (GR), thyroid receptor (TR), pregnane X receptor (PXR) and vitamin D receptor (VDR), employing gene reporter assays. HPRTYale proposed as a pathogenic variant for Lesch-Nyhan syndrome: a case report.

Lesch-Nyhan syndrome (LNS) is an X-linked recessive disorder caused by a deficiency of hypoxanthine-guanine phosphoribosyltransferase (HPRT), an enzyme encoded by the HPRT1 gene. The classic disease phenotype described by Lesch and Nyhan in 1964 includes hyperuricemia, mental retardation, severe motor deficiency, and recurring self-mutilation. Here, we report the case of a family with 4 affected males and several female obligate carriers. In 1989, Fujimori et al. reported on a patient diagnosed with LNS who had an HPRT variant thereafter codenamed HPRTYale. The same patient was studied by Wils

Excision repair is required for genotoxin-induced mutagenesis in mammalian cells.

Certain hexavalent chromium [Cr(VI)] compounds are human lung carcinogens. Although much is known about Cr-induced DNA damage, very little is known about mechanisms of Cr(VI) mutagenesis and the role that DNA repair plays in this process. Our goal was to investigate the role of excision repair (ER) pathways in Cr(VI)-mediated mutagenesis in mammalian cells. Repair-proficient Chinese hamster ovary cells (AA8), nucleotide excision repair (NER)-deficient (UV-5) and base excision repair (BER)-inhibited cells were treated with Cr(VI) and monitored for forward mutation frequency at the hypoxanthine-

Evidence of overestimation of rate constants for the superoxide trapping by nitrones in aqueous media.

Since major disagreements exist regarding the kinetics of superoxide trapping by nitrones, the underlying theory of one of the most popular method used in these studies was reinvestigated. It involves a competition between the nitrone of interest and a superoxide scavenger, and implies that the superoxide spontaneous dismutation, the spin adduct decay, and the consumption of the competitor during the course of the experiments are negligible events. Evidences of the importance of these three unduly neglected reactions are given, and errors connected to their omission are estimated. Hence this S

Molecular, biochemical, and genetic characterization of a female patient with Lesch-Nyhan disease.

Lesch-Nyhan disease (LND) is a rare X-linked recessive disorder caused by virtually complete deficiency of activity of the purine salvage enzyme hypoxanthine phosphoribosyltransferase (HPRT; EC 2.4.2.8). Human HPRT is encoded by a single structural gene located on the long arm of the X-chromosome (Xq26). The classical LND phenotype occurs almost exclusively in males, manifested in excessive purine production and characteristic neurological manifestations, including compulsive self-mutilation, choreoathetosis, spasticity, and occasionally developmental delay. Heterozygous females are usually ph

[Gene therapy for genetic disorders of purine metabolism].

Anderson proposed three genetic diseases deficient in enzymes involved in purine metabolism as candidate for human gene therapy in 1984. Over the past ten years, marked advances in molecular genetics have brought gene therapy to reality. Gene therapy for ADA deficiency was performedin the United States in 1990 and in Japan in 1995 as the first trial. Basic study for HPRT gene transfer has been proceeding toward gene therapy for HPRT deficiency despite the difficulties in gene transfer into the nervous systems.

Transport and accumulation of purine bases by Crithidia fasciculata.

The capacity for the rapid transport of purine bases by Crithidia fasciulata is found only in cells starved for purines. Cells grown in complete medium transport poorly. Rapid transport capability appears and then disappears during growth of purine-depleted cultures. This rapid transport appears to occur by a process of mediated diffusion. Two mechanisms are involved, one of low velocity and high affinity, the other of high velocity and low affinity. Accumulation of the bases within the cell occurs by their rapid conversion to ribonucleotides by phosphoribosyltransferases.

Purine mutants of mammalian cell lines: III. Control of purine biosynthesis in adenine phosphoribosyl transferase mutants of CHO cells.

Spontaneous and mutagen-induced 2,6-diaminopurine-resistant mutants of Chinese hamster ovary (CHO-K1) cells were isolated. Such mutants fell into two classes: spontaneous and ethylmethane-sulfonate-induced mutants had approximately 5% wild-type adenine phosphoribosyl transferase (APRT) activity, whereas ICR-170G-induced mutants had barely detectable APRT activity. Since it has been reported that human hypoxanthine-guanine phosphoribosyl transferase (HGPRT) (Lesch-Nyhan syndrome) and APRT mutants over-produce purines, we examined the control and rate of purine biosynthesis in the Chinese hamste

Effect of Lactiplantibacillus plantarum-mediated lactic fermentation on the characteristic flavor of sea buckthorn juice by volatolomics and metabolomics analysis.

Lactic fermentation enhances sensory quality and nutritional properties of fruit juice, though its comprehensive impact on sea buckthorn through Lactiplantibacillus plantarum fermentation (LPF) remains unreported. This study used volatolomics and metabolomics to explore mechanism of lactic fermentation improving flavor. LPF reduced total acidity (from 24.01 g/L to 15.79 g/L), increased total flavonoid by 20.35% and polysaccharide contents by 55.64%. It also increased the quantity of volatile compounds (from 36 to 41). β-Damascenone and ethyl isovalerate were key aroma compounds, while the incr

Glucocorticoid-induced changes of the gut microbiota and metabolic markers in healthy young men: Outcome of a randomized controlled trial.

Glucocorticoids induce insulin resistance and suppress immunity, but their impact on gut microbiota, which may modulate metabolism and immunity remains under explored. In this 7-day trial, we assess glucocorticoid-induced changes in gut microbiota and metabolic markers in 56 healthy men, randomly assigned to three interventions: oral prednisolone (PO group), intramuscular methylprednisolone acetate (IM group), or saline (CTL group). Shotgun metagenomics reveal that PO glucocorticoid causes shifts in bacterial abundance, increasing Blautia and Collinsella, while decreasing Dysosmobacter welbion

From Theophylline to Adenine or preQ1: Repurposing a DNA Aptamer Revealed by Crystal Structure Analysis.

Aptamers, which are short, single-stranded DNA or RNA, are capable of binding to a wide array of targets with exceptional selectivity. Those with high affinity for theophylline have the potential to serve as biosensors, crucial for tracking theophylline levels in the treatment of respiratory conditions. Despite the extensive structural characterization of the RNA theophylline aptamer, the DNA counterpart's ligand-recognition mechanism has remained unclear. Here, we elucidate the DNA theophylline aptamer's ligand-binding mechanism through high-resolution crystal structures of its complexes with

Small-molecule-catalysed deamination enables transcriptome-wide profiling of N6-methyladenosine in RNA.

The deamination reaction is important to both fundamental organic chemistry and biochemistry. Traditional chemical methods of deamination rely on the use of aryldiazonium salts under harsh acidic conditions, which limits the application scope for most biological substrates. Here we present an N-nitrosation strategy for deamination under mild conditions that DNA and RNA biological macromolecules can tolerate. Cooperative catalysis combining a carbonyl organocatalyst with a Lewis acid catalyst facilitates the formation of a carbon-nitro intermediate from a primary amine, which, on rearrangement

Lesch-Nyhan syndrome a dental approach: case report.

Lesch-Nyhan syndrome was first described in 1964. It is an X-linked recessive disorder caused by the absence of the enzyme hypoxanthine-guanine phosphoribosyl ransferase (HGPRT), which is involved in the metabolism of purines (nitrogenous bases that make up nucleotide). The absence of the enzyme leads to motor and neurological dysfunction, increased uric acid production, and mental retardation. One of the main characteristics of this syndrome is compulsive self-mutilation, such as biting the lips, tongue, and fingers. Men are the most affected and the prevalence is 1/380,000 live births. Insight into the correlation of key taste substances and key volatile substances from shrimp heads at different temperatures.

This study aimed to investigate taste substances of shrimp heads stored at 20 °C, 4 °C, -3 °C, and - 18 °C, and the correlation between taste substances and 25 key volatile substances. Notably, samples stored at 20 °C showed significant changes in bitter amino acids and hypoxanthine, and quickly deteriorated. Samples stored at 4 °C for 14 d or - 3 °C for 30 d facilitated the development of umami amino acids, sweet amino acids, and IMP. Furthermore, samples stored at -18 °C for 30 d demonstrated no significant changes in taste profile. Changes in taste substances through quantitative analysis w

Lesch-Nyhan disease causes impaired energy metabolism and reduced developmental potential in midbrain dopaminergic cells.

Mutations in HPRT1, a gene encoding a rate-limiting enzyme for purine salvage, cause Lesch-Nyhan disease which is characterized by self-injury and motor impairments. We leveraged stem cell and genetic engineering technologies to model the disease in isogenic and patient-derived forebrain and midbrain cell types. Dopaminergic progenitor cells deficient in HPRT showed decreased intensity of all developmental cell-fate markers measured. Metabolic analyses revealed significant loss of all purine derivatives, except hypoxanthine, and impaired glycolysis and oxidative phosphorylation. real-time gluc

Cerebral Venous Sinus Thrombosis in a Child with Lesch-Nyhan Syndrome.

Lesch-Nyhan syndrome is a rare neurometabolic condition characterized by progressive choreoathetosis, intellectual disability, and peculiar manifestations like self-mutilation. Occasional case reports in adults have suggested an association between Lesch-Nyhan syndrome and hypercoagulability; however, no such report of either a venous or arterial stroke in children with Lesch-Nyhan Syndrome exists in literature. We present a 3-year-old boy with global developmental delay, dystonic posturing, choreoathetoid movements, and self-mutilation involving fingers and lips. He had acute worsening of sen
