How Artificial Intelligence Will Transform Clinical Care, Research, and Trials for Inflammatory Bowel Disease.

Artificial intelligence (AI) refers to computer-based methodologies that use data to teach a computer to solve pre-defined tasks; these methods can be applied to identify patterns in large multi-modal data sources. AI applications in inflammatory bowel disease (IBD) includes predicting response to therapy, disease activity scoring of endoscopy, drug discovery, and identifying bowel damage in images. As a complex disease with entangled relationships between genomics, metabolomics, microbiome, and the environment, IBD stands to benefit greatly from methodologies that can handle this complexity.

Insights from populations at risk for the future development of classified rheumatoid arthritis.

Rheumatoid arthritis (RA) develops through a series of stages. In the seropositive subset of classified RA patients, a preclinical stage is present for years before the onset of clinically apparent disease. Relevant preclinical biomarkers include autoantibodies, alterations of lymphoid populations, elevated cytokines/chemokines, genetic/genomic factors, imaging studies, clinical findings, dietary and environmental biomarkers, cardiovascular disease risk assessment, microbiome analyses, and metabolomic changes. Identifying the population of asymptomatic subjects at sufficiently high risk for di

Convergent imaging and genetic signatures of gray matter atrophy in Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by widespread structural brain alterations, yet the specific patterns of brain atrophy and their underlying genetic mechanisms remain incompletely understood. Here, we integrated large-scale neuroimaging meta-analysis with population-scale imaging genetics to systematically characterize the genetic architecture linking gray matter volume (GMV) abnormalities to PD. We first performed a meta-analysis of structural MRI studies comprising 3212 patients with PD and 2056 controls, identifying robust patterns of GMV re

Elucidating the association of obstructive sleep apnea with brain structure and cognitive performance.

BACKGROUND: Obstructive sleep apnea (OSA) is a pervasive, chronic sleep-related respiratory condition that causes brain structural alterations and cognitive impairments. However, the causal association of OSA with brain morphology and cognitive performance has not been determined. METHODS: We conducted a two-sample bidirectional Mendelian randomization (MR) analysis to investigate the causal relationship between OSA and a range of neurocognitive characteristics, including brain cortical structure, brain subcortical structure, brain structural change across the lifespan, and cognitive performa

Genome scan reveals several loci associated with torus palatinus.

OBJECTIVE: Torus Palatinus (TP) is a common trait with an unclear aetiology. Although prior studies suggest a hereditary component, the genetic factors that influence TP risk remain unknown. The purpose of this study is to identify genetic variants associated with TP. MATERIALS AND METHODS: We assessed the TP status of 829 individuals from various ancestral backgrounds using 3D palate scans. We then carried out a genome-wide association study (GWAS) to identify common variants associated with TP. We also performed gene-based tests across the exome to investigate the role of low-frequency codi

Large-scale brainstem neuroimaging and genetic analyses provide new insights into the neuronal mechanisms of hypertension.

While brainstem regions are central regulators of blood pressure, the neuronal mechanisms underlying their role in hypertension remain poorly understood. Here, we investigated the structural and genetic relationships between global and regional brainstem volumes and blood pressure. We used magnetic resonance imaging data from n = 32,666 UK Biobank participants, and assessed the association of volumes of the whole brainstem and its main regions with blood pressure. We applied powerful statistical genetic tools, including bivariate causal mixture modeling (MiXeR) and conjunctional false discover

Ticks of Australia. The species that infest domestic animals and humans.

The book Australian Ticks by F.H.S. Roberts (1970) is a land-mark in Australian tick biology. But it is time for a new and improved book on the ticks of Australia. The present book has identification guides and accounts of the biology and diseases associated with the 16 species of ticks that may feed on domestic animals and humans in Australia. These comprise five argasid (soft) ticks: Argas persicus (poultry tick), Argas robertsi (Robert's bird tick), Ornithodoros capensis (seabird soft tick), O. gurneyi (kangaroo soft tick), Otobius megnini (spinose ear tick); and 11 ixodid (hard) ticks, Amb

Xq26.1-26.2 gain identified on array comparative genomic hybridization in bilateral periventricular nodular heterotopia with overlying polymicrogyria.

Periventricular nodular heterotopia (PNH) with overlying polymicrogyria (PMG) is a recently described, developmental brain malformation; however, the causative genes of this malformation have not yet been identified. We report on a 5-year-old Japanese male with bilateral PNH with overlying PMG. He had mild intellectual disability, distinctive facial features, short stature, and microcephaly, with cardiac disorders. No mutation was identified in Sanger sequences for FLNA and ARFGEF2; however, array comparative genomic hybridization revealed an approximately 0.8Mb gain at Xq26.1-26.2, which incl

Advanced imaging techniques and artificial intelligence in pleural diseases: a narrative review.

BACKGROUND: Pleural diseases represent a significant healthcare burden, affecting over 350 000 patients annually in the US alone and requiring accurate diagnostic approaches for optimal management. Traditional imaging techniques have limitations in differentiating various pleural disorders and invasive procedures are usually required for definitive diagnosis. METHODS: We conducted a nonsystematic, narrative literature review aimed at describing the latest advances in imaging techniques and artificial intelligence (AI) applications in pleural diseases. RESULTS: Novel ultrasound-based techniqu

Characteristic Fetal Brain MRI Abnormalities in Pyruvate Dehydrogenase Complex Deficiency.

Pyruvate dehydrogenase complex deficiency (PDCD) is a disorder of mitochondrial metabolism that is caused by pathogenic variants in multiple genes, including PDHA1. Typical neonatal brain imaging findings in PDCD have been described, with a focus on malformative features and chronic encephaloclastic changes. However, fetal brain MRI imaging in confirmed PDCD has not been comprehensively described. We sought to demonstrate the prenatal neurological and systemic manifestations of PDCD determined by comprehensive fetal imaging and genomic sequencing. All fetuses with a diagnosis of genetic PDCD w

Cardiovascular phenotyping of fetal mice by noninvasive high-frequency ultrasound facilitates recovery of ENU-induced mutations causing congenital cardiac and extracardiac defects.

As part of a large-scale noninvasive fetal ultrasound screen to recover ethylnitrosourea (ENU)-induced mutations causing congenital heart defects in mice, we established a high-throughput ultrasound scanning strategy for interrogating fetal mice in utero utilizing three orthogonal imaging planes defined by the fetus' vertebral column and body axes, structures readily seen by ultrasound. This contrasts with the difficulty of acquiring clinical ultrasound imaging planes which are defined by the fetal heart. By use of the three orthogonal imaging planes for two-dimensional (2D) imaging together w

YOLO-based high-throughput phenotyping pipeline for soybean nodulation traits in genomic research.

The symbiotic interaction between soybean (Glycine max) and Bradyrhizobium japonicum results in the formation of root nodules, specialized organs that house nitrogen-fixing bacteria converting atmospheric N2 into plant-accessible ammonia (NH3). Accurate quantification of nodule traits is essential for understanding host-microbe interactions and genetic determinants of nodulation. However, traditional manual or semi-quantitative approaches are labor-intensive, subjective, and unsuitable for large-scale studies. Here, we present a high-throughput phenotyping pipeline based on the YOLO deep learn

Deep learning for cardiovascular disease: a comprehensive review of detection and risk forecasting.

The biggest health threat to the global population is cardiovascular disease (CVD), which afflicts almost one-third of the global population and causes considerable monetary and social losses. Risk analysis should be performed in a timely and appropriate manner to enhance clinical practice and preventive interventions. The emergence of advanced data modalities, such as wearable sensors, medical imaging, electronic health records (EHRs), and genomic platforms, has led to a paradigm shift in the holistic assessment of CVD risk through multimodal data integration. This systematic review is a meth

A novel potent autophagy inhibitor HM-013 limits production of infectious dengue virus particles.

BACKGROUND: Dengue virus (DENV) infects millions of individuals annually, yet no specific antiviral therapy exists. Autophagy, a conserved cellular degradation pathway, is activated during DENV infection and supports the production of infectious virions. Although autophagy modulation has emerged as a potential antiviral strategy, few small-molecule autophagy inhibitors with both potent anti-dengue activity and low cytotoxicity have been reported. Thus, identifying novel, safe, and effective autophagy-targeting compounds remains an important unmet need for host-directed antiviral development.

Individualized Cell-Free DNA Monitoring With Chromosomal Junctions for Mesothelioma.

INTRODUCTION: The spatially complex nature of mesothelioma and interventions like pleurodesis, surgery, and radiation often complicate imaging-based assessment. Further, cell-free DNA (cfDNA) based monitoring strategies are inadequate for mesothelioma, given the presence of a few recurring nonsynonymous somatic variants. However, patient-specific chromosomal rearrangements are commonly found in mesothelioma. Our study objective was to develop an individualized cfDNA assay to enable blood-based monitoring using circulating tumor DNA (ctDNA) in mesothelioma. We hypothesized that the unique chrom

Novel genome-wide associations for anhedonia, genetic correlation with psychiatric disorders, and polygenic association with brain structure.

Anhedonia is a core symptom of several psychiatric disorders but its biological underpinnings are poorly understood. We performed a genome-wide association study of state anhedonia in 375,275 UK Biobank participants and assessed for genetic correlation between anhedonia and neuropsychiatric conditions (major depressive disorder, schizophrenia, bipolar disorder, obsessive compulsive disorder and Parkinson's Disease). We then used a polygenic risk score approach to test for association between genetic loading for anhedonia and both brain structure and brain function. This included: magnetic reso
